2.Effect of Electroacupuncture on Learning and Memory Ability for Cerebral Ischemia/Reperfusion Injured Rats

Objective To observe the effect of electroacupuncture at Shenting (DU24) and Baihui (DU20) on learning and memory ability,as well as the finding of histopathology and the expression of mRNA of nuclear factor κB p65 (NF-κB-p65) and its inhibitor (IκB) in the hippocampus of rats with cerebral ischemia/reperfusion (I/R) injury. Methods 45 male Sprague-Dawley rats were were randomly diveded into sham control group (SC, n=15), ischemia control group (IC, n=15) and electroacupuncture group (EA, n=15). The latter 2 groups were modeled as focal cerebral I/R injury, and the EA group received electroacupuncture at Shenting (DU24) and Baihui (DU20) for 14 d. They were evaluated with the step down test, their hippocampus was observed under HE staining and the level of NF-κB-p65 and IκB mRNA was determined with RT-PCR 14 d after modeling. Results The frequence of shock was less, and the latency jumping down from the safe platform was longer in the EA group than in the IC group (P<0.001). The damage of the nerve cell and the expression of NF-κB-p65 mRNA significantly decreased and the expression of IκB mRNA increased in the EA group compared with those of IC group (P<0.001). Conclusion The inhibition of NF-kB-p65 might mediate the recovery of learning and memory impairment after electroacupuncture for cerebral ischemia.
3.Expression of Hepcidin-25 in multiple myeloma and its clinical significance

Objective To study the dynamic changes of Hepcidin-25 in different disease process in patients with multiple myeloma (MM) and its relationship with therapeutic effect and prognosis.Methods The expression levels of Hepcidin-25 in 54 MM patients were analyzed by RT-PCR and ELISA.The correlations between dynamic changes of patients' Hepcidin-25 expression and clinical manifestations,clinical predictive value of the prognosis were researched.Results Expressions of Hepcidin-25 mRNA and protein in MM patients were significantly higher than those in normal control group [0.58±0.19 vs 0.08±0.027,P =0.001,(5.2±11.9) ng/ml vs (22.5±7.1) ng/ml,P =0.019].Hepcidin-25 expression of 17 patients remission after treatment was (26.4±7.3) ng/ml,it was significantly lower than that before treatment [(33.4±7.4) ng/ml,t =5.312,P =0.021].But the Hepcidin-25 level of the treatment ineffective patients (37 cases) significantly increased [(55.9±12.7) ng/ml vs (39.1±9.9) ng/ml,t =2.811,P =0.037].There existed obvious differences in survival curves of two groups (P < 0.01).Hb level of patients remission after treatment was 124 g/L,and significantly higher than that before treatment (102 g/L,t =2.113,P =0.035).It had a negative correlation with Hepcidin-25 (r =-0.535,P =0.002).But the Hepcidin-25 level of the treatment ineffective patients significantly increased (46 g/L vs 73 g/L,t =2.730,P =0.036).It also had a negative correlation with Hepcidin-25 (r =-0.642,P =0.001).Conclusions Hepcidin-25 expression levels in patients with MM are high,and are closely related to anemia of chronic disease.High expression of Hepcidin-25 has a predictive value for clinical effect and prognosis.
4.Clinical efficacy of argon- helium knife cryotherapy followed by chemotherapy for advanced non- small cell lung cancer

Objective To explore the short- term efficacy of argon- helium knife cryotherapy followed by chemotherapy in treating advanced non- small cell lung cancer (NSCLC) and to investigate its effect on the long- term survival. Methods During the period from March 2005 to March 2008, a total of 61 patients withⅢb or Ⅳ stage NSCLC received argon- helium knife cryotherapy followed by chemotherapy (study group), and other 52 patients with Ⅲb or Ⅳ stage NSCLC were treated with chemotherapy only (control group). The clinical data were retrospectively analyzed. The pain was evaluated by numeric rating scale (NRS) and the quality of life (QOL) was assessed by functional assessment of cancer therapy- general (FACT- G) scale. The clinical effect was evaluated according to RECIST criteria for solid tumor, and the patient’s survival time was recorded. Results (1) Twenty- six patients had local pain before the treatment, and the pain was relieved in different degrees after cryotherapy. The QOL, including all respects of FACT - G, was significantly improved after cryotherapy in all 61 patients. (2) The remission rate of the study group and the control group was 34.4% and 15.4% respectively, the difference between the two groups was statistically significant (P <0.05). (3) The median survival time of the study group and the control group was 12.9 months and 9.5 months respectively, and the one- year survival rate of the study group and the control group was 53.6% and 35.4% respectively. The differences between the two groups were statistically significant (P < 0.01). Conclusion Argon- helium knife cryotherapy is a safe and effective local treatment for advanced NSCLC, which can quickly reduce the tumor load, relieve the pain and improve patient’s quality of life. Cryotherapy with subsequent chemotherapy is superior to simple chemotherapy in improving the patient’s survival rate.
5.Evaluation of clinical characteristics of genitourinary syndrome in postmenopausal women

One hundred and ten postmenopausal women,complaining vaginal discomfort,recurrent urinary tract infections or painful intercourse and visiting the hospital between March 2014 and February 2015 were enrolled in the study.Among them,58 patients received vaginal administration of estriol estriol ointment (group A) and 52 patients received conventional treatment (group B).There were no significant differences in mean age of menopause and mean menopause duration between two groups.The clinical symptoms,vaginal health scores,pelvic organ prolapse were observed after treatment.After 6-month treatment,the female vaginal health score was significantly improved in women group A,the rate of urinary orifice was reduced (78% vs.7%),and the rate of pelvic organ prolapse was decreased (36% vs.21%).The geritourinary syndrome of menopause is a new term for vulvovaginal atrophy with clinical characteristics of multi-system changes in postmenopausal women,including atrophic vaginitis,female urogenital diseases,and pelvic organ prolapse.The new diagnostic criteria are needed for evaluation and management of geritourinary syndrome.

The therapeutic effects of intensive insulin therapy in treatment of traumatic shock combined with multiple organ dysfunction syndrome (MODS) were investigated. A total of 114 patients with traumatic shock combined with MODS were randomly divided into two groups: control group (n=56) treated with conventional therapy, and intensive insulin therapy group (n=58) treated with conventional therapy plus continuous insulin pumping to control the blood glucose level at range of 4.4-6.1 mmol/L. White blood cells (WBC) counts, prothrombin time (PT), serum creatinine (SCr), alanine aminotransferase (ALT), serum albumin and PaO(2) were measured before and at the day 1, 3, 5, 7 and 14 after treatment. The incidence of gastrointestinal dysfunction, the incidence of MODS, hospital stay and the mortality were also observed and compared. After intensive insulin therapy, the WBC counts, SCr, ALT and PT were significantly reduced (P<0.05), but the level of serum albumin was significantly increased (P<0.05) at the day 3, 5, 7 and 14. In the meantime, the PaO2 was significantly elevated at the day 3, 5 and 7 (P<0.01) after intensive insulin therapy. The incidence of gastrointestinal dysfunction, the incidence of MODS, the length of hospital stay and the mortality were markedly decreased (P<0.01). The results suggest early treatment with intensive insulin therapy is effective for traumatic shock combined with MODS and can decrease the length of hospital stay and the mortality.
7.Study on the Extraction Technology of Phenolic Acids from Anemone altica

OBJECTIVE:To determine the content of phenolic acids from Anemone altica,and optimize its extraction technolo-gy. METHODS:HPLC was used to determine the contents of mono-ferulyl tartaric acid and ferulic acid from A. altica;using the total contents of 2 index components as index,volume fraction of extraction solvent,extraction solvent volume,extraction times and extraction time as factors,orthogonal test was used to optimize extraction technology,and verification test was conducted. RE-SULTS:The contents of mono-ferulyl tartaric acid and ferulic acid were 0.059%,0.0025%,respectively;the optimal extraction technology was as follow as 30% ethanol 600 mL added to 20 g medicinal material,extracted twice,90 min every time. In verifi-cation test,the average contents of 2 components in extract were 0.2971%(RSD=3.64%,n=3),0.0041%(RSD=5.11%,n=3). CONCLUSIONS:A method for contents determination of mono-ferulyl tartaric acid and ferulic acid from A. altica is estab-lished;optimized extraction technology is stable and feasible.
8.Construction and identification of FLT3-ITD/FIV expression vector and acute myelogenous leukemia cell lines

The aim of this study was to construct the lentivirus expression vector of human FLT3-ITD mutation and to screen the AML cells for stable expression of FLT3-ITD.We screened the AML patient with FLT3-ITD mutation by peripheral blood DNA extraction and PCR,and then the ORF of FLT3-ITD was constructed using the method of homologous recombination,which used the BamH Ⅰ and Not Ⅰ double enzyme digestion of the ORF and the FIV vector.Results were confirmed by restriction enzyme identification and PCR.The constructed recombinant vector was co-transfected with VSVG and g/p into HEK293T cells to produce the lentiviral parti cles by transfection reagent.The lentiviral particles were transduced into K562 cells and the infection efficiency was measured by fluorescence microscope.The cells were sorted by the Flow Cytometer.The expression of FLT3 in the stable cell lines was detected by Western blot.There were 42 patients with FLT3-ITD mutation among the 207 patients we collected.We successfully amplified the ORF of FLT3 of the patient with highest mutant/wt ration.Then constructed the lentivirus expression vector FLT3-ITD/FIV and overexpressed it in K562 cell lines.We screened the cells expressed FLT3-ITD/FIV by fluorescence activated cell sorting,and Western Blot results proved the expression of FLT3-ITD.The over-expression 1 entiviral vector of human FLT3-ITD/FIV has been successfully constructed and cell lines for stable expression of FLT3-ITD have been successfully established.This will shed a light on the future study of FLT3 function in AML and provide a new in vitro model for AML.
9.The role and mechanism of SDF-1/CXCR4 signaling pathway in rat model of chronic renal allograft rejection

Objective To investigate the role and mechanism of SDF-1/CXCR4 in the development of chronic rejection (CR) in rat models.Methods CR rat models were established using Fisher 344 to Lewis rats.In the blank control group (n=10),Lewis rats getting isotransplantation were treated with Cyclosporine A.CR rat models were established in positive group (n=10) and the rats were treated with Cyclosporine A.CR rat models were also established in CXCR4 antagonism group (n=10) and the rats were treated with both Cyclosporine A and AMD3100 (1 mg/kg).The serum creatinine levels were monitored every week.Kidney grafts were harvested 12 weeks after transplantation for histological analysis.We evaluated graft injuries using chronic allograft damage index (CADI) scores.Q-PCR and Western blotting were used to measure CXCR4,TGF-β1/Smad3 signaling pathway and α-smooth muscle actin (α-SMA) expression in renal allograft tissues.Results The serum creatinine levels in blank control group and CXCR4 antagonism group were significantly lower than those in positive control group (P<0.05).The blank control group and CXCR4 antagonism group presented milder pathological manifestations of CR.The CADI score in CXCR4 antagonism group was 3.54,which was lower than that of positive control group (P<0.05).The expression of biological markers in TGF-β1/Smad3 signaling pathway and SDF-1/CXCR4 signaling pathway was significantly lower in blank control group and CXCR4 antagonism group than in positive control group (P<0.05).Conclusion SDF-1/CXCR4 signaling pathway may play a crucial role in the development of CR.The usage of SDF-1/CXCR4 antagonist can protect renal allograft by inhibiting the TGF-β1/Smad3 pathway.Therefore,antagonism of CXCR4 may provide a novel way to prevent the development of CR.
