1.Differentiation and regulation of the osteoclast

The osteoclast is a hematopoietic cell derived from CFU GM, which is regulated by systemic hormones, soluble factors in the phase of differentiation, fusion and proliferation. The major singnaling pathways involved in osteoclastic bone resorption and formation in response to cytokine and hormones include cAMP and R PTK. The advances in our understanding of the osteoclast make it possible for us to comprehend the development and the treatment of the bone metabolic diseases.
2.Co-stimulatory molecule CTLA-4 and the potential application in treating rheumatic diseases

Rheumatic diseases are characterized by immunological dysfunction.T-cell medicated immune response plays a very important role in the development and persistence of rheumatic disease.CTLA-4 is an important co-stimulatory molecules expressed on T cells.It has inhibitory effect on T cell-mediated immune response.Plenty of evidences have demonstrated abnormalities in CTLA-4 alleles and proteins in patients with rheumatic diseases.Therefore,CTLA-4 has become an target in treating rheumatic diseases.CTLA-4Ig has been developed as an fusion protein which is composed of extrA-cellular domain of human CTLA-4 and the Fc fragment of human IgG1.So far,only data about CTLA-4Ig in treating rheumatoid arthritis are available.These data showed that CTLA-4Ig could relieve the symptoms of rheumatoid arthritis and retard the development of the disease.It is safe and effective.In addition,effectiveness was demonstrated even in patients who failed to response to anti-TNF monoclonal antibodies.Therefore,CTLA-4Ig is an encouraging therapy for rheumatic diseases.However,because of very short history,long-term study is needed to fully understand its role in the treatment of rheumatoid arthritis.
3.Clinical study on the concept of rapid rehabilitation surgery in perioperative period of colorectal cancer

Objective To explore the application of rapid rehabilitation concept in perioperative period of colorectal cancer.Methods A total of 110 patients with rectal cancer admitted in our hospital from June 2012 to July 2015 were enrolled in the research group(55 cases)and control group(55 cases).The observation group was treated with rapid rehabilitation method,and the control group was treated by traditional perioperative method.Observation of the two groups of patients was in the hands of the amount of bleeding,operation time,the first time the patient exhaust time,postoperative time to get out of bed and other indicators.We observed the two groups of patients with surgical incision infection,intestinal obstruction,pulmonary infection and other complications.Results The difference of the amount of bleeding and the length of the incision in the two groups was statistically significant(t=9.618,7.846,P<0.01).The first time of exhaustive time,the first defecation time,the time of eating,the time of getting out of bed and the postoperative hospital stay of research group were significantly lower than those of the control group(P<0.01).The incidence of complications such as incision infection,intestinal obstruction and pulmonary infection of the research group was lower than those in the control group,and the difference was statistically significant(χ2=4.767,P<0.05).Conclusion The application of rapid recovery concept in perioperative period of colorectal cancer can reduce the incidence of surgical complications.
4.Clinical analysis of autoimmune hemolytic anemia in systemic lupus erythematosus

Objective To determine the clinical and serological characteristics of autoimmune hemolytic anemia (AIHA) in systemic lupus erythematosus (SLE) patients.Methods Seventeen patients were studied who had clinical features of AIHA among 222 hospitalized SLE patients in the Department of Rheumatology of Beijing Chaoyang Hospital were studied.Among the SLE patients without AIHA,34 patients were chosen by random sampling as controls.The patients' demographic data,clinical manifestations,laboratory test results and other examinations were analyzed retrospectively.Student's t test was used for the comparison of continuous variables,and chi-square test was used for the comparison of proportions.Results The prevalence of AIHA was 7.6% in cohort of lupus patients.The positive rate of IgG anticardiolipin antibody in the AIHA group was significantly higher than that in the control group (71% vs 15%,x2=15.366,P<0.01 ).The occurrence of antiphospholipid antibody syndrome between the two groups had no statistical significant difference (x2=0.000,P=1.000).The frequencies of arthritis (12%,x2=4.554,P=0.033),malar rash (6%,x2=4.443,P=0.033),leukocytopenia (12%,x2=8.061,P=0.005) and lymphocytopenia (41%,x2=5.075,P=0.024) were lower in the AIHA group than those in the control group.Three patients in the AIHA group had alveolar hemorrhage in the course of SLE,while there was none in the control group (x2=3.586,P=0.058).Conclusion When lupus patients with AIHA are compared with those without AIHA,the positive rate of anticardiolipin antibody is higher,but the occurrence of antiphospholipid antibody syndrome does not increase in SLE patients with AIHA.And the frequencies of arthritis,malar rash,leukocytopenia and lymphocytopenia are lower in those patients with AIHA,however,patients with AIHA are prone to develop alveolar hemorrhage.
5.Progress in mechanisms of methamphetamine-induced dopaminergic neurotoxicity

Methamphetamine is a powerful stimulant of amphetamine type in the central nervous system (CNS),and recently it has become the major drugs of abuse.A lot of research results show that MA may induce dopaminergic neurotoxicity in the animal and human striatum.The mechanisms include effects on dopaminergic signaling and dopamine oxidation,glutamate induced excitotoxicity,oxidative stress and inflammatory cytokines,disruption of mitochondfia,apoptosis,activation of glial cells and hyperthermia.However the mechanisms by the MA-induced dopaminergic neurotoxicity are not completely understood.The paper reviewed recent progress of study on them to provide reference materials for related research.
6.Effect of ST2325 on cell cycle in erbB2-overexpressing breast cancer MDA-MB-453m1 cells and its mechanism

Purpose:Effect of ST2325 on cell cycle in erbB2-overexpressing MDA-MB-453m1 cells and its mechanism was investigated.Methods:Cell cycle distribution was examined using flow cytometry. The protein expression was detected with Western blot analysis.Results:Under concentrations of 0,6.25,12.5,25.0,50.0,100 μmol/L,the percentrages of G0/G1 in MDA-MB-453m1 were 48.62%, 48.83%, 49.59%, 52.18%, 61.0%,83.96%; the percentages of S were 33.34%, 33.16%, 32.78%,30.77%,25.62%,7.08%;the percentages of G2M were 18.04%, 18.01%,17.63%,17.05%,13.38%,8.96%.After MDA-MB-453m1 cells were treated with different concentrations of ST2325 for 24 hours, Western blot assay showed up-regulation of p27 protein expression and decrease of hyperphosphorylated Rb and cyclin D1 protein expression .Activation of MAPK and AKT in MDA-MB-453m1 was markedly inhibited by ST2325.Conclusions:ST2325 induces G1 arrest in erbB2-overexpressing MDA-MB-453m1 cancer cells. G1 arrest is associated with p27 upregulation,decrease of cyclinD1 protein and hyperphosphorylated Rb.
7.Antisense oligonucleotides targeting TNF-? suppress on Co-Cr-Mo particle-induced osteolysis

[Objective]To investigate the effect of a single subcutaneous dose of an antisense oligonucleotide(ASO) on particle-induced osteolysis.[Method]The murine calvaria osteolysis model was utilized in ICR mice.Bone resorption was measured with the toluidine blue staining.Osteoclasts were detected by tartrate resistant acid phosphatase(TRAP) staining assay and were quantified by a TRAP quantification kit.[Result]Bone resorption was 0.347 ? 0.09 mm2 in animals with particle implantation,and decreased to 0.123 ? 0.05 mm2 and 0.052 ? 0.02 mm2 after ASO treatment in low and high doses,respectively.The bone resorption was reestablished in animals given an additional TNF-?.The number of osteoclasts in animal calvaria treated with ASO was reduced obviously compared with those untreated animals and the quantification results indicated that about 90% osteoclastgenesis was suprressed by the ASO.Additionally,the osteoclastgenesis was reestablished by the addition of TNF-?.[Conclusion]An antisense oligonucleotide targeting an inflammatory factor,TNF-?,has been to suppress the osteolysis induced by particle for the first time.This new finding holds a great promise.It is a therapeutic strategy for the component loosening.
8.Research progress of methamphetamine inducing nerve cell′s autophagy

Methamphetamine ( METH ) , a powerful stimulant of amphetamine type in the central nervous system ( CNS ) , is a new kind of synthetic drugs mainly in psychological dependence. Long-time abuse will lead to strong neurotoxicity and drug de-pendence. Recently it has become the abuse problem in public health. Autophagy is a process of degradation and utilization of the cellular components by the lysosome, which widely exists in eukaryotic cells. Many studies show that methamphetamine can iduce autophagy,but the underlying mechanism is not fully un-derstood yet . Therefore,the mechanisms and signaling pathways of autophagy by methamphetamine and the roles of autophagy in methamphetamine induce neurotoxicity are reviewed in this pa-per, so as to provide reference materials for further research of methamphetamine and autophagy.
9.The diagnostic value of anti-cyclic citrullinated peptide antibody in patients with rheumatoid arthritis

Objective To explore the diagnostic value of anti cyclic citrullinated peptide (CCP) antibody (anti CCP) detected by ELISA in patients with rheumatoid arthritis (RA).Methods The synthesized CCP deduced from the known cDNA sequence was used as substrate in ELISA.The anti CCP was detected by ELISA in 294 RA,132 rheumatic diseases other than RA and 135 non rheumatic diseases.To compare the correlation among the anti CCP and APF (antiperinuclear factor),AKA (anti keratin antibody),RF (rheumatoid factor) and HLA DR4.Results One hundred and thirty seven (46 6%) RA sera,7 (5 3%) other rheumatic disease sera and 2 (1 8%) non rheumatic disease were found to be anti CCP positive by ELISA.The sensitivity of anti CCP was 46 6% with high specificity (96 9%) in RA.Anti CCP was associated with APF,AKA and HLA DR4 except RF.Conclusions Anti CCP presents a reasonable sensitivity (46 6%) with high specificity (96 6%) to RA and it is valuable to RA diagnosis.Anti CCP is associated with APF,AKA and HLA DR4 but not completely overlaps them.Anti CCP could be regarded as a new diagnostic marker in RA.
10.The extraction of filaggrin and its significance in rheumatoid arthritis

Objective To establish the method for extracting the cytokeratin filament aggregating protein (filaggrin) from human epidermis,to detect the anti filaggrin antibody (AFA) in rheumatoid arthritis (RA) and to determine the diagnostic value of AFA in RA.Methods Filaggrin was extracted from human epidermis and partially purified.AFA in 103 patients with RA and 140 controls was detected by Western blotting.Results Filaggrin extracted from the epidermis could be used as the antigen in AFA test by Western blotting.AFA test resulted in diagnosis of 35 9% of 103 RA samples,with a specificity of 93 7%.The result was significantly higher compared with patients′ and normal controls ( P
