Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a highly successful treatment for hematological malignancies,severe congenital immunodeficiencies and some other diseases.Approximate 50 ％ of the disease-free survival patients suffered from long-lasting severe cognitive impairment after allo-HSCT.Pathological evidences showed parenchymal lymphocytic inflammation,microglia activation,and mild cerebral angiitis-like changes in allogeneic transplanted animals and patients' autopsy.Pay much importance attention to the pathological changes of central nervous system and its impact on cognitive function and take suitable measures would help the patients to achieve a state of complete physical mental and social well-being.

Objective To investigate the clinical efficacy of electroacupuncture at specific points in treating different degrees of urgency-frequency syndrome. Methods Sixty patients with urgency-frequency syndrome were randomly allocated, according to the symptom score, to groups A (the symptom score≤ 24) and B (the symptom score>24), 30 cases each. Both groups received electroacupuncture at specific points (four sacral points and four abdominal points). In the two groups, the symptoms were scored before and after treatment and the clinical therapeutic effects were evaluated.Results There was a statistically significant pre-/post-treatment difference in the symptom score in the two groups (P<0.01). The total efficacy rate was 83.3% in group A and 80.0% in group B; there was no statistically significant difference between the two groups (P>0.05).Conclusion Electroacupuncture at specific points is an effective way to treat urgency-frequency syndrome.

The retinoblastoma-protein-interacting zinc finger proteinl (RIZ1),a methyltransferase,contains the characteristic PR zinc finger domain.RIZ1 can methylate H3K9 of the histone,acted as a transcription suppression factor of cancers.Increasing numbers of human cancers are reported to hold decreased or absent RIZ1 expression,which is closely related to the progression of cancer.RIZ1 is defined as a candidate anti-oncogene.The mechanisms of the suppression involved in both oncocytogenetics and epigenetic changes.

Objective To investigate the effects of ketamine in relieving postoperative pain after laparoscopic cholecystectomy(LC).Methods Forty patients undergoing selective LC from July to August 2006 were randomly divided into ketamine group and control group,with 20 patients in each group.Incision pain and non-incision pain was assessed at 1,2,4,6,12,24 h and once each day for the first 3 days postoperatively with visual analogue pain scale(VAS)and sedation scale.Adverse effects and analgesic requirements were recorded.Results Patients in ketamine group had significantly lower scores in incision pain and non-incision pain compared with those of control group(F=22.805,P =0.000;F=18.109,P=0.000).Incidence of postoperative nausea and vomiting was not significantly different(P=1.000)between ketamine(55%)and control group(60%).No significant difference was noticed in analgesic requirements between ketamine group(3 cases)and control group(9 cases)(P=0.082).Conclusions Preoperative administration of ketamine significantly improves postoperative analgesia after laparoscopic cholecystectomy,but does not reduce the incidence of postoperative nausea and vomiting.

Objective To discuss the techniques and efficacy of Ho:YAG laser incision by using uteroscopy for ureterostenosis.Methods From July 2004 to April 2007,52 patients with ureterostenosis received ureteral incision by using Ho:YAG laser under a endoscope.Two double pigtail stents(F5 or F6) were placed in the ureters after the operation and left indwelling for 8 to 12 weeks.Ultrasonography and excretion urography were performed 3 to 6 months after extubation.Results Follow-up was available for 3 to 24 months(mean,17 months) in 46 patients,of which 40(87%) were cured after the treatment.In the cured patients,hydronephrosis,ureteral dilation,and ureterostenosis were improved,and the pain in the kidney region was relieved;none of them showed signs of infection.In the other 6 patients,4 were improved after the treatment(no deterioration of the symptoms of hydronephrosis,ureteral dilation,and pain in the kidney region,and no infection);and 2 failed(the symptoms of hydronephrosis and ureteral dilation deteriorated,and pain in the kidney region and infection were developed).Conclusion Endoscopic ureteral excision using holmium laser combined with indwelling of two double pigtail stents is effective and safe for ureterostenosis.

One hundred and seventy one gastric biopsy specimens from the patients undergoing gastroscopic examination from March to November 2010 were reviewed . HE, sliver and immunohistochemistry stains were performed in the specimens; the correlation of histopatholigy and the density and distribution of Helicobacter pylori in gastric mucosa was analyzed .The results displayed that spiral H.pylori mainly appeared on the surface mucus and in the gland lumen of gastric small concave , some were attached to the epithelial cells; coccoid H.pylori appeared within the apical cytoplasmic part of the epithelial cells and in the mucous membrane stroma .The depth of H.pylori infection was correlated with mucosal erosion , inflammation activities and amount of interstitial lymphocytes .The results suggest that spiral H.pylori infection may facilitate the invasive depth of coccoid H.pylori in gastric mucous membrane and the density of H.pylori infection is correlated with the extent and severity of inflammation .

Objective To evaluate the curative effect of percutaneous compression plating system in treatment of intertrochanteric fractures of the elderly patients.Methods The clinical data of 18 elderly patients(aged 68-89 years)with intertrochanteric fractures were retrospectively analyzed.According to Evans classification,there were seven patients with type Ⅰ fractures,nine with type Ⅱ fractures and two with type Ⅲa fractures.Percutaneous compression plating system was applied to all patients from June 2007 to June 2009,and the therapeutic effect was evaluated by Harris hip function evaluation criteria.Results All the fractures were healed,with excellent results in 15 patients and good results in the rest three patients.Conclusions The percutaneous compression plating system is easy to manipulate with minimal invasion,has minor side effect and is beneficial to reducing surgical complications in the elderly patients with intertrochanteric fractures.

Objective To investigate the advantages, indications and efficiency of video-assited thoraco-scopic sugery in spontaneous pneumothorax. Methods 31 patients with spontaneous pneumothorax underwent video-assited thoracoscopic sugery, performed with endoscopic manual suture without use of endoscopic cutters. Results All patients were cured without recurrence. Conclusions Video-assited thoracoscopic sugery is the first choice in treating spontaneous pneumothorax,with minimal trauma,less bleeding. Endoscopic manual suture can obviously re-duce the treatment cost.

Objective To study compatibility stability of Danshen injection combined with dopamine injection in 5% glucose injection. Methods The relative contents of tanshinol sodium, protocatechuic aldehyde, rosmarinic acid,salvianolic acid B in Danshen injection and Dopamine hydrochloride in dopamine injection were determined by HPLC in 6h after the compatibility in the above infusion at 5 ℃,25℃,35 ℃ under dark and light,and the changes in pH value,appearance and insoluble particles were also observed.ResuIts There were no significant changes in appearance and pH value at 5℃,25℃,35 ℃ under dark,insoluble particles were in line with Chinese Pharmacopoeia,and the relative content of tanshinol sodium, protocatechuic aldehyde, rosmarinic acid,salvianolic acid B and Dopamine hydrochloride were above 99%(compared with 0h) in 6h.In the light conditions,as the temperature raised,the extension of storage time,the relative content of Dopamine hydrochloride was not significantly decreased,but the content of tanshinol sodium, protocatechuic aldehyde, rosmarinic acid and salvianolic acid B had declined,while pH value also had a certain degree of reduction,the insoluble particles had no obvious change.ConcIusion The mixture of Danshen injection and dopamine injection in 5%glucose injection is stable under dark conditions in 6h, illumination and temperature maybe the main factor affecting the stability of Danshen injection combined with dopamine injection.

Objective: To investigate the effects of up-regulating or silencing DJ-1 gene expression on the apoptosis, migration and invasion of colorectal cancer (CRC), and to explore the possible molecular mechanism. Methods: The expression level of DJ-1 in CRC tissues and cells was detected by immunohistochemistry, Western blotting and real-time fluorescent quantitative PCR, respectively. The SW480 and HCT116 cells were transfected with the recombinant lentiviral vector carrying human DJ-1 gene to obtain DJ-1 overexpressed SW480/OE-DJ-1 and HCT116/OE-DJ-1 cells, while the cells transfected with the empty vector was as the negative control (OE-NC). The SW480 and HCT116 cells were transfected with the recombinant lentiviral vector carrying the specific shRNA targeting DJ-1 gene to generate the SW480/shDJ-1 and HCT116/sh-DJ-1 cells with stable knockdown of DJ-1, while the cells transfected with the empty vector was as the negative control (sh-NC). Subsequently, the expressions of DJ-1 and p53 protein and mRNA were detected by immunohistochemistry and real-time fluorescent quantitative PCR, respectively; and their relationship was analyzed. The expressions of p53 and its downstream apoptosis-related proteins Bax and Bcl-2 in SW480 and HCT116 cells with DJ-1 over-expression or knockdown were detected by Western blotting. The effects of overexpressing and silencing DJ-1 gene expression on the invasion and migration abilities of SW480 and HCT116 cells were detected by Transwell chamber assay. The epithelial-mesenchymal transition (EMT) of CRC cells was induced by transforming growth factor-β1 (TGF-β1), then the expression levels of DJ-1 and EMT-related markers (N-cadherin, β-catenin, vimentin, E-cadherin) were analyzed by Western blotting. Results: DJ-1 was highly expressed in 34 CRC tissues (24/34, 70.59%) (P < 0.001). The overall survival time of the patients with DJ-1 high expression was significantly shorter than that of the patients with DJ-1 low expression (P < 0.001). The high expression of DJ-1 was correlated with TNM stage, tumor location, lymph node metastasis, and degree of differentiation (all P < 0.05). There was a negative correlation between DJ-1 and p53 expressions (r =-0.428, P = 0.015). Silencing DJ-1 increased the expression level of p53 and its downstream apoptotic protein Bax, decreased the expression of anti-apoptotic protein Bcl-2 (all P < 0.05), and decreased the invasion and migration capacities of SW480 and HCT116 cells (both P < 0.01); Conversely, overexpressing DJ-1 decreased the expression level of p53 and Bax, increased the expression of Bcl-2 (all P < 0.05), and increased the invasion and migration capacities of SW480 and HCT116 cells (both P < 0.01). Overexpression of DJ-1 induced by TGF-β1 increased the expressions of N-cadherin, β-catenin and vimentin, and decreased the expression of E-cadherin in the process of EMT (P < 0.05). Conclusion: DJ-1 promotes the apoptosis and invasion of CRC cells by negatively regulating the p53 signaling pathway.
