1.The Clinical Study of the H1N1 Influenza Virus with Fanggan Decoction

[Objective]To discuss the clinical effect of Fanggan decoction on H1N1 Influenza virus.[Method]Randomly divided 95 cases into 2 groups,treatment group 48 and control one 47.Treatment group takes Fanggan decoction,the control one uses reducing fever tablet or anti-virus drugs if needed.[Result]The effective rate was 89.6% for treatment group and 85.1% for the control one.The treatment group was better than the control one on the reducing fever time,cough duration and cold symptom time.[Conclusion]Fanggan decoction has a good effect on H1N1 influenza virus.It also can be one of the therapies to relieve influenza patients in Zhejiang Province.
2.Effect of fibronectin on infection of primary human fetal hepatocytes with HBV in vitro

Objective This experiment was carried out to study the effect of fibronectin on the infection of primary human fetal hepatocytes with hepatitis B virus (HBV) in vitro. Method Human fetal hepatocytes were inoculated with HBV positive serum in culture dish coated with fibronectin (Fn) in vitro. HBsAg and HBeAg in supernatant were assayed with ELISA, HBcAg in nuclei by immunohistochemistry, HBV DNA in cells by fluorescence quantitative PCR, and cccDNA in cultured cells by nest PCR. The results were compared with the cells cultured in dishes without fibronectin coating. Results HBsAg and HBeAg in supernatants were positive on the fifth day in dishes without Fn coating, and on the first day in dishes with Fn coating. HBcAg in nuclei was positive on the second and the first day in dishes without and with Fn coating, respectively, and the positive rates were about 10%~15% and 90%, respectively. HBV DNA was detected in cells on the fourth day and first day in dishes without and with Fn coating, respectively. CccDNA in cells was positive on the eighth day and the first day in dishes without and with Fn coating, respectively. Conclusion Fibronectin coating could facilitate HBV infection in primary fetal hepatocytes in vitro. The infection time of HBV was shortened, and the number of infected cells was increased.

4.Effect of heme oxygenase-1 on hypoxia-reoxygenation injury in cultured neonatal rat cardiomyocytes

Objective To investigate the effect of heme oxygenase-1 (HO-1) on hypoxia-reoxygenation (H/R) injury in cultured neonatal rat cardiomyocytes. Methods Primary cultured neonatal rat cardiomyocytes were randomly divided into4 groups: Ⅰ control group (group C), Ⅱ H/R group, Ⅲ hemin group and Ⅳ hemin+zinc protoporphyrin (ZnPP) group. In group H/R, the cardiomyocytes were exposed to 2 h of hypoxia followed by 6 h of reoxygenation. Hemin was added to the culture medium with a final concentration of 20 μmol/L 24 h before hypoxia and immediately before hypoxia in group hemin. Hemin and ZnPP were added to the culture medium with final concentrations of 20 μmol/L 24 h before hypoxia and immediately before hypoxia in group Henin + ZnPP. The cells were incubated in 35 mm culture dishes (2 ml in each dish) and 50 ml culture flasks (3 ml in each flask)(45 dishes and 3 flasks in each group). The HO-1 expression, cell survival rate, LDH activity, malondialdehyde (MDA) level and superoxide dismutase (SOD) activity were detected after the end of reoxygenation. Results Compared with group C, the LDH activity, MDA level and HO-1 expression were significantly increased, while the cell survival rate and SOD activity decreased in the other three groups ( P < 0.05). Compared with group H/R,the LDH activity and MDA level were significantly decreased, while the HO-1 expression, cell survival rate and SOD activity increased in group hemin ( P < 0.05 ), but no significant change was found in group hemin + ZnPP (P >0.05). H/R injury was obvious in group H/R and hemin + ZnPP, but was significantly attenuated in group hemin. Conclusion HO-1 can protect cardiomyocytes against H/R injury in neonatal rats.
5.gyrA and parC genes mutations in quinolone-resistant clinical isolates of Pseudomonas aeruginosa

Aim To study gyrA and parC mutations of clinical Pseudomonas aeruginosa strains. Methods MIC values of 55 clinical P.aeruginosa isolates were determined by agar dilution test and 1 sensitive strain and 8 resistant strains were selected with standard sensitive strain ATCC27853 as control, the quinolone determining region (QRDR) of the gyrA and parC genes were amplified by PCR, the lengths of PCR products were 351 bp and 397 bp. The gyrA PCR products(351 bp) were digested with enzyme sacⅡ. The gyrA and parC gene were sequenced. Results In this study, gyrA genes of all resistant strains had an ACC to ATC mutation in codon 83, leading to the amino acid substitution of an isoleucine for a threonine, and three high level resistant strains also showed a GAC to GGC mutation in codon 87, leading to the substitution of a glycine for an aspartic acid. In addition, four resistant strains also had an TCG to TTG mutation in codon 87 of parC gene, leading to the amino acid substitution of a serine for a leucine. The strains with both gyrA and parC mutations were two to sixteen times more resistant than the strains which had only gyrA mutations. At the same time, a silent mutation (CAC to CAT) in codon 132 of gyrA gene and a silent mutation(GCT to GCG) in codon 115 of parC gene occured, which did not lead to amino acid change. Conclusion The mutations of 83 and 87 codons of gyrA and the mutatations of 87 codon of parC gene were related to fluroquinolone resistance, and the mutations of the 83 codon of gyrA gene were more important.
6.Reduction of hemoglobin induced by linezolid therapy: a retrospective study

The medical records of 197 patients on linezolid therapy were collected retrospectively.The prevalence and risk factors for linezolid-induced reduction of hemoglobin were identified.The incidence of linezolid-induced reduction of hemoglobin was 31.98%.The patients with reduction of hemoglobin had older age [(68 ± 14) vs.(59 ± 18) years,P =0.000] and lower creatinine clearance (CCr) [(72 ±42)vs.(98 ± 52) ml/min,P =0.000].Multivariate logistic regression analysis showed that age ≥65 years,treatment duration ≥ 15 days,CCr < 65 ml/min and pre-treatment hemoglobin value < 95 g/L were significant risk factors for linezolid-induced reduction of hemoglobin.

The experiment was performed at the Institute of Treatment and Prevention of Tumor of Jilin Province and Department of Etiology, Jilin Medical College from October 2004 to June 2005. The normal umbilical cord blood (UCB) was provided by a healthy lying-in woman from Changchun Hospital with the permission of the pregnant woman. Full-term normal delivery and UCB of healthy pregnant women was collected by aseptic venepuncture after fetal disengagement, and with Natrium Citricum anticoagulation. After the UCB mononuclear cells were isolated by Ficoll-paque (relative volume mass: 1.077?0.22) density gradient centrifugation, long-term liquid culture system was used for UCB adherent cells, and co-cultured with UCB mononuclear cells. The adherent cells were observed and mononuclear cell cycles were tested with flow cytometer. It showed that adherent layer of UCB increased UCB mononuclear cells expansion as compared with that of non-adherent layer of UCB after co-culture for 7 days. AIM: To study the effect of the radix sophorae flavescentis on cellular immunity in rats with Pneumocystis Carinii Pneumonia (PCP) induced by long-term use of immunosuppressant, and explore the action of traditional Chinese medicine for the immunological regulation and infectious prevention after organ transplantation. METHODS: The experiment was conducted at Department of Pathobiology, Jilin Medical College from May 2005 to March 2006. Forty adult healthy female SD rats were selected from Harbin Medical University (Certification: 02473146) and randomly divided into experiment group and control group, with 20 rats in each. The model of PCP was set up by glucocorticoid injection subcutaneously to SD rats (25 mg once, 2 times/week). The mixture of sophorae flavescentis was given to stomach with tube in experiment group (3 mL/kg, 2 times/day), and was consisted of radix sophorae flavescentis, ash bark, amur cork-tree, malt, milkvetch root and danshen root. Six weeks later, all the rats were anesthetized and broncholveolar lavage fluids were collected.①Alveolar washing fluid was concentrated 10 times and the levels of the soluble interleukin-2 receptor (sIL-2R) were examined by double antigen sandwich ELISA.②Blood was sampled from rat eyes and the count of lymphocytes in peripheral blood were detected.③The percentages of CD3+ and CD4+ subgroups were assessed with erythrocyte chaplet kit sensitized by antigen. 9.Effects of enteral nutrition and lactalbumin on muscle mass and function in the elderly

Objective To explore the effect of calories supplement or protein supplement on the skeletal muscle in the elderly.Methods The ambulatory elderly (≥65 yrs) with reduced handgrip strength and/or gait speed were enrolled,who did not suffer from any neurological or arthritic disease impairing mobility,or any severe pulmonary or heart disease,or chronic kidney failure.The subjects were recruited with their written informed consent from clinical practices in five hospitals in China.The subjects were randomized to control group(n=26),nutrition supplementation (Nutren 400 kcal/ d) group (n=27) or protein supplementation(Biopro Protein 20 g/d) group (n=21).All the subjects were given healthy lifestyle education.The physical performances i.e.gait speed and handgrip strength were measured at baseline and at 2 and 3 months of followed up.Lean soft tissue mass (muscle mass) was measured by BIA at baseline and at 3 months of follow-up.Results A total of 74 subjects completed the trial,including 26 in controls,27 in Nutren,and 21 in protein group.At the end of the trial,the changes from baseline in appendicular muscle mass were-490.0 g (95%CI:-801.9--245.5,P=0.001)in control group,90.0 g (95%CI:-346.8--442.0,P=0.241) in Nutren group,and 290.0 g (95%CI:-746.6-20.1,P=0.007) in protein group.There were no significant difference in above indices among the three groups (P=0.051),except between Nutren group and control group (P=0.020).The handgrip strength and gait speed in the three groups were improved obviously as compared with those at baseline,but not statistically significant between different groups.Conclusions Caloric supplementation group may have greater effect in maintaining muscle mass than the protein supplement group in the elderly.Both the caloric supplement and the protein supplement produce no additional effect on muscular function as compared with healthy lifestyle education.
10.Comparison of pathophysiological mechanisms in different subtypes of patients with functional dyspepsia

Objective To explore whether the pathophysiological mechanisms are same or not in different subtypes of functional dyspepsia (FD) patients. Methods Severity of symptoms and (44.2± 14. 6) years. Multi-gastrointestinal functions were also tested. Results No statistical difference was found among epigastric pain syndrome (EPS), postprandial distress syndrome (PDS) and mixed type in the data of gastric emptying function, gastric myoelectrical activity, autonomic nerve function,simultaneous introgastric pressure and water loading test, and psychological status assessment as well (P>0.05). There was a positive linear correlation between postprandial fullness and half gastric emptying time (correlation coefficient=11.5, P= 0. 043). Early satiation was negatively related to maximal water intaking volume (correlation coefficient=-6. 8, P=0. 033). Conclusions In this study, it suggested that the FD patients with different pathophysiologic mechanisms cannot be distinguished efficiently according to Rome Ⅲ criteria, and then be treated in a well-targeted manner.
