1.A comparative study of intravenous thrombolysis and late intracoronary stenting in treatment of acute ischemic stroke onset using 99mTc2ECD SPECT imaging

Objective To evaluate and compare the therapeutic effect among intravenous rt2PA thrombolysis,thrombolysis plus delayed intracoronary stenting for acute ischemic stroke onset using 99mTc2ECD SPECT imaging.Methods 45 cases of acute ischemic stroke onset were divided into two groups which were the thrombolysis group( n = 23 ) ,thrombolysis plus delayed stenting group (n = 22), 99mTc2MIBI SPECT imaging was performed 3 weeks after thrombolysis therapy or 1 week after intracoronary stenting. The score of 99mTc2MIBI brain uptake were analyzed semiquantitatively and the total score of myocardial 99mTc2ECD uptake was calculated. Results The scores of the 2 groups were( 12. 4 ± 4. 6) and( 9. 7 ± 3. 8 ) respectively, Significant difference was shown in the thrombolysis plus delayed stenting group compared with the thrombolysis group (P < 0. 01 ). Conclusion 99mTc2ECD SPECT imaging had been proved to be an objective parameter for evaluating the therapeutic effect of the various treatments of AMI. Thrombolysis plus delayed stenting was more effective than thrombolysis.
2.Effect of MAPKK/MEK-MAPK/ERK Signal Transduction Pathway in Rat Neural Stem Cells

Objective To incubate the rat neural stem cells with specific inhibitor (PD98059) of MAPKK/MEK to clarify the effect of MAPKK/MEK-MAPK/ERK signaling pathway in neural stem cells (NSC).Methods NSCs derived fiom E15-16 rats were isolated and cultured.After treated with different concentration of PD98059,they were subjected to the detection of cell proliferation,Nesn'n, BrdU and β-tubulin-Ⅲ by WTS-8 assay,immunofluorescent staining or Western Blot respectively.Results PD98059 had an effect on the survival,proliferation and differentiation of NSC at a concentration-dependent manner.The viability of the NSC was significantly decreased than the control (P<0.05),whereas the numbers of BrdU-positive and β-tubulin-Ⅲ positive cells were notably fewer than the control (P<0.05) after incubated with the higher concentration of PD98059.The ERK expression was blocked by PD98059 at a concentration-dependent manner.Conclusion The MAPKK/MEK-MAPK/ERK signaling pathway played a vital role in the survival, proliferation and differentiation of NSC.
3.Investigation in awareness of prevention of upper respiratory tract infection in parents of leukemia children and the effect of health education

Objective To explore the effect of health education on improving awareness of preven-tion of upper respiratory tract infection of parents of leukemia children. Methods 25 parents were ran-domly chosen, and comprehensive health education interventions were provided, including lectures, advisory guidance and brochures. The awareness of prevention of upper respiratory tract infection of parents was e-valuated before and after health education,and the results underwent χ<'2> test. Results After health educa-tion, the awareness of prevention of upper respiratory tract infection of parents significantly improved com-pared with that before. Conclusions Health education can improve awareness of prevention of upper res-piratory tract infection of parents and decrease the incidence of upper respiratory tract infections.
4.Comparison of the efficacy of vasopressin and epinephrine during cardiopulmonary resuscitation in mouse model

Objective To compare the effects between vasopressin and epinephrine during cardiopulmonary resuscitation(CPR)in a mouse model of cardiac arrest(CA).Method Transoesophageal cardiac pacing was performed so as to elicit cardiac arrest in 30 Kunming male mice.Four minutes after the initiation of cardiac pacing,the animals were prospectively randomized into three groups in equal number(n=10/group),namely,control group(saline 0.2 mL intra-arterial),vasopressin group(vasopressin 0.4U/kg intra-arterial)and epinephrine group(epinephrine 0.04 mg/kg intra-arterial),then CPR was initiated.Restoration of spontaneous circulation (ROSC)was observed.If ROSC failed to appear at 10 minutes after CPR,resuscitation was discontinued.Electrocardiogram and blood pressure of resuscitated animals were invasively monitored for an additional 60 minutes.Electrocardiogram and blood pressure.and the restoration of spontaneous respiration and survival time were observed and recorded.Results Rates of ROSC in vasopressin group and epinephrine group were significantly higher than those in saline group(9/10,10/10 vs.3/10,P<0.05,P<0.01 respectively),and there was signilieant difference between vasopressin and epinephrine group.All resuscitated mice treated with epinephrine restored sponlaneous respiration after ROSC,while only 4 of 9 animals trealed with vasopressin did(P<0.05).Survival time of anireals in epinephrine group was longer than that in vasopressin group or in saline group(P<0.05,P<0.05,respectively).Conclusions Both vasopressin and epinephrine increase the rates of ROSC.Epinephrine 0.04 mg/kg improved respiratory function and results in a longer survival time compared with vasopressin 0.4 U/kg in this mouse model.and the precise mechanism is not clear and further investigation is required.
5.Impact of point spread function on image quality and SUV in FDG PET imaging

Objective To discuss the impact of an iterative reconstruction algorithm (True X) implemented with point spread function (PSF) on image quality and SUV in 18F-FDG PET, and compare with other reconstruction algorithms.Methods A total of 79 hypermetabolic lesions from 37 cancer patients (23 males, 14 females, age (63.38±15.25) years) were retrospectively studied.The PET images were reconstructed using the following six reconstruction algorithms, including FBP, FBP+TOF, iteration, iteration+TOF, True X, True X+TOF.Paired t test was used for statistical analysis.Results All six reconstruction algorithms showed significant difference in lesion SUVmax.The adoption of PSF increased SUVmax by 15%-16%, and decreased the dispersion of SUV in the liver parenchyma remarkably.Conclusions Lesion SUVmax calculated from different reconstruction algorithms has significant differences.Caution should be taken in using PSF, as it can improve image quality but may also increase lesion SUVmax.
6.Application of carbon nanoparticle in the axillary sentinel lymph node biopsy for breast cancer

Objective To explore the clinical effect of carbon nanoparticle in the axillary sentinel lymph node tracer of breast cancer.Methods Fifty-seven patients with breast cancer underwent lymphatic tracing with carbon nanoparticle.The carbon nanoparticle suspension was subcutaneously injected around the areola (points at 4 o'clock) and tumor surface skin at 30 min before operation.The first lymph nodes blacked were named sentinel lymph node.Then performed modified radical mastectomy,observing the application value of carbon nanoparticle.Results The detection rate of sentinel lymph node was 98.2% (56/57).The correct,sensitive and false negative rates were 98.2%(55/56),95.2%(20/21) and 4.8%(1/21),respectively.The false positive rate was 0.Conclusions The carbon nanoparticle suspension injection subcutaneously around areola in sentinel lymph node biopsy is helpful for accurately evaluating the axillary lymph node condition.Being simple to operate,accurate positioning,dyeing time and is worth popularizing.
7.THE CHEMOPROPHYLACTIC EFFECTS OF PROCYANIDINS FROM LOTUS SEEDPOD ON DMBA-INDUCED BUCCAL-POUCH CARCINOMAS IN GOLDEN HAMSTERS

Objective: To study the contents and constituents of LSPC (procyanidins from Lotus Seedpod) and its chemoprophylactic effects on 9,10-dimethyl-1,2-benzanthracene (DMBA)-induced golden hamsters buccal-pouch carcinomas. Methods: ESI-MS was used to analyze LSPC, and the change of body weight, mortality during test, and the value of serum MDA, GSH-Px, T-SOD,the gross and pathological change of buccal-pouch mucosa were investigated when golden hamsters were via gastric intubation or the buccal pouch mucosa was smeared with 100mg/(kg bw?d)LSPC. Results and conclusion: The contents of LSPC exceeded 98% and mono-, di-, tri-, tetrameric procyanidins as well as di-, trimeric galic ester were constituted of LSPC with molecular weight ranging from 290-1154. LSPC had chemoprophylactic effects on DMBA-induced golden hamsters buccal-pouch carcinomas, and the effect was superior through smearing LSPC rather then via gastric intubation .
8.Effects of thymosin alpha 1 combined with neoadjuvant chemotherapy in breast cancer

Objective To study the impact of the thymosin alpha 1 on the toxicity and celluar immune function during neoadjuvant chemotherapy of breast cancer.Methods 83 patients of Ⅱ b-Ⅲ a stage breast cancer were randomly divided into two groups:study group(40 patients,neoadjuvant chemotherapy of CEF combined with thymosin α 11.6mg HQD) and control group(43 cases,neoadjuvant chemotherapy of CEF alone).Results Rates of gastrointestinal reaction and bone marrow depression in study group were significantly lower than those in control group.The levels of CD3,CD4,CD4/CD8 and NK in study group were significantly higher than those in control group after chemotherapy.Conclusion The combination of thymosin alpha 1 and neoadjuvant chemotherapy for breast cancer can reduce the toxicity,improve the tolerance,enhance cellular immune function and improve the quality of breast cancer patient's life.
9.The application of ultrasound scalpel in axillary lymph node dissection in modified radical mastectomy for breast cancer

Objective To study the effect of the ultrasound scalpel and electric knife in axillary lymph node dissection in modified radical mastectomy for breast cancer.Methods The clinical data of 429 patients with modified radical mastectomy for breast cancer were analyzed retrospectively.The axillary lymph node dissection in 178 cases were performed with the ultrasound scalpel (ultrasound scalpel group) and 251 cases were done with the electric knife(electric knife group).The blood loss,operative time,extubation time,the total amount of drainage,the incidence of seroma were compared.Results Two groups were successfully operated,no axillary vessels and long thoracic nerve,chest nerve injury.The blood loss,operative time,extubation time,the total amount of drainage in ultrasound scalpel group were significantly better than those in electric knife group [(98.0 ± 10.2) ml vs.(152.0 ± 11.5) ml,(125.0 ±9.6) min vs.(155.0 ± 12.3) min,(6.0 ± 2.1) d vs.(9.0 ± 4.3) d,(227.0 ± 10.6) ml vs.(319.0 ± 15.8) ml,P< 0.05 or < 0.01].The incidence of seroma was no significant difference between two groups (P > 0.05).Conclusions Using ultrasound scalpel in axillary lymph node dissection for breast cancer can significantly reduce operative time,and is consistent with less blood loss,less extubation time and the total amount of drainage.
10.The early protective effect of NADPH on OGD/R injury of human umbilical vein endothelial cells

Objective To study the early protective effect of NADPH on human umbilical vein endothelial cells (HUVECs) and the expression of occludin and MMP9 induced by oxygen glucose deprivation and reoxygenation (OGD/R). Methods HUVECs were cultured and divided into blank control group, OGD/R group and OGD/R+NADPH 20 μmol/L group. The proliferation of HUVECs after treatment was detected by CCK-8 assay. The cytotoxicity was detected by LDH release assay. The morphological changes of HUVECs were observed by inverted microscope. Superoxide dismutase (SOD MDA) activity and malondialdehyde (MDA) were detected by commercially available kit. The expressions of occludin and MMP9 were detected by Western blot assay. Results Compared with the OGD/R, NADPH enhanced the cell viability significantly (P<0.05), reduced the release of LDH (P<0.05), promoted the maintance of HUVECs morphology, reduced MDA generation (P<0.05) and increased SOD activity (P<0.05). Following OGD/R,the treatment of NADPH can inhibit MMP9 level (P<0.05) and promote the recovery of occludin level (P<0.05). Conclusion NADPH can protect HUVECs from the damage induced by OGD/R by reducing oxidative stress and regulating the expressions of MMP9 and occludin.
