1.The clinical effect of Pudilan combined with ribavirin in treatment of children with HFMD

Objective To investigate the effect of Pudilan combined with ribavirin in treatment of the hand-foot-mouth disease(HFMD). Methods 70 patients were randomly divided into treatment group ( Pudilan combined with ribavirin) and control group (ribavirin only). Results Compared with the control group,the cure rate and the effective rate in treatment group had statistically significant difference ( all P < 0.05 ). Compared with the control group,the rash disappeared time and the fever subsiding time in treatment group were shorter obviously ( all P <0. 05 ). The both groups had no obvious side effect. Conclusion Pudilan combined with ribavirin in the treatment of HFMD had a good therapeutic effect and were a safe and effective method.
3.Establishment of an in-vitro root canal-apical complex model

Objective To establish an in-vitro root canal-apical complex model for studying the mechanisms of the infected root canal to apical periodontitis and periapical biofilm .Methods Single rooted premolar extracted for orthodontic ,was sealed in sterile vial containing LB solid medium ,and the culture medium covered apical thirds .Totally 25 root canal-apical complex models were prepared .Five models were randomly selected for the bacteria detection in periapical by PCR at 1st day .The remained 20 models were randomly subjected to a control group(n=10) and experimental group(n=10) .Extracted teeth were opened in experimental group and control group with no treatment .All models were exposed in air .At 21st day ,bacteria were detected through PCR in root canal and apical;endotoxin content in apical was assayed by chromogenic end-point limulus test .Results In apical ,bacteria was not found in all groups ,but not for the experimental group .The mean endotoxin content was (8 .913 ± 0 .614)EU/mL in control group and (10 .525 ± 0 .981)EU/mL in experimental group .The endotoxin content was increased significantly in experimental group ,when compared with control group(P<0 .01) .Conclusion Root canal-apical complex was established in vitro through this method .Bac-teria was not easy to reach the apical when the infected root canal was not disturbed .Bacteria in the infected root canal caused apical periodontitis through the secretion of virulence factors such as endotoxin .

5.Ultrasound-mediated microbubble destruction enhances GFP gene expression in human gingival fibroblasts cells in vitro

Objective To investigate whether ultrasound-mediated microbubble destruction can effectively deliver pEGFP-N1 plasmid into human gingival fibroblasts (HGFs).Methods The primary cultured HGFs were divided into 4 groups,that is,plasmid,microbubble+plasmid,ultrasound+plasmid,and ultrasound+microbubble+plasmid groups.pEGFP-N1 plasmid was used to transfect to HGFs as a gene marker with ultrasound-mediated microbubble destruction.The last group was further divided into subgroups to optimize the transfection conditions.After 48 h,phase-contrast fluorescent microscopy was employed to evaluate the expression of green fluorescent protein (GFP).The cell vitality was measured by the MTT assay.Results The transfection efficiency of the ultrasound+microbubble+plasmid group was higher than other experiment groups.Optimal gene expression was found when ultrasound was radiated at 1.5 W/cm2 for 60 s,microbubble was at a concentration of 10% and plasmid was at a concentration of 6.67 ?g/ml,and the transfection efficiency was highest under this condition.Conclusion Under specific conditions,ultrasound mediated microbubble destruction enhances the reporter gene transfection and expression in HGFs.
6.Clinical validation of detection results of three different blood routine analyzers

Objective To evaluate whether the comparability of 3 automatic blood cell analyzers meet the clinical requirements by conducting the comparative study on the detection results of these instruments.Methods With the Sysmex 2100 automatic blood cell analyzer as the reference instrument,Sysmex 1000i and Abbott 1800 as the experimental instrument,the original quality control provided by the instrument factory and the patient′s fresh anticoagulant blood samples in the laboratory were adopted to monitor for continuous 40 d by these three instruments and the detection results of WBC,RBC,HGB,HCT and PLT were analyzed.Results The detection results of these 3 instruments were statistically tested by the F test,the differences showed no statistical significance (P >0.05)and the bias was in 1/2 of the maximum permissible error range in America department clinical test revised regulations (CLIA′88).Conclusion The detection results by these 3 instruments are comparable and can meet the clinical requirements.
8.Mechanical and biodegradable properties of absorbable hydroxyapatite/poly-DL-lactide screws

BACKGROUND:Through compounding hydroxyapatite and poly-DL-lactide, the mechanical properties, physical and chemical properties of the implants can be enhanced.
OBJECTIVE:To detect the effect of hydroxyapatite/poly-DL-lactide composite internal fixation screws in canine femoral condyle cancel ous bone fracture repair.
METHODS:Forty-two beagle dogs were operated to bilateral femoral condyle fracture models. The left side was fixed using hydroxyapatite/poly-DL-lactide screws as experimental group;while the right side was fixed with pure poly-DL-lactide screws as control group. After 2, 4, 8, 12, 24, 36 and 48 weeks, conditions of fracture were observed using X-ray, femoral and screw specimens were observed histopathological y, and the bending strength and the average molecular weight were detected. The biological absorption rate, intensity decay rate and biodegradation rate were calculated.
RESULTS AND CONCLUSION:After 2-48 weeks, bilateral fractures were fixed wel and new bone grew wel , but the biological absorption rate of the screws in the experimental group was lower than that in the control group (P ≤0.01). In the first 2, 4 weeks, the bending strength of the experimental screws was higher than that of the control screw (P ≤0.05), but the biodegradable speed was slower in the former one (P ≤ 0.05 or P ≤ 0.01). The pathological changes were similar in the two groups. After 48 weeks, the fractures were healed and bone tissue reconstruction was completed. Compared with the pure poly-DL-lactide screws, the hydroxyapatite/poly-DL-lactide composite internal fixation screws have better fixation effects, biocompatibility, mechanical properties and biodegradability.

Objective To investigate the therapeutic effects of hepatic resection plus Roux-en-Y choledochojejunostomy in combination with reflux prevention on intrahepatie bile duct stone. Methods The clinical data of 230 patients with intrahepaticbile duet stone surgically treated in our hospital in the past 18 years were retrospectively analyzed. Of the 230 patiets, 110 underwent hepatectomy plus Roux-en-Y choledochojejunostgmy in combination with reflux prevention and the others received other surgeries. The complications of pleural effusions, infection of incisional wound, leakage of bile and retained calculus after operation were studied. Meanwhile, the recurrent rate of stone and long-term effects were determined. Results The occurrinjg rate of pleural effusion, infection of incision wound,leakage of bile was not significantly different among different groups. However, the retained calculus and recurrent rate of stone in the group of patients underwent hepateetomy plus Roux-en-Y choledochojejunostomy in combination with reflux prevention were significantly decreased as compared with other group (P<0.05). So was the effect in long-term (P<0.05). Conclusion Hepatectomy plus Roux-en-Y choledochojejunostomy in combination with reflux prevention could have better effect on patients with intrahepatic bile duct stone.
