1.Regulation of transforming growth factor alpha on glutamate transpoter of retinal M(u)ller cells in mice

Objective To observe the regulation effect of transforming growth factor alpha(TGFα) on expression of glutamate transporter(GLAST)and ingestion activity of retinal M(u)ller cells in mice.Methods To take the retinal tissue of Kunming mouse at postnatal 7~10 day.and then cultured M(u)ller cells according to literature.The 3~4 generation cultured cells of the same primary cell were divided into two groups at random:①TGFα group:maintained in different concentrations of TGFα as 50,75,125 and 150 ng/ml,3 holes in each concentration;②Control group:cultured by Eagle culture medium which improved from Dulbeccon and contained 20%fetal calf serum.The influence of different concentrations TGFα on GLAST activity in M(u)ller cells were observed by L-3H-glutamate uptake detection;the expression of GLAST mRNA in M(u)ller cells was determined by RT-PCR;the expression of GLAST protein was detected with immunocytochemical staining.Results With the increase of TGFα concentration.both L-3H-glutamate uptake and GLAST mRNA expression were increased.The L-3H-glutamate accumulation had got to the maximum uptake at concentration of 125 ng/ml,which was 266% of that in control group,meanwhile,the expressions of GLAST mRNA also got to the maximum as 4 times of control group.Immunocytochemical staining indicated that the effect of 125ng/ml TGFα on expression of GLAST protein was higher than that in the control group,the differences between two groups were statistically significant(P<0.05).Conclusion TGF-α can increase GLAST activity through up-regulating the expression of GLAST mRNA and protein.
2.Treatment of fresh Monteggia fracture with steel-wire loop in adults whose annular ligament needs repairing

Objective To assess the treatment of fresh Monteggia fracture with a steel-wire loop for adults whose annular ligament needs repairing. Methods From May 2006 to March 2008, we treated 15 adult patients with fresh Monteggia fracture whose annular ligament needed repairing with a steel-wire loop. A retrospective study was performed for the 15 cases, including 9 males and 6 females. Their ages ranged from 19 to 46 years. According to the Bado classification system, there were 6 cases of type Ⅰ, 4 type Ⅱ, 5 type Ⅲ. The ulnar fractures were treated with open reduction and compression plate fixation. Next, radial head dislocations were reduced openly and the reduced heads were hitched to the proximal end of the ulna with a steel-wire loop of 0.8 mm in diameter around the radial neck. Last, the broken annular ligaments were repaired. The elbow functional trainings were carried out 48 hours after operation and the steel-wire was removed 3 weeks postoperatively. Results The patients were followed up for 8 to 11 months (mean,9. 5 months). The functional recovery evaluated by Broberg and Morrey evaluation system showed excellent results in 11 patients, good in 2, fair in 2 and poor in 0. The excellent to good rate was 86. 7%.Conclusion The steel-wire loop treatment is a good strategy for adults with fresh Monteggia fracture whose annular ligament needs repairing.
3.Research progress and prognostic significance of EZH2 in acute myeloid leukemia

Objective:To investigate the relationship between DNA methyltransferase (DNMT1) and EZH2 gene expression levels and their clinical significance in patients with acute myeloid leukemia (AML). Methods:The mRNA expression levels of DNMT1 and EZH2 in 50 AML cases and 30 healthy controls were quantified through real-time PCR. The relationship among DNA methyltransferase (DNMT1) and EZH2, clinicopathological factors, and prognosis was analyzed. Results:The mRNA expression of DNMT1 in the AML cases (2.72 ± 0.73) was significantly higher than that in the healthy controls (0.89 ± 0.27) (P<0.01). The mRNA expression of EZH2 in the AML cases (4.39±1.06) was also significantly higher than that in the healthy controls (1.87±0.33) (P<0.01). The expression of DNMT1 was positive-ly associated with that of EZH2 (r=0.51, P=0.002). The expression of DNMT1 was also associated with PB%and WBC≥50 × 109/L (P<0.05). The overall survival of the group with a high-mRNA-expressing DNMT1 was 15 months (95%CI=9-19 months). This period was significantly shorter than that of the group with low-mRNA-expressing DNMT1 (32 months, 95%CI=27-40 months;P=0.006). Conclu-sion:DNMT1 and EZH2 expression levels were downregulated. These levels were associated with poor AML prognosis. The expression of DNMT1 was also positively associated with that of EZH2.
4.Analysis of distribution and drug resistance of pathogens in premature infants with ventilator-associated pneumonia

Objective To investigate the distribution and characteristics of drug resistance of pathogens in premature infants with ventilator‐associated pneumonia(VAP) ,and explore the therapeutic measures for premature infants with VAP ,so as to provide references for clinically reasonable administration of antibacterial agents .Methods A total of 54 cases of premature infants diag‐nosed with VAP in the First Affiliated Hospital of Xiamen University from January 2013 to June 2014 were enrolled in this study . Specimens of respiratory tract secretion were collected ,and species identification of pathogens and drug sensitivity test were per‐formed by VITEK 2 Compact system .Results A total of 69 strains were isolated .Gram negative bacteria was accounted for 94 .20% ,and gram positive bacteria was accounted for 5 .80% .Four of the most common pathogenic bacteria were Klebsiella pneu‐monia (29 strains ,42 .03% ) ,Enterobacter aerogenes (12 strains ,17 .39% ) ,Pseudomonas aeruginosa (10 strains ,14 .49% ) and Stenotrophomonas maltophilia(7 strains ,10 .14% ) .The results of antibiotics‐sensitivity tests showed that these strains were multi‐drug resistant to most commonly used antibiotics ,but sensitive to only a few antibiotics .Conclusion Gram‐negative bacteria are the major pathogens for premature infants with VAP in this hospital .Mixed infection and multidrug resistance are common .Therefore , the rational use of antibiotics according to drug sensitivity tests′results is critical in treating VAP .
5.Suppression of RANKL/OPG pathway activation in FLSs in interfacial membrane through up-regulation of HIF-1α induced by Ti particles

Objective To explore the correlation between HIF-1α expression and RANKL/OPG pathway activation of fibroblast-like synoviocytes(FLSs)induced by Ti particles in aseptic loosening interfacial membrane. Method FLSs were extracted from the synovial tissue collected in surgeries and then co-cultured with Ti particles. QRT-PCR and Western blotting were conducted to measure the mRNA and protein expression of RANKL ,OPG and HIF-1αin FLSs at different concentration and time. Results Genes and protein expression levels of RANKL/OPG and HIF-1α were up-regulated with the increase of concentration of Ti particles. Expression of HIF-1α gene and protein increased time-dependently;the mRNA and protein expression of RANKL/OPG increased firstly and then declined alongside the increase of HIF-1αexpression. Conclusions Ti particles induce the up-regulation of HIF-1α expression and activate RANKL/OPG pathway. Up-regulation of HIF-1α may suppress the activation of RANKL/OPG pathway in FLSs of the interfacial membrane induced by Ti particles.
6.Biomechanical comparison of 4 plate internal fixation methods for simulated fractures of acetabular posterior column

Objective To evaluate the stability of 4 different methods of plate internal fixation for simulated posterior column fracture of the acetabulum.MethodsTwenty preserved cadaveric semipelvic specimens were divided into 4 groups randomly.Models of isolated posterior column fractures of acetabulum were established and then fixed with 1 of following 4 methods of plate internal fixation:2 screws at the each end of plate(group A),3 screws at the each end of plate(group B),1 screw each side in the closest allowable hole to the fracture and 2 screws at the each end of plate(group C),and 4 screws on each side of the fracture(group D).Under vertical compression load of 3 times' body weight,the displacements of the fracture site were measured and the shear rigidity of different internal fixation methods were compared.ResultsThe displacements of the fracture site in Group A,B,C,and D were 4.68?0.35,2.80?0.25,2.12?0.11 and 1.58?0.17 mm respectively,with a decreasing trend from Group A to D.The shear rigidity of internal fixation methods in Group A,B,C,and D were 129.42?9.60,228.91?12.05,285.21?26.16 and 414.71?34.29 N/mm respectively,with an increasing trend from Group A to D.The differences of both displacements and shear rigidity between each 2 groups were significant(P
7.Effect of early rehabilitation on the hip joint function in patients with comminuted posterior wall fractures of the acetabulum after internal fixation

BACKGROUND: Treatment for acetabular fracture becomes a hot point in orthopaedic trauma, but ignorance of rehabilitative treatment that effect therapeutic efficacy is to be seen repeatedly. Normative rehabilitation therapy is one indispensable procedure of acetabular fracture treatment.OBJECTIVE: To investigate the effect of early rehabilitation on the hip joint function in patients with comminuted posterior wall fractures of the acetabulum after internal fixation.METHODS: Forty patients of comminuted posterior wall fractures of the acetabulum at the Department of Orthopedics, Third Hospital of Hebei Medical University, were selected, including 36 males and 4 females, aged 23-61 years, mean aged 35.3 years. All patients were randomly divided into rehabilitation and control group, with 20 patients in each group. All the patients were fixed with reconstruction plate, the patients in the rehabilitation group received early standard rehabilitation exercises intervention and those in control group received usual care. The clinical result was evaluated with the scores of Merle d'Aubigne and Postel.RESULTS AND CONCLUSION: The scores of Merle d'Aubigne and Postel of the rehabilitation group were (13.10±2.05),(14.50±1.32), and (15.80±1.36) points at months 3, 6, and 12 after operation, which were greater than those of the control group (11.20±2.80), (13.00±2.10), and (14.25±1.74) points, respectively (P<0.05). The early rehabilitation exercises may improve the hip joint function for patients of the comminuted posterior wall fractures of the acetabulum after surgery.
8.Model establishment of posterior column fracture of the acetabulum and biomechanical evaluation of the stability between plate and lag screw internal fixation

BACKGROUND:Surgery has become gold standard to treat fracture of acetabulum.The incidence of posterior column fracture of affected acetabulum is high.The appropriate treatment for posterior column fracture of acetabulum remains controversial.OBJECTIVE:To establish the model of posterior column fracture of acetabulum,and evaluate the stability of internal fixation with the plate and lag screw for the posterior column fracture of acetabulum.METHODS:A total of 20 preserved cadaveric semipelvic specimens were randomly divided into two groups.Models of isolated posterior column fracture of acetabulum were established.The models were fixed with plate or lag screw.Under vertical compression load,the horizontal displacements of the fracture site were measured and the shear rigidity of two fixation conditions was compared.RESULTS AND CONCLUSION:The horizontal displacements of the fracture site fixed with plate and lag screw were (1.64±0.17) mm and (1.70±0.20) mm.The shear rigidity fixed with plate and lag screw were (392.40±41.22) N/mm and (386.33±50.07) N/mm.The differences of both horizontal displacements and shear rigidity between the two groups were not significant (P>0.05).The fracture model provides an ideal tool for biomechanical evaluation of the stability of internal fixation for the posterior column fracture of acetabulum.There was no significant difference in the stability of internal fixation with the plate and lag screw for the posterior column fracture of acetabulum.
9.Physicochemical and ecological characteristics of the granular sludge during start-up of Anammox reactor.

The anaerobic granular sludge from an Internal Circulation (IC) reactor of a paper mill wastewater treatment plant were seeded in an Anammox upflow anaerobic sludge blanket reactor. After 185 days operation, the reactor was finally started up by increasing the influent ammonium and nitrite concentrations to 224 mg/L and 255 mg/L, respectively, with volumetric nitrogen removal rate increasing to 3.76 kg/(m3·d). The physicochemical characteristics of the cultivated Anammox granules were observed by scanning electron microscope, transmission electron microscope and Fourier Transform infrared spectroscopy (FTIR). Results suggested that during the start-up course, the granular sludge initially disintegrated and then re-aggregated. FTIR spectra results revealed that the Anammox granular sludge contained abundant functional groups, indicating that it may also possess good adsorption properties. The ecological structure of the granular sludge, analyzed by the metagenomic sequencing methods, suggested that the relative abundance of the dominant bacterial community in the seeding sludge, i.e., Proteobacteria, Firmicutes, Bacteroidetes, significantly reduced, while Planctomycetes which contains anaerobic ammonium oxidation bacteria remarkably increased from 1.59% to 23.24% in the Anammox granules.

10.Clinical analysis of hematologic disorders complicated with herpes zoster after treating with arsenic trioxide

Objective To explore the increasing risk of herpes zoster and its possible mechanisms for hematologic disorders treated with arsenic trioxide (ATO). Methods The cases were divided into study group (with ATO) and control group (without ATO). The incidence rate of herpes zoster was compared between the two groups, and then the average cycles of chemotherapy were compared between the patients complicated with herpes zoster or not in study group. Results The rate of herpes zoster was significantly higher in study group than that in control group (χ2 =4.492, P =0.034). The rates of herpes zoster were 23.95 % (23/96) in study group and 7.89 % (3/38) in control groups. Patients in study group with herpes zoster had received 7.60 cycles and those without herpes zoster 7.72 cycles of chemotherapy on average (Z=0.976, P=0.296).Conclusion The risk of herpes zoster complication in hematologic disorders was increased after ATO treatment which probably activated varicella-zoster virus.
