1.The expression and significance of ERCC1 in the tissue of nasopharyngeal carcinoma

Objective To study the expression and significance of excision repair cross complementing genes 1 (ERCCI) in the tissue of nasopharyngeal carcinoma(NPC).Methods ERCC 1 proteins was assayed with immunohistochemical method in the tissue of 103 cases of NPC and 50 cases of chronic inflammation of nasopharyngeal epithelium(CINE).Results The expression rate of ERCC1 was 45.63% in NPCs,which was significantly lower than that in CINE (70.00%) (χ2 =8.037,P < 0.05).The expression rate of ERCC 1 in NPCs had relationship with T,N staging and clinical staging as well as the presence of recurrence and metastasis (χ2 =9.007,9.992,8.343,5.792,all P < 0.05),while no relationship with age and gender(χ2 =0.026,0.871,all P > 0.05).The expression of ERCC1 in group of recurrence and metastasis was significantly lower than those in group of no recurrence and metastasis.Conclusion The expression of ERCC1 was significantly decreased in NPCs.ERCC1 will probably become prognostic biological parameters of NPC.
2.The study on the effect of psychotherapy in patients with methadone maintenance treatment

Objective To explore the effect of psychotherapy in patients with methadone maintenance treatment.Methods 60 patients accepted drug and 69 patients accepted drug with psychotherapy treatment were evaluated with Self-Rating Anxiety Scale(SAS) and Self-rating Depression Scale(SDS) in the beginning of psychotherapy therapy and 3 months later.All of them were assessed for heroin use at 3 months after the end of a 3-month treatment period.Results SAS、SDS did not show any significant differences between the two groups in the beginning(P >0.05).After 3 months of psychotherapy,the differences in SDS and SAS between the two groups showed statistically significant(t =- 2.858,- 2.465,all P <0.05).After 2 months of psychotherapy,the number of relapsing again patients of group psychotherapy treatment was significantly lower than control group(x2 =6.906,5.054,5.100,4.247,all P <0.05).Conclusion The mental healthy status of patients with heroin dependence is poor.Psychological treatment plays an important role in short-term prevent hernoin addicts from relapse again.
3.Potential mechanism of myocardium apoptosis of right ventricle in rat under chronic hypoxia

AIM: We used an animal model of chronic hypoxia to mimic right ventricular hypertrophy and try to study the potential mechanism of myocardium apoptosis of right heart in rat under chronic hypoxia.METHODS: Rat hypoxia models were established by exposing the rats to normobaric chronic hypoxia(oxygen levels were maintained at 9.5%-10.5%).Sixty rats were separated into two groups: one exposed to hypoxia and the other serving as control.Ten rats,randomly selected from each group were killed at 14,21,28 d after hypoxia.The apoptosis was determined.The changes of RV weight to left ventricle and interventricular septum weight ratio[RV/(LV+S)],the RV weight to body weight ratio(RV/BW) were also observed.The ?-MHC,bcl-2 and bad mRNA levels in right ventricle were detected by semi-quantitative RT-PCR assays and expression of ?-MHC,Bcl-2 and Bad protein levels were detected by Western blotting.RESULTS: The RV/(LV+S),RV/BW and apoptosis index in chronic hypoxia group were higher than those in normal control group(P0.05).Finally,a decreased bcl-2/bad ratio in chronic hypoxia group was found compared with control group(P
4.gyrA and parC genes mutations in quinolone-resistant clinical isolates of Pseudomonas aeruginosa

Aim To study gyrA and parC mutations of clinical Pseudomonas aeruginosa strains. Methods MIC values of 55 clinical P.aeruginosa isolates were determined by agar dilution test and 1 sensitive strain and 8 resistant strains were selected with standard sensitive strain ATCC27853 as control, the quinolone determining region (QRDR) of the gyrA and parC genes were amplified by PCR, the lengths of PCR products were 351 bp and 397 bp. The gyrA PCR products(351 bp) were digested with enzyme sacⅡ. The gyrA and parC gene were sequenced. Results In this study, gyrA genes of all resistant strains had an ACC to ATC mutation in codon 83, leading to the amino acid substitution of an isoleucine for a threonine, and three high level resistant strains also showed a GAC to GGC mutation in codon 87, leading to the substitution of a glycine for an aspartic acid. In addition, four resistant strains also had an TCG to TTG mutation in codon 87 of parC gene, leading to the amino acid substitution of a serine for a leucine. The strains with both gyrA and parC mutations were two to sixteen times more resistant than the strains which had only gyrA mutations. At the same time, a silent mutation (CAC to CAT) in codon 132 of gyrA gene and a silent mutation(GCT to GCG) in codon 115 of parC gene occured, which did not lead to amino acid change. Conclusion The mutations of 83 and 87 codons of gyrA and the mutatations of 87 codon of parC gene were related to fluroquinolone resistance, and the mutations of the 83 codon of gyrA gene were more important.

Objective To compare efficacy of pemetrexed (MTA) combined with cisplatin (DDP) and docetaxel (TXT) combined with DDP in treatment of non-small cell lung cancer (NSCLC).Methods 50 patients with NSCLC from May 1, 2013 to May 1, 2015 in the hospital were seleced and randomly divided into two groups:MTA group (n=25) were received MTA combined with DDP, and TXT group(n=25) were received TXT combined with DDP, for a course of twenty-one days.After treatment, the efficacy of two therapy as well as adverse reactions were compared.ResuIts After treatment, the soluble intercellular adhesion molecule 1 (sICAM-1) and soluble vasccular cell adhesion molecule 1 (sVCAM-1) levels in MTA group were significantly lower than those before treatment (P<0.05), but there were no statistical differences in TXT group, and the above two indicators in MTA group were lower than those in TXT group(P<0.05).There were no significant difference of total efficacy between MTA group and TXT group (20.00% vs.16.00%, χ2 =0.136,P>0.05).The adverse reaction of nausea and vomiting, anemia, low platelet count, neutropenia and alopecia in MTA group was lower than that in TXT group (40.00%vs.72.00%,χ2 =5.195,P<0.05).ConcIusion The efficacy of MTA combined with DDP and TXT combined with DDP in treatment of NSCLC is similar, but the adverse reactions of fomer one is lower.
7.Analysis of risk factor and intervention of intracranial hemorrhage in the premature infants

Objective To explore the risk factors and seek effective intervention of intracranial hemorrhage in the premature in-fants .Methods Clinical data of the premature infants in our hospital from January 2009 to December 2013 was retrospectively ana-lysed and single factor analysis of 20 relevant factors was done for cases with intracerebral haemorrhage and without intracerebral haemorrhage .Logistic regression analysis were done for some influence factors of intracranial hemorrhage .Results 1 726 cases of premature babies were included in the study ,including 264 cases of intracranial hemorrhage .Logistic regression analysis results shown that the neonatal transport network and integrated active transport models are protective factors of intracranial hemorrhage in the premature infant .We found that basic-level hospital transport was an independent risk factor .Between January 2009 and De-cember 2011 ,142 of 714 premature infants were intracranial hemorrhage ,including 88 cases from 348 patients transported from bas-ic-level hospital ,the incidence of intracranial hemorrhage was 25 .29% ,and compared with the incidence of intracranial hemorrhage (14 .75% ) of our hospital ,the difference was statistically significant (P<0 .05) .From January 2012 ,we established perfect neonatal transport network and implementation of comprehensive active transport model .122 of 1 012 premature infants were intracranial hemorrhage ,including 75 cases of 490 patients from basic-level hospitals .The incidence was statistically significant different com-pared with the incidence of intracranial hemorrhage(9 .00% ) transported from our hospital(P<0 .05) .The incidence of intracranial hemorrhage in the premature infants transported from basic-level hospitals were statistically different before and after neonatal transport network and comprehensive active transport model was established (P<0 .01) .Conclusion It will effectively reduce the incidence of intracranial hemorrhage in the premature infant by establishing the perfect regional neonatal three-level network trans-port system and comprehensive active transport models .
8.Determination of gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction

AIM: To determine gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction(Semen Persicae,Flos Carthamietc.) METHODS: To analyse three constituents by HPLC on a Hypersil BDS C_(18) column and gallic acid detection wavelength was set at 271 nm.The mobile phase was methanol-water(5∶95) (adjusted to pH=3.0 with H_3PO_4) at a flow rate of 1.0 mL/min. Safflor yellow detection wavelength was set at 403 nm.The mobile phase was methanol-water(80∶20) (adjusted to pH=3.0 with H_3PO_4) at a flow rate of 1.0 mL/min. Paeoniflorin detection wavelength was set at 230 nm.The mobile phase was methanol-KH_2PO_4 at a flow rate of 1.0 mL/min. RESULTS: Gallic acid showed a good linear relationship at the range of 4.547?10~(-3)-1.455?10~(-1) ?g/?L(r=0.999 3).Paeoniflorin showed a good linear relationship at the range of 0.010-0.200 ?g/?L(r=(0.999 8)) and safflor yellow showed a good linear relationship at the range of 2.34?10~(-2)-7.50?10~(-1) ?g/?L(r=(0.999 4)).The average recoveries were 98.0% for gallic acid,98.4% for paeoniflorin and 99.2% for safflor yellow.The RSD of the method were 1.8%,0.69% and 0.88%,respectively. CONCLUSION: The method is proved to be fast,feasible,simple,and can be used for quality control of gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction.
9.Clinical and laboratory investigation of hematological malignancies with t(11;19)(q23;p13.1)

Objective To investigate the clinical and laboratory characteristics of hematological malignancies with t(1 1;19)(q23;p13.1).Methods Chromosome specimens of bone marrow cell from a patient with hematological malignancy were collected.After short-term culture,and chromosome karyotype analysis was carried out by R banding technique.Results The chromosome karyotype of the patient diagnosed with AML M4+ was t (11;19)(q23;p13.1).The patient did not obtain complete remission after application of MA regimen chemotherapy.Conclusions t(11;19)(q23;p13.1) translocation is a rare and recurring chromosome abnormality,which is related with a specific type of AML.The prognosis of the AML patients with this chromosome abnormality is poor.
10.Detection of multi-leaf collimator leaf position errors in implementing static intensity-modulated plans and its effects on dose distribution

Objective To design a method for detecting multileaf collimator (MLC) leaf position accuracy in implementing a static intensity-modulated plan and to analyze the impacts of leaf errors on dose of targets and normal organs.Methods Static intensity-modulated planning for twenty lung cancer cases through dose verification was sorted in an ascending order according to the number of segment,and then the first and the last 10 plans were sorted as the simple plan group and the complex plan group,respectively.These plans were transmitted to a Varian 600CD accelerator and implemented by it.Photos were taken with PV aS500 electronic portal imaging device (EPID) and actual position of leafs was determined by gradient algorithm to calculate the pass rate for leaf verification.MLC files were modified according to examination results and the plans were re-calculated while keeping other parameters unchanged.Thus,difference of targets and normal organs dose distribution before and after the appearance of leaf errors were obtained.Results The dose distribution of most organs after leaf errors were increased or decreased,and the maximum dose of spinal cord in the sixth and thirteen cases exceeded the limit of 45 Gy.In the group of simple plan only the changes of maximum dose to the spinal cord were statistically significant(t =-3.08,P < 0.05),while in the group of the complex plan all changes of D95% of PGTV and PTV,maximum dose of the spinal cord,V20 of lung and V40 of heart were statistically significant(t =-1.89,-1.99,-2.36,-2.55,-1.85,P < 0.05).Conclusions To ensure the safety and effects,it was necessary to detect leaf position,particularly the complex intensity-modulated planning.Electronic portal imaging devices and treatment planning system could detect leaf positions during the implementation of a plan and obtain the actual dose of targets and normal organs.
