1.Current concept and overview of the technique of computer-assisted surgery(CAS) for total knee arthroplasty

Accurate alignment of knee implants is essential for the success of total knee replacemem.Recently,computer-aided navigation systems have become available,and the results showed more accurate and consistent installation of knee implants.The technology has evolved rapidly with widespread and continue to be developed.The current computer environmem and graphical user interface will become more user friendly and thus gain more widespread acceptance among experienced surgeons With this technology growing and systems evolving,computerized navigation will become the standard of care in knee reconstructive surgery.The key of future studies is to evove less invasive tracker anchoring systems,electronic balancers and computerized saws.A hybrid technique between image-based and image-free methods may develop to produce image-enhanced navigation.It should be an intermediary step between current technology and a fully robotic system.
2.Discussion About Development Strategy of Chinese Materia Medica

OBJECTIVE: To discuss about the key and entrance points of the development strategy of Chinese materia medica in the new century.METHODS: Some important items,such as difficult problems which Chinese materia medica facing to at present,new way to research into its effective substance bases,innovation on the theory and “ molecular targets” of Chinese materia medica,were analyzed.RESULTS & CONCLUSION: Chinese materia medica industry would become a most important pharmaceutical industry in China and really walk up to the whole world if traditional Chinese herb medicine supplies“ international language” for its exchange,qualitative and quantitative standards on judging the essence of its prescriptions and builds up its“ earmark” of Chinese materia medica industry.Based on the train of thought of this paper,suggestions how to promote the development of Chinese materia medica in the new century are put forward.

Since the first session of Chinese anti-inflammatory immunopharmacological society in 1982,many breakthroughs of immunological research have been made in western countries,such as identification of pathogens by natural immune cells,regulation of immune response,lymphocyte differ?entiation and development. At the same time,much progress has been achieved in anti-inflammatory and immunological research in China,especially in the field of basic scientific issues in immunology, frontier and hot topics,immunological mechanisms of major diseases and related drugs. The course of development of Chinese anti- inflammatory immunopharmacological research has experiences various stages,such as ″follow ″ in the past to ″work together ″ and to ″lead ″ in some research areas in today′s international immunology.
5.PH Domain and Cells Sense of Direction

The cells sense direction is closely related with the proteins that contain PH (pleckstrin homology) domain. PH domain has been found in about 60 proteins, many of which could activate the sequent events of signal transduction via combining with the related binding sites on the surface of chematactic cel ls. The characteristics of this combining are: a.rapid and transient; b.It is only related to the concentration gradient of surroundings outside cells, whi ch is the base of spatial model; c.The distribution of the binding sites on th e cell membrane changes when the researchers altered the position of the chemoat tractant. This is the base of temporal model. To deeply investigate the effects of all kinds of proteins which contain PH domain on cells sense of direction will greatly promote the research of this field, and hence, has great theoretica l significance.
6.Experimental Study on the Effects of Panax Notoginsengnoside on Neutrophil Functions in Rats With Air-vesicle Synovitis and Its Mechanisms

AIM:To study of effects of Panax notoginsengnoside(PnGL) on neutrophil(Neu) functions in carrageenin(Car) induced subcutaneous air-vesicle synovitis in rats and its mechanisms METHODS:Rat air-vesicle synovitis model was established with sc Car(25mg/kg) Neu count,the phagocytosis of Neu,the realease of O2 ?- from Neu and the level of Neu-i were measured with test tube diluting method,staphylococcus aureus phagocytosis method,cytochrome C reduction method and Fura-2 fluorescence technique respectively RESULTS:At 12h after Car used,Neu count of exudates elevated,phagocytosis rate and index of Neu and the release of O2 ?- from Neu increased obviously,suggesting that Neu was in an activated state Mea_nwhile,the level of Neu-i elevated significantly PnGL 60~240mg/kg ip,30min before and 6h after Car sc,reduced Neu count of exudates,inhibited phagocytosis functions and the increase of i level of Neu in a dose-dependent manner There was a significant correlation between the reduction of Neu-i and the inhibition of Neu phagocytosis functions and the release of O2 ?- from Neu Dexamethason had similar effects on Neu functions to PnGL,but with no effect on the level of Neu-i CONCLUSION:The reduction of Neu-i level and the subsequent inhibition of Neu functions is one of the important anti-inflammatory mechanisms of PnGL,which is different from that of Dex
7.Effects of hepatitis B virus X protein on the induction of cytochrome P450 3A4 in HepG2 cells

Objective To investigate the effect of hepatitis B virus X protein (HBx) on the induction of eytochrome P450 (CYP) 3A4 by 1α, 25-(OH)2D3 in HepG2 cells in vitro. Methods HepG2 cells were transiently transfected with plasmid pEGFP-N1 (control) or co-transfected with recombinant HBx eukaryotic expression plasmid pcDNA3-X and pEGFP-N1. All HepG2 cells were divided into four groups: control group (without transfection), plasmid pEGFP-N1 transfection group, plasmid pEGFP-N1 transfection plus 1α ,25-(OH)2D3 group, plasmid pcDNA3-X and pEGFP-N1 co-transfection plus 1α ,25-(OH)2D3 group. The expression of CYP3A4 in HepG2 cell was induced by 0.35 μ mol/L 1α ,25-(OH)2D3 for 72 h, and mRNA levels and protein levels of CYP3A4 in the cells were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) assay and Western-blot assay, respectively. The comparison between groups was done by F test. Results CYP3A4 mRNA level in plasmid pcDNA3-X and pEGFP-N1 co-transfection plus 1α ,25-(OH)2D3 group was 1.52 folds of control group, while that in plasmid pEGFP-N1 transfection plus 1α, 25-(OH)2D3 group was 3.97 folds (F= 4.72, P<0. 05). Similarly, intracellular CYP3A4 protein expression in plasmid pcDNA3-X and pEGFP-N1 co-transfection plus 1α , 25-(OH)2D3 group increased to 2.1 folds of control group, while that in plasmid pEGFP-N1 transfection plus 1α,25-(OH)2D3 group increased to 5.9 folds (F=4.68, P<0.05). Conclusion HBx interferes with the induction of CYP3A4 by 1α , 25-(OH)2D3 in HepG2 cell line, which suggests that HBx has suppressive effect on the expression of CYP3A4.
8.Hypoglycemic effect of the mixture of kelp extract, wolfberry, and Chinese yam to diabetic rats

Objective To investigate the therapeutic effect and its mechanism of pathogenicity of streptozotocin diseased rats treated by the mixture of kelp extract, wolfberry, and Chinese yam. Methods Streptozotocin-diabetic rat model were taken the mixture of kelp extract, wolfberry and Chinese yam with the ratio of 1:1:1 by intragastric administration. positive control group was fed metformin of 83.5mg/kg, the control group and the model group were given the same amount of distilled water. The safety of high dose (1500 mg) of the mixture therapy for the normal rat were evaluated, their blood glucose levels, glucose tolerance, body weight and insulin levels were observed and its possible mechanism were analyzed. Results Compared with the model group, the disease diabetic rats of the administered group (250mg/kg,500mg/kg,1500mg/kg) were significantly alleviated. First of all, coat color was more shiny, the behavior were more physical agility. Followed by the decrease of blood glucose levels and AUC, and body weight and insulin levels were elevated. In cell experiments, the number of islet cells was increased in the administered group by the mixture in a dose-dependent manner. Conclusion The mixture of kelp extract, wolfberry, and Chinese yam had obvious hypoglycemic effect. The hypoglycemic activity of the mixture may be attributed to increase insulin secretion through inhibit apoptosis of islet β-cells.
9.Study of influence of chronic cerebral hypoperfusion on cognitive function and mechanisms in rats of pomegranate juice

Objective To explore the influence of pomegranate juice on cognitive behavior of chroniccerebral hypoperfusion in rats and its mechanism.Methods 48 healthy male Wistar rats were selected and randomly divided into 3 groups, sham operation group (SO), permanent bilateral persistence commancarotid artery occlusion or two-vesselocclusion (2VO) group, pomegranate juice group.Chronic cerebral hypoperfusion model was prepared in rats by permanent bilateral common carotid artery ligation.Given the intervention after operation 1 weeks, before the 2 groups were given by sugar water intragastric ( every 100 mL syrup containing 25 g,sucrose 2 g, fructose glucose 2 g) with pomegranate juice, pomegranate juice group ( polyphenol content of 2.4-4.8 mg/mL) were given by gavage,2 mL/d, a total of 7 weeks.8 th weeks after surgery, cognitive behavior ability of 3 groups was tested by space exploration target quadrant swimming time after an interval of 1day;CA1 region of 8-hydroxy deoxyguanosine (8-OHdG), 4-hydroxyl nonenal ( 4-HNE ) expression of 3 groups in cortex and hippocampus detected by immunohistochemical method; observed whether it could improve the chronic cerebral hypoperfusion in rats learning, memory ability, or had antioxidative stress injury.Results On the 4th day and 5th day, the time of rats in SO to find the platform and the shortest time, group 2VO for the longest time, compared with statistical significance between the three groups (P<0.01); the 3 groups of rats in the target quadrant emergence time in sham operation group, the longest (10.05+1.688)s, 2VO group has the shortest time (6.343 ±1.220) s, and there was statistical significance (P<0.05).The integral optical density of positive cells in 3 groups of rats cortex and hippocampal CA1 region of 4HNE, the value of the 8-OHdG IOD is the lowest in the sham operation group, 2VO group was the highest, with statistical significance.Pomegranate juice intervention, cortex of rats and hippocampal CA1 region of 8-OHdG, 4-HNE decreased significantly ( P <0.05).Conclusion Chronic cerebral hypoperfusion may cause oxidative stress injury, pomegranate juice can improve the low irrigation learning and memory ability of rats with chronic cerebral, decrease 8-OHdG and 4-HNE expression may play a protective function through antioxidative stress pathway.
10.Study of the injurious effects of neutrophils on myocardium and their medicinal therapy in rabbits

It was found that there was a significant increase of the number of circulating leucocytes 2h after a subcutaneous injcetion of isoprenaline and severe myocardial necrosis occurred after consecutive injections for 3 d.Cytoxan-induced depletion of leucocytes resulted in an obvious decrease of the percentage of myocardial necrosis.In the control.the neutrophilic phagocytosis was enhanced and their release of acid phosphatase elevated greatly,which indicates that the neutrophils are in an activated state.After the.neutrophils were inhibited with dexamethasone or indomethacint significant reduction of the contents of prostaglandin E2 and malondialdehyde and an increase of superoxide dismutase activty in the myocardium were observed and consequently myocardial necrosis was greatly alleviated.Our findings suggest that neutrophils might play an important role in the pathogenesis of myocardial infarction through the release of lysosomal enzymes and the production of oxygen-derived free radicals and certain metabolites of arachidonic acid,and proper regulation of the neutrophilic functions could exert prophylactic effects on myocardial infarction.This might be a promising direction to look for more effective drugs to treat myocardial infarction.
