Objective:To investigate 7-11-years-old children's emotional responses, emotional attributions, coping styles to success and failure situation in learning. Methods: With clinical interview method ,90 pupils from seven-years-old to elevelen-years-old were asked to inference to the dramatis personae's affect reaction, emotional attribution in the given situation.Results:Children's emotional response and emotional attribution in learning context is self-regarded, coping style also decline to self effort .There were no age and sex differences in emotional responses to successful situation,there were age differences to failure situation,and there were no sex difference in emotional attribution.Conclusion:Children's emotional responses and emotional attribution to success and failure in learning have different features.

Osteosarcoma is the most common primary malignant tumor that develops in bone. Since chemotherapy was introduced into the multi-modality treatment regimen for high-grade osteosarcoma, its prognosis has been impressively improved; about two-thirds of all patients could achieve long-term survival. This review summarized the history of chemotherapy for osteosarcoma and current chemotherapeutic treatment strategies for classical osteosarcoma. The review also emphasized the necessity that the patients should be treated in specialized centers and prospective, multi-institutional trials are needed.

Objective To establish a method for quantitative detection of hepatitis B virus covalently closed circular DNA(HBV cccDNA ) in infected cells. Methods The transfected cell line HepG2.2.15 which can consistently produce Dane particles was maintained in DMEM containing 380 ?g/ml G418 and 10% fetal bovine serum. Cells in the exponential period were harvested from flasks, then intracellular HBV cccDNA was extracted from pellet containing 1?10~6 cells with mini plasmid extraction kit (QIAGEN).The extraction product was further purified by mung bean nuclease to remove HBV relaxed circular DNA possibly remained. HBV cccDNA was quantitatively detected by fluorescent PCR with selective primer set and Taqman MGB probe. Culture medium before exponential period, HBV DNA positive and negative serum samples from patients with chronic hepatitis B (mild) were amplified simultaneously to test the specificity of the fluorescent PCR method. Plasmids containing whole HBV genome were amplified with the same primer set and fluorescent probe to determine the sensitivity of the method. Results HBV cccDNA was detectable in HepG2.2.15, and the average quantity was 18 copies per cell approximately. No detectable fluorescent signal was observed when culture and serum samples were amplified. The detectable HBV cccDNA was as low as 10~3 copies per ml at least by this method. Conclusions This method is convenient, highly specific and highly sensitive. It can be utilized in the quantitative detection of intracellular HBV cccDNA as well as in the screening and evaluation of antiviral agents.

BACKGROUND:The long-term contraction of skeletal muscle can lead to the decrease of resting potential, affect its contraction, and result in the occurrence of fatigue. Whether acupuncture and injury-free magnetic lifting needle are effective on the recovery of resting potential? OBJECTIVE: To observe and compare the influence on the recovery of resting potential of skeletal muscle by taking acupuncture and magnetic needle as the recovery means.DESIGN: A controlled study.SETTINGS: College of Physical Education, Shanxi Normal University;Binzhou College of Shandong Province.MATERIALS: Thirty experimental toads, which were provided by Beijing University of Physical Education, were randomly divided into acupuncture group (n=10), magnetic needle group (n=10), magnetic needle+pressing group (n=10). Left and right semitendinosus muscles were taken as controls.METHODS: The in vitro toad's semitendinosus muscles (experimental side and control side) were treated with electrostimulation for 40 minutes,and then the resting potential of skeletal muscular cells were measured with the intracellular glass microelectrode technique. After the electrostimulation, filiform needle acupuncture, magnetic needle, magnetic needle plus pressing were also given.MAIN OUTCOME MEASURES: The resting potentials were observed before exercise, after electrostimulation and other treatments in the groups.RESULTS: ① The resting potentials of skeletal muscular cells after electrostimulation were decreased as compared with those before electrostimulation [experimental side: (59.48±7.37), (84.54±5.72) mV; control side:(59.43±7.14), (83.62±5.86) mV]. ② After the treatments of acupuncture,magnetic lifting needle, magnetic lifting needle plus pressing, the decreased resting potentials after electrostimulation were obviously recovered (P ＜ 0.01). ③ The recovered percentage of resting potential in the magnetic needle group was obviously smaller than those in the acupuncture group and magnetic needle+pressing group [(42.50±12.41)%, (53.71±10.93)%,(49.10±8.39)%, P ＜ 0. 01].CONCLUSION: Acupuncture, magnetic lifting needle, magnetic lifting needle plus pressing plays a role in promoting the recovery of resting potential of skeletal muscular cells after long-term electrostimulation, and the effect of simple magnetic needle is smaller than that of acupuncture and magnetic needle plus pressing.

Objective Protein chip for mucin detection was prepared to detect the serum levels of mucin1(MUC1),mucin2(MUC2)and micin5ac(MUC5AC)of colorectal cancer patients,and to discuss the relationship between colorectal cancer and the expression of MUC1,MUC2 and MUC5AC in serum.Methods The protein chip was prepared with monoclone and polyclone antibodies for the measuement of the serum levels of MUC1,MUC2 and MUC5AC in 30 patients with colorectal cancer and 30 healthy adults as control,using the method of immuno-fluorescence.Results The serum levels of mucin and CEA in colorectal cancer group were higher than that in control group.The differences were significant between the two groups concerning MUC1(5.25?2.37 vs 3.65?1.82U/ml,P0.05).The positive expression of mucin was related with the cancer differentiation.The positive expression of MUC1 and MUC5AC was higher when cancer differentiation was low as compared with high and median differentiation,but the positive expression of MUC2 was higher in high and middle cancer differentiation.When the serum level of mucin was used as an indicator for the diagnosis of colorectal cancer,the sensibility and specificity were both higher.The sensibility and specificity of MUC1 were 82.1% and 78.1%,respectively,while of combined mucins(at least 2 in 3 mucins were positive)were 96.2% and 85.3%,respectively.Conclusion The application of protein chip for mucin detection on the measurement of serum levels of MUC1,MUC2 and MUC5AC is of higher sensibility and specificity in the diagnosis of colorectal cancer.The establishment of protein chip has provided a new approach to early diagnosis of colorectal cancer,and help estimate the prognosis.

Objective To explore the surgical indication, the operative procedure and effect of breast-conserving surgery plus combined treatment for the patients with early breast cancer. Methods 51 patients with early breast cancer were treated with breast-conserving surgery (BCS) plus radiotherapy, chemotherapy and hormonotherapy postoperatively. The result of breast-conserving group was compared with 56 patients treated by modified radical mastectomy (control group). Results 25 patients underwent quadrant resection plus axillary lymph node dissection (ALND), 15 patients were subjected to local resection plus ALND, and 11 patients received quadrant resection only. All patients were followed-up for an average of 16.9 months. In BCS group, one patient was found to have local recurrence, and the distant metastasis rate was 0%. In the control group, no patient had local recurrence and the distant metastasis rate was 3.6%. The operative complication rate in BCS group and control group was 13.7% and 33.9% respectively (p﹤0.05). Conclusion Breast-conserving surgery plus combined treatment is safe and satisfactory for early breast cancer. It can be considered as the first choice for the treatment of patients with breast cancer in early stage.

Objective To enhance the understanding of chest primitive neuroectodermal tumor (PNET) and improve the results of management. Methods The results of 10 cases of chest PNET, which had been surgically treated and pathologically confirmed from 1999 to 2004 were retrospectively analyzed. Results There were 6 intrathoracic tumors, 2 in the chest wall, 1 in the pericardium, and 1 in the posterior mediastinum. Two intrathoracic tumors were completely resected, 2 partially resected, tumor and the involved lung were removed in 2 cases. Two had tumor with the affected ribs removed and the chest wall reconstructed. Tumor and the invaded pericardium were removed in 1 case. The PNET in the posterior mediastinum, invading into spinal canal as a bell tumor, was excised by the thoracic surgeon cooperating with neurosurgeon. There was no hospitalization death. All the specimens demonstrated positive reaction to CD99 and negative to LCA. 4 cases received postoperatively adjuvant chemotherapy, 4 radiotherapy and chemotherapy, the other 2 cases had none adjuvant therapy. 7 patients died during 6 years of follow-up, with 9 to 27 months survival. 3 patients are alive and have over 12 months survival so far. Conclusion PNET is an aggressive and invasive malignant tumor with poor prognosis. Difficulties exist in diagnosis. Besides common microscopic examination, pathological histology and immunochemistry, even electron microscopic examination may be required in differential diagnosis. Complete removal of tumor and involved tissue plays an important role, postoperative adjuvant therapy, including radiotherapy or combination with chemotherapy, may prolong survival. Modalities that are more effective should be developed to improve the treatment results.
