Objective To study COX-2 expression in bladder transitional cell carcinoma in order to explore the relationship between the immunohistochemical staining and biological behavior of bladder cancer. Methods The expression of COX-2,Ki-67,and VEGF was examined in 50 cases of bladder transitional cell carcinoma and 10 normal bladder mucosa by means of immunohistochemical technique.Results The expression of COX-2 in BTCCs was higher than that in normal bladder mucosa.The positive rate of COX-2 was 80% in BTCCs while it was not detected in normal bladder mucosa.COX-2 was highly positively related to the grade,stage and prognosis of tumors.The expression of COX-2 in poorly differentiated,invasive or re- curring BTCCs was higher than that in well differentiated superficial or nonrecurring ones.But there was not significant difference between the solitary tumour and multiple ones.There were positive correlation both be- tween COX-2 and Ki-67 and between COX-2 and VEGF.Conclusion The high expression of COX-2 may benefit increase in bladder cancer cell proliferative activity.VEGF expression in COX-2 positive tumor was significantly higher than that in COX-2 negative tumor.

OBJECTIVETo study the effect of melatonin on the gastrointestinal motility and plasma levels of the stress hormone in overtraining rats.

METHODThirty adult SD rats were randomly divided into three groups (n = 10): control group, over-training group, melatonin intervention group. 30 min before each training, rats in the control and over-training groups were fed with normal saline (15 mg/kg) once a day and 5 times per week, while rats in the melatonin intervention group were administrated with melatonin, perfusion in the intervention group (15 mg/kg). Excessive training group and melatonin intervention group rats were subjected to excessive training at 5 times a week for 6 weeks. After 6 weeks, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma motilin (MTL) and calcitonin gene-related peptide (CGRP), cortisol (CORT) and catecholamines (CA) were observed in all groups.

RESULTSCompared with the control group, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma MTL, CORT and CA were increased significantly (P < 0.01) while the content of CGRP was reduced (P < 0.01) in over-training group. After treated with melatonin, this trend was reversed, that was, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma MTL, CORT and CA were surpressed significantly (P < 0.01) while the content of CGRP was improved obviously (P < 0.01) in over-training group.

CONCLUSIONMelatonin plays an important role in protecting gastrointestinal tract from dysfunction, in which MTL, CGRP, CORT and CA are all involved.

Objective To investigate the pathogen distribution and drug sensitivity in childhood bacillary dysentery,and to guide clinically the selection of reasonable antibiotics.Methods Bacterial drug susceptibility test was performed by standard Kirby-Bauer method.The results were interpreted according to NCCLS 2002.Results Of the 98 cases,there were two types of positive bacterial species:sh.flexneri(n = 77)and sh.sonnei(n = 21).Both sh.flexneri and sh.sonnei were sensitive to cefoperazone,eeftazidime,ceftriaxone,cefoperazone/sulbactam and fura- zolidone,and insensitive to ampicillin and co-trimoxazole.Conclusion sh.flexneri was the major pathogen of child- hood bacillary dysentery.The third generation cephalosporins were the first choice for shigella infections.

Objective To evaluate the diagnosis, treatment and prognosis of occult breast cancer(OBC). Methods Clinical data of 12 OBC cases from June 1995 to June 2006 were retrospectively analyzed.Results There were 12 female; the mean age was 52.7 years(39～66). Tumor of all cases was in one of breasts, left(10 cases) or right(2 cases), 4 cases in pro- menostasis, 8 cases in post-menopause, and inci-dence of OBC in all kinds of breast cancer was 0.5% (12/2385). All cases were found because of lump in axillary fossa, the biopsy of 10 showed cases were lymph node adenocarcinoma metastatic, the fine needle aspiration of 2 cases were lymph node adenocarcinoma metastatic. Three cases were positive in the determi-nation of estrogen receptor of lymph node adenocareinoma metastatic. Two cases, suspecting breast cancer,could be found fine calcification in the examination of breast X-ray molybdenum target in 10 cases. Buhra-sound in 11 cases and near infrared ray scan in 10 cases did not detect cancer in breast; chest X-ray and Bultrasound in 10 cases and CT and all over the body bone scan in 5 cases did not found any cancer besides breast. Ten cases used modified radical mastectomy for breast cancer, one case radical mastectomy for breast cancer, one palliative operation. Pathological diagnosis of 9 cases after operation found breast primary le-sion, 6 cases were infiltrated duct cancer, 3 eases were intraductal cancer. All cases received radiotherapy and chemotherapy. Three cases which were positive in the determination of estrogen receptor of lymph node adenocarcinoma metastatic received TAM. Followed-up was made from 3 to 10 years, 11 cases were not found recurrence or metastses, and 5 cases had been living over 5 years. Conclusions OBC is one of rare special breast cancers. The powerful infiltrating capability leads to lump in axillary fessa in prophase and dis-rant metastses. The biopsy and fine needle aspiration would be used for diagnosis. Modified radical mastecto-my for breast cancer is the usual therapy method.

Purpose:To evaluate the different dose distribution created by three different plans on a nasopharyngeal carcinoma case with simultaneous integrated boost intensity modulated radiation therapy (SIB-IMRT). Methods:we designed three different SIB-IMRT plans on a nasopharyngeal carcinoma case, one was from anterior direction of the head, 5 non-equally spaced fields, another was from posterior direction of the head, 5 non-equally spaced fields also, third was 5 equally spaced fields around the head. Each plan had its dose distribution to the target and main critical structures.Results:No significant difference of dose distribution to the target when comparing the previous three different IMRT plans. However, dose to the parotids delivered from 5-equally spacd was obviously lower than the other plans, dose to the optic chiasm was much lower created by the plan designed from the posterior direction of the head with 5 non-equally spacea field.Conclusions:For a nasopharyngeal carcinoma case, there is no significant difference of dose distribution to the target compared with three different 5-beam orientation SIB-IMRT plans. To protect parotids, we recommend designing a plan with equal beam direction around the head. When the target is very close to the optic chiasm, plauning from posterior direction of the head should be better.

Ipomoea pes-caprae, the plant of Ipomoea (Convolvulaceae), is the marine medicine growing in the intertidal zone of coasts. It is not only the Jing nationality medicine in the southern coastal areas of China, but also the folk medicine in the tropical and subtropical countries such as Australia, Mexico, Thailand, Brazil, and Pakistan. It is resource-rich and widely used. The main compounds contain resin glycosides, terpenoids, phenolic acid, flavonoids, volatile oils, and steroids. Pharmacological studies have been reported that I. pes-caprae has a variety of pharmacological activities, such as antitumor, antibiosis, anti-inflammatory and analgesic, anticollagenase, anti-oxidation, immun regulation, and other activities. The recent study on chemical composition and pharmacological activities of I. pes-caprae are summarized in this paper to provide reference for the clinical application, quality control and product development of the marine medicine.

Objective To develop a new method, capillary electrophoresis(CE) based on molecular beacon(MB),for rapid detection of polymerase chain reaction(PCR).To explor the roles of the IL-13 gene exon4 A2044G single nucleiotide polymorphism(SNP) in the pathogenesis of bronchial asthma.Methods The IL-13 exon 4 was amplified by PCR with genomic DNA used as templates from 20 healthy persons and 32 patients with dominantly allergic familial history living in the north of china.Then separating the amplification with capillary electrophoresis followed by hybridization of molecular beacons into the PCR product which were sequenced in the end. Results Detected with CE-MB method,there was significant difference in the distribution of A/G in IL-13exon 2044,A allele frequency was higher in asthma compared with normal controls,same as sequencing. Conclusion The method of capillary electrophoresis based on molecular beacon(MB-CE) is able to be developed as a clinical detecting method for genetic variation diseases.IL-13 A2044G SNP is important in the asthmatic mechanism.

Objective To construct prokaryotic vector and express human herpes virus 8 (HHV8) small capsid protein open reading frame 65 (ORF65) in Escherichia coli.Methods DNA was extracted from BCB-1 cells (HHV8-positive but EBV-negative).HHV80RF65 coding sequence was amplified by PCR and inserted into the prokaryotic expression vector pThioHisA.Recombinant plasmid was transformed into E.coli BL21 to express fusion protein induced by IPTG.The expressed products were purified by affinity chromatography on Ni-NTA resin.The antigenieity and the specificity of the recombinant proteins was identified by Western blot with HHVS-positive serum samples.ELISA coating with the recombinant proteins was used to screen 568 serum samples,which were simultaneously detected by IFA kit(Biotrin) and ELISA kit(Qiagen).Results Gene sequencing showed that the target gene was identical with that of HHV8 standard species,the 31 500 fusion protein was found in SDS-PAGE.ELISA coating with the recombinant proteins had a good agreement with IFA kit(Biotrin) and ELISA kit(Qiagen).The detection for the clinical samples showed the ELISA kit was feasible,applicable and consistant.Conclusion The recombinant proteins showed good antigenicity,and it is valuable for further study on HHV8 specific antibody detection.

Objective:To investigate the effect of eucaryotic expression plasmid DNA on activation of macrophage and the mechanism Methods:NO production by macrophages,the expression of surface molecules on macrophage by FACS and the cytotoxicity of macrophage were measured by Griess method,FACS and MTT respectively Results:The results showed that macrophages were activated to produce NO by plasmid pCH510 and pcDNA3 1 respectively in vitro or in vivo The expression of MHC Ⅱand B7 1 on macrophage and the cytotoxicity of macrophage was increased Plasmids DNA in combination with IFN ? but not with CH50 augmented the activation of macrophage in vitro In contrast, the activation of the macrophage were enhanced when the stimulated macrophages by plasmid DNA in vivo were treated by addition of CH50, but not IFN ? Conclusion:The results suggested the plasmid DNA could activate macrophage in vitro or in vivo but the mechanism of activation was different In vitro, the plasmid DNA directly activated macrophage and had synergistic effect with IFN ?,while in vivo macrophages were mainly indirectly activated and the activation of macrophages was enhanced in the presence of CH50 When eucaryotic expression plasmid pCH510 was used in gene therapy of tumor, besides the function of its products expressed, the plasmids showed the significant role in tumortherapy by its direct immune activation of macrophages
