Objective To determine the association of glutathione-stransferase M1(GSTM1), T1(GSTT1) genetic polymorphisms, smoking and alcohol consumption with the risk of gastric cancer. Methods 1:1 matched case-control study and molecular epidemiology technique were used to analysis the genetic polymorphisms of GSTM1, GSTT1 genotype among 60 cases of gastric cancer patients and sex-and age-matched controls, who were selected from Nanjing Zhongda Hospital and Jiangsu Province Cancer Hospital. Statistical analysis was performed to elucidate interaction of GSTM1. GSTT1 genotype and smoking and alchol consumption in the risk for stomach cancer. 0.05). Conclusion Compared with the Whiteside′s line, the transepicondylar axis seems to be more reliable to determine the rotational alignment of femoral component in total knee arthroplasty. The posterior femoral condylar angle of the Chinese people is larger than that of the Caucasians. If the widely used way of 3? lateral rotational orientation of the femoral component referring to the posterior condylar surfaces of the femur in total knee arthroplasty is adopted, it tends to produce internal malrotation of the femoral prosthesis after operation.

Somatostatin,a cyclopeptide hormone,is generally distributed in vivo.Some studies have indicated that somatostatin can reduce proinflammatory cytokine in hepatic stellate cells and extracellular matrix,and inhibit their proliferation and contraction,promote their apoptosis.It can also regulate the function of Kupffer cells,influence the homing of hepatic stem cells.Somatostain has therapeutic effect on liver fibrosis,especially on the liver fibrosis caused by schistosoma mansoni.
4.Experimental study on the mechanism of nordihydroguaiaretic acid on human prostate cancer PC-3 cells

Objective To investigate the effects of nordihydroguaiaretic acid(NDGA), a general lipoxygenase inhibitor, on the proliferation and apoptosis of human prostate cancer PC-3 ceils and ex-plore the mechanism. Methods The cultured PC-3 ceils were exposured to NDGA at the different concentrations of 0 (control group), 20, 40, 80 and 160 μmol/L, respectively. The effect of NDGA on growth inhibition and apoptosis inducement were measured with morphometry, MTT and TUNEL as-say. The activity of caspase-3 was determinated with flow cytometry. Results The survival rate of PC-3 cell decreased significantly after treated with 40, 80, 160 μmol/L NDGA. The apoptosis indices of PC-3 cell dealt with 0, 20, 40, 80 and 160 μmol/L NDGA for 48 h were (2.9±0.2)%, (3.2±0.3)%, (68.5±0.8)%, (86.2±0.3)% and (86.9±0.6)%, respectively. When PC-3 cells were treated with 0, 20, 40, 80, 160 μmol/L NDGA for 48 h, the positive rates of activated caspase-3 were (3.9±0.0)%, (4.15±0.1)%, (55.5±0.8)%, (75.1±0.3)%, and (76.3±0.7)%, respectively. Compared with the control group, the apoptosis indices and positive rates of activated caspase-3 in the groups of 40, 80, and 160 μmol/L NDGA increased significantly(P<0.05). Conclusions NDGA can inhibit the growth and induce apoptosis of PC-3 cells. The mechanism may be related with intra-cellular activation of caspase-3.

Objective To investigate the value of detection of telomerase activity by fine needle aspiration biopsy (FNAB) of thyroid mass in the preoperative diagnosis of thyroid cancer. 6.Clinical Study on Role of Serum Homocysteine Level in Gastric Cancer and Precancerous Diseases

Background:Recent studies have showed that high homocysteine(Hcy)level can increase the risk of gastric cancer, but no related studies have been reported on role of Hcy in gastric precancerous diseases. Aims:To investigate the role of serum Hcy,folic acid and vitamin B12 in patients with gastric cancer and precancerous diseases. Methods:Eighty-six normal controls,46 atrophic gastritis,46 gastric ulcer,31 gastric polyp,52 gastric cancer patients diagnosed by gastroscopy and pathology were enrolled. Serum levels of Hcy,folic acid and vitamin B12 were determined,and their correlations with clinicopathological features in gastric cancer were analyzed. Results:Compared with normal controls, serum Hcy level in patients with atrophic gastritis and gastric cancer was significantly increased(P < 0. 05);serum folic acid and vitamin B12 levels in patients with gastric ulcer,gastric polyp and gastric cancer were significantly decreased(P <0. 05). Serum Hcy level in patients with gastric cancer was positively correlated with depth of tumor infiltration,TNM staging and lymph node metastasis(P < 0. 05),however,serum folic acid and vitamin B12 levels had no correlation with clinicopathological features. Conclusions:Hcy level is increased in chronic atrophic gastritis,gastric cancer;levels of folic acid and vitamin B12 are decreased in gastric ulcer,gastric polyp and gastric cancer. High level of Hcy is involved in infiltration and metastasis of gastric cancer. Intervention in patients with high level of Hcy,low levels of folic acid and vitamin B12 might be an effective strategy for the prevention and treatment of gastric cancer and precancerous diseases.

Objective To investigate the changes of the cell cycle regulators ATM, Chk2 and p53 and cell cycle arrest in HeLa cells after cisplatin therapy. Methods The proliferation-inhibiting rates of HeLa cells induced by eisplatin of different concentrations were measured by MTT assays. The mRNA and protein expressions of ATM, Chk2 and p53 of HeLa cells with and withont cisplatin were detected by RT-PCR and Western blot, respectively. The cell cycle analysis was conducted by flow cytometric analysis. Results Cisplatin inhibited the proliferation of HeLa cells in a dose- and time-dependent manner. The mRNA and protein expressions of ATM, Chk2 and p53 were increased in HeLa cells treated with cisplatin. The cell cycle was arrested in G2/M phase in HeLa cells treated with cisplatin. Conclusion Activation of ATM, Chk2 and p53 might be critical in determining whether cells survive or undergo apoptesis. Targeting ATM, Chk2 and p53 pathway might he a promising strategy for reversing chemoresistance to clsplatin in cervical cancer.
10.Efficacy of different doses of sufentanil in minimum alveolar concentration of sevoflurane in patients undergoing bronchoscopy

Objective To investigate the effects of different doses of sufentanil on the minimum alveolar concentration (MAC) of sevoflurane for sedation in patients undergoing bronchoscopy. Methods ASA physical status I orⅡpatients of both genders, aged 20 ~ 65, undergoing bronchoscopy under general anesthesia,were randomly divided into 4 groups (n=20 each):control group (group C) and different doses of sufentanil groups (Sl, S2 and S3 groups). Sufentanil 0.1, 0.2 and 0.3 μg/kg in 5 mL of normal saline was intravenously infused before induction of anesthesia in groups of SI S2 and S3 respectively. While 5 mL of the normal saline was given instead in the group C The patients were mechanically ventilated after insert laryngeal mask. Anesthesia was maintained with inhalation of sevoflurane. Each time the concentration of sevoflurane at end expiration increased/decreased in the next patient depending on the concentration of sevoflurane at end expiration with which the former had no cough. The ratio between the two consecutive concentrations was 1.1. The middle point between the positive response and negative response served as a crossover pair. After at least 7 independent crossover pairs were observed in each group. The MAC and 95%confidence interval of sevoflurane were calculated. The time of anesthesia induction and analepsis was recorded. Results The MAC (95%CI) of sevoflurane was 3.0%(2.8%~3.3%), 2.3%(2.1%~2.5%), 1.9%(1.6% ~ 2.2%) and 1.6% (1.3% ~ 1.9%) in groups of C, S1, S2 and S3 respectively. The MAC of sevoflurane was significantly lower in groups of S1, S2, S3 than in the group C, and in groups S3 than in the group S1 (P<0.05). The time of anesthesia induction was significantly shorter in groups of S2, S3 than in the group C and significantly longer in groups S3 than in the group C. Conclusion Sufentanil of 0.1, 0.2, 0.3 μg/kg can significantly decrease the MAC of sevoflurane in patients undergoing bronchoscopy in a dose-dependent manner.
