1.Analysis on the Basic Situation of Medical Service for Inpatients with Mental Illness in Beijing from 2003 to 2015

Objective:Through the analysis of the basic situation of the medical service of the psychiatric inpatients in Beijing from 2003 to 2015,it provided the basis for rational allocation of mental health resources and reasonable utilization of medical expenses.Methods:Descriptive statistical method was used to analyze the annual discharged patients,average hospitalization days and hospitalization expenses of inpatients with mental illness.Results:The annual discharged patients were increasing gradually.The number of discharged patients increased from 7861 to 24898 during 2003-2015.The annual average growth rate was 10.08%.The average length of stay in hospital since 20ll was lower than before,which were about 40 days in recent years.The average hospitalization expenses per day were also increasing.It increased from 131.04 yuan to 461.02 yuan during 2003-2015.The annual average growth rate was 11.05%.Conclusion:The demand of mental illness patients for hospitalization was gradually increasing.The hospitalization demand of patients with mental illness was increasing.The medical expenses quickly increased.It needed to adjust the payment policy to promote the rational and effective utilization of medical expenses.
2.Quality of life study among the female condyloma acuminatum pateints

Objective To explore the quality of life (QoL) among the female condyloma acuminatum patients.Methods 118 female condyloma acuminatum patients were interviewed face-to-face by the well-trained nurses with questionnaire including SF-36 Chinese version,the t-test and x2 test were used to compare the scale scores of SF36 between the patient and control groups.Results There were 101 eligible questionnaire,the scale scores of physical function(PF) and bodily pain(BP) were highest in the patient group,the scale score was (92.9 ± 10.5)points and (75.0 ± 1 1.2) points,respectively.Role limitations due to emotional problems (RE) was the lowest with the scale score only(50.8 ± 13.7)points.All scale scores of SF-36 in patient group,except PF(no statistical significant),were lower than that in the control group (all P < 0.01).Conclusion In the female condyloma acuminatum patients,the QoL is lower significantly than that in the control group,especially in emotional scales.
3.Molecular Typing of Neisseria Gonorrhoeae by Pulsed field Gel Electrophoresis and Random Amplified Polymorphic DNA Analysis

Objective To evaluate the ability of restriction endonuclease analysis by pulsed field gel electrophoresis (PFGE REA) and random amplified polymorphic DNA (RAPD) analysis for molecular typing of Neisseria gonorrhoeae isolates. Methods Genomic DNA from 36 Neisseria gonorrhoeae isolates were digested with SpeI and analysed by contour clamped homogeneous electric fields electrophoresis. The 36 isolates were also characterised by RAPD analysis with arbitrary primer OPA 03. Results 12~19 DNA fragments from 5 to 600kb were obtained among the 36 isolates after PFGE REA. The 36 isolates were discriminated into 20 PFGE patterns. Amplification of Neisseria gonorrhoeae DNA with primer OPA 03 produced 15 different DNA fragments (280~1900bp), and 3~9 fragments per strain could be seen. The 36 isolates showed 18 different RAPD patterns. Strains sharing common auxotypes and antibiotic spectrum could be differentiated by PFGE REA and RAPD analysis. General agreement was found between these two techniques. Isolates from different geographic areas and even some isolates in the same area showed considerable amount of DNA polymorphism. In addition, some isolates shared common or very similar patterns were unique to a given geographic area. Conclusion It is concluded that both PFGE REA and RAPD analysis are useful, sensitive molecular techniques for differentiation of clinical isolates of Neisseria gonorrhoeae. They should be helpful in the investigation of strain origin, clonal relation among strains and spread of antibiotic resistance. Compared with PFGE REA, RAPD analysis is faster, relatively simple and more economical.
4.Caspase 3 siRNA inhibits chondrocytes apoptosis

BACKGROUND: Up to date, studies concerning capspase 3 inhibitor mainly focus on peptide/non-peptide compounds synthesis and detection. Few reports addressing inhibits chondrocytes apoptosis using silenced caspase 3 gene. OBJECTIVE: To inhibit apoptosis of chondrocytes by blocking the apoptotic cascade reaction, gene silencing of caspase 3, and transduction of caspase 3 siRNA into chondrocytes with lentivirus vector.DESIGN, TIME AND SETTING: A single sample observation was performed at the Institute of Traumatic Surgery, Guangzhou Red Cross Hospital, Medical College of Jinan University from June 2008 to June 2009.MATERIALS: Chondrocytes were harvested from SD rats, and caspase 3 shRNA plasimid was constructed by our laboratory.METHODS: Rattus caspase 3 siRNA was synthesized and cloned into pSIH1-H1-copGFP plasmid. pSIH1-H1-copGFP-caspase 3 siRNA lentivirus was generated in 293TN cells by pPACKH1~(TM) Lentivector Packaging Kit and transducted into chondrocytes of rats.MAIN OUTCOME MEASURES: After the lentivirus was transducted into chondrocytes, the caspase 3 mRNA was tested by RT-PCR and the caspase 3 protein was tested by Western blot. Both the transducted cells and untransducted cells were induced apoptosis by tumor necrosis factor α (TNF-α). Cell apoptosis was assessed by flow cytometry, Annevin V/PI.RESULTS: The transduction rate of caspase 3 siRNA was about 90% by lentivirus vector. The expression of caspase 3 mRNA and caspase 3 protein in transducted chondrocytes was lower than the normal chondrocytes (P < 0.01). When the cells induced apoptosis by TNF-α, the apoptosis rate of the negative siRNA- chondrocytes was 7 times higher than that of caspase 3 siRNA-chondrocytes.CONCLUSION: The caspase 3 siRNA could inhibit caspase 3 expression and decrease drug-induced apoptosis of the chondrocytes.
5.The relationship between the stress-related psychosocial factors and the psychosomatic adjustment after surgery(II):The effect of Qi-Gong relaxation training

Objective To study the effect of Qi-Gong relaxation training on the operation-related psychological stress and the post-operative psychosomatic adjustment in upper-abdominal surgery patients. Methods Forty-two patients undergoing upper-abdominal surgery in intervention group were instructed to do Qi-Gong relaxation training, and forty patients matched were controls without Qi-Gong relaxation training. On the day before surgery, a self-developed patients' pre-operative appraisal questionnaire, a self-developed patients' pre-operative behavior rating questionnaire for completion by medical workers, and State Anxiety Inventory (SAI) of State -Trait Anxiety Inventory (STAI) were used in two groups. In an hour before operation,the shift of the mean value of pulse and systolic pressure of every case compared with the base line measured at his (or her) admission was also recorded. After surgery, patients were assessed with a self-developed patients' post-operative appraisal questionnaire, a self-developed patients' post-operative behavior rating questionnaire for completion by medical workers and SAI of STAI. Besides, the indexes of physical rehabilitation, including intestinal aerofluxus, total dosage of analgesia and so on, were recorded. Data were analysed in t-test with SPSS. Results Compared with controls, the patients in intervention group had more positive anticipation about the following operation, lower pre- and post-operative anxiety, and lighter post-operative pain experience. There was no difference in the indexes of physical rehabilitation between two groups. Conclusions Qi-Gong relaxation training could lead patients to better psychological adjustment both before and after operation. This finding suggests that Qi-Gong relaxation training could be used as an effective psycho-behavior intervention in abdominal surgery patients.
7.Study of a Bio-oss and rhBMP-2 composite repairing the alveolar bone defects around heterogenous implanted teeth

Objective: To evaluate the reliability and effects of a compound bone substitute materials Bio-oss artificial bone grafts with recombinant human bone morphogenic protein-2(rhBMP-2) filling the alveolar bone defects around heterogenous replanted teeth. Methods:3 dogs′ bilateral mandible 3rd premolars(6 sites) were extracted. The defect sites without buccal and lingual alveolar bone were prepared for the heterogenous implanted teeth and the compound of Bio-oss and rhBMP-2 filled the alveolar bone defects around. The bone regeneration was observed by naked eye, radiograph, histology and histomorphometric analysis after 2, 3 months. Results:The new alveolar bone showed osteogenesis and calcification after 2 months, and intergrated with implanted teeth root under the light microscope and X ray. 8.The Role of Prostaglandin E_2 on Pathogenesis of Candidal Vaginitis

In order to study the role of prostaglandin E2(PGE_2)on the pathogenesis of candidal vaginitis,the effect of Candida albicans and histamine on the production of PGE_2 by peripheral blood mononuclear cells from patients with candidal vaginitis and normal controls,the effect of PGE2 on germ tube formation of Candida albicans,and the effect of PGE2 on Candida albicans adhering to vaginal ep- ithelial cells were observed.The results showed that:Candida albicans and histamine induced PGE2 synergistically,and the production of PGE2 increased markedly.PGE2 enhanced germ tube formation in Candida albicans,increased the number of Candida albicans adhering to the vaginal epithelial cells, and the pathogenicity of C.albicans.The above results indicate that PGE_2 plays a role in the pathogene- sis of candidal vaginitis.
9.Experiment research of natural killer cells amplification in vitro and the killing effect on ovarian cancer cells

Objective To amplify natural killer (NK) cells in vitro and explore its killing effect on ovarian cancer cells.Methods (1) The separation of NK cells and identification.A total of 20 ml peripheral blood of one healthy volunteer was collected in Nov.2015,Peking University People's Hospital.The peripheral blood mononuclear cells of normal volunteers were isolated,cultured in vitro and amplificated cultivation for 14 days with K562 cells transfected and expressing interleukin 21 (IL-21-K562) as nourish cells.The number and dynamic state of the growth cells were monitored during the cultured process.Cells were harvested and counted after 14 days cultured.The NK cells phenotypes were detected by flow cytometry.(2) The killing effect of NK cells on ovarian cancer cells:the ratio of effector cells (NK cells) and target cells (ovarian cancer cells and its control) was 50∶ 1,20∶ 1,10∶ 1,5∶1 or 1 ∶ 1,NK cells killing effect on ovarian cancer cells was detected by the lactate dehydrogenase (LDH) release experiments.Results (1) The results of NK cells establishment and phenotypic characterization:the cells were induced in vitro for 14 days by amplification culture.With the extension of incubation time,the number of NK cells increased constantly,from 2.0× 107 on day 0 to 5.1 × 109 on day 14.Obvious amplification of the total number of cells were detected for 255 times.Living cells unstained by trypan blue eventually reached 95% above.Before and after the induction and amplification in vitro,the percentage of NK cells (CD3-CD56+cells) in CD3-cells were 2.33% and 85.32%,respectively (P<0.01),which covered the whole lymphocytes 1.06% and 69.42%,respectively (P<0.01),which showed that NK was the main cell type in the amplificated lymphocytes.(2) The killing rate of NK cells on ovarian cancer cells in vitro:the results detected by LDH release experiments showed that NK cells could performed strong nonspecific killing effect on ovarian cancer cell lines SKOV3,HOC1A,3AO and CAOV3,as well the normal ovarian cell line T29 and NK sensitive cell line K562,and the killing effect increased significantly along with the increase of effector cells and target cells ratio (P<0.01).When the ratio was 1 ∶ 1,the killing rate was 37% for K562,while the rate of killing of other cells was around 10% (P<0.05).When the effect-target ratio was 20∶1 and 50∶ 1,in addition to CAOV3 cells (more than 70%),NK cells had a kill rate of more than 80% for other ovarian cancer cells lines and their control cell K562 and T29 cells (P>0.05).Conclusion NK cells could be established in vitro and have a good non-specific killing effect on ovarian cancer cells.
10.Nursing observation of adverse reactions of advanced lung cancer treated with Apa imatinib mesylate

Objective Study on 60 cases of advanced non-small cell lung cancer patients in the Apa imatinib mesylateadverse reactions after treatment , and to develop nursing methods for these adverse reactions. Methods Non-small cell lung cancer patients who were treated in our hospital from September 2015 to September 2016 were treated with Apa imatinib mesylate and observed the adverse reactions during the course of treatment. Results Adverse reactions occurred in 45 patients, and the incidence of adverse reactions was 75.00%.29 cases of patients with skin rash, 24 cases occurred in patients with hypertension, 30 cases of patients with diarrhea, nausea and vomiting occurred in 21 patients, 5 cases occurred in patients with proteinuria, 7 cases of patients with oral mucositis.Conclusion Non-small cell lung cancer patients had adverse reactions after treatment with Apa imatinib mesylate and nursing intervention can improve the quality of life of patients.
