3.The study of a differential induced FOXP3 expression by transforming growth factor-β1 between CD4+and CD8+ T lymphocytes
Chinese Journal of Hepatobiliary Surgery 2013;(4):301-306
Objective To study the different induced expressions of FOXP3 by transforming growth factor-β1 (TGF-β1) between CD4+ and CD8+ T lymphocytes. Methods After the cultures of CD4+ cells and or CD8 + cells under an anti-TCR stimulation condition with an addition of TGF [β1 or not for 4 days respectively, the induced FOXP3 expressions were detected through the flow cytometric intracellular staining.Meanwhile we also examined the proliferative activity of TGF β1 induced FOXP3 expressing lymphocytes through a CFSE labeling experiment and the effect of TGF-β1 on cell apoptosis by annexin V staining. Results TGF-β1 selectively induced the CD4+ T lymphocytes to express FOXP3 (PBMC:29.66±3.624 vs 7.430±0.643; NIL:31.74±2.612 vs 8.637±1.146); The induced cells also had a proliferative activity (94.39 ± 1.179) and could secrete IFN-γ efficiently after activation (39.58±1.611); TGF-β1 could reduce the apoptosis rate of CD8+T lymphocytes(25.39±2.158 vs 9.320±0.3219).Conclusions Consistent with the discovery that >95% of the FOXP3+lymphocytes in TIL (tumor infiltrating lymphocytes) of HCC (human hepatocelluar carcinoma) patients were concentrated in CD4+ T lymphocytes (97.15±0.3807,n=10),TGF-β1 induced the CD4 +T lymphocytes to express FOXP3 in preference to CD8+T lymphocytes; But with different to natural Tregs, the induced cells still could proliferate and secrete IFN-γactively after an effective stimulation;CD8+ T lymphocytes were more easily suffering cell apoptosis after activation than CD4+ T lymphocytes, and TGF-β1 could rescue the more cell apoptosis.
4.The experimental study of effect of electric vagal stimulation on the gene expression of injured myocardium from ischemia/reperfusion rat model
Journal of Chinese Physician 2014;16(8):1060-1063
Objectives To investigate the effect of electric vagal stimulation on the gene expression of injured myocardium from ischemia/reperfusion rat model and explore the involved molecular mechanism. Methods Twenty Sprague Dawley male rats were randomly selected and divided into 2 groups evenly:ischemia/reperfusion group (I/R group),and vagus nerve stimulation group (STM group). The left anterior descending coronary artery (LAD) was ligated and subjected to 30 min of myocardial ischemia followed by 2 h of reperfusion. In addition,10 min before reperfusion, left cervical vagus nerve of STM group was subjected to electronic stimulation at 5 V,2 ms and 1 Hz for 20 min. After 120 min of reperfusion, every group was randomly divided into two parts. One part that myocardium was collected from left ventricle was applied to determine the area of myocardial infarction. The RNA isolated from another part of the ischemic myocardium collected from left ventricle was hybridized to get gene expression profiles and the quality of hybridized RNA from both I/R and STM group was assessed and analyzed.GeneSpring software was applied to screen out the genes, which show significant difference between groups I/R and STM.Real-time polymerase chain reaction (RT-PCR) was applied to analyze the expression of important genes. Results (1)The area of myocardial infarction of STM (25.5 ± 3.9) % was significant reduced relative to L/R group (45.5 ± 4.8) % (P < 0.05).(2)The expression levels of 186 genes were changed significantly, and analyzed by Gene ontology (GO) software, there were 3 kinds of genes were affected. The upregulated genes were reported to show protective effect on myocardium. The downregulated gene was relative to inflammation.(3)The RT-PCR result confirmed the genechip assay. Conclusions Electric vagal stimulation can reduce myocardial I/R injury in rats.Significant change of the gene expression was detected between groups I/R and STM.The results suggest that activation of cholinergic anti-inflammatory pathway be involved in the mechanism.
5.A Clinical Study on Gut Barrier Function Change after Acute Intraperitoneal Hemorrhage
Journal of Chinese Physician 2001;0(06):-
12h interval from injury to operation had significantly higher concentration of plasma D-lactate and endotoxin at above metioned 4 time points. Plasma concentration of D-lactate and endotoxin in the last two groups declined to normal range at 72h after operation, while in the first two groups reduced to normal range before leaving hospital. Conclusion Acute intraperitoneal hemorrhage could damage gut barrier function, and injury degree was related with interval from injury to operation and blood loss amount.

This review focuses on PET reporter gene imaging which has been studied intensively and applied widely in the research on imaging monitoring of cellular immunotherapy for cancer, with the purpose to provide innovative clues for the preclinical study and clinical translation research.
7.Pedicle screw fixation for treating thoracolumbar fracture:related factors influencing vertebral height restoration
Chinese Journal of Tissue Engineering Research 2015;(39):6315-6320
BACKGROUND:Pedicle screw fixation in the treatment of thoracolumbar fracture can effectively restore vertebral body height and physiological radian, and are widely used in the clinic. In the clinic, we found that different surgical methods on restoration of vertebral height are different. Simultaneously, there are differences in relevant factors before repair, which causes that the recovery of vertebral body height is not consistent.
OBJECTIVE:To explore the related factors influencing the recovery of vertebral height of pedicle screw fixation system for treating thoracolumbar fracture.
METHODS:Clinical data of 63 patients with thoracolumbar fracture, who were treated by posterior pedicle screw fixation system from September 2012 to March 2015, were summarized. The fracture types were A3 or A4 type in the AO spine thoracolumbar injury classification system, including 32 cases of transpedicular screw fixation (group A) and 31 cases of beyond-injured-vertebral fixation (group B). The related factors including age, course of disease (within 3 weeks), bone mineral density, preoperative degree of vertebral compression and preoperative sagittal Cobb’s angle were recorded. The degree of vertebral height restoration was also observed. Intergroup analysis was conducted using independent samplet-test. Intragroup analysis was performed using bivariate regression analysis in single factor analysis. Multivariate linear regression analysis was applied for multivariate analysis so as to explore the factors related to vertebral height restoration.
RESULTS AND CONCLUSION:No significant difference in preoperative rate of vertebral compression or Cobb’s angle was detected between the two groups (P > 0.05). Postoperative rate of vertebral height restoration of group A was significantly greater than group B (P < 0.05). Univariate analysis within the group showed that course of disease, preoperative degree of vertebral compression and bone mineral density were strongly associated with the restoration of vertebral height (P < 0.05). In contrast, there was no significant correlation between age, preoperative sagittal Cobb’s angle and the restoration of vertebral height (P > 0.05). Multiple linear regression analysis showed that the preoperative degree of vertebral compression and the course of disease were the predominant factors affecting the vertebral height restoration. The standardized partial regression coefficients were 0.225, -0.621, respectively. These results demonstrate that transpedicular screw fixation can more effectively restore vertebral height than traditional beyond-injured-vertebral internal fixation. The preoperative vertebral compression and course of diseases are the predominant factors affecting the vertebral height restoration, which can predict postoperative vertebral height restoration.
8.Mechanisms of DNA Damage Induced by Hexavalent Chromium Compounds
Journal of Environment and Health 1993;0(01):-
Hexavalent chromium[Cr(VI)] is a well-know environmental toxicant which can cause many types of DNA damage. However Cr(VI) does not react directly with DNA itself, it penetrates the cell membrane through the nonspecific phosphate/sulphate anion channel and undergoes a reductive metabolism to a series of lower oxidation state including quinquevalent chromium[Cr(V)], quadrivalent chromium[Cr(IV)] and trivalent chromium[Cr(III)]. Cr(III) can form variety of Cr-DNA adducts; moreover oxidative Cr(V) and free radicals generated during the reduction cause DNA oxidative damage. The process of how Cr(VI) induce DNA damage after entered the cells and types of DNA lesions were briefly reviewed in the present article.
9.Identification of low-molecular weight prostate-specific antigen(PSA) and lactoferrin in the prostatic secretion of benign prostatic hyperplasia
Journal of Peking University(Health Sciences) 2003;0(06):-
Objective:To investigate the expression of low-molecular-weight PSA(lw-PSA) and lactoferrin in the expressed prostatic secretion(EPS) from both benign prostatic hyperplasia(BPH) and normal prostate. Methods: Forty human EPS samples obtained from 20 BPH patients and 20 normal males were subjected to two-dimensional gel electrophoresis(2-DE).Mass spectrometry was performed to confirm the nature of the secreted proteins in EPS.Results: One uniquely expressed protein in BPH was detected and mass spectrometry determined its nature as lw-PSA(molecular weight 10?10~3,pI 8.5-(9.3)).More importantly,Western blotting analysis also revealed that lw-PSA detected in BPH-EPS,but was undetectable in BPH-free EPS.In addition,up-regulation of Lactoferrin(molecular weight 35?10~3,pI 7-7.5) in BPHEPS,as compared with BPH-free EPS,was also observed.More interestingly, lactoferrin was absent in prostate cancer tissues. Conclusion: Our results indicate lw-PSA may be produced specifically by BPH epithelium and it has a potential to be used as a specific biological marker for the diagnosis of BPH.In addition, benign prostatic epithelium can produce more lactoferrin while prostate cancer tissues go without its lactoferrin secretion.
10.Technology of breast cancer screening:current situation and prospect
Chinese Journal of Health Management 2016;10(4):321-325
Breast cancer is the first malignant tumor of women, and the incidence is still rising. Although the overall survival rate of breast cancer is high, the difference in prognosis is significant with different stage. Finding the patients at early stage and commencement of therapy timely by breast cancer screening is the key to reduce the incidence and improve the survival rate. At present, clinical breast examination, ultrasonography, mammography and MRI are still the most common methods for breast cancer screening, but in view of the specific technology and specific population it remains controversial as to which techniques are the best, especially the value of ultrasonography still needs to be confirmed in randomized clinical trials. With the constant improvement of technologies and programs, the breast cancer screening work will be more standardized and optimized.
