1.Clinical Observation on 150 Patients with Diabetes Type Ⅱ Treated by Qiyao Xiaoke Capsule Combined with Metformin Hydrochloride Tablets:A Multi-centered Random-Controlled Study

Objective To observe the therapeutic effect of Qiyao Xiaoke Capsule(Capsule for Diabetes) on the diabetes type Ⅱ.Methods Totally 240 diabetes type Ⅱ patients were randomized into a treatment group(n=150),and a control group(n=90);and other 30 diabetes patients who didn't regularly take hypoglycemic drugs were set as opening group.The treatment group was administered Qiyao Xiaoke Capsule plus Metformin Hydrochloride Tablets,while the control group was prescribed Xiaokeling Capsule in addition to Metformin Hydrochloride Tablets,and the opening group was given Qiyao Xiaoke Capsule only,with a leading period of two weeks.The patients were treated for 4 weeks.Before treatment and at the end of the 4 weeks' treatment,the fasting blood glucose(FBG),2 hours postprandial blood glucose(2hPG),and glycosylated hemoglobin(HbA1c) of all groups were determined,and scores of TCM syndromes and symptoms were observed to evaluate the therapeutic effect.Results The total effective rate of the treatment group was 74.67%,of the control group 72.22%,and in the opening group 70.00%.Except for the 2hPG of the opening group,the FBG,2hPG and HbA1c of 3 groups were decreased(P

Liver diseases post great threats to human public health globally.Lacking of appropriate small animal models largely impeded the translational studies on human liver diseases, especially on viral hepatitis and related cirrhosis, hepatocellular carcinoma, etc.By human hepatocyte transplantation, the liver-humanized mice have significantly contribu-ted to the researches of human liver diseases.This review summarizes the currently widely used and representative human-ized mouse models, including uPA, FAH, TK-NOG, AFC8 mice and their applications in studies of human liver diseases.
3.Effects of Survivin-T34A mutant on breast cancer cell in vitro and in vivo.

To study the effect of the proliferation and apoptosis of Survivin-T34A mutant on breast cancer MCF-7 cell, we adopted the method of cell culture in vitro to observe the proliferation and apoptosis of the cell. In the experiment, MCF-7 cells were randomly divided into three groups and transfected with normal saline, PORF-9-null and Survivin-T34A, respectively. Breast cancer nude mouse models were established to study anti-tumor effect of Survivin-T34A in vivo. The activity of the cells in the Survivin-T34A-transfected group was lower than that in PORF-9-null group. The increase of cell apoptosis was observed under electron microscopy, meanwhile the apoptotic rate was obviously higher than that in PORF-9-null control by flow cytometry. Tumor inhibition effects of the mouse which received the injection of Survivin-T34A intratumoral injection were apparent, and the inhibition ratio was as high as 47.1%. In conclusion, Survivin-T34A mutant has anti-tumor effect through efficiently inhibiting the growth of breast cancer MCF-7 cell and actively promoting apoptosis of cancer cells.

Purpose:To study the expression of survivin a nd its relationship with the expression of bcl-2,p53,PCNA in the dysplasia of gastric mucous epithelium and gastric carcinoma.Methods:The expression of survivin,bcl-2,p53,PCNA was detected in 60 cases of gastric carcinoma, 30 cases of dysplasia of gastric mucous epithelim and 8 cases of normal gastric mucous tissue by using immunohistochemical SP method. Results:The positive rates of survivin were 10%,13%,80% and 82%, respectively, in mild, moderate and svere dysplasia and gastric carcinoma. The positive rates of survivin in mild and moderate dy splasia were significantly lower than in severe dysplasia and carcinoma (P0 05). ObjectiveTo evaluate the effect of health education to women of childbearing age on knowledge of birthdefents.MethodsRandomly selected 200 case of maternal pre-pregnancy check-up in the hospital outpatient department were divided into two groups,The observation group of 100 cases,100 cases of the control group.the control group received rule progestation knowledge education,the observation group enforce health education intervention,apply the way of questionnaire survey,and the results were statistically analyzed.ResultsThe observation group answer correct rate is obviously higher than the control group ( all P < 0.05 ).ConclusionTaking a variety of measures of health education for pregnant women during the physical examination for effective interventions,could significantly improve the population birth defects knowledge rate had positive significance to the prevention of birth defects.
9.Effect of basic fibroblast growth factor on endogenous neural stem cell in rat cerebral cortex with global cerebral ischemia-reperfusion.

The present paper is aimedto investigate the effect of basic fibroblast growth factor (bFGF) on proliferation, migration and differentiation of endogenous neural stem cell in rat cerebral cortex with global brain ischemia-reperfusion. A global brain ischemia-reperfusion model was established. Immunohistochemistry was used to observe the pathological changes and the expression of BrdU and Nestin in cerebral cortex. RT-PCR was used to measure the NSE mRNA in brain tissue. The results of measurements indicated that in sham operation group, there was no positive cell in cerebral cortex, and the content of NSE mRNA did not change. In the operation group, the expression of BrdU and Nestin increased significantly at the end of the 3rd day, and peaked on the 7th day. NSE mRNA expression did not significantly increase. In bFGF group, compared with sham operation group and model group, the number of BrdU-positive and Nestin-positive cells increased significantly at each time point (P<0. 05), and peaked at the end of the 11th day, and the content of NSE mRNA increased significantly (P<0. 05). This research demonstrated that the proliferation of endogenous neural stem cells in situ could be induced by global cerebral ischemia and reperfu- sion, and could be promoted and extended by bFGF. In additiion, bFGF might promote endogenous neural stem cells differentiated into neurons.

10.Using quadratic regression universal rotary combination design to optimize the fluorescence labeling condition of lycium barbarum polysaccharide

Objective Quadratic regression universal rotary combination design was used to optimize the fluorescence labeling condition of lycium barbarum polysaccharide (LBP).Methods The fluorescence labeling condition of LBP presented, when the covalent coupling of LBP and tyramine was reacted with fluorescein isothiocyanate (FITC). Filter the best labeling condition via using quadratic regression universal rotary combination design experiment on the relationship of labeling efficiency among pH value of buffer solution, reaction time, temperature and the dose of tyramine.Results The regression equation was:Y=0.085 41 - 0.002 82X1 - 0.015 68X2 + 0.008 11X3 + 0.005 01X4 + 0.008 75X1X2 - 0.005 75X1X3 - 0.001 75X1X4 + 0.010 63X2X3 + 0.000 125X2X4 + 0.000 25X3X4 - 0.021 44X12 - 0.008 89X22 - 0.001 984X32 + 0.003 66 X42, and the variables fromX1 toX4 represented pH value, reaction time, temperature and dose of tyramine, respectively. The goodness of fitting of regression equation was statistically significant. The condition of labling LBP was optimized when the temperature was at 50℃, timing of labling at the fourth day and pH value was 8.5.Conclusion The condition of labling LBP was optimized with suitable temperature, pH value and extended timing.
