Objective To analyze the impact of tripterygium glycosides (TG) on the expression of CXCR3 and CCR4 gene in peripheral blood mononuclear cells from experimental autoimmune thyroiditis (EAT) rats, and to explore the possible mechanism for regulation of Th1/Th2 balance by TG. Methods EAT was induced in 20 rats, which were randomly divided into two groups, i.e., control group and TG group, to receive intragastric physiological saline and TG suspension (5.5 mg/kg) daily, respectively. The rats were killed 4weeks later, and total RNA was extracted from the peripheral blood mononuclear cells of rats and used for cRNA synthesis. The gene expressions of CXCR3 and CCR4 were detected with a Th1-Th2-Th3 microarray gene chip and realtime RT-PCR. Results Compared with the control group, there was a significant decrease in the mRNA expression of CXCR3 (0.52 ± 0.10 vs. 1.05 ± 0.17, P < 0.01 ) and CXCR3/CCR4 ratio (0.39 ± 0.22 vs. 1.04 ± 0.12, P< 0.01) in the TG group, together with an increase in the mRNA expression of CCR4 (1.56 ±0.13 vs. 1.02 ± 0.09, P < 0.01 ). Conclusions TG could regulate the number of Th1 and Th2 cells as well as their cytokine expression ratio likely by reducing CXCR3 and enhancing CCR4 expression, so as to modulate the Th1/Th2 balance in EAT rats.

Objective To investigate the improvement role of aspirin combined with statins on carotid plaque of patients with cerebral thrombosis.Methods80 cases of patients with cerebral thrombosis in our hospital from May 2014 to May 2016 were selected, these patients were divided into aspirin combined with statins treatment group (combined treatment group, 40 cases) and single aspirin treatment group (single treatment group, 40 cases) two groups according to the treatment methods, the NIHSS scores, Barthel indexes, clinical curative effects, IMT, plaque areas of the two groups were statistically analyzed.ResultsThe NIHSS score of the combined treatment group was significantly lower (P<0.05), the Barthel index was significantly higher (P<0.05), the total treatment efficiency 95.0% (38/40) was significantly higher than the single treatment group 80.0% (32/40) (P<0.05), the IMT was significantly shorter (P<0.05), the plaque area was significantly smaller than the single treatment group (P<0.05).ConclusionAspirin combined with statins can more effectively improve the carotid plaque of patients with cerebral thrombosis than aspirin alone, so is worthy of promotion and use in the clinical.

OBJECTIVE: To explore the effects of Liangxue Huayu therapy (LXHYT), a traditional Chinese herbal therapy for cooling blood and dissipating blood stasis, on rat model of experimental autoimmune thyroiditis (EAT) and its impact on tumor necrosis factor-alpha and interleukin-10 mRNA expressions in peripheral blood mononuclear cells. METHODS: Forty Wistar rats were randomly divided into five groups: normal control group, untreated EAT group, cyclosporine A (CyA)-treated group, Tripterygium glycosides-treated group and Liangxue Huayu Recipe (LXHYR)-treated group. The interventions were given by gavage to the rats in different groups once a day. All rats were sacrificed after 4-week treatment, and the level of serum thyroglobulin antibodies (TgAb) and the changes of histological grade of thyroid specimen were assessed by blind evaluation. Expressions of tumor necrosis factor-alpha (TNF-alpha) and interleukin-10 (IL-10) mRNAs in peripheral blood mononuclear cells were assessed by real-time reverse transcription polymerase chain reaction method. RESULTS: There was a significant increase in the serum TgAb level and severe inflammatory infiltration in the untreated group. Expression of TNF-alpha mRNA in peripheral blood mononuclear cells was increased, while the expression of IL-10 mRNA was decreased in the untreated group (P<0.01). Compared with the untreated group, CyA, Tripterygium glycosides and LXHYF could decrease the serum TgAb level (P<0.05), but the three interventions showed no significant improvement in thyroid inflammation (P>0.05). TNF-alpha mRNA expression was decreased, while IL-10 mRNA expression was increased as compared with the untreated group (P<0.05). CONCLUSION: LXHYT can decrease the serum TgAb level and recover the balance of Th1/Th2. This may provide an experimental basis for further research of assessing the antipsoriatic effect of Chinese herbal drugs with a rat model of EAT as an alternative model of psoriasis in vivo.

Objective To explore the expression and significance of Annexin Ⅱ protein in ovarian adenocar-einoma.Methods The expressions of Annexin Ⅱ protein were detected in 48 cases of ovarian adenocarcinoma and 10 cases of ovarian adenoma by SP immunohistochemistry.Results The positive rates of expression of Annexin Ⅱprotein in ovarian adenoma were 50.00% (5/10), and were 81.25% (39/48) in ovarian adenocareinoma (χ2 =4.414, P <0.05), in well and moderately-differentiated, poorly differentiated ovarian serous adenocarcinoma were 93.33% (14/15) and 78.95% (15/19) respectively (χ2 = 1.383 ,P0.05 ), and on the Ⅰ and Ⅱ stage, Ⅲ and Ⅳ stage of ovarian serous adenocarcinoma were 60.00% (6/10) and 95.83% ( 23/24 ) respectively (χ2=7.226, P <0.05).The positive rate in the group of ovarian serous adenocarcinoma with lymph node metastasis was 90.00% (27/30), while 50.00% (2/4) in the group without lymph node metastasis (χ2=4.502, P<0.05).Conclusion The positive expression of Annexin Ⅱ protein may be correlated with the carcinogenesis, development and lymph node metastasis of ovarian serous adenocarcinoma.

OBJECTIVE:To investigate the clinical efficacy and safety of aerosol inhalation of magnesium sulfate and salbuta-mol sulfate in the treatment of acute attack of severe bronchial asthma in children. METHODS:85 children with acute attack of se-vere bronchial asthma were divided into control group(45 cases)and intervention group(40 cases)in accordance to even and odd number of hospitalization time. Based on routine treatment of glucocorticoid intravenous injection,oxygen uptake,sedation and nu-tritional support,control group was given aerosol inhalation of salbutamol sulfate,0.6 ml each time,once every 20 min,for 1 h;intervention group was additionally given aerosol inhalation of magnesium sulfate,0.6 ml each time,once every 20 min,for 1 h, on the basis of control group. Therapeutic efficacy,improvement time of shortness of physical examination indexes (breath, wheeze,coughing),lung function indicators and ADR were observed in 2 groups,and discharge time was recorded in 2 groups. RESULTS:The total effective rate of clinical efficacy of intervention group was 95.00%after 1 h treatment,which was significant-ly higher than 77.78% in control group,with statistical significance (P<0.05);the time of shortness of breath improvement, wheeze improvement,coughing improvement and discharge in intervention group were significantly shorter than in control group, with statistical significance (P<0.05);PEF and FEV1 of intervention group were improved significantly compared to control group,with statistical significance(P<0.05). Both groups suffered from slight ADR,there was no statistical significance in the in-cidence of ADR between 2 groups (P>0.05). CONCLUSIONS:On the basis of conventional treatment,the aerosol inhalation of magnesium sulfate and salbutamol sulfate shows good clinical efficacy and safety in the treatment of acute attack of severe bronchi-al asthma in children.

Objective To evaluate the inhibitory effect of chidamide combined with curcumin on the proliferation of cutaneous T-cell lymphoma (CTCL) cell line Hut78,as well as their promotive effect on its apoptosis,and to explore their therapeutic mechanisms in CTCL.Methods Some Hut78 cells were treated with different concentrations of chidamide (0.3,0.6,1.2,2.4 μmol/L) and 10 μ mol/L curcumin alone or the combination of 1.2 μ mol/L chidamide and 10 μ mol/L curcumin for 24,48 and 72 hours separately.MTS assay was conducted to estimate cell viability at each time point.After selection of chidamide concentrations,some Hut78 cells were treated with chidamide (0.6 and 1.2 μmol/L)and curcumin (10 μmol/L) alone or in combination (1.2 μmol/L chidamide and 10 μmol/L curcumin) for 24 hours,then,flow cytometry was performed to detect cell apoptosis and analyze cell cycle,real-time (RT)-PCR and Western-blot analysis were conducted to quantify the mRNA and protein expressions of apoptosis-associated genes Fas,caspase 8,nuclear factor (NF)-κB p65 as well as cell cycle-associated genes P21,CDK2 and cyclin E respectively.Statistical analysis was carried out by repeated-measures analysis of variance,one-way analysis of variance and the least significant difference (LSD)-t test.Results Chidamide could significantly inhibit the proliferation of Hut78 cells in a dosedependent and time-dependent manner (F =266.558,564.966,respectively,both P ＜ 0.001).After 48-and 72-hour culture,the combination of 1.2 μmol/L chidamide and 10 μmol/L curcumin showed significantly stronger inhibitory effect on cell proliferation compared with 1.2 μmol/L chidamide or 10 μmol/L curcumin alone (all P ＜ 0.001).As flow cytometry showed,the percentage of apoptotic cells was significantly higher in the combined treatment group than in the 0.6-,1.2-μmol/L chidamide groups and 10-μmol/L curcumin group (all P ＜ 0.001).Compared with the 0.6-μmol/L chidamide group and 10-μmol/L curcumin group,the combined treatment group and 1.2-μmol/L ehidamide group both showed significantly increased proportion of cells at G0/G1 phase and mRNA expressions of Fas,caspase 8 and P21,but decreased proportion of cells at S phase or G2/M phase and mRNA expressions of NF-κB p65,CDK2 and cyclin E (P ＜0.05 for proportion of cells at different phases,P ＜ 0.001 for mRNA expressions of different genes).Furthermore,the mRNA expression of Fas was significantly higher in the combined treatment group than in the 1.2-μmol/L chidamide group (P ＜ 0.001),while no significant differences were observed in the mRNA expressions of caspase 8,P21,NF-κB p65,CDK2 and cyclin E or the proportion of cells at any phase between the combined treatment group and 1.2-μmol/L chidamide group (all P ＞ 0.05).Western-blot analysis showed that protein expressions of Fas,caspase 8 and P21 significantly increased,but those of NF-κB p65,CDK2 and cyclin E significantly decreased in the combined treatment group compared with the 0.6-,1.2-μmol/L chidamide groups and blank control group receiving no treatment,which were in accordance with the above changes in mRNA expressions of these genes.Conclusion Chidamide can inhibit the growth of the CTCL cell line Hut78 by directly decelerating cell proliferation and inducing cell apoptosis,and the combibation with curcumin can markedly enhance the inhibitory effect of chidamide on the growth of Hut78 cells.

Objective To evaluate the clinical and imaging characteristics of osteoid osteoma in femoral neck and to improve diagnostic accuracy of this disease.Methods Twenty-one patients (18 males and 3 females,age,7-26 years,median age,13 years) with pathologically proven osteoid osteoma of the femoral neck were retrospectively analyzed for their clinical profile and radiologic features.CT and X-ray examinations were performed in all patients,10 of them pefformed post-contrast CT scan and 4 of them performed MRI examinations.Results Nineteen patients had hip pain (pain worse at night in 11,and 8 received salicylates treatment with good response),and 2 patients only with intermittent claudication.The duration ranged from 2 months to 54 months (median duration 12 months).X-ray: Nidus was seen on plain film in 10 cases,18 cases showed different degrees of bone sclerosis of the nidus.CT: Nidus was demonstrated in all cases.Among them,8 were intracortical,6 were subperiosteal,7 were endosteal.Twenty cases showed different degrees of bone sclerosis of the nidus-extra-articular anteromedial cortical surface of the femur neck.Nineteen cases showed vascular groove sign.MRI: Nidus was seen in 4 cases.Bone sclerosis was low signal on all sequences.Three cases had joint effusion,4 cases had bone marrow edema,and 2 cases had synovial thickening.Conclusions Although osteoid osteoma of femoral neck has non-specific clinical features,the radiographic findings are usually typical.The nidus of osteoid osteoma is often located within the joint.Bony sclerosis occurs at the area of extra-articular anteromedial cortical surface of the femur neck.CT examination remains an optimal method to identify the nidus.

Objective:To probe the different influence of pericardial devascularization by preserving vagus trunk(VTPPD) and pericardial devascularization (PD) on portal hypertensive gastropathy (PHG).Methods:77 patients with portal hypertension were divided into VTPPD and PD group,the VTPPD group included 36 cases,and PD group included 41 cases.Varices of esophagus and fundus of stomach and PHG were observed by gastroscopy before and 3 weeks after operation in all cases,and compared postoperative incidence of PHG in the 2 groups.Results:In all cases,Varices of esophagus and fundus of stomach disappeared or relieved obviously.The incidence of PHG in VTPPD group before operation was 55.6%(20/36),and that after operation was 69.4%(25/36),the former was not higher statistically(P=0.224);the incidence of PHG in PD group before operation was 61.0%(25/41),and that after operation was 87.8%(36/41),the former was not higher than the latter statistically(P=0.005);and the postoperative incidence of PHG in PD group was higher significantly than that in VTPPD group (P=0.048).There were 8(22.2%,8/36)patients whose degree of PHG aggravated in VTPPD group,and there were 19(46.3%,19/41)patients whose degree of PHG aggravated in PD group,the rate of the former was significantly lower than that of the latter(P=0.027).Conclusion:Comparing with the classic portoazygous devascularization,VTPPD can reduce the incidence and the degree of PHG.
