1.Influences of retrorsine on mouse hepatocyte proliferation after liver injury

Objective:To compare the influences of retrorsine on hepatocytes proliferation in mice and rats after liver injury,so as to investigate the feasibility of using retrorsine for establishment of liver cell transplantation model in mice.Methods: Male mice and rats were pretreated with 2 injections of retrorsine(70 mg/kg for mice and 30 mg/kg for rats)(as retrorsine-treated group,n=30) at 2 weeks interval or saline(as nontreated group,n=30).A single injection of carbon tetrachloride(CCl_(4), 0.5 mg/kg) was given to all animals 4 weeks after the final injection of retrorsine or saline.At 0(before administration),1,2,3,4,6,15 d after CCl_(4) administration,the animals were sacrificed and their livers were subjected to H-E staining and Ki-67 antibody immunohistochemistry analysis to evaluate the pathological changes and hepatocyte proliferation.Results: The liver in rats treated with retrorsine displayed obvious megalocytosis,small bile duct hyperplasia,and small hepatocyte hyperplasia(forming nodule);no such changes were found in non-treated group.However,the liver in mice treated with retrorsine displayed hepatocyte degeneration and necrosis in the perivenous areas and the same was true to the liver in non-treated mice.Ki-67 immunohistochemistry analysis showed that in rats treated with retrorsine,the positive hepatocytes,mainly found in small hepatocyte nodules,were obviously less than those in control group 3 d after CCl_(4)administration(P

3.Effect of atorvastatin on the levels of serum IL-6、ET and MCP-1 in patients with acute cerebral infarction

Objective To explore the effect ot atorvastatin on the levels of serum Interleukin-6 ( IL-6),Endothelin(ET) and MCP-1 in patients with acute cerebral infarction(ACI).Methods 100 patients with ACI were randomly divided into the atorvastatin therapy group(n =50) and the conventional therapy group(n =50).At the basic of conventional therapy,the atorvastatin therapy group was treated with Atorvastatin 20mg/d.Both groups were treated for 4 weeks.The levels of serum IL-6,ET and MCP-1 were measured and the scores of neurological deficit ( NDS ) were evaluated before and after treatment.A healthy control group which approximately matched the experimental group included 30 persons.Results The levels of serum IL-6,ET and MCP-1 in both two groups after 14d treatment were significantly lower than those before treatment(t =26.38,49.10,34.51,25.77,32.23,34.68,P <0.05),and the levels of serum IL-6,ET and MCP-1 in atorvastatin therapy group was significantly lower than that in conventional therapy group( t =10.10,13.18,23.85,P <0.05 ).NDS of both groups after treatment was lower than those before treatment(t =18.38,6.98,P <0.05 ),and the score of atorvastatin therapy group was significantly lower than that of conventional therapy group( t =12.00,P < 0.05).Conclusion Atorvastatin could significantly decrease the levels of serum IL-6,ET and MCP-1,and it could promote the neurological fuction recovery in the patients with ACI.
4.Clinical observation of Rhubarb and mirabilite with comprehensive nursing in treatment of abdominal surgery in patients with abdominal distension

Objective To study the clinical effect of complex treatment combined with external application of Rhubarb and Mirabilite on abdomen of patients with abdominal distension after abdominal surgery .Methods 90 patients with abdominal distension after gynecological abdominal surgery from June 2014 to September 2015 were randomly divided into conventional group(45 patients) and comprehensive group(45 patients).Patients in the conventional group were given routine gynecological postoperative care, and patients in the comprehensive group were given complex treatment combined with external application of Rhubarb and Mirabilite on abdomen.The abdominal distension duration, anal exhaust time, time of resuming peristaltic sound, first defecation time, treatment effect, depression and anxiety emotions, changes of life quality of the two groups before and after intervention were compared.Results The comprehensive group had higher treatment effect than the comprehensive group ( P<0.05 ) .The abdominal distension duration, anal exhaust time, time of resuming peristaltic sound, first defecation time of the comprehensive group were shorter than that of the conventional group(P<0.05).The comprehensive group had better depression, anxiety emotions and life quality compared to the conventional group (P<0.05).Conclusion Complex treatment combined with external application of Rhubarb and Mirabilite on abdomen has a very significant clinical effect on patients with abdominal distension after abdominal surgery, it can improve patients’negative emotions and the symptom of abdomen distension, promote life quality.
5.The effect of diabetes on the bone metabolism

The relationship between diabetes and osteoporosis has gradually become a new research hotspot , but is still much controversy .High concentration of blood glucose can be bond to varieties of proteins in vivo and form glycosylation end products in col -lagen tissue and reduce bone strength , besides it can inhibit RANKL induced osteoclast formation and bone resorption by repressing redox-sensitive NF-kappa B activity .A series of diabetes mellitus complications can induce less local bone nutrition supply and calcium and phosphorus sorption , reduce muscle tension and bone mineral density , which leads to the increase of fracture risk .Type 2 diabetes patients with overweight or obesity often have increased secretion of inflammation factors , adiponectin and leptin , which promotes the formation of osteoclasts and encourages bone marrow stromal stem cells to differentiate into adipocytes ;On the other hand , adiponectin and leptin can facilitate osteoblasts proliferation , differentiation and activity , and promotes bone formation .This article will discuss how diabetes works on bone metabolism and the underlying pathophysiological and physiological mechanism .
6.The potential therapeutic effect of BDNF on GABAergic neuroplasticity dysfunction in late-life anxiety

Late-life anxiety has a high prevalence and usually co-morbidity with cerebral-and cardiovascular diseases which in turn increases the disability and mortality in old people.The pathology of late-life anxiety is difference from adult with the character of GABAergic neuroplasticity dysfunction.The therapeutic strategy presently can only alleviate the anxious symptom but not ameliorate the neuroplasticity.Brain-derived neurotrophic factor (BDNF) is a critical signaling molecule which regulates the GABAergic neuroplasticity,reduction of BDNF along with aging can induce GABAergic dysfunction which contributes to late-life anxiety.BDNF may exert anxiolytic effects by restoring the GABAergic plasticity and can be a potential therapeutic strategy of late-life anxiety.
7.Progress on the Study of Human Herpesvirus-8

Human herpesvirus-8 (HHV-8) also referred to as Kaposi’s sarcoma-associated herpes-virus (KSHV),is a novel tumor causing virus,which is thought to be the etiologic agent of KS,and also is associated with primary effusion lymphoma(PEL) and multicentric castleman’s disease(MCD) HHV-8 encodes a number of viral proteins, including the proteins associated with HHV-8 latent infection and lytic infection,and the proteins unique to HHV-8,which are critical players in the development of KS and HHV-8-associated diseases. The proteins encoded by HHV-8 and association between HHV-8 and KS were reviewed.
8.Dvelopment of a DNA-based microarray for detection of human adenovirus serotypes

Objective To develop a chemiluminescence imaging DNA microarray method for simultaneous,quick and accurate detection of serotypes of human adenovirus (HAdV ),namely,HAdV3,HAdV7,HAdV11,HAdV14 and HAdV55.Methods Based on the specific gene sequences in the conserved region of adenovirus from GenBank, oligonucleotide primers and probes were designed and synthesized to prepare the oligonucleotide microarray.The specific genomic sequences were amplified by multiplex PCR method.The multiplex PCR amplification products were hybridized with the specific probes of microarray that was scanned after washing and chemiluminescenceb before the result was analyzed.After optimization of the multiple PCR system,hybridization reactions and conditions of chemiluminescence,the specificity,sensitivity and reproducibility of the chip were evaluated.Results The microarray displayed high sensitivity, specificity and reproducibility.The minimum detection limit of plasmidg DNA was 3 ×103 copies/reaction.The microarray detection results of 38 clinical samples were approximately consistent with those using the direct sequencing method(37 /38).Conclusion A chemiluminescence imaging DNA microarray method for quick,sensitive and specific detection of five serotypes of HAdV is established,which can provide a new means for detecting serotypes of HAdV.
10.Effect of domperidone on enhancement of colonic motor activity

Objective To observe the prokinetic effect of domperidone on colon and compare with that of mosapride and cisapride.Methods ① In viva experiment:forty rats were divided into control,domperidone,mosapride,and cisapride groups with 10 in each group.Stran gauges were planted both in proximal and distal colons and colonic motor activities were recoded in conscious rats.② In vitro experiment:the prokinetics effects of dopamine or dopamine combined with domperidone on the contraction of isolated rat colon strips were recorded by tone-transducers in the thermostatic muscle bath.Results In viva experiments:① in the interdigestive period of resting,activities of rhythimic phasic contraction were recoded in colon at conscious rats,and ② it had been showed that dornperidone significantly enhanced the contraction of colon in dose-dependent manner.Compared with control group,the mean contraction amplitude of proximal colon and distal colon increased by 84.61% ± 7.26% and 76.37 % ± 8.47% in domeridone group,respectively,which was higher than those in mosapride group (50.32%±8.16% and 45.13%±7.16%,respectively),but as same as those in cisapride group (92.55% ± 8.37% and 81.27% ± 9.95%,respectively).In vitro experiments:① perfusion of dopamin (40 mg/ml) could significantly decrease the contraction of isolated colon strip by 91.56% ± 10.24% in comparison with Krebs-Ringer solution,and ① domperidone could block the relaxant effect of dopamin on the isolated colon strip.Conclusions Domperidone can significantly enhance the colonic motor activities via antidopaminergic action.The effects of domperidone are similar as eisapride and greater than mosapride.
