Objective The experiment aims at probing the best condition of the isolation and purification of rat islets. Methods The islets were isolated from rat pancreas by hepatopancreatic duct perfusing with collagenase and purified with Ficoll 400 discontinuous density gradient centrifugation. Then the purified islets were subjected to histological staining, electron microscopy and radioimmunoassay for identification of specificity and viability. Results The histological staining revealed that the viability and the purity of the purified islets were above 95% and 85% respectively.Electron microscopy showed that the purified islets were morphologically intact with clear membrane and plenty of secreting granules.Radioimmunoassay demonstrated the secreted insulin concerntration between low-glucose groups and high-glucose groups varied significantly, which verified the good function of the islets. Conclusion Hepatopancreatic duct perfusing with collagenase is a good method for digestion. There are many factors that influence the quantity and quality of the acquired islets, such as the completed expansion of pancreas, the concentration and viability of collagenase and the digested time, etc.

Objective To investigate telomerase activity in patients with psoriasis. Methods The telomerase activity in the skin lesions of patients was detected by TRAP (Telomeric repeat amplification protocol)——ELISA which is based upon PCR amplification of the initial telomerase product and detected by ELISA. Results Telomerase activity was detected in the lesions of psoriatic patients, but the level of it was lower in comparison with that in tissues of malignant tumor and K562 cell line. Conclusion These findings support that the increase of telomerase activity may be linked to the pathogenesis of psoriasis and that the telomerase activity can be detected not only in malignant tumors but also in nonmalignant skin diseases.

Objective To measure the expressions of Toll like receptors (TLRs) in lesions of condyloma acuminatum (CA). Methods Tissue samples were resected from lesions of 25 patients with CA and normal skin of 16 human controls. Frozen sections were prepared and the expressions of TLRs 1 - 10 were measured by immunohistochemical staining. Results In normal skin samples, there was a strong expression of TLR 6 and 9, medium expression of TLR 1, 3, 10 and weak expression of TLR 2, 4, 5, 7, 8 in the stratum corneum and stratum granulosum, but a weak or negative expression of TLRs in the stratum spinosum and stratum basale. In contrast, a strong expression of TLR 1, 3, 6, 9, medium expression of TLR 4, 8, 10 and weak expression of TLR 2, 5, 7 were detected in keratinocytes of almost all epidermal layers except for stratum basale in CA lesions. Conclusions The expressions of TLRs, especially TLR 1, 3, 6, 9, are markedly enhanced in proliferated keratinocytes in CA lesions compared with normal skin, which is likely to be associated with anti-viral immunity triggered by HPV infection.

Objective To investigate the the effect of long-term glucocorticoid on bone mineral density (BMD) and bone turnover makers in patients with glomerular diseases. Methods The dual-energy x-ray absorptiometry (DXA) was used to measure the bone mineral density of lumbar spine (L1~L4), femoral neck and trochanter of the 97 patients treated with glucocorticoid and the 20 patients in the control group. In addition , ELISA assay was used to measure the concentrations of bone turnover makers including serum PINP and CTX-I. Results (1)Compared with the control group, the BMD of lumbar spine (L1 ~ L4), femoral neck and trochanter and the concentrations of PINP were significantly lower, while the concentration of CTX-I was increased (P < 0.05);(2) Following up the passage of time and the accumulation of the amount of GC application , the BMD of lumbar spine , femoral neck and trochanter , and the concentrations of PINP decreased , while the concentration of CTX-I steadily increased (P<0.05);(3)Multiple linear regression analysis indicated that the BMD of lumbar spine and trochanter were negatively correlated with the time of GC application (P<0.01),and the concentrations of PINP and CTX-I were correlated with the cumulative doses of GC (r = -0.310 vs 0.221, P < 0.05);(4)The incidence of bone abnormalities in patients received vitamin D and calcium was markedly reduced (P<0.05). Conclusion The long-term glucocorticoid treatment for the patients with glomerular diseases can lead to bone mass reduction or osteoporosis , it can be helpful for early prevention and treatment of glucocorticoid osteoporosis with bone mineral density and bone turnover makers.
