1.Clinical efficacy of Roukouwuweiwan in treatment of schizophrenia and its effect on serum SOD, MDA and NO

Objective To study the clinical efficacy of Roukouwuwei Wan in the treatment of schizophrenia and its effect on serum superoxide dismutase, malondialdehyde and nitric oxide.Methods 84 cases of patients with schizophrenia were enrolled in our hospital from September 2015 to June 2016,according to coin-casting method divided into observation group and control group,42 cases in each group.The control group was treated with conventional regimen, and the observation group was treated with Rouku Wuwei Pills on the basis of control group.The quality of life, serum SOD, MDA and NO were measured before and after treatment,the clinical efficacy and side effects were analyzed.Results After treatment, the total effective rate of the observation group was significantly higher than the control group (P<0.05).After treatment,the scores of physical life,social function,mental health and physical health of the observation group were significantly higher than the control group ( P <0.05 );The levels of superoxide dismutase, malondialdehyde and nitric oxide in the observation group were significantly lower than the control group (P<0.05).The adverse reaction rate in the observation group was significantly lower than the control group (P<0.05).Conclusion The treatment of schizophrenia patients with Roukouwuwei Wan can reduce the levels of SOD,MDA and NO, improve the quality of life of patients, the clinical efficacy is good and reduce the incidence of adverse reactions.
3.Clinical observation of orbicularis muscle shortening improved treatment for senile entropion

To analyze the clinical results of orbicularis muscle shortening surgery and eyelid skin orbicularis muscle resection for senile entropion.
●METHODS: From January 01, 2006 to December 31, 2012, senile patients with lower eyelid were divided into two groups according to different surgical methods. Group A with the orbicularis muscle shortening improved operation in 20 cases (38 eyes), group B used the eyelid skin orbicularis muscle resection in 21 cases (36 eyes). The effects of surgery were followed-up postoperatively.
●RESULTS: Patients were followed up for 1 - 2a, 20 patients ( 38 eyes ) in group A were cured without recurrence; 7 eyes recurred in group B, the recurrence rate was 19%, the recurrence rate after surgery both groups were statistically significant (P<0. 05).
●CONCLUSlON: The effect of orbicularis muscle shortening improved operation for senile entropion is good and stable.
4.CT guided the preventation and treatment of hemorrhage after renal cancer cryoablation

Objective To explore the methods of the hemorrhage prevention and the measures of hemostasis by CT guided after renal cancer cryoablation.Methods Retrospective analysis of 8 patients of renal cell carcinoma patienthave been done,all the patients were underwent cryoablation.The basic model included 10 min frozen,3 min thawed and 1 cycle were repeated.Before the cryoprobe puncture,a CT scan was taken to observe the blood supply of the tumor and the relationship between the border of the tumor and renal cortex,renal pelvis.Besides,the path of reducing the injury of normal renal cortex and renal pelvis was chosen,and the decreased number of probes was adopted by taking conformal cryoablation to reduce the damage to the renal capsule,in theensurance of the treatment efficacy.Intraoperative CT scan was performed discontinuously,to observe the covering status of the range of ice ball on the lesion and to monitor the perirenal bleeding.Postoperative ECG and blood pressure changes was observed 24 h after operation.Besides,CT scan or blood routine examination is necessary to monitor whether active bleeding happened.Results According enhanced CT image positioning puncture path,the path of probes were selected in the shortest path between the tumor margins and the renal cortical which avoid the renal pelvis.Probes were CT-guided conformal puncture into the lesion successfully for 8 patients,according to freezing basic mode to complete cryoablation.Timely intraoperative CT scan can clearly observe that a low-density ice ball wrapped lesion completely without involving the renal pelvis and normal tissue surrounding the kidney.There was no bleeding in the procedure of cryoablation.After the probe was pulled,CT scan showed:3 of the 8 patients didn't show any sign of henorrhage,in which 1 patient had heavily bleeding,about 150 ml due to the biopsy after cryoablation.The bleeding was stopped by interventional embolization.A little hemorrhage was found in two patients,about 10 ml outside renal capsule.Medium hemorrhage took place in two patients,30 nl in one patient which underwent several cycles of thawing and 60 ml in another.The patients of mild and medium hemorrhage was treated with intravenous injection of Reptilase and strict bed rest.There was a great hemorrhage in one patient,about 200 ml.The bleeding was stopped by interventional embolization.A small and moderate bleeding,it necessary to execute intravenous infusion 2 IU reptilase and strict bed rest.There was no significant active bleeding from the CT scan after 24 h operation and no change in the blood routine examination of blood hemoglobin and red blood cell count.Conclusions Before the puncture,it is necessary to perform an enhanced CT scan to understand the position,which can determine the path of the puncture.It can reduce the damage of renal cortex,renal capsule and surrounding tissue and prevent and reduce the risk of bleeding.For larger amounts of intraoperative and postoperative bleeding subjects,the embolization should be given immediately,as for the small or moderate amount of bleeding,the conservative treatment may be given firstly.

6.Imaging evaluation of response of rectal cancer to preoperative chemoradiotherapy.

In recent years, preoperative therapy has become the standard procedure to improve radical resection rate and local control for advanced rectal cancer. Tumor responses to chemoradiotherapy, however, vary considerably, thus increasing the demand for both functional and morphologic radiologic evaluation of response to chemoradiotherapy to distinguish responders from nonresponders. MR imaging is considered the most accurate tool for the primary staging of tumor extent, and can be used to evaluate the efficacy of chemoradiotherapy. Functional imaging modalities including DW-MRI and PET-CT have shown promising prospect in the early evaluation of the response of rectal cancer to preoperative chemoradiotherapy. However, wide clinical application will take some time.

7.Induction of apoptosis by celecoxib through activation of cytochrome C pathway in HT-29 cell

Objective To study the effect of selective cyclooxygenase-2 inhibitor celecoxib on cell apoptosis of human colorectal cancer cell line HT-29 and the probable mechanism involved by detecting the expressions of cytochrome C, Caspase-9 and poly ADP-ribose polymerase(PARP) at protein level. Methods Apoptosis was determined by Acridine orange and Ethidium bromide staining under fluorescence microscope and flow cytometry. The protein expression of cytochrome C, Capsase-9 and PARP were examined by Western blotting.Results Celecoxib induced apoptosis of HT-29 human colorectal cancer cells in a concentration and time-dependent manner from 0 to 120 ?mol/L. Sub-G 1 peak was detected by flowcytometry, and the apoptotic rate was between(7.31?2.37)%-(48.30 ?2.86)%. Celecoxib induced cytochrome C release into the cytosol from mitochondria, then activated Caspase-9 and consequently triggered PARP cleavage.Conclusion Celecoxib can induce apoptosis through a cytochrome C-dependent pathway in human colorectal cancer cell line HT-29.
8.EFFECT OF SOYASAPONINS IN PREVENTION OF HYPERLIPIDEMIA IN MICE AND ITS MOLECULAR MECHANISM

Objective: To investigate effect of soyasaponins in the prevention of hyperlipidemia in mice and its molecular mechanism. Methods: 56 healthy mice were randomly divided into 7 groups according to their levels of TC in serum: normal control, high fat control, high fat with 20mg/kg.d gypenosides,high fat with soyasaponins of four doses(5 mg/kg.d, 10mg/kg.d, 20mg/kg.d, 30 mg/kg.d). Results: As compared to high fat control soyasaponins significantly reduced the serum TC, LDL-C, TG concentrations, and increased the HDL-C levels distinctly. They significantly deceased MDA content, increased the activities of SOD and LPL in liver. The results of RT-PCR showed that high fat feeding could induce the reduction of LPL mRNA expression, while soyasaponins could increase it. Conclusion: Soyasoponins prevent hyperlipidemia through upregulation of LPL mRNA expression and increase of antioxidative capacity.

10.The Effect of a Selective COX-2 Inhibitor on Cell Growth in Human Colorectal Cancer Cell Line

Objective To study the effect of a selective COX-2 inhibitor Celecoxib on cell proliferation and apoptosis of human coloretal cancer cell line HT-29 to seek an effective and safe drug for colon cancer chemoprevention. Methods Using MTT assay, flow cytometry(FCM), acridine orange and ethidium bromide staining, the effect of celecoxib on the proliferation and apoptosis of HT-29 cells were investigated. Results The growth of HT-29 cells was inhibited by celecoxib in a dose- and time- dependent manners. FCM analysis showed that the treated HT-29 cells had typical Sub-G 1 peak, the apoptotic rate of which was (7 31?2 37)%~(48 3?2 86)%. The cell ratio of G 0/G 1 phase increased, whereas the cell ratio of S and G 2/M phases decreased after treatment, which was in a dose-dependent manner as well. The treated HT-29 cells exhibited some morphological features of apoptosis, including cell shrinkage, nuclear condensation, DNA fragmentation, and the formation of apoptosis bodies, the apoptotic index of which was in a dose- and time- dependent manners. Conclusions Celecoxib inhibited proliferation and induced apoptosis of human colorectal cancer cell line HT-29, which may be related to blocking the cell cycle progress of HT-29 cells.
