Objective To investigate the morphologic characteristics of neuron-like cells induced from the rat mesenchymal stem cells(MSCs). Methods The rat MSCs from bone marrow were cultured by being adhered to the culture dish.The thirdgeneration of the MSCs were induced by the neonate rat brain homogenate supernatant for 48 hours.The morphological characteristics and the ultrastructures of both non-induced and induced MSCs were observed under the inverted microscope and the electron microscope.The property of the induced cells was identified by using immunocytochemical method. Results Under the inverted microscope MSCs showed spindle or polygon-shaped cell bodies and the nuclei with one or two nucleoli were located in the middle of the cells.After the inducement the cells appeared neuron-like with axonlike and dendrite-like processes.The neuron-like cells were neuron special endolase(NSE),neurofilament protein(NF) positive and glial filbrillary acidic protein(GFAP) negative by irnmunocytochemical staining.Under the electron microscope the MSCs had plentiful cytoplasm and organelles and had an obvious nuclei containing a nucleoli.There were a lot of microvilli on the surfaces of the MSCs.The neuron-like cells had plentiful cytoplasm and irregular nuclei with one to three nucleoli.There were also plenty of microvilli on the cell surfaces.Conclusion MSCs are one kind of multipotent stem cells and can differentiate neurons with matured organelles induced by the neonate rat brain homogenate supernatant.

Objective To evaluate the role and mechanism of miR-150 in cardiac fibrosis after MI.Methods A rat model of MI was established by up-regulating miR-150 through overexpressing miR-150 lentivirus.Real-time PCR and Western blot were applied in detecting the expression of collagen 1 α 1 and α-SMA protein in infarction area border.Masson coloration was applied in measuring fibrosis.Cardiac fibroblasts were isolated and cultured.UTR was used to report the carrier and lentivirus.And c-Myb siRNA was used to verify the relationship between c-Myb and microRNA-150.Results In vivo,MiR-150 was down-regulated in myocardium border zone in 14 day and 28 day after infarction (P ＜ 0.001,P ＜ 0.05),and overexpressing miR-150 promoted myocardial fibrosis (P ＜ 0.001),and inhibited the expression of collagen1α 1 and α-SMA (P ＜ 0.01,P ＜ 0.05).In vitro,c-Myb was the direct target gene of miR-150,and inhibited the expression of c-Myb resulting in the down regulation of collagen1α 1 and α-SMA,suggesting that the role of miR-150 was achieved by regulating c-Myb.Conclusions MiR-150 was down-regulated in myocardium border zone,and myocardial fibrosis can be improved by targeting c-Myb.

Objective To investigate the association between diet during pregnancy and hypertensive dis-order complicating pregnancy ( HDCP ) , so as to provide a theoretical basis guiding appropriate diet during pregnancy.Methods Using 1∶2 matched case-control study method, we selected pregnant women delivered in Anhui Women and Child Health Care Hospital from January to December 2014, and interviewed them with food frequency questionnaire.The relationship between intake frequency of various food and HDCP was analyzed. Results A total of 543 women were included in this study, including 181 cases and 362 controls.Mann-Whit-ney U test results showed that the frequencies of meat and fish, eggs, beans, nuts, milk, and pickled food con-sumption were significantly different between the cases and the controls (all P<0.05);while the differences in cereals, vegetables, and fruits consumption frequency between cases and controls were not statistically signifi-cant (all P>0.05).Multivariate Logistic regression analysis showed that higher intake frequencies of beans (OR=0.746, 95%CI:0.645-0.862), eggs (OR=0.789, 95%CI:0.693-0.898), and milk (OR=0.822, 95% CI:0.725-0.931) were associated with lower risk of HDCP; in contrast, higher intake fre-quency of pickled food was associated with higher risk of HDCP (OR=1.190, 95% CI:1.054-1.344). Conclusions Beans, eggs, and milk may be protective factors for HDCP, while pickled food may be risk fac-tor of HDCP.Diet during pregnancy should be appropriate and following scientific guidelines.

OBJECTIVE:To provide reference for clarifying the pharmacodynamic material basis of anti-inflammation effect of Uygur medicine Hyssopus cuspidatus. METHODS:HPLC was conducted to establish the fingerprint of 8 extracts with different po-larities. Using macrophage RAW264.7 as object,the inhibitory effect of extracts with different polarities on inflammatory factor ni-tric oxide(NO),TNF-α and IL-6 in supernatant of cell culture medium was detected,the differences of anti-inflammatory activity in vitro were compared. Grey correlation analysis method was used to analyze the spectrum-effect relationship of peak area of common peaks and anti-inflammation activity. RESULTS:The fingerprint of 8 extracts with different polarities showed obvious differences. Anti-inflammation in vitro results suggested that 30% ethanol extract had the strongest inhibitory effect on inflammatory cytokines in vitro. There were 14 common peaks in the established HPLC fingerprints,the 5 common peaks that was closely related to the in-hibitory effect of inflammatory factors in vitro were peak 8,6,5,10,13,respectively;peak 8 was rosmarinic acid. CONCLUSIONS:The established fingerprint and its spectrum-effect relationship with anti-inflammation activity in vitro can provide certain reference for its pharmacodynanic material basis study.

Background and purpose: Plumbagin is the main active components of traditional Chinese medicine of plumbago zeylanica. The present studies show that plumbagin has a killing effect on tumor cells. This study aimed to investigate the function and primary mechanism of plumbagin on invasion and metastasis of human liver cancer SK-hep-1 cells. Methods:With the treatment of plumbagin in vitro, cell proliferation and adhesion of SK-hep-1 cells were detected by MTS staining, cell cycle of SK-hep-1 cells were detected by lfow cytometry, the self-renewal and propagation abilities of SK-Hep-1 cells were conducted by colony formation assay , invasion in cells were performed using transwell invasion assay, and the p21 and MMP-2/9 mRNA levels were detected by real-time RT-PCR. Results:With the treatment of plumbagin, SK-Hep-1 cells proliferation was decreased with plumbagin concentration-dependency and the IC50 value of plumbagin in SK-Hep-1 cells was 22.04 mmol/L. The colony formation ability of SK-Hep-1 cells was decreased and the percentage of cells in G0/G1 phase was increased in a dose-dependent manner, as compared to control. The cell adhesion and invasion abilities were decreased. The real-time RT-PCR showed that p21 mRNA expression was increased and the MMP-2/9 mRNA was decreased. Conclusion:Plumbagin could suppress the proliferation and invasiveness of human liver cancer SK-hep-1 cells in vitro, and these effects may be by up-regulation of p21 and down-regulation of MMP-2 and MMP-9.

Objective To determine the transfection of PTEN gene in human ovarian carcinoma SKOV3 cells by ultrasound mediating and contrast agent′s enhancing, and to explore the mechanism of PTEN gene inhibiting the tumor cells′ invasion. Methods After addition of PTEN gene and 5 ?l lipofectamine,human ovarian carcinoma SKOV3 cells on 6-well plates were explored to ultrasound for 60 s in the presence(200 ?l) of ultrasound agent. The invasiveness of transfected cells was measured quantitatively by Matrigel invasion assays(Transwell chamber) after 48 h. Results The invasiveness of SKOV3 cells transfected by ultrasound-mediated PTEN gene was significantly declined compared with that of the controls. Conclusions The transfection of PTEN gene into SKOV3 cell can inhibit their invasion.The gene transfection can be enhanced by ultrasound and contrast agent,which may be expected to be a new means of clinical gene therapy.

Objective To evaluate the open-set auditory speech perception abilities, to characterize the auditory speech development of children with cochlear implants.Methods 27 congenitally deafened children with cochlear implants were evaluated with Mandarin monosyllable and disyllable lexical neighborhood test(M-LNT-Monosyllable & M-LNT-Disyllable) and Mandarin hearing in noise test for children(MHINT-C) according to the order from M-MLNT easy test,M-MLNT hard test, M-LNT easy test ,M-LNT hard test, MHINT-C quiet condition, MHINT-C noise front condition, MHINT-C noise non-implant side condition to MHINT-C noise implant side condition in sound field.Results 27 CI children have completed M-LNT/M-MLNT, 9 of them could be tested in quiet condition with MHINT, and 7 could be tested in noise with MHINT. There were significant differences between scores of easy and hard words lists of M-LNT and M-MLNT(P

Objective To investigate the ability of decision-making under risk condition in benign epilepsy children with centrotemporal spikes (BECT).Method Seventy-eight BECT patients and seventyfive healthycontrols were studied by Iowa Gambling Task (IGT) with inexplicit probability and Game of Dice Test (GDT) with explicit probability.Results (1) There was a significant difference in IGT task scores between the two group (t=-2.56,P＜0.05).The total number of advantageous choices in IGT were lower in BECT group (49.35±5.38) than that in healthy control group (52.24±6.23).BECT group (-1.41 ± 10.60) performed worse on task than healthy control group (4.43±11.88),and the difference was statistically significant (t=-3.21,P＜0.05).The control group scored higher on Blockl,Block4 and Block5 than that of BECT group (P＜0.05).(2) There was no significant difference between the BECT group (4.42±7.74) and the control group(5.49±6.81) in the GDT test scores (t=-0.91,P=0.37).(3) Pearson correlation analysis showed that there was no significant correlation between the age,age of onset,education,seizure frequency and onset duration and the score of IGT and GDT (r =-0.091-0.274,all P ＞ 0.05).Conclusion There is no significant decision-making disability in children with BECT under the condition of certain risk probability,while there is significant impairment under the condition of uncertain risk probability.The decision-making disability may be associated with impaired frontal lobe function.
