Objective To study the inhibitory effect and mechanism of fluorofenidone on the proliferation of the rat lung fibroblasts induced by TGF-β1 . Methods Pulmonary fibroblasts cell were cultured and purified by using differentially adherent method, the third generation was used in the experiment. Cells were pretreated with 1,3,10 mmol?L-1 fluorofenidone, respectively. Pulmonary fibroblasts were identified by immunocytochemistry, and interfered by transient transfection. Cell proliferation was measured by BrdU marking. The mRNA and protein expression of eIF3α and p27 were analyzed by real-time PCR and Western blot. Results Immunocytochemistry staining identification of vimentin was positive in the third generation of pulmonary fibroblasts .TGF-β1 could obviously promote the eIF3α expression and proliferation of pulmonary fibroblasts. EIF3a siRNA was transfected with lipofectamine into cells, which obviously inhibit eIF3α expression induced by TGF-β1 , reversed cell proliferation induced by TGF-β1 , and could obviously promote p27mRNA expression induced by TGF-β1 . Fluorofenidone (3, 10 mmol?L-1 )could obviously inhibit cell proliferation induced by TGF-β1 and reduce eIF3α expression of pulmonary fibroblasts promoted by TGF-β1 . The inhibition rate was 66.7% and 78.8% respectively. Fluorofenidone (3 mmol?L-1 ,10 mmol?L-1 ) could also obviously reverse p27 expression inhibited by TGF-β1 , the reverse rate was 75. 5% and 71. 3% respectively. Conclusion eIF3α/ p27 signal pathway involves in pulmonary fibroblasts proliferation induced by TGF-β1 .Fluorofenidone can inhibit pulmonary fibroblasts proliferation induced by TGF-β1 by inhibiting eIF3α expression, enhancing p27 expression and/ or increasing p27 directly.

Objective To compare the effect of application of constant speed injection pump and manual injection on hysterosalpi-nography (HSG).Methods A total of 220 infertile patients were selected and classified into experimental group and control group. Then the patients were scheduled to undergo HSG.The experimental group was undergone constant speed pump injection and control group manual injection.Observe the effectiveness of the two methods on patients in two groups,respectively.Results The excellent rate of the images in experimental group (94.55%)was significantly higher than the control group (85.45%)and the risk of adverse reactions were significantly decreased in the former.There was statistical difference between the two groups.Conclusion Constant speed infusion pump is a convenient,safe and effective method in hysterosalpinography compared with manual injection,which is worthy of application in clinic.

BACKGROUND:Posttraumatic osteomyelitis is bone infection after surgery of open fractures or other joint, which leads to more secretions in antrum or bone exposed wounds, long treatment cycle, delayed healing of fracture, even disunion. The key link in the treatment is complete debridement, adequate drainage, and the use of external fixators with low interference and far away from lesions. ＜br> OBJECTIVE:To observe the clinical efficacy of external fixator combined vacuum sealing drainage (VSD) in the treatment of post-traumatic osteomyelitis, and compared with conventional catheter drainage. ＜br> METHODS:Since June 2010 to June 2013, 21 patients of post-traumatic chronic osteomyelitis were included in this study and divided into VSD group (n=11) and conventional catheter drainage group (n=10). Al patients in VSD groups underwent debridement and VSD was used to fil dead space, then the wounds were sutured. Cases appeared nonhealing fractures underwent external fixation, 10 cases using external fixators and 1 case retained original external fixator. Al patients achieved the closed wounds at the second phase, 9 cases were directly sutured, and 2 cases received skin flap transplantation. Among them, 1 case received autologous bone graft at the third phase because of bone defects. In the conventional catheter drainage group, al patients underwent debridement and conventional catheter drainage to rinse the wounds after surgery, 3 cases failing to close the wounds changed to receive ordinary dressing and skin flap transplantation at the second phase. Cases appeared nonhealing fractures underwent external fixation. Local swel ing, pain and fever were observed after treatment. The time of total hospitalization stay and fracture healing was recorded. ＜br> RESULTS AND CONCLUSION:The VSD group quickly control ed preoperative symptoms such as swel ing and pain and fever than the conventional catheter group (P＜0.05). The length of hospital stay showed no significant difference between the two groups (P>0.05). Al patients were obtained through clinic service and fol owed up for 24-36 months. The healing time in the VSD group was shorter than that in conventional catheter group (P＜0.05). Until the end of fol ow-up, no patients in the VSD group appeared infection recurrence, and two cases in the conventional catheter group had recurrent infection. The results suggest that VSD combined with external fixator can repair post-traumatic chronic osteomyelitis due to rapid control of infection, easy care, shortened treatment time, and reduced rate of postoperative recurrence. In addition, external fixator can be used as the final fixation for post-traumatic chronic osteomyelitis.

For the existing measurement methods of residual voltage which can't turn the power off at peak voltage exactly and simultaneously display waveforms, a new residual voltage detection method is put forward in this paper. First, the zero point of the power supply is detected with zero cross detection circuit and is inputted to a single-chip microcomputer in the form of pulse signal. Secend, when the zero point delays to the peak voltage, the single-chip microcomputer sends control signal to power off the relay. At last, the waveform of the residual voltage is displayed on a principal computer or oscilloscope. The experimental results show that the device designed in this paper can turn the power off at peak voltage and is able to accurately display the voltage waveform immediately after power off and the standard deviation of the residual voltage is less than 0.2 V at exactly one second and later.

Objective To investigate treatment and related prognostic factors of tongue carcinoma .Methods Clinical and patho-logical parameters of 1 171 cases of tongue carcinoma treated in Department of Oral and Maxillofacial Surgery ,School of Stomatolo-gy ,Harbin Medical University and Central South University from 2001 to 2007 were retrospective analyzed ,which included age , gender ,location ,pathological grade ,clinical TNM stages ,and treatment modality .Results After 3 to 5 year of follow-up ,survival rate of patients with tongue body carcinoma was obviously higher than that on the base of tongue .High clinical stage ,low cell dif-ferentiation degree of the postoperative patients had lower survival rate .The prognosis of patients with positive surgical margins and positive lymph node metastasis was poorer .The survival rate had no correlation with excision of mandible .When the tongue was re-stored with flaps ,the safety margins were increased to reduce recurrence .Conclusion Clinical TNM stages and surgical margin are independent prognostic factors for tongue cancer .Early diagnosis and treatment are vital to increase the survival rate of patients .

Objective To study the effects of methylprednisolone on the permeability of blood-spinal cord barrier (BSCB) and claudin-5 expression after spinal cord injury in rats. Methods The rat model of spinal cord injury was estab?lished using modified Allen method. SD rats were randomly divided into sham-operated group, spinal cord injury group and methylprednisolone pretreatment group. The permeability of BSCB and expression of claudin–5 were assessed at 12 h, 1, 3, 5, and 7 d after the onset of spinal cord injury (five animals per each time point). RT-PCR and Western blot were used to detect the expression of claudin-5. Results The success rate of the model was 84.0%. EB content was sig?nificantly higher in spinal cord injury group than in sham-operated group at each time point (F value 27.732,P<0.05). EB content was lower in methylprednisolone pretreatment group than in spinal cord injury group (F value 48.149,P<0.05). The mRNA expression of claudin-5 was lower in spinal cord injury group than sham-operated group at each time point (F value 12.248,P<0.05). The mRNA expression of claudin–5 was higher in methylprednisolone pretreatment group than in spinal cord injury group at each time point (Fvalue 15.316,P<0.05). The protein expression of claudin-5 was lower in spinal cord injury group than in sham operated group at each time point (Fvalue 18.108,P<0.05). The pro?tein expression of Claudin-5 was higher in methylprednisolone pretreatment group than spinal cord injury group at each time point (F value 20.247,P<0.05). Objective To investigate the clinical effect of standard large decompressive craniotomy in treatment of intracranial hematoma induced by severe traumatic brain injury (sTBI) and its influence on neuron-specific enolase (NSE) and inflammatory cytokines.Methods A total of 168 patients with sTBI-induced intracranial hematoma were assigned to standard large decompressive craniotomy (standard group) and traditional treatment (control group),with 84 patients per group.Glasgow outcome scale (GOS) and levels of serum NSE,TNF-α,IL-6 and IL-10 were compared between the two groups after surgery.Results GOS was better in standard group than in control group after surgery (P ＜0.05).Standard group presented significantly lower levels of serum NSE,TNF-α and IL-6 (P ＜ 0.05),significantly higher level of IL-10 (P ＜ 0.05) and a dropped ratio of IL-6 to IL-10 after surgery.However,there was no significant change in control group.Mean survival time of the patients in standard group was (33.11 ± 0.62) months.Two-year and three-year survival rate were significantly higher in standard group than in control group.Conclusion Standard large decompressive craniotomy cna enhance the clinical effects in patients with sTBI-induced intracranial hematoma,improve their neurologic deficits and daily living ability and decrease their body inflammatory stress level and injury.
