To elucidate the processing mechanism of Herba Epimedii from the new view of the intestinal absorption and metabolism.From analyzing the literatures of processing mechanism and togethering with the research of our lab,a new idea of processing mechanism of Herba Epimedii was brought:the pharmacologically active and easier absorbed flavonoids might be present more in the herbs when changing the heating processing parameters and thereby increased or maintained the efficacy.This thesis first pointed the new idea and method that the intestinal absorption and metabolism of herbs should be considered when studying the mechanism of processing.
3.Absorption and transportation of flavonoids in Herb Epimedii across Caco-2 monolayer model

Objective To study the absorption and transportation of flavonoids in Herb Epimedii by using Caco-2 monolayer model. Methods Caco-2 cell monolayer model was used to study the bi-direction transport of icariin, epimedin A, epimedin B, epimedin C, and baohuoside Ⅰ. The concentration of the five flavonoids in cell culture medium was measured by UPLC and the apparent permeability coefficients (Papp) were calculated. Results The absorptive permeability coefficients (PAB) of icariin, epimedin A, epimedin B, epimedin C, and baohuoside Ⅰ were 5.91?10-7, 3.22?10-7, 2.76?10-7, 4.23?10-7, and 1.46?10-6 cm/s, respectively. Except baohuoside Ⅰ, the other four flavonodes had lower permeabilities, and the secretive permeabilities (PBA) of all the flavonoids were larger than their absorptive permeabilities. Among them, the PBA of baohuoside Ⅰ was 9.8 times as much as the PAB. Conclusion The results suggest that the intestinal absorption of the five flavonoids is lower, which might have efflux mechanism by transporters, and the absorption of monloglycoside (e.g. baohuoside Ⅰ) is better than that of diglycoside (e.g. icariin) and triglycoside (e.g. epimedin A, epimedin B, and epimedin C).
4.Evaluation on fi ngerprint characteristic of Prunella Species with cluster analysis method

Objective:To establish the HPLC fingerprint of Prunella Species from different places and evaluate it by cluster analysis.Methods:The analysis method of HPLC ngerprint was established.Alltima C18(4.6mm?250mm,5?m) was utilized for separation.The mobile phase consisted of methanol(A) and 0.5% glacial acetic acid(B) with a ow-rate of 1.0ml/min.The detection wavelength was 280nm and the injection volume was 20?l.The column temperature was 30℃.The HPLC ngerprints of Prunella Species from three di erent places were determined and analyzed by SPSS.Results:There is a big di erence among ngerprints of Prunella from di erent places.Prunella from Hubei,Anhui,Jiangxi,Guizhou,Guangxi,Sichuan were divided into a class;Henan,Jiangsu,Zhejiang were divided into another class.Conclusion:the eatsblished ngerprint showed characteristics of Prunella Species from di erent places obviously,and the attribute of Prunella Species can be distinguished e ectively by cluster analysis method.
5.CT and MRI Diagnosis of Rare Cystic Disease of the Pancreas

Objective To investigate CT and MRI findings of rare cystic disease of the pancreas.Methods Elven cases with rare cystic disease of the pancreas underwent plain and contrast-enhanced CT and MR imaging before operation were reviewed.The clinical presentations and imaging findings were analysed.Among eleven cases,four were epidermoid,four were lymphepithelial cyst and three were lymphangioma.Results (1) Epidermoids located in the tail of the pancreas with smooth wall,the density of parenchyma of the lesons was the same as spleen at CT plain scan.On contrast-enhanced CT and MRI,the parenchyma of the lesions showed the same enhanced pattern with spleen.(2)Lymphepithelial cysts often occurred in olderly men.The lesions appeared as multilocular masses with definite border,isodensity at CT plain scan,and mixed iso-hyperintensity on both T_1WI and T_2WI images.After injection of contrast medium,the wall and septum of the lesions were enhanced.(3)Lymphangiomas were multilocular cystic lesion in the body-tail of the pancreas,hypodense at CT plain scan,and long T_1 and T_2 signal intensity at MRI plain scan.The septum and wall of the lesions were slightly enhanced on contrast-enhanced images.The lesions were not communicated with the pancreatic duct but pancreatic ducts were compressed and slightly shifted.Conclusion CT and MRI findings of rare neoplastic cystic disease of the pancreas are of certain characteristics.
6.GC-MS Analysis on Chemical Compositions of the Volatile Oil of the Overground Parts of India Wormwood Herbs

OBJECTIVE:To analyze the chemical compositions of the volatile oil on the overground parts of India wormwood herbs.METHODS:The chemical compositions of the volatile oil of the overground parts of India wormwood herbs extracted by wet distillation were analyzed by capillary GC-MS method and the relative component percentage of each component was determined by GC area normalization method.RESULTS:52peaks were separated by capillary GC-MS and their corresponding compounds were identified.The main chemical compositions were as follows:?—Caryophyllene(8.245%);10,10—Dimethyl—2,6—dimethylenebicyclo[7.2.0]undecane—5—?—alcohol.(7.089%);6,10,14—trimethyl—2—Pentadecanone(5.199%);Caryophyllene oxide(4.808%),etc.CONCLUSION:This study can be served as a scientific basis for the further exploitation and utilization of India wormwood herbs.
7.Study on in situ intestinal absorption kinetics of icariin in rats

To investigate the absorption kinetics characteristics of icariin for various intestinal segments.Methods: To establish the situ intestinal perfusion model and to study the absorption of icariin for various intestinal segments.Results: Icariin was metabolized quickly in the four intestinal segments.The permeability coefficient(P*eff) were(4.27?0.28),(5.25?0.17),(1.99?0.09),(0.80?0.03) at duodenum,jejunum,ileum,colon,respectively.Icariside can be detected in the four intestine segments in rats by HPLC.Conclusion: Icariin is metabolized into icariside,and then icariside can be absorbed.
8.Study on the Extraction and Purification Process of Schisandrin

OBJECTIVE: To optimize the process for extraction and purification of Schisandrin in Fructus Schisandra Chinensis. METHODS: The orthogonal test L9( 34) was adopted to optimize the water extraction process using Schisandrin and its extracts as indicators. The parallel test was used to optimize the parameters of the alcohol precipitation technique. RESULTS: The optimized process for extraction and purification of Schisandrin was as follows: extracting three times using 10- fold water, 1. 5h each time, with filter liquids concentrated to 1. 4g of crude drug mL- 1 then precipitated to 80% ; filtering alcohol liquids, and adjusting pH value to 7. CONCLUSIONS: This method can be used as a reference for the extraction of Fructus Schisandra Chinensis.
9.Study on the Effect of Various Polarity Fractions Extracted from Sancaofang and Its Combination on Human Lung Adencarcinoma SPC-A-1 Cells

OBJECTIVE: To study the effect of various polarity fractions extracted from Sancaofang (SCF) and its combination on proliferation of human lung adenocarciaoma SPC-A-1 cells for bolting the most effective position of anti-tumor and suitable compatibility regimes.METHODS: Ethanol extraction and water extraction were adopted to prepare 95%,60% and 30% ethanol extract portion,water extract portion of SCF and compound decoction.The MTT assay was used to determine the effect of various polarity fractions of SCF,decoction and its combination on SPC-A-1 cells proliferation.RESULTS: IC50 of 60% ethanol extract was the smallest for SPC-A-1 cells.60% ethanol extract combined with 95% ethanol extract acts as a stimulus to anti-tumor activity significantly.CONCLUSION: The best suitable compatibility regimes were 95% ethanol extract combined with 60% ethanol extract.The liposolubility extract of SCF can be applied for anti-tumor.
10.Determination of Anticancer Activity Components of Flavonoids from Hedyotis diffusa by HPLC

OBJECTIVE:To establish the method for the simultaneous determination of anticancer activity components of flavonoids from Hedyotis diffusa,i.e. quercetin,kaempferol. METHODS:HPLC was applied to determine the contents and performed on Alltima C18(250 mm?4.6 mm,5 ?m) column. Mobile phase consisted of methanol(A)-0.5% glacial acetic acid,(gradient elution). The detection wavelength was aet at 350 nm. RESULTS:The linear range of quercetin was 0.006 2~0.244 0 ?g(r=0.999 8)and that of kaempferol 0.007 8~0.310 6 ?g(r=0.999 9). The average recovery of quercetin was 101.84%(RSD=1.79%,n=6) and that of kaempferol 99.04%(RSD=2.90%,n=6). CONCLUSIONS:The method is simple,accurate and reproducible for the quality control of H. diffusa.
