2.MRI study of the non-thyroid-related exophthalmos

ObjectiveTo investigate the manifestation and diagnostic value of MRI in the patients with non-thyroid-related exophthalmos.Methods One hundred and sixty-eight patients with non-thyroidrelated exophthalmos confirmed by histopathology underwent MRI examination using 8-channel phased array head coil.Of 168 patients,165 underwent contrast-enhanced MRI.Results Of the 168 cases,68 were hemangioma,34 were lymphoma,14 were pleomorphic adenoma of the lacrimal gland,12 were inflammatory pseudotumor,6 were adenoid cystic carcinoma of the lacrimal gland,6 were schwannoma,5 were dermoid cyst,5 were meningioma,3 were solitary fibrous tumor,2 were optic nerve glioma,2 were embryonal rhabdomyosarcoma,2 were lipoma,et al.Of the 165 enhanced cases,the MRI diagnosis was in accordance with surgical pathology findings in 111 cases (67.3% ).The tumor was hemangioma in 65,lymphoma in 22,pleomorphic adenoma of the lacrimal gland in 10,inflammatory pseudotumor in 5,meningioma in 3,adenoid cystic carcinoma of the lacrimal gland in 3,optic nerve glioma in 1,schwannoma in 1,and dermoid cyst in 1.Eleven cases simultaneously had dynamic contrast-enhanced (DCE) MRI,the MRI diagnosis was consistent with pathological findings in 10 cases,which included 7 hemangioma,2 lymphoma,and 1 meningioma.The time-intensity curves of DCE-MRI showed persistent enhancement type,rapidly enhancing and rapid wash-out and plateau-shaped type,and rapidly enhancing and rapid wash-out,respectively.Three cases showed soft tissue mass on only plain MRI.ConclusionsThe conventional orbital MRI can explore the causes of proptosis,which is helpful to determine the location and nature of the mass.Enhanced and DCE MRI can further observe blood supply of the lesions.
3.Topical drugs for the treatment of psoriasis

In recent years, great progress has been made in the pathogenesis of psoriasis, which has provided multiple new targets for the treatment of psoriasis, and ushered the treatment, especially topical treatment, of psoriasis to a new era. Some new targeted topical drugs with action mechanisms different from those of traditional drugs have been applied in clinical practice, which are mainly small -molecule inhibitors targeting cell signaling pathways or enzymes involved in the pathogenesis of psoriasis, such as the Janus kinase/signal transducer and activator of transcription(JAK/STAT) pathway, phosphodiesterase type 4 (PDE4), mitogen-activated/extracellular-signal-regulated kinase kinase (MEK), vascular endothelial growth factor receptor (VEGFR), tropomyosin receptor kinase A (TrKA), retinoic acid receptor-related orphan receptor γ (RORγ), and so on. At present, many phaseⅠ and Ⅱ clinical trials are conducted for these drugs. Some completed trials have shown that they have good effectiveness, safety and tolerance, providing more options for the topical treatment of psoriasis.
4.Bridging effect of nuclear factor kappa B on upregulation of vascular endothelial growth factor expression in hypoxia-inducible factor-1 alpha gene-modified neural stem cells

BACKGROUND: Numerous studies have shown that nuclear factor-KB (NF-kB) was positively correlated with vascular endothelial growth factor (VEGF) in the cerebral hypoxia region. Thus, we assumed whether NF-kB in the pathway of hypoxia-inducible factor-1 α (HIF-1α) upregulating VEGF and plays a bridge effect.OBJECTIVE: To study the effects of NF-kB in HIF-1α pathway of increasing expression of VEGF using HIF-1α-modified neural stem cells (NSCs) as vectors.DESIGN, TIME AND SETTING: In vitro observation of cytology was conducted at the Neuroscience Institute, Jiamusi University from March to December 2008.MATERIALS: Wistar rats aged less than 24 hours of both genders were used.METHODS: NSCs were transfected after amplification of adenovirus vector HIF-1α (AdHIF-ia)-green fluorescent protein (GFP).Fluorescence detection was used to determine HIF-1α-GFP and blank vector Ad-GFP expression in NSCs. Protein was extracted from transfected NSCs, blank vector NSCs and normal NSCs, separately. Subsequently, NF-kB specific inhibitor pyrrolidine dithiocarbamate (PDTC) (50,150, 300 μmol/L) was added in HIF-1α-modified NSCs. Western blot analysis was used to determine changes in VEGF expression.MAIN OUTCOME MEASURES: The following parameters were measured: expression of HIF-1α, VEGF and NF-kB in NSCs in each group; VEGF expression in NSCs following treatment of NF-kB specific inhibitor.RESULTS: Gene expression was associated with MOI and transfected time following AdHIF-1α-GFP transfected with NSCs. After transfected AdHIF-1α-GFP in NSCs, HIF-1α, VEGF and NF-kB expression was positively correlated. Expression of VEGF was reduced in AdHIF-1α-GFP-modified NSCs following treatment of NF-kB inhibitor PDTC in a concentration-dependent fashion.There were significant differences in VEGF expression between each concentration group (P < 0.05-0.01).CONCLUSION: NF-kB in signaling pathway of HIF-1α-upregulated VEGF expression and played a bridging effect.
5.Association of plasma cystatin C and intima-media thickness of carotid artery in patients with early type 2 diabetes mellitus

Ninty-eight patients with newly diagnosed type 2 diabetes mellitus were divided into atherosclerosis(AS) group and non-AS group.Fasting plasma cystatin C level( CysC ) was determined.The results showed that there existed a significant correlation between CysC level and the number of carotid arteries plaque ( r =0.432,P<0.01 ).CysC was an independent risk factor( OR =2.21,95% CI 1.88-3.02 ) of carotid artery intimamedia thickening in patients with type 2 diabetes.
6.Expressions of Notch1 and Cyclin D1 in mammary ductal hyperplasia and ductal carcinoma

Objective To investigate the relationship between mammary ductal hyperplasia and ductal carcinoma by detecting the expressions of Notch1 and Cyclin D1 in various histotypic mammary ductal diseases.Methods A total cases of 44 normal ductal tissues,44 usual ductal hyperplasias.40 atypical ductal hyperplasias,42 ductal carcinomas in situ and 51 invasive ductal carcinomas were included in this study.Expressions of Notch1 and Cyclin D1 were detected by S-P immunohistochemistry.Results There was significant difference of expression of Notch1 among the five subgroups of this study(P<0.01);there were significant differences between every two subgroups(P<0.05)except for normal ductal tissues versus usual ductal hyperplasias and ductal carcinomas in situ versus invasive ductal carcinomas(P>0.05).There was also significant difference of expression of CychnD1 amongthe five subgroups of this study(P<0.01),and there were significant difference between every two subgroups(P<0.05)except for normal ductal tissues versus usual ductal hyperplasias and normal ductal tissues versus atypical ductal hyperplasia(P>0.05).The correlation between Notch1 and Cyclin D1 was negative (r=-0.428,P<0.01).Conclusions There is a relationship between mammary ductal hyperplasia and ductal carcinoma.Decrease of Nowhl or increase of Cyclin D1 expressions may promote nmnmmry ductal hyperplasia advancing to manunary ductal carcinoma.
7.The Diagnosis and Treatment of Colon Cancer Presenting Appendicitis

[Objective]To investigate the reasons of misdiagnosis of colon cancer complicated with appendicitis and to sum up the experience of diagnosis. [Methods] Between May 1995 and September 2005, 1060 cases with appendicitis were undergone appendectomy. However, of them, 9 cases complicated with colon cancer had confirmative diagnosis during the operation or after, performed successfully suitable operations. [Results] All of 9 cases were misdiagnosed as appendicitis and undertook appendectomy for similar clinical traits. According to the signs of exploration in the appendectomy and the analysis of patient history, clinic characteristic and some radiologic features, we acquired confirmative signs of carcinoma and put up reasonable surgical intervention. [Conclusion] There are some imperative measures to decrease misdiagnosis by analyzing clinical manifestations comprehensively, observation dynamically, exploration in the appendectomy, and effective examination. Among them, the key to reduce misdiagnosis rate is exploration during the period of appendectomy.
9.Study on Quality Standard for Compound Kangganling Granules

Objective To establish the quality standard of Compound Kangganling Granules. Methods TLC method was used for qualitative identification of Lonicerae Japonicae Flos and Forsythiae Fructus, and HPLC method was used for determination of chlorogenic acid in the preparation. The HPLC separation was performed on Diamonsil C18 column. The mobile phase was consisted of acetonitrile-0.4%phosphoric acid (20∶80, V/V), and the flow rate was 1.0 mL/min. Results TLC identified chlorogenic acid in Lonicerae Japonicae Flos and phillyrin in Forsythiae Fructus. The linear relationship of chlorogenic acid in the preparation measured by HPLC was A=30461C-5938.8, r=0.9998, RSD=1.01%, showing that the linear range of chlorogenic acid was 10.2-102.0μg/mL. Conclusion The method is accurate and rapid, with good stability, reliability and reproducibility, and can be used for the quality control and evaluation of the preparation.
10.Effect of vitamin E on detrusor protection in BOO rabbits

Objective To explore the protective effect of vitamin E on the rabbit bladder after partial outlet obstruction artificially setup. Methods A total of 28 New Zealand white male rabbits were divided into 4 groups (group A in 6,group B in 6,group C in 8 and group D in 8).Group A,B and C were fed a regular diet,and group D were placed on a diet enriched with 600 mg vitamin E.After 4 weeks partial outlet obstruction was created in groups C and D,while group B underwent sham operation. After 4 weeks of obstruction each rabbit was sedated and cystometry was repeated.After cystometry the bladder was weighed.The gene expression of sarcoplasmic endoplasmic reticulum,calcium,magnesium,adenosine triphosphatase (SERCA2) in bladder was detected by using RT-PCR assay,while the protein level of SERCA2 was measured by Western blot analysis. Results All parameters measured were approximately identical in nomal rabbits(Group A) and shum operation rabbits(group B).Thus,these 2 groups were combined as the control group(Group A and B).Partial outlet obstruction resulted in bladder weight increased significantly in obstructed groups given vehicle group C(13.07?1.71)g and those vitamin E group D(11.80?2.01)g,4-fold higher than in the control group (2.81?0.30)g(P
