Objective:To construct a lentiviral vector overexpression of micrRNA-29b and investigate the biological characteristics in mouse neuronal cell lines GT1-7.Methods:We chemically synthesized two oligonucleotide single-stranded,complete the comple-mentary by bridging extension into DNA double-stranded to form miR-29b precursor structure.The restriction enzyme digested vector plasmid FUGW was ligated to the precursor structure ofmiR-29b by homologous recombination to construct the corresponding lentiviral vector of microRNA-29b overexpression,and the stable cells were obtained in the mouse neuronal cell line GT1-7 by bleomycin drag screening.RT-PCR was used to detect the expression level of related genes at mRNA transcription level,Results:The recombinant lentiviral expression plasmid f-F-miR-29b was successfully constructed,and the expression level was about 30 times higher than that of the control group.The expressions of DCX,Vdac1 and pten were inhibited,have no changes in sex developmental related genes LH-β,kiss-l,Inshulin,IGF-I,GPR54,GnRH and leptin-R.Conclusion:Using the method of lentivirus screening,the microRNA-29b overexpressing stably transformed cells was successfully obtained in mouse neuron GT1-7 cells,which laid a foundation for the study of biological characteristics ofmicroRNA-29b.

Objective To investigate the effect of Dexamethasone on microtubule - associated protein 1 light chain 3(LC3)expression of cells and neurons in cerebral cortex of juvenile Wistar rats with sepsis. Methods Models of juvenile Wistar rats with sepsis were established through cecal ligation and puncture(CLP). Totally 60 cases of 30 -day - old juvenile male Wistar rats were randomly divided into sham - operation group(10 cases),non - treated group (25 cases)and Dexamethasone group(25 cases). Twelve hours after CLP,rats in Dexamethasone group were injected with Dexamethasone(1 mg / kg)via tail vein every other day,with a total of 3 times. The same dose of saline was used in the non - treated group. All rats were killed at the age of 40 days. Expressions of LC3 and neuronal nuclei(NeuN)of cells in cerebral cortex of rats were detected by using immunofluorescence assay,and the number of positive cells was calculated by using image analysis system software. Expressions of LC3 - Ⅰ and LC3 - Ⅱ protein were measured by a-dopting Western blot. Results Three hours after CLP,rats appeared to be curled up and showed piloerection and shi-vering and the neurobehavioral score in non - treated group was significantly lower than that in sham - operation group (t = 9. 895,P = 0. 000). Twelve of 25 rats in Dexamethasone group died in 10 days after CLP(48% ),while 8 of 25 rats in non - treated group died(32% ),and the difference was not statistically significant between the 2 groups(χ2 =1. 333,P = 0. 248). Conclusions The LC3 expres-sion of cells in the cerebral cortex of juvenile Wistar rats with sepsis was up - regulated,LC3 - Ⅱ/ LC3 - Ⅰ ratio in-creased,and the number of LC3 positive neurons also increased,while Dexamethasone could have inhibitory effect on them.

Objective: To expatiate the application of int er ventional treatment in emergency management of life-threatening bleeding follow ing maxillofacial and jugular trauma. Method : Three cases of ma xillofacial and jugular serious injury with life threatening bleeding were treat ed by external carotid arteriography and embolization at branch of external car otid artery. Result:External carotid arteriography and emboliza tion could effectively stop life-threatening bleeding of maxillofacial and jugu lar serious injury. Preoperation arteriography could define diagnosis and direc t the treatment. 2 cases survived after treatment and 1 died of blood lose durin g treatment. Conclusion:External carotid arteriography and embo lization is effective in the treatment of maxillofacial and jugular serious inj ury with life-threatening bleeding.

Objective To investigate self-management behaviors of kidney transplantation recipients after discharge.Methods A total of 97 kidney transplantation recipients were recruited from a teaching hospital of Peking University From September 2009 to June 2012,and all eligible subjects were then assigned to 3 groups:post-transplantation less than 6 months group (group A),post-transplantation 6-12 months group (group B),and post-transplantation more than 12 months group (group C).The subjects were required to complete self-management scale for renal transplant recipients.ANOVA was used for data analysis.Results There were 32 patients in group A,31 in group B,and 34 in group C.Self-management behavior score of the participants of the three groups was 100.1 ± 7.0,99.0 ± 7.3 and 91.3 ± 5.8,respectively (F =3.53,P =0.03).In terms of diet,group B got the highest score (F =16.41,P =0.00).However,group A showed excellence in physical activities (F =11.50,P =0.00).For three groups,score of drug effect and side effect was 2.00 ±0.00,2.03 ±0.18,and 2.41 ±0.50; score of routine laboratory values was 2.00 ±0.00,2.05 ± 0.16 and 2.82 ±0.39; and score of skin protection was 3.09 ±0.30,3.03 ± 0.91,and 2.85 ±0.36,respectively.Conclusions Post-transplantive self-management behavior of patients who completed the surgery for more than 12 months may not be better than others.Healthcare professionals need to improve patients' self-management through health education.

To build a reversed phase ion-pair chromatography to determination content of Dencichine from Panax notoginseng. Using Tetrabutyl ammonium hydroxide ions by the combination of reagent and HPLC method without derivatization to test the content of dencichine directly. The optimum conditions of supersonic extraction were solid-to-liquid ratio 1: 20, Continuous ultrasonic extraction: twice, each time 15 minutes; 3,500 r · min⁻¹, then centrifuging 15 minutes. Dencichine in different age, place, part and the different Processing mode were examined. The method is simple with sound separation degree and stability, which can facilitate the determination of dencichine content directly and provide the basis in quality standard of raw material.

Objective To solve the problems of low power laser instrument with single wavelength and instable output power as well as its incapability of studying the biological effect of low intensity laser and clinical treatment. Methods The AT89S52 singlechip and PID algorithm were used. Results The desired instrument could output the wave of 532nm green laser which could be adjusted continuously from 0mW to 30mW and the wave of 650nm red laser which could be adjusted continuously from 0mW to 50mW. The error of power could be controlled within 0.1mW. Conclusion The instrument can be used to study the biological effect of low intensity laser and clinical treatment with small volume,easy operation and high stability of laser output.

AlM: To compare the two methods for the measurement of glaucoma patients' intraocular pressure ( lOP) between Goldmann applanation tonometer ( GAT) and non-contact tonometer ( NCT) and find the laws of the two methods.?METHODS: The lOP of 108 glaucoma patients ( 206 eyes) were measured by GAT and NCT respectively.?RESULTS: ln 108 glaucoma patients, the average lOP of 206 eyes was 29. 77 ± 10. 27mmHg by GAT and 24. 59 ± 8. 58mmHg by NCT. There was significant difference between GAT and NCT (P<0. 01). The higher of lOP, the difference between GAT and NCT was greater.?CONCLUSlON: The measurement results with NCT were lower than that of GAT. The higher of lOP, the difference between GAT and NCT was greater. lt's better to measure lOP by GAT for the glaucoma patients, in order to avoid the misdiagnosis and mistreatment of glaucoma.

AlM:To evaluate the clinic efficacy of protein-free calf blood extract ophthalmic gel on healing children dry eye.METHODS: One hundred patients ( 50 patients each group) with children dry eye were divided into two groups randomly: control group was dropped 1g/L sodium hyaluronate eye drop four times per day and experimental group was dropped protein-free calf blood extract ophthalmic gel four times per day. Both groups received the depathogeny treatment at the same time. Symptom scores, Schirmer l test values, tear break-up time ( BUT ) , cornea fluorescein staining scores were recorded on the prior treatment and 30d post-treatment, then they were analyzed.
RESULTS:There were statistically significant difference (P<0. 05) for the comparison in the symptom scores, Schirmer l test values, BUT, cornea fluorescein staining scores of prior treatment and post-treatment in each groups. And there were statistically significant difference (P<0. 05) in the same items of post-treatment between the two groups.
CONCLUSlON: There is a clearly beneficial effect of protein-free calf blood extract ophthalmic gel on healing children dry eye.

Objective To explore curative effect of Xiyanping and vidarabine in treatment for children with viral encephalitis and its impact on T cells subgroup.Methods Methods In June 2012~October 2014, randomly selected 106 cases of children patients with viral encephalitis, as the research object.Randomized divided into observation group (n=53) cases, control group(n=53).Both two group were performed routine therapy, and then control group was given Xiyanping treatment, observation group was given Xiyanping combined with vidarabine treatment.1 continuous week treatment, compared two groups of T cell subgroup number and symptoms disappear time.Results In the two groups after treatment of T cell subgroup CD3 +, CD4 +,CD8 +was significant increase in the number of observation group increased number was significantly higher than the control group,and statistically significant differences ( P<0.05 ) .The observation group’s antifebrile time ( 2.5 ±1.1 ) d; headache, vomiting disappear time ( 3.6 ±2.2 ) d;disturbance of consciousness disappear time (2.6 ±1.3) d and length of hospital stay (9.3 ±2.4) d were significantly lower than the control group (4.7 ±2.8) d, (6.5 ±2.3)d, (4.3 ±2.2) d, (14.2 ±3.6) d, which were statistically significant differences (P<0.05).Observation group’s curative effect for instituting accounted for 73.58%, good rate 92.45%, were significantly higher than the control group 52.83%, 77.36%.which were statistically significant differences (P<0.05).The complication rate of observation group was 16.98%, mortality was 0%,were significantly lower than that of 33.96%, 9.43% of the control group;Cure rate of observation group (90.57%) was significantly higher than that of 49.06% of control group. Observation group severe sequela incidence 11.32% was significantly lower than that of 39.62% of control group and statistically significant differences (P<0.05).Conclusion Xiyangping combined with vidarabine in treatment for children with infantile viral encephalitis can significantly increase the number of T cell subgroup, improve immune function in children with, and curative effect is remarkable and high security.
