1.Fingerprint Establishment of Amino Acids Composition of Jinlong Capsule

Objective To establish the fingerprint of amino acids composition of Jinlong capsule. Methods Using 6-acetamido-4-hydroxy-2-methyl quinoline (AQC) as the derivating agent, 17 kinds of amino acids were analyzed by UPLC at the same time and the fingerprint of amino acids composition of Jinlong capsule was established. Result Totally 22 co-possessing peaks were selected as the fingerprint peaks, 17 kinds of amino acids were identified. The fingerprint similarity value of 10 batches was above 0.9. Conclusion The fingerprint is characteristic and can be used for the quality control of Jinlong capsule.

6.Antiarrhythmic effect of TJ0711.

To study the antiarrhythmic effect of the newly developed alpha/beta-blocker TJ0711, a variety of animal models of arrhythmia were induced by CaCl2, ouabain and ischemia/reperfusion. Glass microelectrode technique was used to observe action potentials of right ventricular papillary muscle of guinea pig. The onset time of arrhythmia induced by CaCl2 was significantly prolonged by TJ0711 at 0.75, 1.5 and 3 mg x kg(-1) doses. TJ0711 (1.5 and 3 mg x kg(-1)) can significantly shorten the ventricular tachycardia (VT) and ventricular fibrillation (VF) duration, the incidence of VF and mortality were significantly reduced. On ischemia-reperfusion-induced arrhythmic model, TJ0711 (0.25, 0.5, 1 and 2 mg x kg(-1)) can significantly reduce the ventricular premature contraction (PVC), VT, VF incidence, mortality, arrhythmia score with a dose-dependent manner. At the same time, rats serum lactate dehydrogenase (LDH) and creatine kinase (CK) activities decreased significantly by TJ0711 (1 and 2 mg x kg(-1)). Ouabain could cause arrhythmia in guinea pigs, when TJ0711 (0.375, 0.75, 1.5 and 3 mg x kg(-1)) was given, the doses of ouabain inducing a variety of arrhythmia PVC, VT, VF, cardiac arrest (CA) were significantly increased with a dose-dependent manner. In the TJ0711 0.1-30 micromol x L(-1) concentration range, guinea pig right ventricular papillary muscle action potential RP (rest potential), APA (action potential amplitude) and V(max) (maximum velocity of depolarization) were not significantly affected. APD20, APD50 and APD90 had a shortening trend but no statistical difference with the increase of TJ0711 concentration. TJ0711 has antiarrhythmic effect on the sympathetic nerve excitement and myocardial cell high calcium animal arrhythmia model. Myocardial action potential zero phase conduction velocity and resting membrane potential were not inhibited by TJ0711. APD20, APD50 and APD90 were shortened by TJ0711 at high concentration. Its antiarrhythmic action mechanism may be besides the action of blocking beta1 receptor, may also have a strong selective blocking action on alpha1 receptor and reducing intracellular calcium concentration.

8.Role of NF-kB in EAN and the effetct of nasal tolerance

Objective:To explore the role of nuclear transcription factor and the effect of nasal tolerance on EAN.Methods:75 male Wistar rats were divided into five groups: Group NS, EAN, EANL, EANM, EANH. The expression of IFN-?, TGF-?1, IL-10 mRNA and P65 protein were detected in spleen by using of RT-PCR and western blot.Results:The expression of IFN-? mRNA were the highest at early phase, those of TGF-?1 and IL-10 augmented significantly at recovery phase. In Group EANM and EANH, the severity of clinical signs and the infiltration of inflammatory cells decreased. IFN-? mRNA and P65 protein expression was lower than that in Group EAN, however, IL-10 and TGF-?1 mRNA increased markedly. At the same time, the expression of P65 protein were positively correlated with those of IFN-? mRNA, negatively correlated with those of TGF-?1 and IL-10 mRNA.Conclusion:NF-?B may play a vital role in EAN. Nasal administration of antigen may effectively prevent Wistar rat from inducing EAN in some degree, which may associate with the inhibition of NF-?B.
9.Hypoxic tumor microenvironment and immune response

The tumor microenvironment is closely related to the occurrence and development of tumor. Hypoxia is considered to be one of the most important factors in tumor microenvironment.Formation of hypoxic microenvironment can be found in most of malignant tumors,which can inhibit the anti-tumor immune response. Recent studies have indicated that immunosuppressive cells,tumor stem cells and circulating tumor cells in hypoxic tumor microenvironment can mediate immune suppression and immune tolerance,and then promote development of tumor.The new immune therapy will focus on normalizing tumor vasculature,reconstructing the tumor microenvironment,avoiding immune suppression and averting tumor immune tolerance.
10.Clinical Observation of Wenjing Shexue Recipe in Treating Polycystic Ovarian Syndrome

Objective To observe the clinical efficacy of Wenjing Shexue Recipe ( WSR) in treating polycystic ovarian syndrome ( PCOS) with liver-kidney deficiency, and to explore the possible therapeutic mechanism. Methods Seventy-four PCOS patients were randomized into observation group ( N=38) and control group ( N=36). The observation group was given oral use of WSR and the control group was treated with Ethinylestradiol and Cyproterone Acetate Tablets orally. Both groups received treatment for 6 months, and were followed up for 3 moths after drug withdrawal. After treatment, the therapeulic effect was evaluated , and the changes of body mass index ( BMI) , hairiness. acne, and sexual hormone levels were monitored before and after treatment. Results ( 1) The total effective rate of the observation gorup was 92.11% and that of the control group was 75.00%, the difference being significant ( P<0.05). ( 2) After treatment for 6 months, body mass index (BMI), the scores of hairiness and acne, and serum levels of testosterone (T) and luteotrophic hormone (LH) were decreased in the observation group (P<0.05 compared with those before treatment). The control group had lower scores of hairiness and acne and serum levels of T and LH than those before treatment ( P<0.05) , but BMI showed no obvious decrease (P>0.05) . The results of follow-up showed that BMI, the scores of hairiness and acne, and serum levels of T and LH were still lower in the observation group than those before treatment (P<0.05), but the above indexes except acne scores were increased in the control group (P>0.05 compared with those before treatment) and were higher than those of the observation group (P<0.05). Conclusion WSR exerts better effect for the treatment of PCOS with liver-kidney deficiency than Ethinylestradiol and Cyproterone Acetate Tablets, and the possible mechanism is probably related with the regulation of endocrine disturbance by decreasing serum T and LH levels.
