1.Determination of Icariin in Chuanlong Guci Pian by HPLC

Objective To establish the method for detemining the content of Icariin in Chuanlong Guci Pian by HPLC. Methods Diamonsil ODS1 C18 Column was used with acetonitrile-water (27∶73) as the mobile phase, detection wavelength as 270 nm, and flow rate was 1.0 mL/min. Results The calibration curve was linear at the range of 0.125~1.00 ?g for Icariin and linear equation was Y=81 601X+10 356, r 2=0.999 7. The average recovery was 99.12% and RSD was 0.65% (n=6). Conclusion This method was simple, accurate and proper, and the reduplication of the result was good, which provides scientific quantitative analysis of Icariin in Chuanlong Guci Pian.
2.Summary of the nation-wide autoantibodies test quality survey in 2006

objeetive To consecutively understand the current national clinical testing quality and enforce quality-control of auto-antibody detection.Methods Hospitals or departments were recruited by letters or telephone communications:The autoantibodies examined for quality control survey included anti-nuclear antibodies (ANA),anti-double-stranded DNA (A-dsDNA)antibody,anti-extractable nuclear antigens(A-ENA)antobodies,anti-mitochondria antibody(AMA)/anti-smooth muscle antibody(ASMA),and anti-CCP antibody.Each autoantibody was tested in 3 samples, and altogether 15 samples in total for testing.Sample designation and testing results data analysis were double-blinded.Results Fifltv-five hospitals/departments participated in this survey.The accuracy rates for this survey were 92%,89%,96%,72%respectively for ANA,A-dsDNA,AMA/ASMA,and anti-CCP.Anti-ENAs were further divided into anti-RNP,Sm,SSA,SSB and Scl-70 subgroups,and the accuracy rates were 98%,89%,92%,75%and 77% respectively.Conelusion Compared to the previous 3 national surveys.accuracy rates in our country's autoantibody testing is increaseing steadly with more testing items included each year.This indicats that the quality of auto-antibody testing is improving across the country.
3.Modulation of CXCR4,and MCF-7 cells invasion potential in vitro by norcantharidin

Purpose To investigate the effect and mechanism of norcantharidin(NCTD)on invasion of human breast cancer cell line MCF-7 in vitro.Methods MTT assay Was used to determine MCF-7 cell proliferation,Transwell kit was used to determine cell migration,treated with NCTD on 200,400,600 μmol/L,to detect the expression of CXCR4 and CXCL12 in the control and treatment groups by real time PCR and Western blot.Results NCTD had inhibitive effects on proliferation and invasion of human breast cancer cell line MCF-7 in a dose-dependent manner,with the IC_(50) value of 582 μmol/L at 24 h,and the expression of CXCR4 Was deCreased in NCTD groups in vitro.Conclusion NCTD in vitro inhibits invasion and metastasis of human beast cancer cell line MCF-7 throush the down regulation to the expression of CXCR4.
4.Influence of thickness of cancellous bone and cortical bone in stress distribution in vicinity of an implant

Objective:To investigate the stress distribution surrounding an implant under different thickness of cancellous bone and cortical bone,and to analyze the influence of thickness ratio and total thickness of bone tissues on the reliability of an implant.Methods:By using the commercial finite element method software Abaqus, a simplified three-dimensional model of a jawbone consisting of a cancellous bone,a cortical bone,an implant,and a ceramic crown was constructed,and then the computation was performed.Under the condition that the system was loaded by lateral and normal stresses, the influence of thickness ratio and total thickness of cancellous bone and cortical bone in the stress distribution surrounding the implant was studied,where the thickness ratios were 3∶1, 2∶1, 1∶1, 1∶2, and 1∶3;the total thickness were 0.5, 1.0, 2.0, 3.0 and 4.0 mm, respectively. Results:The maximum stresses on the cortical bone,the cancellous bone as well as the implant were all found to decrease with the increasing of the total thickness of cortical and cancellous bones,with a higher decreasing rate in the range between 0.5-2.0 mm and a lower decreasing rate between 2.0-4.0 mm. More importantly, the maximum value of stress in the cortical bone within the neck region of the implant was observed to increase dramatically via reducing the total thickness below 2 mm, while it was increased insignificantly when the total thickness was above 2.0 mm. Conclusion:The thickness ratio and the total thickness of cancellous bone and cortical bone have strong influence in the stress distribution surrounding the implant.In dental implantation surgery, the total thickness of cancellous bone and cortical bone should be at least 2 mm, and therefore 2 mm is an optimal value.
5.The correlations of single photon emission computed tomography joints scan and bone metabolic markers in active rheumatoid arthritis

Objective To explore the correlation between quantitative value of joint bone scan by single photon emission computed tomography (SPECT) and serum bone metabolic markers in patients with active rheumatoid arthritis (RA).Method Clinical data of 60 newly diagnosed RA patients were retrospectively collected in Department of Rheumatology and Immunology from May 2013 to December 2014,including 28 cases with medical history less than 6 months and 32 from 6 months to 2 years.Serum Cterminal telopeptide of type Ⅰ collgen(CTX-Ⅰ)and N-terminal propeptide of type Ⅰ collagen(PINP) were tested by chemiluminesence.SPECT whole body bone and target joint scan before treatment was done.Thirty-nine healthy subjects as control group received ultrasound,electrocardiogram,X-ray,and whole body bone imaging examination.Quantitative values of joint imaging were calculated for the statistical analysis.Result Demographic data between RA group and control group were comparable (P > 0.05),including sample,sex,age and BMI.The joint SPECT value,CTX-Ⅰ and PINP levels were all significantly higher than those of control group (P < 0.01),which were 6.48 ± 1.98 versus 3.73 ± 1.t6;(0.66 ± 0.37) mg/L versus (0.58 ±0.21) mg/L;(46.35 ±28.15) mg/L versus (30.47 ± 13.75) mg/L respectively.Joint SPECT values had positive correlations with serum CTX-Ⅰ levels in all RA patients,as well as PINP in patients with disease duration 6mon-2years.And the according correlation coefficients were 0.513,0.495,0.402 (P < 0.05).But SPECT value had no correlation with CTX-Ⅰ (P =0.081) in patients with disease duration less than 6 mon.The correlation coefficient was 0.336.Conclusion SPECT imaging quantitative values were positively correlated with serum bone metabolic parameters.Thus SPECT imaging alone or combined with bone markers are helpful in diagnosing active RA.

7.Expression and its significance of Cyclin G2 in laryngeal squamous cell carcinoma

OBJECTIVE To study the correlation between the expression level of Cyclin G2 and the laryngeal squamous cell carcinoma.METHODS The clinical data and paraffin-embedded tumor samples of 81 patients with LSCC treated at Shenyang 463 Hospital between Jan. 2000 to Dec.2000 were retrieved.Cyclin G2 expression was analyzed immunohistochemically in tumor tissues and adjacent normal laryngeal mucosa.The relationship between Cyclin G2 expression and clinical data including age,gender,tumor stage,pathological type,and prognosis were analyzed using software SPSS12.0.RESULTS The positive rate of Cyclin G2 in laryngeal squamous cell carcinomas was 66.7% and it was correlated with tumor differentiation,lymph node metastasis(both P value less than 0.05).CONCLUSION The expression of CyclinG2 correlates with the malignant degree and development tendency of LSCC negatively.Cyclin G2 is an important biological factor affecting the prognosis.
9.Positron emission tomography-CT evaluation of therapeutic effect on lung cancer: a comparative study

Objective To compare the PET response criteria in solid tumors (PERCIST) and response evaluation criteria in solid tumors (RECIST) in the evaluation of therapeutic response in 49 non-small-cell lung cancer(NSCLC) patients.Methods Forty-nine NSCLC patients who received chemotherapy but no surgery were studied.Therapeutic responses were evaluated using 18 F-FDG PET and CT according to the RECIST and PERCIST methods.The PET-CT scans were obtained before chemotherapy and about 2 or 6 weeks after completion of chemotherapy.Firstly the reduction rates of tumor diameter and reduction rates of tumor standardized uptake value were compared with paired t-test.Then the response was classed into 4 levels according to RECIST and PERCIST:PD and PMD =1,SD and SMD =2,PR and PMR =3,CR and CMR =4.Pearson and Chi-square test was used to compare the proportion of four levels in RECIST and PERCIST.Finally one target lesion and two target lesions were compared for RECIST therapeutic evaluation in 33 cases with two target lesions.Results The diameter was (3.52 ± 1.65) cm before the therapy and (2.39 ± 1.43)cm after the therapy.The standardized uptake value was 8.78 ±4.18 vs.5.06 ±3.62 before and after therapy for the first target lesions in 49 patients.The differences of reduction rates between tumor diameter and standardized uptake value were not significant because of selection bias (0.32 ± 0.27 vs.0.28 ±0.64,t =0.176,P >0.05).However,there was a significant difference when the reduction rate was compared only in 39 patients in which the standardized uptake values were reduced after therapy (0.39 ± 0.29 vs.0.52 ±0.28,t =-4.08,P <0.01).The results of classification were 1/13 for CR/CMR,25/16 for PR/PMR,22/15 for SD/SMD,1/5 for PD/PMD,and 33 cases had no consistent results from RECIST and PERCIST.There was a significant difference in response classification between RECIST and PERCIST (x2 =16.252,P < 0.01).No significant difference was found between one target lesion and two target lesions for RECIST evaluation results in 33 cases (x2 =1.171,P > 0.05),but results of response classification were changed in 5 cases.Conclusions PERCIST criteria may be more sensitive in NSCLC therapeutic evaluation comparing to RECIST criteria.There is a higher proportion with CMR and PMD in PERCIST,but its effect on the prognosis is not yet clear.The number of target lesions may affect the results of therapeutic evaluation with RECIST criteria.
10.Effect of enalapril combined with folate acid on endothelial function and urine microalbumin in patients with hypertension

Objective To explore the effect of enalapril combined with folate acid on endothelial function and urine microalbumin(UMA) in patients with hypertension.Methods 120 patients with hypertension were randomly divided into two groups:control group (n =60) was given enalapril 10.0mg/d,observation group (n =60) received enalapril 10.0mg + folic acid 0.4mg/d.The total treatment period was 8 weeks.Blood pressure,plasma homocysteine (Hcy),flow mediated dilation (FMD) and UMA were examined.Results The efficacy of pressure releasinghad no significant difference between two groups.Hcy[(10.2 ± 5.8) μmol/L vs (16.6 ±-8.1) μmol/L,t =3.641],FMD[(14.8 ±5.4)% vs (8.2±3.5)%,t =7.325] and UMA[(14.8 ±5.4)mg/L vs (31.6 ±9.5)mg/L,t =8.221] of two groups were significantly different after treatment.Conclusion Combination therapy of enalapril and folate acid can decrease plasma Hey and UMA,restore vascular endothelium function in patients with hypertension.
