Inhibitor of growth 2 (INC2 ) is a member of INC family, and cooperates with p53 in regulating cell cycle, apoptosis, and aging. It can also facilitate chromatin remodeling and transcription regulating through histone modifications. Researches suggest that INC2 protein is an important factor in the genesis, metastasis and prognosis of many kinds of tumors. Resent studies indicate that INC2 can not be simply classified as oncogene or tumor suppressor. At present, the function of INC2 in tumorigenesis and tumor development involves interaction with varied molecules, but the mechanisms remain largely unknown, which still require further investigation.

Objective To investigate the effects of 1,25-dihydroxyvitamin D3[1,25(OH)2D3] on regulatory T cells(CD4+CD25+ Foxp3+ T cell)and airway inflammation in asthmatic rats. Methods Asthmatic models were established in 28 rats with ovalbumin as sensitinogen,and four groups were randomly divided(n=7).1,25(OH)2D3 was orally administered 1 h before challenge in treatment group 1,dexamethasone was subcutaneously injected 1 h before challenge in treatment group 2,both 1,25(OH)2D3 and dexamethasone were given 1 h before challenge in treatment group 3,while no medicine was administered 1 h before challenge in group 4(asthmatic control group).The levels of IL-4 and IL-10 in bronchoalveolar lavage fluid(BALF) were detected,the total cell numbers were obtained and the eosinophil count was performed.The ratios of Treg to CD4+ T cells in peripheral blood and spleen mononuclear cells were measured with flow cytometry,and the mRNA expression of IL-10 and Foxp3 in lung tissues was detected.Wistar rats without asthma were served as normal controls(n=7). Results There were significant differences between asthmatic control group and normal control group in each parameter.Compared with asthmatic control group,the total cell number and eosinophil count decreased,the level of IL-4 decreased and the level of IL-10 increased in BALF in treatment groups.The ratios of Treg to CD4+ T cells in peripheral blood and spleen mononuclear cells significantly increased,and the mRNA expression of IL-10 and Foxp3 in lung tissues increased in treatment groups compared with asthmatic control group.There was no significant difference between treatment groups and normal control group in each parameter. Conclusion Administration of 1,25(OH)2D3 increases the proportion of Treg and expression of Foxp3 mRNA in peripheral blood and spleen in asthmatic rats.1,25(OH)2D3 has a regulatory effect on Treg,and may serve as a potential therapeutic approach against asthma.

?AIM: To discuss the effect of laser exposure on visual acuity and macula.?METHODS: Retrospective and consecutive case series. A retrospective analysis of 11 patients (11 eyes) with laser retinal injury was carried out from January 2014 to June 2015 in Ophthalmology Department of No. 474 Hospital of Chinese PLA. All individuals underwent visual acuity, best corrected visual acuity ( BCVA ) , and spectral-domain optical coherence tomography ( SD-OCT) for macular at first visit, and fundus fluorescein angiography ( FFA ) , visual field, and multifocal electroretinogram ( mf ERG ) were perform if necessary. Symptomatic therapies, supportive therapies and pars plana vitrectomy ( PPV ) were performed depended on the patient’s condition. The patients were followed-up at 1, 3 and 6mo after the first visit, and patients were undertaken visual acuity, BCVA, macular SD-OCT and so on.?RESULTS: Eight patients ( 73%) were under 18 years old and all patients were young males, who were injured by laser pointers when playing. Three patients ( 27%) over 18 years old were injured accidentally at work. Ten (91%) patients’ BCVA were ≤0. 3, while one ( 9%) patient’s BCVA was higher ≥0. 3. Full-thickness macular holes ( the diameter 224-519 μm ) were detected in 10 patients (91%), while sub-foveal RPE changes and IS/OS injury in 1 patient ( 9%) . Macular hole with traction or cystoid edema in 6 eyes (55%) were received PPV, while the other 5 eyes ( 4 eyes with stable macular hole and 1 eyes with RPE injury ) received conservative treatment. Macular hole closed successfully in 1 eye ( 17%) after PPV, while macular hole in the other 5 eyes ( 83%) were stable after PPV of which the cystoid edema faded. The 4 patients with macular hole and 1 patient with RPE injury were stable during follow-up period. However, the BCVA in all patients had no significant improvement at end.?CONCLUSION: Exposure to laser devices could lead to severe macula injury that could reduce central vision, which is permanent.

Objective To explore the risk factors of prehypertension and the relationship between blood pressure and atherosclerosis.Methods The data of 456 cases in the Department of General Medicine were cross-sectional analyzed,including 174 subjects with normotension and 282 with prehypertension.The information was consisted of demographic characteristics,blood pressure,blood biochemical metabolism index,brachia-ankle pulse wave velocity (baPWV),ankle-brachial index (ABI) and carotid intima media thickness (CIMT).Results Compared to normotension group,the levels of systolic pressure,diastolic pressure,pulse pressure,body mass index,fasting blood glucose,glycosylated hemoglobin,triglyceride,uric acid,and C-reactive protein in prehypertension group were significantly increased (P ＜ 0.05) ; and high-density lipoprotein cholesterol was significantly decreased (P ＜0.05) ; the body mass index(OR =1.185),triglyceride(OR =1.302),and fasting blood sugar (OR =1.690) were the independent risk factors of prehypertension ; baPWV and CIMT in prehypertension group were higher,but ABI and artery atheromatous plaque rate were not obvious changed.When baPWV or CIMT were set as the dependent variables,multiple linear regression analysis showed that systolic blood pressure(β =0.226,P =0.007),fasting blood glucose(β =0.209,P =0.018),and age(β =0.279,P =0.002) were risk factors of baPWV;systolic blood pressure(β =0.118,P =0.015),body mass index(β =0.109,P =0.001),and age(β =0.396,P =0.001) were risk factors of CIMT.Conclusions Body mass index,triglycerides,and fasting blood sugar were the independent risk factors of prehypertension.The early subclinical damage of hardening of the arteries was occurred in the prehypertension cases,and systolic blood pressure was closely related with baPWV or CIMT.

Objective To investigate the diagnosis of a case with 7p15.3p22.1 microdeletion by applying array-based comparative genomic hybridization (array-CGH) and to analyze the relationship between the clinical manifestations and 7p15.3p22.1 microdeletion. Method Array-CGH technique was used to detect genomic copy number variations (CNVs) in an infant with normal karyotype after conventional chromosomal karyotyping. Results Array-CGH detected 7p15.3p22.1 deletion (chr7: 6777262-23981753), which was confirmed as pathogenic CNV after comparative analysis with database. Conclusion Array-CGH could serve as a useful complement for G-banding to be used in the clinical cytogenetic diagnosis.

Objective To investigate ING2 expression in normal gastric tissue,gastric carcinoma and precancerous lesions, and to explore correlation between ING2 expression and the carcinogenesis and progression of gastric carcinoma and the clinicopathological signficance as well as the correlation between ING2 and raP53 protein. Methods The expression of ING2 was measured by PV9000 two-step immunohistochemical staining. In total, 188 gastric cancer(GC), 128 matched normal gastric mucosa,35 chronic atrophic gastritis (CAG), 87 intestinal metaplasia (IM) and 36 dysplasia (DYS) samples were analyzed. Expression of mP53 was measured in 40 samples from the 188 gastric carcinomas mentioned above. Results Positive ING2 expression was significantly more frequent in CAG (74.29%) ,IM (91.95%) ,DYS (75.0%) and GCs (70.21%) than in normal gastric mucosa (36.72% ,P ＜0.05). Among GCs, ING2 expression varied by Lanren's classification. A significantly higher rate of positive ING2 expression was observed in intestinal type GCs (80. 56%) than in diffuse (64.49%) and mixed (55.56%, P ＜ 0.05) type GCs. Furthermore, positive ING2 expression was significantly more frequent in well-to-moderately differentiated adenocarcinoma(80. 60%) than in poorly differentiated adenocarcinoma (62. 62%, P ＜ 0. 05). No correlation was found between ING2 expression and mP53 expression in GC (P ＞ 0. 05). Over-expression and mislocalization of ING2 may be involved in the development of CC, especially intestinal-type GC. Further investigations are needed to explore the relevant molecular mechanism.

BACKGROUND: Three-dimensional printing technology is a new technology which can quickly and accurately transform the virtual computer-aided design into the three-dimensional physical prototypes. Three-dimensional printing physical model method can replace the method of traditional preoperative planning and repair surgical simulation, with the characteristic of repeatable, which has been deepened day after day in clinical application of spine surgery. OBJECTIVE: To summarize the application status of three-dimensional printing technology in spine surgery and look forward to its future development directions. METHODS: The articles regarding the application of three-dimensional printing technology on clinical applications in spine surgery were retrieved from PubMed databases, Google Scholar, China National Knowledge Infrastructure and Wanfang Database from January 2000 to July 2015. The key words were 3D printing technology, rapid prototyping technology, spine, vertebra, department of orthopedics, fracture, joint, hand and foot, bone tumor, trauma, cervical vertebrae, thoracic vertebrae, lumbar vertebrae, sacral vertebrae, pedicle of vertebral arch, vertebral body, intervertebral disc, and clinical application. A total of 50 articles with a good representation were selected and discussed after repetitive studies and reviews were excluded. RESULTS AND CONCLUSION: The three-dimensional printing technique has been applied in preoperative diagnosis, individualized orthosis customerization, the communication between doctors and patients, teaching, the formulation of individualized and high-accurate repairing plan, intraoperative navigation and individualized implant customization. These results suggest that with the rapid development of medical imaging, digital medicine and technologies of the cel and tissue culture and new materials, three-dimensional printing technology wil have a wide range of applications in spine surgery.

Objective To establish a novel flow cytometric crossmatch assay allowing detection of complement-fixing donor specific anti-HLA IgG alloantibodies (Flow cytometry complement-dependent crossmatch,Flow-CDC). Methods One hundred pretransplant crossmatchings were performed using Flow-CDC and NIH-CDC between 62 patients awaiting renal transplantation and 33 donor cells.These crossmatchings were divided into two groups according to PRA.Group 1 consisted of 25 sera with negative PRA,and group 2 consisted of 75 sera with positive PRA.All of the sera were pretreated with DTT to inactivate IgM. Lymphocytes were isolated from peripheral blood (or,in a few instances,from the spleen) of the cadaveric donors. The correlation between different techniques for detection of donor specific anti-HLA antibodies was evaluated.The effect of both methods on clinical transplantation outcome was observed. Results In group 1,NIH-CDC and Flow-CDC were negative for all 25 sera.In group 2,24 (32.0%) had a positive NIH-CDC,31 (41.3%) had a positive Flow-CDC.There was a significant difference between two methods (?2=5.14, P= 0.016 ).Overall concordance between both tests was 93% with 69 concordant negatives and 24 concordant positives. The correlation coefficient (r) was 0.80.In group 2,5 patients received transplantation. One of them with negative NIH-CDC and positive Flow-CDC suffered from acute rejection after transplantation and lost the graft,and the other patients with negative NIH-CDC and Flow-CDC had good outcome. Conclusions Flow-CDC can detect specifically complement-fixing IgG alloantibodies against donor HLA and is more sensitive than NIH-CDC.Additionally,the computer printouts represent a permanent record of the crossmatch for retrospective review.Flow-CDC may become the standard crossmatch method as a possible alternative to conventional NIH-CDC testing.
