1.Improved design and biomechanical characteristics of the rotary self-locking intramedullary nail of the humerus

BACKGROUND:Humeral fractures can be treated by using rotary self-locking intramedulary nail. The previous spin in self-locking intramedulary nail has some limitations in application, so it should be improved in its design so as to obtain better biomechanical effect.
OBJECTIVE:To improve the design of the rotary self-locking intramedulary nail of the humerus, and to analyze its biomechanical characteristics and application effect, and to provide reference for the clinical treatment of humeral fractures.
METHODS: (1) The design of the rotary self-locking intramedulary nail was improved and tested by mechanics test. (2) A total of 63 cases of humeral fractures were randomly divided into two groups: control group (31 cases) and experimental group (32 cases). Patients in the control group received conventional rotary self-locking intramedulary nail, and patients in the experimental group received improved nail.
RESULTS AND CONCLUSION:Results of mechanical test concerning anti-axial compression, tensile, torsion and bending were obviously better after improvement than before improvement. No significant difference in range of motion was detected between the two groups (P > 0.05). Compared with that before treatment, the range of motion was significantly increased after treatment in both groups (P < 0.01 orP < 0.05). The range of motion was significantly higher in the experimental group than in the control group (P < 0.05). Simultaneously, the excelent and good rate was significantly higher in the experimental group than in the control group (P < 0.05). Thesedata suggest that the improved rotary self-locking intramedulary nail has better mechanical characteristics and can obtain better clinical effect.

3.THE CHARACTERISTICS OF STEM CELL DIFFERENTIATION IN HUMAN SKINS FROM DIFFERENT DEVELOPED STAGES AND THEIR EFFECTS ON SKIN WOUND REPAIR

To gain insight into the mechanisms of an age related difference in ability of wound healing, the characteristics of stem cell differentiation in skins from fetus, child and adult were investiga ted. Integrin ? 1 and keratin 19 (K19) were used as the biochemical markers for stem cells and transit amplifying cells. Biopsies were taken from fetus (22~24week gestational age), children (4~12year) and adults (35~53year). Immunohistochemistry was used. As for the immunostainings of fetal tissue sections, integrin ? 1 and K19 expressions were observed in all epidermal basal cells. In children skin, the ratio of integrin ? 1 and K19 positive cells in the epidermal basal layer was 60%~80%. In adults, the ratio in the epidermal layer decreased. These results indicate that fetal skin epidermis contains a large number of stem cells and transit amplifying cells, and the proportion of stem cells and transit amplifying cells decreases with age after birth, which maybe a reason of the age associated difference in ability of wound healing.
4.Progress in multidrug resistance of gastric cancer

Chemotherapy is one of the main treatments for gastric cancer, but its drug resistance often limits the effectiveness of chemotherapy, leading to treatment failure. Drug resistance can be divided into primary drug resistance and secondary resistance. It has showed that several factors were involved in the multidrug resistance of gastric cancer, including the expression of drug resistance-related proteins, abnormalities of apoptosis-related genes, dysfunction of DNA damage repair, epithelial-mesenchymal transition and non-coding RNA. The solution to improve the efficacy of chemotherapy is to overcome drug resistance or delay drug resistance. This article will explore the progress in multi-drug resistance of gastric cancer.

6.A COMPARATIVE STUDY ON THE PROLIFERATION AND DIFFERENTIATION CHARACTERISTICS OF EPIDERMAL STEM CELLS IN NORMAL SKIN AND SCAR OF CHILDREN

To study the location and expression characteristics of epidermal stem cells in normal skin and scar epidermis of children, and to explore the relationship between the differences of these two epidermal stem cells and wound healing after burn. ?1 integrin and keratin 19 (K19) were used as the biochemical markers to identify stem cells and transit amplifying cell, keratin 14 (K14) and keratin 10 (K10) were used as the markers for post-mitotic cells and terminally-differentiated cells, respectively. Normal skin and scar tissue were obtained from children of 4 to 12 years of age. Elivision two-step immunohistochemistry was used. The results showed that in the immunostained tissue sections, the positive ?1 integrin and K19 expression cells were observed in 2~3 layers above the basal layer, whereas K10 expression cells were observed in all epidermal cells except basal cell layer in the scar tissue. Observations revealed that the number of stem cells and transit amplifying cells were less in the scar tissue than that in the normal skin, the differentiation process of scar epidermal stem cells was different from that of normal skin, and the proportion between post-mitotic cells and terminally-differentiated cells was abnormal. The results indicated that the self-renewal ability of the scar epidermis was lowered, and the differentiation process of it was deranged, and this might be considered to be a reason of abnormality of structure and function of the epidermis of scar tissue, and its poorer ability in wound healing.

Objective: To establish a HPLC method for the determination of liquiritin、 liquiritigenin and iso liquiritigenin in Licorice Mixture.Methods: A Cromasil C 18 column was used. the mobile phase was methanol 0.5% acetic acid for gradient elution. Liquiritin、 liquiritigenin and iso liquiritigenin were determined by dual wavelength, ? 1=276 nm,? 2=372 nm.Results: The linear range of liquiritin was 50.0~1250.0ng, r= 0.9999; The average recovery was 98.30%, RSD =1.01%( n= 6). The linear range of liquiritigenin was 5.0~125.0 ng, r= 0.9998; The average recovery was 98.01%, RSD =0.68%( n= 6). The linear range of iso liquiritigenin was 3.0~6.0 ng, r= 0.9999. The average recovery was 98.37%, RSD= 0.89%( n= 6). Conclusion: This method is convenient, rapid and accurate. It can be used for quality control in production of Licorice Mixture.
10.Effects of curcumin on secretion of adiponectin and interleukin-6 in human adipose tissues: an in vitro study

OBJECTIVE: To investigate the effects of different concentrations of curcumin on secretion of adiponectin (APN) and interleukin-6 (IL-6) in human adipose tissues cultivated in vitro. METHODS: Seven male patients with kidney stones were admitted. Abdominal subcutaneous adipose tissue and perirenal adipose tissue were collected from the operating-patients, and were cultivated with different concentrations of curcumin (10 and 100 microg/ml) in vitro. The contents of APN and IL-6 in the culture medium of adipose tissue cells were measured by enzyme-linked immunosorbent assay (ELISA) after they were cultured for 6 and 24 hours. RESULTS: Compared with the blank control group, the content of APN in the adipose tissue culture medium was increased by 100 microg/ml curcumin (P<0.05) after 6-hour culture, and the content of IL-6 was significantly decreased by 100 microg/ml curcumin after 6- and 24-hour culture (P<0.05). CONCLUSION: 100 microg/ml curcumin can increase APN secretion and decrease IL-6 secretion in human adipose tissues cultivated in vitro.
