3.Correlation between MRI and clinical profiles of periventricular leukomalacia in children

Objective To study the relationship between MRI and clinical profiles of periventricular leukomalacia (PVL) in children Methods The clinical and MRI findings in 34 cases with PVL were retrospectively analyzed Results (1) Periventricular hyperintensity on T 2WI was more prominent in the preterm group than that in the term group, and P value was 0 000; (2) Cortical lesion and subcortical leukomalacia was seen in 9 of 19 cases in the children with PVL born at term, but detected in only 1/15 in the preterm group P value was 0 020; (3)Seizure was common in term children P value was 0 036; (4) The degree of reduction of periventricular white matter correlated with motor impairment and mental retardation in all children, and P values were 0 002 and 0 000, respectively The thinning of the corpus callosum also correlated with mental retardation and P value was 0 012 The degree of reduction of periventricular white matter correlated with visual impairment in preterm group Conclusion The end stage PVL can been clearly displayed by MRI, and gestational age and clinical manifestation were closely related to the findings of MRI
4.Clinical Observations on Acupuncture Treatment of Post-Stroke Dysphagia

Objective: To seek a treatment that can improve the effect on dysphagia following post-stroke pseudobulbar palsy. Methods: Sixty patients were randomly allocated into observation and control groups, 30 cases each. The observation group was treated with Western drugs plus acupuncture at Tiantu (CV 22) and other three acupoints around Lianquan (CV 23) and the control group with simple Western drugs. Results: The effective rate was 86.7% in the observation group and 30.0% in the control group. Conclusion: Acupuncture can improve the curative effect of Western drugs on dysphagia following post-stroke pseudobulbar palsy.

6.The subjective well-being of type 2 diabetes patients and its influential factors

Objective To investigate the situation of subjective well-being feeling of type 2 diabetes patients and its influential factors.Methods One hundred and twenty-seven patients with type 2 diabetes were recruited as our subjects and all of them were performed the questionnaire investigation about well-being feeling.Influential factors were analyzed through several statistic methods.Results The index of subject wellbeing ranged from 3.70 to 13.70 and the average was 10.67 ± 2.10 of all type 2 diabetes patients.Of which The index of total affectation was 5.11 ± 0.92 and the range was from 2.63 to 6.63.The score of life satisfaction was 5.58 ± 1.22,and the range was from 1.10 to 7.70.Index of subjective well-being significantly associated with education,family income,therapy approach,HbA1 c level,complication,medicine burden.However there was no correlation with gender,age,diabetes duration.Conclusion The main types of subjective well-being feeling of patients with type 2 diabetes were positive effect and positive experience.Many factors could be the influence factor to well-being.Measures should be taken to improve Subjective well-being of type 2 diabetes patients.
7.Maximal sequence length of exact match between members from a gene family during early evolution.

Mutation (substitution, deletion, insertion, etc.) in nucleotide acid causes the maximal sequence lengths of exact match (MALE) between paralogous members from a duplicate event to become shorter during evolution. In this work, MALE changes between members of 26 gene families from four representative species (Arabidopsis thaliana, Oryza sativa, Mus musculus and Homo sapiens) were investigated. Comparative study of paralogous' MALE and amino acid substitution rate (d(A)<0.5) indicated that a close relationship existed between them. The results suggested that MALE could be a sound evolutionary scale for the divergent time for paralogous genes during their early evolution. A reference table between MALE and divergent time for the four species was set up, which would be useful widely, for large-scale genome alignment and comparison. As an example, detection of large-scale duplication events of rice genome based on the table was illustrated.

8.The enantioselective pharmacokinetic study of desvenlafaxine sustained release tablet in Chinese healthy male volunteers after oral administration.

A chiral LC-MS/MS method for the simultaneous analysis of desvenlafaxine (DVS) enantiomers in human plasma was developed and applied to a pharmacokinetic study on 12 Chinese healthy volunteers. d6-Desvenlafaxine was used as internal standard (IS). Chromatographic separation was performed on the Astec Chirobiotic V chiral column (150 mm x 4.6 mm, 5 μm). The assay was linear over the concentration range of 0.500-150 ng x mL(-1) for both enantiomers (r2 > 0.99). The method was successfully applied to a stereoselective pharmacokinetic study of 100 mg desvenlafaxine sustained release tablets on 12 Chinese healthy volunteers under fasting conditions. The results showed that the pharmacokinetic parameters were similar to both enantiomers in Chinese healthy volunteers. The AUC(0-t), and C(max) of the two enantiomers were about 1.5 times higher than those of blacks and whites reported in the literature.

9.Cell biologic changes in thd cells which PTEN gene activated by double-stranded RAN

ObjectiveTo evaluate thd cell biologic changes in thd non-small-cell lung cancer(NSCLC) which PTEN gene were activated by double-stranded RNA(dsRNA).MethodsSpecific dsRNA was designed.First,the promoter region of PTEN gene was determined by Promoter 2.0 program,then the CpG island in the promoter was found by CpGisland searcher software and the possible target non-CpG sequence that dsRNA might activate were defined by SiRNA Target Finder software.dsRNA were synthesized at Genechem Company( Shanghai,China).Then the specific dsRNA was transfected into A549 and H292 cells which were stored in our laboratory using Lipofectamine 2000 ( Invitrogen,USA) according to manufacture's instruction.Total celluar RNA was isolated.The expression of PTEN mRNA in transfected,control and mock group were determined by real-time quantitative polymerase chain reaction.Cell profiferation was investigated on days 1 to 5 by using Cell Counting Kit-8 according to the manufature's technical manual.Cell invasion ability was assessed by Transwell method that transmembrane cells were counted,and cell bycle distribution were studied by flow cytometer(FCM) using CycleTESTTM PLUS DNA Reagent Kit.ResultsAfter the introduction of dsRNA into the A549 cells,the PTEN mRNA expressin was upregulated to (4.35 ±0.42) folds compared with the mock and control cells.And in H292 cells,the mRNA expression of PTEN was upregulated to (3.92 ± 0.20) folds.It confirmed the RNA activation phenomenon in the PTEN gene in NSCLC cells.Compared with the control group,the number of alive transfected cells did not decreased in the cell proliferation assay.In the cell invasion test we found that the transmembrane A549 cells were 122.4 ±11.2 vs.150.7 ±13.1 in transfected group and control group respectively.In the cell cycle distribution we found dsRNA in duced part ofthe transfected cells arrested in G1 phase and a corresponding decrease in S-phase population was observed,though this change was not statistically significant.Conclusion The expression of PTEN mRNA could by enhanced by inducing the specific dsRNA into the A549 and H292 cells,though no evidence was found that after the activation of silenced PTEN,the cell proliferation and invasion ability were significantly changed.
10.Comparison of multi-slice CT findings between malignant focal pulmonary ground-glass opacity nodules and solid nodules of 3 cm or less

Objective To compare the MSCT findings of malignant focal pulmonary ground-glass opacity nodules (fGGO) and solid nodules of 3 cm or less, and try to find specific signs in fGGO. Methods Clinical data (sex ratio, age), size of lesion and MSCT findings (shape, margin, interface, internal characteristics, adjacent structure) of 105 cases pathologically confirmed to have solid lung cancers and 48 cases with fGGO less than 3 cm were retrospectively analyzed. Differences were analyzed by using the Fisher exact test or Mann-Whitney U test. Results The male and female ratio of solid lung cancer(60:45) were higher than that of fGGO (18:30, X~2 value 5.09, P<0.05). But no differences were found in age and size of lesion (P value 0.200,0.673). For solid lung cancer, the incidence of round shape (n=101), irregular (n=4), speculation (n=60), vacuole sign (n=12) and air bronchograms (n=0) were significantly different from those of fGGO (38, 10, 19,25 and 7, respectively), and the corresponding (X~2 values were 11.48,4.07,29.70 and 22.38 respectively, P<0.05). No differences were found in lobulation, cusp angle, spine-like process, well-defined, coarse, ill-defined interface, honey-combing, pleural indentation sign and blood vessel cluster sign (there are 85,0,33,5,100,0,0,59,35 cases for solid cancer, and 42,1,15,3,45,0,2,32,16 for fGGO (X~2 values 1.00,2.20, 0.00,0.15, 4.43,1.50, 0.00, P>0.05). Conclusions Malignant fGGO and solid lung cancer manifest mostly similar MSCT features. The frequency of irregular shape, vacuole sign and air bronchograms was higher in fGGO than in solid lung cancer to some degree, but speculation is more infrequent in fGGO, which may be attribute to thepathological type and basis of tumor.
