1.Role of inflammation in and effects of saponin of Panax Notoginseng on formation of foam cells

Objective To study the role of zymosan in and the effects of total saponin of Panax notoginseng (PNS) on the formation of foam cells originating from peritoneal macrophages. Methods The peritoneal macrophages of mice were cultured with zymosan, PNS and oxidized low density lipoprotein (ox-LDL) for 48 h. The shape of the foam cells was shown with the staining of red oil O dye. The content of total cholesterol (TC) in the foam cells was determined. The concentration of TNF? and IL-1 in the supernatant of the cell culture was measured. Results The TC content and the concentration of TNF-? and IL-1 were significantly higher in the zymosan group than in the PNS group. Conclusion Inflammation can accelerate the formation of foam cells through the pathway of inflammatory cytokines but PNS can reduce the formation with the inhibition of inflammation.
2.Investigation of the insulin dose-correlated factors in treating patients with type 2 diabetes

Objective To investigate the association of insulin dose with clinical factors of type 2 diabetes. Methods We reviewed the data of 214 type 2 diabetic patients who received insulin regimens.We compared the insulin dose and period at the targeted blood glucose levels among 6 groups according to different type of oral hypoglycemic agents. Results There existed statistically significant correlations of the glucose-targeted insulin dose with course,FPG,BMI,combination therapy with oral hypoglycemic agent(P<0.1,test level P=0.1).Merely metformin failure group took less insulin than the group admitted due to combination therapy failure (P=0.016, 0.53(0.35~0.62)U/kg and 0.63(0.51~0.75)U/kg respectively).The longest targeted period existed in the later. Relationship between the targeted bedtime insulin NPH dose and FPG level can be demonstrated by equation Y=0.255X+7.8. Conclusions The targeted insulin dose is influenced by synthetic factors. Patients admitted due to combination oral hypoglycemic drug failure have the maximal targeted insulin dose and the longest titration period. The final bedtime insulinNPH dose can be predicted by FPG level.

A strain producing pullulanase was isolated from soil and mutated by UV and EMS with the increase of pullulanase activity from 1.6 to 5.4 u/mL. The fermentation conditions and medium were studied and optimized. The enzyme activity can reach 8.8 u/mL under the optimal fermentation conditions and medium. The optimal pH and temperature for enzyme are 4.6 and 75℃ respectively. It shows a high stabiliby during pH4.0~8.0 at 55℃.
4.Clinical study on preserve the intercostobrachial nerve after radical operation for breast carcinoma

Objective To evaluate the clinical value of preserving the intercostobrachial nerve after radical operation for breast carcinoma.Methods The clinical data of patients with breast cancer were analyzed retrospectively,all cases were divided into the observation group and the control group.all cases were followed up after operation from six months to six years in order to observe the abnormal sensation of the skin and the recurrence rate.Results The abnormal sensation of the skin:1 case in the observation group(4.5%),17cases in the control group(73.9%),Two groups have significant difference(x2=20.79 P<0.01),both groups had not found partial recurrence.Conclusion Preseving the intereostobmchial nerve in modified radical mastectomy for patients with breast cancer,dichat increase the partial recurrence,can increase the life quality of patients after operation,decrease the abnormal sensation of the skin.promote the health both body and mind.
5.The effect of rosiglitazone combined with insulin on plasma aldosterone in diabetes patients

Objective To investigate if edema caused by rosiglitazone is associated with the change of aldosterone level. Methods Plasma aldosterone levels in different periods from patients treated with rosiglitazone combined with insulin were measured.Type 2 diabetes patients who have been treated with insulin alone were divided into two groups: patients treated with rosiglitazone(4mg/d) combined with insulin(RSG+insulin group), or treated with insulin alone (insulin group)(n=10). Aldosterone levels, incidence of edema and weight gain were monitored every two months during six months. Results The incidence of edema and weight gain were higher in RSG+insulin group than in insulin group. After two months, aldosterone levels were higher in RSG+insulin group than in insulin group[(77.8±25.9)vs(60.2±27.6)pg/ml,P>0.05],but it had no statistical difference; after four months, aldosterone levels were higher in RSG+insulin group than in insulin group[(87.2±27.1)vs (61.5±25.6) pg/ml,P<0.05]. After six months, aldosterone level recovered gradually,it had no statistical difference compared with that of control[(77.0±21.0)vs(69.6±21.7) pg/ml,P>0.05]. Conclusions The cause of early edema in type 2 diabetic patients treated with rosiglitazone combined with insulin was probably assosiated with elevated aldosterone level.
6.CTLA-4 and PD-1:the potential immunotherapeutic targets for malignant lymphoma

The human immune system has the regulatory functions of eradicating pathogens and limiting excessive inflammation, which protect the surrounding tissue from being damaged. The immune bal-ance of self-limiting is mainly controlled by complex interactions between antigen-presenting cells ( APCs ) and T cells. However, the immune balance is destroyed in cancer, which results in immune evasion and tumor metastasis or promotes the development of drug resistance. Immune checkpoints play critical roles in the immune system. Therefore, blocking tumor immune evasion by targeting the immune checkpoints has be-come a research focus in the treatment of relapsed or refractory malignant tumors. Currently, in the studies of malignant lymphomas, some phaseⅠ/Ⅱclinical studies of immune checkpoint inhibitors have achieved sur-prising results. This review will discuss the regulation and immunotherapy of immune checkpoints in malig-nant lymphomas.

8.Preparation and identification of monoclonal antibodies against pgp3 of Chlamydia trachomatis

Objective:To express secreted protein-pgp3 of Chlamydia trachomatis(Ct)plasmid,produce monoclonal antibodies (mAbs)and identify their basic biological characteristics.Methods: Construction pGEX-6p2-pgp3 prokaryotic expression vector,then expressed GST-pgp3 fusion protein in E.coli as antigen used to immune BALB/c mice, spleen cells were fused with SP2/0 mouse myeloma cells.The hybridoma cell lines of screening mAbs were secreted by ELISA,and mAbs specificity,type,class and titer were identified.Results:GST-pgp3 fusion protein was successful expressed,5 strains stable hybridoma cell lines that secreted mAbs were screened out,including 4 strains secreted anti-pgp3 mAbs(P1B3,P2A1,P2B6,P2C2),mAbs type were IgG1/κ,the other strain secretion anti-GST mAbs(P3B4),mAbs type was IgG2b/κ.The titer of P1B3,P2A1,P2B6,P2C2,P3B4 were 1∶6 400,1∶3 200,1∶12 800,1∶6 400 and 1∶6 400 respectively.Conclusion:Successful prepared anti-pgp3 mAbs,and lay a foundation for further study the function of Ct plasmid protein pgp3 and the establishment of Ct detection method.
9.Effects of Total Saponins of Panax Notoginseng on Oxidative Stress During Atherosclerogenesis in Rabbits Accelerated by Persistent Inflammation

OBJECTIVE:To study the effects of Total sasponins of Pannax Notoginseng(PSN)on oxidative stress during atherosclerogenesis(AS)in rabbits accelerated by persistent inflammation.METHODS:A total of 24 Japanese big-eared rabbits were divided into control group,high lipid group,persistent inflammatory group,and PSN group to be treated accordingly.Serum levels of C reactive protein(CRP),nitric oxide(NO)and malondialdehyde(MDA)as well as the activity of superoxide dismutase(SOD)were given a dynamic monitoring;and aortic atherosclerotic plaque areas were measured.RESULTS:In the persistent inflammatory group,serum levels of CRP and MDA were increased significantly,but SOD activity and level of NO were decreased significantly,aortic atherosclerotic plaque area was increased obviously,and CRP level was positively correlated with both MDA content and plaque area,showing significant difference as compared with the high-lipid group(P

[ ABSTRACT] AIM:To investigate the protective effect of 1, 3-dicyclopentyl-1, 2, 3, 6-tetrahydropyrimidine-4, 5-dicarboxylic acid diethyl ester (ZL-5015) on lethal endotoxin-challenged mice and to explore the underlying mechanism. METHODS:Mouse model of lethal endotoxin challenge and endotoxemia were established by intraperitoneal administration of lipopolysaccharide (LPS) at a dose of 70 mg/kg to the C57BL/6J mice.Mouse peritoneal macrophages stimulated with LPS (10 mg/L) were used as an in vitro inflammatory model.The levels of interleukin-1β( IL-1β) , interleukin-10 ( IL-10) and tumor necrosis factor-α(TNF-α) were measured by enzyme-linked immunosorbent assay (ELISA).Real-time PCR was used to evaluate the mRNA expression of the cytokines.RESULTS:Prophylactic treatment of the mice with ZL-5015 (100 and 200 mg/kg, ig) slightly increased the survival rate, extended the survival time, decreased the serum levels of IL-1βand TNF-α, and increased the serum level of IL-10 in the early stage of endotoxemia as compared with model group.The results of in vitro study demonstrated that treatment of the endotoxin-stimulated mouse peritoneal macrophages with ZL-5015 (10, 20 and 40μmol/L) inhibited the expression of IL-1βand TNF-αat both mRNA and protein levels but promoted the expression of IL-10 at both mRNA and protein levels.CONCLUSION: The tetrahydropyrimidine derivative ZL-5015 shows a moderate anti-endotoxin effect by increasing the survival rate and extending the survival time of the mice challenged by endotoxin, which may result from inhibition of the expression of pro-inflammatory cytokines such as IL-1βand TNF-α, and promotion of the expression of anti-inflammatory cytokine IL-10.
