1.The influence of Intra-abdominal pressure of carbon dioxide on pregnancy outcome and malformation ratio in pregnant rats

Objective To study the influence of carbon dioxide pneumoperitoneum on the pregnancy outcome and newborn rat malformation in pregnancy rats. Methods SD rats were randomly divided into control group (group A and group B) and the experimental group (group C; group D; group E). There were 10 rats in each group, the general situation,abortion rate, premature delivery rate,newborn rats numbers,weights and lengths and malformation rates were recorded. Results There was no malformation in newborn rats in all groups (P > 0. 05 ). Conclusion It was safe to undergo laparoscope operation in metaphase pregnancy with low CO2 pneumoperitoneum,which has no influence on abortion rate and malformation rate.
2.Studies of the Effects of Hydrogen Peroxide on the Ultrastructure and Permeability of Candida Albicans

The Effects of hydrogen peroxide(H2o2) on the ultrastructure, cellular pea-meability barrier and 3H-TdR-DNA leakage of C. albicans were studied. Damage of the cell, especially leakage of cytoplasm were found under transmission electron microscope. Morphological changes and small hole like structures were also found under scanning electron microscope. Radioactivity assay of the leakage of 3H-TdR-DNA leakage of C. albicans further suggested that the 3H-TdR-DNA leakage of C. albicans was directly due to the damage of cellular permeability barrier. Negative correlation (r=+0.8392--0.9915)between the extent of 3H-TdR-DNA leakage and the number of surviving colony-forming units was found. This indicated that the damage of cellular permeability barrier and leakage of DNA of C. albicans caused by H2o2 was one of the most importent causes of cell killing.

4.Detection of human mammaglobin in diagnosis and treatment of breast cancer

Human mammaglobin (hMAM) is a recently discovered breast tissue specific protein with a high sensitivity and specificity. The expression of hMAM in breast cancer and its relationship with breast cancer indicate that hMAM is a novel breast specific marker that can induce cytotoxic actions from immune system and therefore may have great potentials in breast cancer treatment.
5.Engineered Protein Nanopore for Real-time Monitoring Single-molecule Reaction between Cadmium Ion and Glutathione

The chelating reaction between glutathione peptides and divalent cadmium ion was used as a typical model for investigating the coordination chemistry of SH-containing peptides and heavy metal ions, which was essential to understand the mechanism of intracellular cadmium detoxification.Here, the mutant (CM113R)7 αHL protein nanopore equipped with a first synthesized per-6-quaternary ammonium-β-cyclodextrin (p-QABCD) was used as nanoreactor and detector to investigate the single-molecule reaction of GSH molecules and Cd2+ ions.Different reaction pathways, intermediates, and products could be recognized.Cd2+-GSH reaction was highly dependent on the solution pH.The Cd(GSH)2 was formed at pH 7.4, while the Cd(GSH)2 and Cd2(GSH)2 were formed at pH 9.0.Cd2(GSH)2 was formed by two possible pathways: (1) A Cd2+ ion primarily coordinated with the thiol group of two GSH molecules to form Cd(GSH)2, and then the second Cd2+ ion quickly incorporated with the deprotonated amino group of Cd(GSH)2 to form Cd2(GSH)2;(2) Two Cd2+ ions separately coordinated with the thiol and deprotonated amino group of one GSH molecule to form Cd2(GSH)1, and the second GSH molecule quickly bounded to Cd2+ ions to form Cd2(GSH)2.This work studied the chelating reaction between metal ions and bioactive glutathione at single-molecule level without labeling and chemical modification, and would be important to further understand intracellular mechanisms of detoxification of heavy metals and greatly expand the research field of nanopore single-molecule technique.
6.The application of ASL in assessing cerebral blood flow in healthy volunteers

Objective To evaluate the correlation between cerebral blood flow(CBF)with age and gender with arterial spin labe-ling in magnetic resonance imaging.Methods All thirty-seven healthy volunteers were divided into A(20-29 years,n=10),B(30-39 years,n=9),C(40-49 years,n=9)and D(50-59 years,n=9),underwent conventional MRI and ASL(Q2TIPS)examination. Cerebral blood flow (CBF)maps were obtained and reviewed.ROIs were respectively selected in bilateral cerebral hemispheres and CBF values were generated.The comparison of CBF value between the left and right hemispheres and within the four age group differences were performed.The relationships between CBF values with age and gender were also analyzed.Results There was no statistical difference of CBF in white or gray matter between bilateral hemispheres in all groups (P > 0.05).A significant positive correlation was found between age and CBF values of the left whole brain (r=0.378,P =0.020)in group C,while no significant difference of CBF in bilateral gray matter,white matter or whole brain was detected among all age groups (P > 0.05).There was no statistical difference of CBF between male and female volunteers (P > 0.05).Conclusion Quantitative estimates of CBF can be obtained with ASL sequence.The preliminary results suggest that CBF values do not significantly associated with age or gender.

8.Controlled clinical studies on drug-induced liver injury in the elderly Chinese patients: a literature review

Objective To review the clinical features of drug-induced liver injury (DILI) in elderly Chinese patients.Methods A literature search was conducted with the key wordsdrug-induced liver injury ordrug-induced liver disease andelderly; four Chinese medical databases were used:including Chinese Biomedical Literature Database (CBM),Chinese Science and Technology Periodical Database (VIP),Wanfang Database and China National Knowledge Infrastructure (CNKI),from when the bases established to December 2011.Data from retrieved literatures of retrospective controlled studies of DILI were analyzed by SPSS 14.0 software.Results Five studies met the inclusion criteria,including 275 elderly patients (aged group) and 602 non-elderly patients (non-aged group) with DILL There was no significant difference in gender between the two groups (P > 0.05).Clinical manifestations of jaundice,fatigue,anorexia,and skin itching were more commonly seen in the aged group (P < 0.01).Cholestatic hepatic injury occurred more frequently in the aged group than in the non-aged group [24.9% (61/245) vs.18.8% (102/245),P < 0.05],while hepatocellular type occurred more frequently in the non-aged group [64.3% (350/544) vs.55.1% (135/245),P < 0.05].More DILI were induced by Chinese herb medicine and cardiovascular drugs in the aged group than in the non-aged group [47.0% (79/168) vs.35.9% (93/254) and 8.3% (14/168) vs 1.2% (3/259) respectively,P < 0.05].As for the prognosis,there was no significant difference in clinical cure and improvement rate between the two groups [84.4% (141/167) vs.81.5% (291/357),P >0.05].Conclusion The elderly are more sensitive to the drugs and have a high risk of DILI.Prevention and timely diagnosis and treatment of DILI are particularly important for elderly patients.

Antineutrophil cytoplasmic antibody-associated vasculitis (AAV) refers to the groups of potentially life-threatening autoimmune disease.Antithyroid drug is one of the causes.Usually the involved organs are skin,kidney,and (or) lung.Early diagnosis and treatment of AAV is essential.Timely cessation of antithyroid drugs is the first step.If necessary,glucocorticoids and (or) immunosuppressive agents should be used to delay the progression of the disease.
10.Effects of hippocampal neurogenesis in spatial learning and memory in rats

Objective To investigate the effects of hippocampal neurogenesis in spatial learning and memory,especially in spatial long-term memory.Methods Totally 24 wistar rats were randomly divided into two groups:normal control group (NC group,n =12) and irradiation group (IRR group,n =12).We use low-dose irradiation at subgranular zone to inhibit adult hippocampal neurogenesis and new neurons were investigated by 5-Bromo-2-deoxyUridine/neuron specific nuclear protein double-labeling.Four weeks after irradiation,Morris water maze,including navigation test and space exploration assay,was used to detect spatial leaming and memory.Then,in the day 8 and day 15 after the navigation test,space exploration assay was repeated to detect spatial long-term memory.The expression levels of postsynaptic density protein 95 and synaptophysin were evaluated using western blot and real-time quantitative PCR.Results Hippocampal neurogenesis was inhibited by low dose irradiation(2.80 ± 0.44 vs 23.50 ± 1.12,t =20.21,P < 0.01).After the inhibition,the escape latency did not change,while the time spent in target quadrant was significantly declined in the day 2((14.76 ±.2.04) s vs (20.47 ± 1.29) s),day 8((11.95 ± 1.34) s vs (19.52±1.43) s)and day 15((11.79 ±1.35) s vs (21.58 ±1.07) s) after the navigation test (t=2.45,P<0.05; t =3.76,P<0.01; t =5.39,P<0.01).The postsynaptic density protein 95 and synaptophysin expressions were significantly decreased in IRR group.As to the RNA levels,there was also a significant difference between the two groups.Conclusion Hippocampal neurogenesis in the dentate gyrus plays an important role for the formation of spatial long-term memory.
