3.Effects of Different Extracts of Rupixiao Granules on Mouse Primary Dysmenorrhea Model

Objective:To research the anti-dysmenorrhea effect of different extracts of Rupixiao granules. Methods:The anti-dys-menorrhea effect of different extracts of Rupixiao granules was studied by writhing test induced by oxytocin in mice. Results:Compared with the model group, the extracts of Rupixiao granules could reduce the writhing times induced by oxytocin significantly (P<0. 05,P<0. 01 or P<0. 001), and the water-soluble extract showed the best effect (P<0. 001) without significant difference when compared with the positive control group(P>0. 05). Conclusion:The water-soluble parts of Rupixiao granules have significant anti-dysmenor-rhea effect.
4.Antibacterial Activities of Cefoperazone-sulbactam Against Gram-negative Bacilli

OBJECTIVE To evaluate the antibacterial activities of cefoperazone combined with sulbactam against Gram negative bacteria,and compare with other antimicrobial agents.METHODS The antibacterial activities of 10 frequently used antibiotics against 1 670 strains of clinical isolated Gram negative bacteria were studied by using agar dilution methods according to the NCCLS 2002.RESULTS The results showed imipenem was the most active tested against Enterobacteriaceae such as Escherichia coli and Klebsiella pneumoniae,followed by cefoperazone-(sulbactam),which had similar activities as imipenem against the non-fermentative strains such as Pseudononas aeruginosa,and Acinetobacter spp,but cefoperazone-(sulbactam) had higher susceptibility to imipenem-(resistant) Stenotrophomonas maltophilia.CONCLUSIONS Cefoperazone-sulbactam has good and broad spectrum(antibacterial) activities especially against Gram negative(bacteria,) and is expected to have a bright prospect in the treatment of severe hospital infections induced by Gram negative bacteria.

6.High and stable expression of an analog of human basic fibroblast growth factor in Escherichia coli

AIM: To obtain a high and stable expression analog of human basic fibroblast growth factor by genetic engineering. METHODS: The cysteins 78 and 96 of natural hbFGF polypeptide was substituted with serines by means of site-directed mutagenesis. Using pET- 3c as vector, the mutated polynucleotide was cloned and then transferred into BL21 (DE3)plysS. After induction by IPTG, the analog was obtained and analyzed by SDS - PAGE. RESULTS: After purification the form of soluble mutant increased remarkably but the forms of dimmer and higher multimer were reduced greatly to no more than 8% of the total recombinant protein. By MTT assay, the analog showed the same biological activity. This new analog represented a desirable complementation for native hbFGF to develop pharmaceutical drug in clinical use. CONCLUSION: Substitution of certain amino acids of polypeptide without altering native protein' s bioactivity to get the analog is an effective means to increase stability of foreign protein and its solubility in E. coli.

8.Analysis on the cooperative intentions and needs of Chinese and Association of Southeast Asian Nations medical universities/colleges in research

Objective:The study aims to gain information on medical research status and cooperative needs of medical universities/colleges from China and Association of Southeast Asian Nations (ASEAN) and provide evidence to guide the regional cooperation and development in medical reserach.Methods:A self-constructed questionnaire survey was adopted to collect information on the cooperative intentions, expertise and challenges of the 43 member institutions of China-ASEAN University Consortium on Medicine and Health in medical research.Results:In total, 20 Chinese members and 10 ASEAN members completed the survey. The study found that both Chinese and ASEAN medical universities/colleges were particularly interested in cooperation in academic conferences, multi-center clinical trials and joint research projects /flagship projects, followed by large cohort studies and medical journal/publication. In terms of research priority, both Chinese and ASEAN institutions were very interested in big data, followed by cancer precision medicine; while ASEAN members were more interested in natural drugs and dengue than Chinese ones.Conclusions:Both Chinese and ASEAN medical universities/colleges had strong interest in international cooperation in medical research. They should further explore the advantages and characteristics of both sides in medical research based on the interests of two sides, adopt appropriate scientific research cooperation methods, and establish sound scientific research cooperation mechanism so as to deepen cooperation in medical research, promote the development and improve the outcome of regional medical research, and facilitate the building of a community of common health for mankind.
9.A dynamic observation of pathologic and ultrastructural changes of perihematoma in intracerebral hemorrhage patients

Background In recent years,some researches had been conducted on the pathologic changes of the secondary injury of perihematoma in animal experiments,but only a few studies had been done on the dynamic pathologic and ultrastructural changes of the perihematoma in ICH patients. The unique contribution of our study is to investigate the dynamic pathologic and ultrastructural changes of the perihematoma in ICH patients and provide significant insights into how the pathophysiology and ultrastructures changed after ICH.Methods The written informed consents were obtained from the ICH patients or their relatives. 30 patients (the supertentorial hemotoma volume>30 mi and the cerebellar hemotoma volume >10 mi) were divided into 8 groups according to the time passed after ICH:<6 h (6 patients), 6 ~ 12 h (7 patients), 12 ~24 h (5 patients), 24~48 h (3 patients), 48 ~72 h (3 patients), 3 ~4 days group (3 patients), 5 days group (2 patients) and 8 days group ( 1 patient) and subjected to craniotomy for hemotoma evacuation. During the operation for the hemotoma's evacuation, a small amount of tissues that must be removed, which located at 1 cm near the hematoma, were taken as experimental groups; And the same tissues of 7 patients (<12 h), which were far from the hemotoma on the operational way, were taken as control group. The pathologic and ultrastructral changes were observed.Results The tissues of the control group were almost normal while the damages of the tissues from the experimental groups were slight in <6 h groups, more severe after 6h and got to the maximum between 24 ~48 h , recovered gradually after 72 h, became similar to the 6 ~ 12 h group on 5 th day, got better on 8 th day and resembled the 6 h group.Conclusions The injury of the perihematoma occurred in early stage, reached the peak level between 24 and 48 hours after ICH; which was consistent to the clinical nervous functional deficits in the ICH patients.
10.Effects of vitamin D3 adjuvant therapy for clinical efficacy, peripheral blood eosinophils and serum IL-4 levels in patients with allergic rhinitis

OBJECTIVE To explore the possible role and mechanism of vitamin D on the pathogenesis of allergic rhinitis by adjuvant therapy with vitamin D3. METHODS A total of 60 allergic rhinitis patients and 30 healthy subjects were selected as the combination therapy group, standard treatment group and control group. Vitamin D3 nasal drops to nasal cavity and oral Desloratadine Citrate Disodium at the same time 8.8 mg once a day were given in combined treatment group patients. Standard treatment group patients received oral Desloratadine Citrate Disodium 8.8 mg once a day. The total course of treatment was 4 weeks and then to assess the changes of serum 25(OH)D concentration, eosinophils, IL-4 levels and the therapeutic effect before and after treatment. RESULTS The serum 25(OH)D levels before and after treatment in combination therapy group were(23.67±4.47)ng/ml and (47.57±2.83)ng/ml, the IL-4 levels were (14.576 ± 4.472)pg/ml and (10.381 ± 3.41l)pg/ml, and the eosinophils of peripheral blood counts were (0.71±0.14)× 109/L and (0.34 ± 0.09)×109/L. The total effective rate was 97%. The serum 25(OH)D levels before and after treatment in standard treatment group were (23.42±3.83)ng/ml and(31.51 ±2.95)ng/ml, the IL-4 levels were (15.187±5.144)pg/ml and (12.794 ±5.396)pg/ml, and the eosinophils of peripheral blood counts were (0.67±0.12)×109/L and (0.41 ±0.10)×109/L. The total effective rate was 84%. CONCLUSION Nasal vitamin D3 drops combined with antihistamines for allergic rhinitis can effectively improve the clinical symptoms and reduce the related inflammatory markers.
