1.A Study on the Perioperative Clinical Efficacy of Restrictive Blood Transfusion Combined with Hyperbaric Oxygen Preconditioning on the THA and TKA

Objective:To compare the clinical effects of restrictive blood transfusion combined with hyperbaric oxygen preconditioning (HBOPC) and restrictive blood transfusion in the treatment of hip,knee arthroplasty (THA,THA).Methods:40 patients in the period of epidural anesthesia,femoral nerve hysteresis hip and knee arthroplasty were selected and randomly divided into two groups:restrictive transfusion group (maintain 80 g/L≤ Hb <100 g/L,n=20) and restrictive blood transfusion combined with HBOPC (HBOPC+maintain 80 g/L =Hb <100 g/L,n=20).The red blood cell transfusion,red blood cell transfusion rate,perioperative Hb,blood oxygen saturation (SO2),the incidence of hypotension during operation,hospitalization time and postoperative cerebral infarction,acute pulmonary embolism,pneumonia,myocardial infarction,wound infection rate and 90 days mortality rate were compared between two groups.Results:Compared with the restrictive transfusion group,the postoperative Hb,blood oxygen saturation (SO2) of restrictive blood transfusion combined with HBOPC group were significantly increased(P < 0.05);the red blood cell transfusion,red blood cell transfusion rate,incidence of pneumonia,wound infection rate were significantly decreased (P<0.05).Conclusion:Restrictive blood transfusion combined with hyperbaric oxygen preconditioning could improve the anoxic state of the hip,knee arthroplasty patients,which could effectively reduce red blood cell transfusion,reduce postoperative complications,has good clinical curative effect.
2.Comparison of LOXL-2 Expression between Active Systematic Scleroderma and Active Mixed Connective Tissue Disease

Objective To investigate the expression of lysyl oxidase like protein?2(LOXL?2)in the sera of patients with active systemic scleroder?ma(SSc)and mixed connective tissue disease(MCTD). Methods An enzyme?linked immunosorbent assay was adopted to measure LOXL?2 in the serum of 20 patients with active SSc,20 patients with active MCTD,and 20 healthy controls. The measurements among different groups was com?pared,and correlations between LOXL?2 levels and clinical manifestations of SSc and MCTD were examined. Results The levels of LOXL?2 ex?pression in MCTD and SSc groups were significantly higher than those in control group(P<0.05 for all groups). LOXL?2 expression is also related to the presence of skin lesions in SSc(r=0.982 P=0.001). Conclusion High serum level of LOXL?2 in these patients with active SSc and active MCTD suggests that LOXL?2 may be involved in the process of fibrosis and the resulting vasculitis in multiple organs.
3.Association between four SNPs of eNOS gene and EH in Ningxia Hui population *

Objective To investigate the correlation between 4 SNPs of eNOS gene and EH in Ningxia Hui population ,and to provide theory basis for taking precautions against .Methods Polymerase chain reaction and restriction fragment length polymor-phism(PCR-RFLP) genotyping method were carried out to examine SNPs at rs2070744(T>C) ,rs1799983(G> T) ,rs1800780(A>G) and rs3918181(A>G)sites of eNOS gene ,including 134 EH patients and 115 healthy individuals in Ningxia Hui population .For EH patient-control haplotype analyses ,SHEsis online haplotype analysis software was applied .Results The genotypic frequencies of rs1800780 and rs1799983 had significant difference between patients group and normal group (P<0 .05);The allele frequency of rs1799983 had significant difference between patients group and normal group(P<0 .05);and the frequency of G allele in EH group was less than healthy controls ,which OR value was 3 .851(95% CI:2 .236 -6 .631) .According to the four SNP sites 15 kinds of haplotype were detected ,of which haplotype CGAG ,TTAG ,TGGG ,TTGG ,TTGA in Ningxia Hui healthy people and in patients with EH had a statistics difference(P<0 .05) .haplotype CGAG ,TGGG OR of 0 .352 ,0 .600 ,95% CI less than 1;haplotype TTAG OR 2 .689 ,95% CI greater than 1 .Conclusion haplotype with the emergence of CGAG ,TGGG may reduce the risk of EH Hui na-tionality .haplotype the emergence of TTAG may increase the risk of EH Hui nationality .
4.Evaluation of reliability and validity of Diabetes Diet-Related Quality-of-Life Scale

Objective To evaluate the reliability and validity of Diabetes Diet-Related Quality-of-Life Scale (DDR-QOL) in Chinese version,in order to provide an efficient instrument for assessing the quality of life of diabetes in diet therapy.Methods Translating and edit the DDR-QOL scale initially,275 eligible subjects were invited to the study.DDR-QOL scale and 36 item Short Form Health Survey (SF-36) were used to collect data which were conducted for evaluating the internal consistency,reproducibility,construct validity,convergent and discriminant validity of DDR-QOL.Results The total Cronbach's α was 0.86.The factor analysis suggested good construct validity of DDR-QOL.Significant correlations were found between DDR-QOL and SF-36.The quality of life of diabetes who is on the job or not can be identified by DDR-QOL.Conclusions The Chinese version of DDR-QOL is a valid and reliable tool to assess the quality of life of diabetes who are in diet therapy.
5.Effect of poly (isopropylacrylamide)-based copolymer structure on protein adsorption resistance

BACKGROUND: Non-specific protein adsorption resistance is the most important factor for biocompatibility; pre-adsorption of hydrophilic polymer on artificial material surface is one of the effective methods to inhibit protein adsorption. OBJECTIVE: To study pre-adsorption and protein adsorption resistance of poly (isopropylacrylamide) (PNIPAM)-based amphiphilic comb-block copolymers on polystyrene (PS) surface, and to understand the effect of the copolymer structure, i.e. the chain length and the number of hydrophilic polyvinylpyrrolidone (PVP) or polyethylene oxide (PEO) comb-branches, on protein adsorption resistance. DESIGN, TIME AND SETTING: An observational study was performed at College of Materials Science and Engineering, Beijing University of Chemical Technology from November 2007 to November 2008. MATERIALS: Monodisperse PS microsphere was employed to simulate surface of hydrophobic materials, and lysozyme was used as protein model. METHODS:① Qualitative analysis: Aqueous suspension of PS microspheres (0.1 g/L) was treated with PNIPAM-based copolymer (0.1 g/L) at room temperature for 24 hours to allow pre-adsorption of the copolymer on PS surface to build up a hydrophilic layer. Lysozyme (0.1 g/L) was mixed with the PS suspension at 37 ℃ and the mixture was kept for 24 hours. Apparent particle size and turbidity of the suspension were measured at 37C to observe coagulation or flocculation of PS microspheres, which related to the extent of protein adsorption on PS surface. ②Quantitative analysis of protein adsorption: The PS suspension containing lysozyme was subjected to ultracentrifuge (15 000 r/min) to collect clear aqueous solution. The lysozyme concentration in the clear solution was measured by spectrophotometry at 280 nm. The amount of protein adsorbed on PS surface was calculated based on the decrease in the protein concentration in the supernatant solution. MAIN OUTCOME MEASURES: Apparent particle diameter, turbidity, mass concentration of aqueous phase residual protein. RESULTS: ① Suspension of bare PS microspheres flocculated in the presence of lysozyme at 37℃ due to protein adsorption,which caused significant decrease in the turbidity of the suspension. Bare PS surface could adsorb 25.5 mg lysozyme per square meter.② PS microsphere surface pre-adsorbed by PNIPAM-based amphiphilic comb-block copolymer adsorbed much less protein. No flocculation and only limit coagulation was observed when the suspension of the surface modified PS microspheres was mixed with lysozyme, due to less protein adsorption on the PS surface. CONCLUSIONS: ① Pre-adsorption of water-soluble PNIPAM-based comb-block copolymer on PS microsphere surface can inhibit protein adhesion on the surface. ② The structure of the copolymer strongly affects the performance of protein resistance. A proper amount of hydrophilic branch unit, VP or EO, is required to exhibit good protein resistance of the copolymer. However, excess hydrophilic branches in the copolymer results in worse protein resistance probably due to less stability of the pre-adlayer of the copolymer on PS surface.
6.The idiosyncratic hepatotoxicity of Polygonum multiflorum based on endotoxin model.

The liver injury induced by Polygonum multiflorum Thunb. (PM) was investigated based on idiosyncratic hepatotoxicity model co-treated with lipopolysaccharide (LPS) at a non-hepatotoxic dose. Sprague-Dawley (SD) rats were intragastrically administered with three doses (18.9, 37.8, 75.6 g crude drug per kg body weight) of 50% alcohol extracts of PM alone or co-treated with non-toxic dose of LPS (2.8 mg·kg(-1)) via tail vein injection. The plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were assayed and the isolated livers were evaluated for histopathological changes. The dose-toxicity relationships of single treatment of PM or co-treatment of LPS were investigated comparatively to elucidate the idiosyncratic hepatotoxicity of PM. The results showed that no significant alterations of plasma ALT and AST activities were observed in the groups of solo-administration of LPS (2.8 mg·kg(-1), i.v.) or different dosage (18.9, 37.8 and 75.6 g·kg(-1), i.g.) of PM, compared to normal control group (P > 0.05); while significant elevations were observed in the co-administration groups of PM and LPS. Treatment with LPS alone caused slight infiltration of inflammatory cells in portal area but no evident hepatocytes injury. Co-treatment with LPS and PM (75.6 g·kg(-1), i.g.) caused hepatocyte focal necrosis, loss of central vein intima and a large number of inflammatory cell infiltration in portal areas. When further reduce the dosage of PM, significant increases of plasma ALT and AST activities (P < 0.05) were still observed in co-administration groups of LPS and PM (1.08 or 2.16 g·kg(-1)), but not in LPS or PM solo-administration groups. Nevertheless, the co-treatment of low dosage of PM (0.54 g·kg(-1)) with LPS did not induce any alteration of plasma ALT and AST. In conclusion, intragastric administration with 75.6 g·kg(-1) of PM did not induce liver injury in normal rats model; while the 2 folds of clinical equivalent dose of PM (1.08 g·kg(-1)) could result in liver injury in the LPS-based idiosyncratic hepatotoxicity model, which could be used to evaluate the idiosyncratic hepatotoxicity of PM.

Moreover, this study aims to help naval officers and soldiers to take protective measures in training life to reduce the incidence of LBP.

T cells engineered with chimeric antigen receptor (CAR) have been successfully applied to treat advanced refractory B cell malignancy.However,many challenges remain in extending its application toward the treatment of solid tumors.The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CAR-T efficacy.One negative regulator of T cell activity is lymphocyte activation gene-3 (LAG-3).We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture.LAG-3 knockout CAR-T cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model,which is comparable to standard CAR-T cells.Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.
