Selenium, iodine and zinc are the essential trace elements for human body and the important components of many kinds of hormones, proteins and enzymes. The three elements can play important biological roles by affecting the hormones, proteins and enzymes in human body, one of the three elements can influence the biological effects of the other two. For preventing and curing some deficiency diseases and supernutrition, to study the correlated biological effect of the three trace elements will demonstrate the important theory significance and practical value.
2.Comparison of two kinds of HLAˉB27 detection method in diagnosis of ankylosing spondilitis

Objective To investigate the clinical application value of fluorescence PCR in vitro diagnosis(IVD)reagent kits in the HLA-B27 detection by comparing 2 kinds of HLA-B27 IVD reagent kit approved by CFDA.Methods A total of 573 clinical blood samples were collected and detected for HLA-B27 by the approved reagent kits based on the fluorescence PCR technique and the flow cytometry.The samples with inconsistent testing results by the two kits were further confirmed by the PCR sequencing.At the same time,about 5% samples of the positive results detected by the fluorescence PCR method were extracted for conducting the re-testing.Results Among 573 samples,191 samples were HLA-B27 positive and 382 cases were HLA-B27 negative by flow cytome-try;the same samples had 194 cases of HLA-B27 positive and 379 cases of HLA-B27 negative by real-time PCR.With flow cytome-try as reference of the final results,the positive coincidence rate of the two kinds of kit was 96.33%(184/191),the negative coinci-dence rate was 94.76%(362/382),27 samples had inconsistent results from the two kinds of assay(accounting for 4.71% of the to-tal number of samples),the total coincidence rate was 95.29% [(184+362)/573],the Kappa value was 0.896(P =0.02);the chi-square test P =0.021,the two kinds of testing method had the high consistency,but the differences existed in the testing results. The re-testing results by PCR sequencing(including 27 samples with inconsistent results by two kinds of kit)were entirely consist-ent with the fluorescence PCR testing results.Conclusion Compared with the authority method flow cytometry for HLA-B27 tes-ting in clinic,the fluorescence PCR kit may present more accurate judging ability for the HLA-B27 testing on the basis of ensuring the higher consistency of the testing results,is easier compared with the sample preparation and operating procedures,and has the stronger clinical application value and prospects s.
3.Effects of extracorporeal shock wave therapy on interleukin-1beta and matrix metalloproteinase-13 expression in rabbits with knee osteoarthritis

BACKGROUND:Interleukin-1βand matrix metal oproteinase-13 can promote the metabolism of chondrocytes, inhibit the capacity of synthesizing and repairing, induce the degradation of extracellular matrix, and play a crucial role in the occurrence of osteoarthritis.
OBJECTIVE:To observe the effects of extracorporeal shock wave therapy on the interleukin-1βand matrix metal oproteinase-13 expression in rabbits with experimental knee osteoarthritis.
METHODS:Thirty New Zealand rabbits were randomly and equal y divided into treatment group, model group and control group, with 10 rabbits in each group. Model of knee osteoarthritis was established in both the treatment group and model group, using modified plaster cast in extension position for 6 weeks. Then the rabbits of treatment group were treated with extracorporeal shock wave therapy, each 1 000 impulse, at the energy flux density of 0.1 mJ/mm2. There were no treatments in the control group. The rabbits in each group were sacrificed at 4 weeks after treatment, the knee synovial fluid and articular cartilage were col ected from the rabbits. The pathological changes of knee joint were detected using hematoxylin-eosin staining and toluidine blue staining. The interleukin-1βand matrix metal oproteinase-13 expression in the synovia was detected using ELISA and immunohistochemical staining respectively.
RESULTS AND CONCLUSION:The interleukin-1βconcentration in the synovial fluid was significantly higher in the treatment group and model group than the control group (P<0.01), and the treatment group after treatment showed a lower concentration than the model group (P<0.05). Mankin scores in treatment group and model group were significantly increased compared with the control group (P<0.01), and the treatment group after treatment showed a lower score than the model group (P<0.05). The interleukin-1βand matrix metal oproteinase-13 positive expression rates in the treatment group and model group were significantly increased compared with the control group (P<0.01), and the treatment group after treatment showed a lower rate than the model group (P<0.05). The extracorporeal shock wave therapy can downregulate the expression of interleukin-1βand matrix metal oproteinase-13, promote the synthesis of new col agen.
4.Effects of Selenium on Endothelial Cell Growth Cultured in vitro

Objective To research the growth of human umbilical vein endothelial cell(HUECV) cultured with different dose of selenium. Methods HUVEC were cultured with selenium in various concentration (50、100、500、1000、2000 nmol/L) for 48 h in vitro. The morphological changes of cells were observed, activity of cell was measured by MTT method, and at the same time the levels of SOD, GSH-Px, MDA, NO, iNOS, eNOS in the culture were determined. Results At the doses of 1 000-2 000 nmol/L, the cells were damaged and revealed related morphological change. 5.Ureaplasma urealyticum infection induced the production of calculus in urinary and reproductive system of male rats

Objective:To investigate affection of ureaplasma urealyticum (UU) infection on the production of calculus of the urinary and reproductive system in male rat. Methods:Sprogue-dawley(SD) rats were infected with UU4 (serotype 4) through repeated natural sexual intercourses for 8 weeks. The urinary and reproductive system were detected . Results:Twelve point five percents of rate infected with UU had soft calculus in urinary tracts, while 27.5 percents of rate infected with UU had soft or hard calculus. Conclusion: Infection with UU may lead to the production of calculus in urinary and reproductive system in male rats
6.Effect of Extracorporeal Shock Wave on Expression of Interleukin-1β and Tumour Necrosis Factor-α in Cartilage in Rabbits with Knee Osteoarthritis

Objective To explore the effect of extracorporeal shock wave (ESW) on expression of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in cartilage in rabbits with knee osteoarthritis. Methods 30 rabbits were randomly divided into control group, model group and treatment group, with 10 rabbits in each group. Model of knee osteoarthritis was established with modified plaster cast in extension position for 6 weeks except the control group, and the treatment group was treated with ESW (each 1000 impulse, the energy flux density of 0.1 mJ/mm2). The rabbits were sacrificed respectively 4 weeks after ESW treatment, the general and histological changes of articular cartilage were examined and immunohistochemical staining was used to observe the expression of IL-1β and TNF-α in cartilage. Results The expression of IL-1β and TNF-α were significantly lower in the treatment group than in the model group (P<0.01). Conclusion ESW can down-regulate the expression of IL-1β and TNF-α in chondrocytes with osteoarthritis.

Ubiquitination on target proteins is the signal of cellular protein degradation.Ubiquitin ligase E3 is one of the key enzymes in ubiquitination,it recognizes a specific substrate protein and recruits an ubiquitin conjugating enzyme E2,mediating the ubquitin transfer from the E2 to the substrate protein.Ubiquitin ligase E3 can be divided into two subfamilies according to their different structure characters and function mechanisms,the HECT(homologous to E6AP C terminus) family and the RING-finger family.Members of the HECT E3 share the common HECT catalytic domain,which can bind to an E2 and load the ubiquitin on themselves before catalyzing the transfer of ubiquitin to the target proteins.While the RING-finger E3 all contain an similar E2 binding domain and a unique substrate binding part,mediating direct ubiquitin transfer from the E2 to the substrate.The most recent progresses in the stuctural and functional studies of these two E3 famlies were summarized.

9.Research on the antitumor actions of extracts from the fruiting body of coriolus versicolor

Objective: To investigate the antitumor actions of polysaccharide extracts from the fruiting body of coriolus versicolor (CVE). Methods:Hepatoma HepA cells were injected into mice subcutaneously. Different doses of CVE were given by gavage. On the 7 th and 14 th day, tumor inhibitive rates were calculated. ELISA was performed to measure the serum IgG level; MTT was used to examine CVE′s effects on the proliferation of T lymphocytes of thymus. Immunohistochemistry was used to determine CVE′s influence on the expression of tumor related genes P53 and VEGF in liver. Results: CVE may evidently inhibit the growth of the transplanted HepA tumors. Its effects on the serum IgG level and on the proliferation of T lymphocytes of thymus were also significantly. Also, CVE markedly decreased the expression of P53, VEGF genes in liver. Conclustion: CVE had significant antitumor effects in vivo . The mechanisms may involve immune modulation effects and antimetastasis actions.
10.Clinic significance of serum Cystain C level in patients with chronic heart failure

Objective To explore serum Cystain C (Cys C) level in chronic heart failure (CHF) patients and its relation?ship with cardiac function and ventricular remodeling. Methods Patients with heart failure (n=75) and healthy adult (n=35) were enrolled in this study. According to the NYHA classification,CHF patients is divided into NYHA Ⅱ, NYHAⅢand NYHAⅣ;CHF patients can also be divided into 2 groups base on their level of serum Cys C:Cys C>0.95 mg/L is defined as elevated serum Cys C group, serum Cys C≤0.95 mg/L is defined as normal serum Cys C group;According to left ventricu?lar ejection fraction (LVEF), CHF patients can also be divided into 2 groups:LVEF≥0.50 is defined as ejection fraction re?served group, LVEF<0.50 is defined as the ejection fraction reduced group. Cys C, N terminal B-type natriuretic peptide (NT-proBNP), serum creatinine(Scr),UREA and Echocardiography were analyzed in all groups and their correlation was studied. Results Serum Cys C level in CHF patients was obviously higher than that in healthy controls, and it increased with NYHA heart function classification;compared with normal Cys C group,Cys C elevated group presented higher NT-proBNP level but lower eGFR and LVEF (P<0.05);compared with ejection fraction reduced group, ejection fraction reserved group demonstrated lower NT-proBNP, LVEDD, LVMI and Cys C levels (all P<0.05);Correlation analysis revealed that Cys C is correlated with age, NT-proBNP, LVEF, LVEDD, Scr and UREA (r=0.411, 0.658, 0.465, 0.310, 0.552, 0.486, P<0.01). Conclusion Serum Cys C can be used to evaluate cardiac function of heart failure, and is associated with ventricular remodeling.
