3.Development of advanced gastric cancer treated with weekly administration of paclitaxel
China Oncology 2006;0(10):-
Benefits in survival and quality of life have been demonstrated in patients with advanced gastric cancer receiving chemotherapy plus optimal supportive care versus optimal supportive care alone. The taxanes, which are among the most promising cytotoxic agents in clinical use have considerable activity in a number of tumor types.Paclitaxel, especially low-dose weekly administration, has shown encouraging activity in the treatment of advanced gastric cancer as a single agent.Patients treated with the low-dose weekly paclitaxel regimen had a statistically superior response rate and time to disease progression as well as clinically significant prolongation of survival. And it offers a new option for those advanced gastric patients such as the aged, poor performance status(PS) or unsuitable for combination chemotherapy, and those even with no indications for rountine chemotherapy. This review is focused on the clinical reports of low-dose weekly administration of paclitaxel for patients with advanced gastric cancer.
4.The experimental study of effect of electric vagal stimulation on the gene expression of injured myocardium from ischemia/reperfusion rat model
Journal of Chinese Physician 2014;16(8):1060-1063
Objectives To investigate the effect of electric vagal stimulation on the gene expression of injured myocardium from ischemia/reperfusion rat model and explore the involved molecular mechanism. Methods Twenty Sprague Dawley male rats were randomly selected and divided into 2 groups evenly:ischemia/reperfusion group (I/R group),and vagus nerve stimulation group (STM group). The left anterior descending coronary artery (LAD) was ligated and subjected to 30 min of myocardial ischemia followed by 2 h of reperfusion. In addition,10 min before reperfusion, left cervical vagus nerve of STM group was subjected to electronic stimulation at 5 V,2 ms and 1 Hz for 20 min. After 120 min of reperfusion, every group was randomly divided into two parts. One part that myocardium was collected from left ventricle was applied to determine the area of myocardial infarction. The RNA isolated from another part of the ischemic myocardium collected from left ventricle was hybridized to get gene expression profiles and the quality of hybridized RNA from both I/R and STM group was assessed and analyzed.GeneSpring software was applied to screen out the genes, which show significant difference between groups I/R and STM.Real-time polymerase chain reaction (RT-PCR) was applied to analyze the expression of important genes. Results (1)The area of myocardial infarction of STM (25.5 ± 3.9) % was significant reduced relative to L/R group (45.5 ± 4.8) % (P < 0.05).(2)The expression levels of 186 genes were changed significantly, and analyzed by Gene ontology (GO) software, there were 3 kinds of genes were affected. The upregulated genes were reported to show protective effect on myocardium. The downregulated gene was relative to inflammation.(3)The RT-PCR result confirmed the genechip assay. Conclusions Electric vagal stimulation can reduce myocardial I/R injury in rats.Significant change of the gene expression was detected between groups I/R and STM.The results suggest that activation of cholinergic anti-inflammatory pathway be involved in the mechanism.
5.Study of Candida albicans vaginitis model in Kunming mice.
Journal of Huazhong University of Science and Technology (Medical Sciences) 2007;27(3):307-10
The model of vaginal candidiasis in Kunming mice was constructed in order to search for the optima construction conditions and provide an economic animal model of Candida albicans (C. albicans) vaginitis. Estrogen benzoate (E2) was given to mice at different concentrations ranging from 0.0 to 0.05 mg/mouse (4 levels) beginning 72 h prior to vaginal inoculation, then mice were inoculated intravaginally with various concentrations of stationary-phase C. albicans blastoconidia (ATCC90028) (5 levels) in 20 microL of phosphate-buffered saline (PBS) in each E2 level. General state, scores of genital pathology, the hyphae and vaginal fungal burden (CFU) in vaginal lavage fluid, the hydrops rate of uterus and vaginal tissues for pathological section in mice were observed and obtained at day 2, 4, 7, 14 and 21 after inoculation. The results showed the infection rate in mice was related to the dosage of E2 and concentration of C. albicans blastoconidia. Additionally there was better cross-effect between the two treated factors. The infection rate was about 80% on the day 4, and could reach 100% on the day 7 until the end of experiment after inoculated intravaginally in groups of E2I3, E2 0.025 mg/mouse injected hypodermically and inoculated intravaginally with 5 x 10(4) C. albicans blastoconidia, and large amount of hyphae and blastoconidia could be observe in superficial layer tissue and canal of vaginal by PAS. From the results in our experiment it was concluded that E2I3 was the optima construction condition in kunming mice.
