Objective:To study the therapeutic effect of warming needle plus tuina for degenerative knee osteoarthritis.Method:Randomize 120 cases into a treatment group and a control group,60 cases in each group by the visiting sequence,and then apply warming needle plus tuina for the cases in the treatment group and articular cavity injection of Sodium Hyaluronate for the cases in the control group.Results:The total effective rates in the treatment group and control group were 93.3%and 83.3%respectively,showing a significant difference (P＜0.05).Conclusion:Warming needle plus tuina works well on degenerative knee osteoarthritis.

Objective To investigate the effects of serum from burned rats on the gene expression in cultured rat mesenchymal stem cells (MSCs). Methods Bone marrow was extracted from sacrificed Wistar rats, and MSCs were then incubated for 24h in F-12 medium in the presence of normal rat serum (group N) or serum obtained from burned rats 3 days after injury (group B). Total RNA was extracted from both groups.The mRNA was isolated.An Oligo microarray containing 5705 genes was used to compare the differences of gene expression between two groups. Results There were four genes which expressed differently in two groups.In comparison with group N, the expression of steroid sensitive gene 1 was decreased, but that of fibroblast growth factor-4, dihydroxyacetonephosphate acyltransferase and a EST, which was moderately similar to Bmp2-inducible kinase, were increased in group B. Conclusion Serum from burned rats was able to change the gene expression of MSCs, and it might play the key role in wound repair.

Objective To investigate the effects of serum from rats with burn injury on the differentiation of bone marrow mesenchymal stem cells(MSCs). Methods Bone marrow was extracted from sacrificed Wistar rats and MSCs were then incubated in F-12 medium in the presence of fetal calf serum (FCS) (group F),normal rat serum (group N),or serum harvested from rats with burn injury(group B). Cells of the fifth passage were harvested to investigate the expression of CK and FⅧ by immunohistochemistry and flow cytometry. Results With immunohistochemical staining,both CK and FⅧ were positive in MSCs of group B and negative in the cells of the other two groups. The flow cytometry analysis showed that the positive rates of CK and FⅧ staining MSCs of group B were higher than that of the other two groups ( P 0.05). Conclusions The serum obtained from rats with burn injury could induce differentiation of MSCs to both epithelial cells and vascular endothelial cells,implying that serum after barm injury might promote the MSCs to participate in the process of wound repair.

Objective To investigate the expression of heat shock protein (HSP) 60 and analyze the significance in cervical carcinoma. Methods The cervical tissue samples of 57 cases of cervical cancer and 57 normal cervical tissues were collected. The expression of HSP60 were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). The protein expressions of HSP60 were determined by Western blot analysis. To observe the relationship between HSP60 expression and the occurrence and development of cervical cancer. Results HSP60 mRNA expression in normal cervical tissue was significantly lesser than that in cervical cancer tissue (t=2.65, P0.05). Objective To investigate the effect of bFGF and MSCs autografting on expression of PCNA and Ⅷ-R Ag of porcine skin defect wound. Methods Six male minipigs were used to produce 48 full-thickness skin wounds, each wound area about 2.54 cm~ 2 on both sides of the back (8 wounds per pig). These 48 wounds were randomly treated with bFGF, MSCs, bFGF+MSCs, and normal saline (12 wounds for each treatment). The tissue from the wounds was harvested on day 3, 7, 14 after surgery, then used for HE staining and immunohistochemical staining for proliferating cell nuclear antigen (PCNA) and factor Ⅷ related antigen (Ⅷ-R Ag). Results Granulation tissue was presented in all groups on day 3 and 7 after wound, most abundant in the wounds treated with bFGF+MSCs, as well as blood capillary formation. PCNA expression increased after skin injury, reached the peak on day 7, most in the granulation tissue, and lasted to day 14, then began to decrease. On day 3, 7, 14 after skin injury, PCNA expression was the most highest in the wounds treated with bFGF+MSCs. Objective To investigate the changes of the cell cycle regulators ATM, Chk2 and p53 and cell cycle arrest in HeLa cells after cisplatin therapy. Methods The proliferation-inhibiting rates of HeLa cells induced by eisplatin of different concentrations were measured by MTT assays. The mRNA and protein expressions of ATM, Chk2 and p53 of HeLa cells with and withont cisplatin were detected by RT-PCR and Western blot, respectively. The cell cycle analysis was conducted by flow cytometric analysis. Results Cisplatin inhibited the proliferation of HeLa cells in a dose- and time-dependent manner. The mRNA and protein expressions of ATM, Chk2 and p53 were increased in HeLa cells treated with cisplatin. The cell cycle was arrested in G2/M phase in HeLa cells treated with cisplatin. Conclusion Activation of ATM, Chk2 and p53 might be critical in determining whether cells survive or undergo apoptesis. Targeting ATM, Chk2 and p53 pathway might he a promising strategy for reversing chemoresistance to clsplatin in cervical cancer.

Objective To explore the psychosocial influencing factors of suicide attitudes among new recruits in training camp.Methods Cluster sampling was conducted and 5 200 new recruits in 11 training camps were investigated with suicide attitude questionnaire(QSA),symptom checklist 90(SCL-90) and self-designed questionnaire.Influencing factors associated with suicide attitudes were analyzed with muhiple linear regression analysis.Results The score of awareness of property of suicide behavior dimension of QSA were (3.71±0.58),and (2.77±0.47) of the attitude to the suicide dimension,(2.43±0.43) of the attitude to family members of the suicide dimension and (3.13±0.86) of the attitude to euthanasia dimension.The results of multiple linear regression analysis indicated that recruits who came from single parent family,had a history of family violence,had a introvert character,had poor emotional stability,had a history of psychiatrist visiting,took Sedatives more often,had more negative life events in past year,tended to use unhealthy cathartic method of emotion,had more difficulty in adjusting to the army life,had lower factor score of obsessivecompulsive symptoms in SCL-90,and higher factor score of depression and paranoia,tended to hold an admissive attitude to suicide behavior.Conclusion The recruits in training camp don' t accept suicide behavior as a whole,and their suicide attitudes were influenced by mental health states and many other psychosocial factors.Suicide prevention education should be targeted.

Aim To construct eukaryotic expressing plasmid of hi FGF2 ( high molecular weight isoform fi-broblast growth factor-2,hi FGF2) gene and to investi-gate its effect on apoptosis after its overexpression in HEK293 cells. Methods The DNA template primer was designed and synthesized. The pDsRed1-N1 plas-mids were digested by the restriction enzymes of Nhel and Hind III. The hi FGF2 was ligated with linearized pDsRed1-N1 by T4 DNA Ligase. The recombinant plasmid was identified by endonuclease digestion and sequenced. The recombinant hi FGF2 plasmid was transient transfected into HEK293 cells by Lipofectami-neTM 2000 Reagent. The transfection efficiency was de-tected by fluorescence inversion microscope. The cell apoptosis was detected by Annexin V-FITC/PI apopto-sis detection kit with flow cytometry analysis. Results The pDsRed1-N1 eukaryotic expression vector was consistent with the design. The recombinant hi FGF2 plasmid was transfected in HEK293 cells. The trans-fection rate was more than 70%. The FITC/PI dyeing rate in hi-FGF2 over-expression HEK297 cells was a-bout ( 29. 12 ± 2. 81 )%. Conclusions pDsRed1-N1 eukaryotic expression vector is successfully constructed and transfected into HEK293 cells. Over-expression of hi FGF2 induces cell apoptosis.

Purpose To describe clinicopathological features, diagnosis and differential diagnosis of Castleman′s disease. Methods Retrospective analyses of the clinical data, clinicopathology and immunohistochemistry were conducted in ten cases of Castleman dis-ease and reviewed of literature. Results There were 8 cases of unicenrtic Castleman′s disease and 2 cases of multicentric Castleman′s disease. Pathologically, there were 6 cases of hayline vascular types, one case of plasmatcyic type and 3 cases of mixed type in all Castleman′s disease. Immunohistochemically, all cases were negative for BCL-6 and CD10, and Ki-67 expression was less than or e-qual to 30%. There were 4 cases with complete follow-up data, including one case of intermediate type, 3 cases of hyaline vascular type which were healed by surgical resection without recurrence. Conclusions Castleman′s disease is a rare and lymphoproliferative disorders with unknown cause, it is not easy to diagnose before the operation. Whether immunohistochemical features reflect abnormal immune function or play unknown role in the pathogenesis of Castleman′s disease is also demanded further study.

Objective To explore the reliability and validity of the Chinese version of the 36-item Short Form Health Survey (SF-36) for evaluating the health-related quality of life of patients with chronic soft tissue pain, (CSTP). Methods Two hundred and ten CSTP patients were rated with the SF-36 and the Medical Outcomes Study Pain Measurement (MOSPM) instrument and retested 2 weeks later. The internal consistency, test-retest relia-bility and parallel validity of the SF-36 were analyzed. Results The SF-36 showed good internal consistency and test-retest reliability. Most repeated reliability internal correlation coefficients were between 0.44 and O. 66 (P≤0.01) except that of the SF sub-scale of the SF-36, which was O. 336 (P≤0.05). All the internal consistency Cronbach's a coefficients were >0.70. In the assessment of validity, there was significant correlation among all eight sub-scales of the SF-36, and between items 4-11 and total MOSPM scores. The correlation coefficients were between -0.145 and 0. 635 (P≤0.05 or P≤0.01), except between GH of the SF-36 and item 4 of the M OSPM (pain in-fluence on walking) and between MH and item 10 (pain influence days). Conclusion The Chinese version of the SF-36 has good reliability and validity in assessing the health related quality of life of CSTP patients.
