2.Establishment and Application of Information System for Hospital Infection Management

OBJECTIVE To establish and apply the hospital infection information system,and study the risk factors and occurrence pattern hospital infection,to control the incidence of hospital infection.METHODS The hospital infection management software was developed by the hospital with the Hope-Bridge Technology Company of Guangzhou.RESULTS This system can joint with the hospital information system(HIS) seamlessly,as a subsystem of HIS,and be utilized in the real-time monitoring for all inpatients.It has the function of inquiry,statistics and analysis,it can monitor correctly and comprehensively on infection case,hospital environmental hygiene,usage of antibiotics and intervening operations.CONCLUSIONS The hospital infection management software is convenient and easy to operate,which can enhance the efficiency and effect of the management of hospital infection.
5.Primary culture of human vascular smooth muscle cells and their calcification mode

Objective To establish a calcification mode in vitro of human vascular smooth muscle cells (HVSMCs) induced by β-GP. Methods Primary HVSMCs were obtained from human embryo by plant method and confirmed by stain with α-sm-actin antibody. The cells after 4-6 passages were divided into two groups.The control group was incubated with normal DMEM medium while the calcification group was incubated with the medium containing 10 mmol/L β-glycerophosphate for 10 days.Calcification was confirmed by alizarin red S stain and alkaline phosphatase(ALP) assays. Results The primary cells observed by S-P stain were positive and the cells after being stained were pale yellow. After being induced with β-GP, the cells of calcification group began concentric growth and formed vesicles. Alizarin red S staining showed that the reaction of calcifying nodules was red,ALP activity was higher than that of controls at various time points(4 d,6 d,8 d and 10 d,P<0.01 ).Conclusion The HVSMCs could be induced into calcification in vitro by β-GP, and this model contribates to further studies of vascular diseases.
6. Effect of STAT 5 decoy oligodeoxynudeotide on tumorigenicity of K 562 cells in nude mice

Objective: As a potential gene therapeutic method, transcription factor decoy strategy has been used to block the transcription function of multi-transcription factors. This study aims to further verify the inhibitory effect of signal transducer and activator of transcription 5 (STAT 5) decoy oligodeoxynucleotides (ODN) on tumor growth in nude mice. Methods: The xenografted tumor model was established by subcutaneously injecting leukemia K 562 cells in nude mice. Liposome-ODN complex was directly injected into tumor body once daily every 3 days. The tumor growth and tumor size were measured everyday. Nude mice were sacrificed at the end of the experiment. The tumor was removed and weighed. RT-PCR and Western blotting were performed to detect mRNA and protein expression of bcl-xL, cyclin D1, and c-myc, respectively. Results: The tumorigenicity of K 562 cells was significantly inhibited by decoy ODN. The tumor weight was markedly reduced in decoy ODN group than mutant ODN group [(0.485 ± 0.178) g vs (0.928 ± 0.223) g, P < 0.05 ]. The tumor growth inhibition rate was 47.7%. RT-PCR and Western blotting showed that the mRNA and protein expression level of bcl-xL, cyclinD1, and c-myc were down-regulated. Conclusion: Transcription factor decoy strategy effectively blocks transcription of STAT 5 target genes in xenografted tumor of nude mice.
7.STUDIES ON THE EXPRESSION CONDITION OF SEGMENT OF GABA_A RECEPTOR a1 SUBUNTT IN E. COLI

In order to improve the expression level of segment of GABAA receptor a1 subunit in E. coli, growth conditions of the recombatant, which influence the final yield of protein expression, including growth medium, inoculation ratio, temperature, pH, rotation speed, inducing time and concentration of IPTG and so on, were studied in shaking flasks. The results indicated that, with 3% inoculation ratio, cultured 3.5 hours at 37℃, and then induced 5 hours by IPTG at 32℃, the yield of GABAA receptor protein was 95mg/L and the biomass was 3.25 g/ L. In contrast, using a 16 L stirred fermentor instead of shaking flasks, the highest level of the protein expression, 136mg/L with 4.95g/L of biomass, was achieved after fermenting 5.5 hours.

With its important functions as load-bearing, shock absorption, stabilization, lubrication and congruency, the meniscus plays an important part in the complex biomechanics of the knee joint. Therefore it is still a challenging question for knee surgeons to reconstruct the function of meniscus which was injured or resected. This review details current efforts to transplant or to replace a meniscus.
9.The relationship of the expression of thymic stromal lymphopoietin in nasal polyps tissues and Th2 inflammatory response.

OBJECTIVE:
To explore the relationship of the expression of thymic stromal lymphopoietin (TSLP) in nasal polyps tissues and the Th2 inflammatory response.
METHOD:
Sixty patients with nasal polyps were collect ed. The immunohistochemical staining method was used to detect the expression of TSLP in nasal polyps tissues and ELISA method to the expression of IL-4, IL-5, IFN-gamma, IL-13 and analyzed the correlation between them.
RESULT:
The expression of TSLP in nasal polyp tissues was higher than that in normal inferior turbinate mucosa (P < 0.05). The expression level of IL-4, IL-5, IFN-gamma and IL-13 in nasal polyps tissues were significantly higher than those in control group (P < 0.05). TSLP staining was a statistically significant positive correlation with IL-4, IL5 and IL-13 (r = 0.475, 0.594 and 0.582, respectively, P < 0.01), while inverse correlation with IFN-gamma (r = -0.614, P < 0.01).
CONCLUSION
The high expression of TSLP might promote T cell differentiation towards Th2, and participated in the occurrence/development of nasal polyps, aggravated the nose Th2 inflammatory response.
