1.Effect of glutaminase 1 inhibitor on carbon tetrachloride-induced liver fibrosis in mice

Objective To investigate the effect of glutaminase 1(GLS1)specific inhibitor BPTES[bis-2-(5-phenylacetamido-1,3,4-thiadiazol-2-yl)ethyl sulfide]on the liver fibrosis in the mouse model of liver fibrosis induced by carbon tetrachloride(CCl4).Methods Male C57BL/6J mice were intraperitoneally injected with olive oil(control group),10%CCl4(10 μL/g,model group)or 10% CCl4(10 μL/g)+ BPTES(10 mg/kg,treatment group),with 10 mice in each group,two doses a week for four weeks to establish liver fibrosis model. Collagen deposition in mouse liver tissue was observed by Sirius red staining. The expression levels of actin alpha 2(Acta2),collagen typeⅠalpha 1(Col1a1)GLS1 and GLS1 protein were detected by qRT-PCR and immunohistochemical staining.Results Compared with the control group,the liver tissue of mice in the model group was generally enlarged,the surface was not smooth and granular,and the ratio of liver mass to tibia length significantly increased(t = 2. 979,P < 0. 05);The Sirius red positive area of collagen deposition increased signifi-cantly in the liver tissue of mice in the model group(t = 7. 661,P < 0. 01),the relative expression levels of Acta2 and Col1a1 significantly increased(t = 4. 335 and 5. 319,respectively,each P < 0. 01),and the mRNA and protein levels of GLS1 significantly increased(t = 5. 319 and 9. 725,respectively,each P < 0. 01). However,compared with the model group,the BPTES treatment group had a reduction in liver mass,a significant reduction in the Sirius red positive area of collagen deposition in liver tissue(t = 7. 427,P < 0. 01),and a significant reduction in the relative expressions of Atca2 and Col1a1(t = 3. 713 and 2. 628,respectively,each P < 0. 05).Conclusion Inhibition of GLS1activity can significantly improve the degree of liver fibrosis induced by CCl4,providing a new idea for the treatment of liver fibrosis.

6.Therapeutic progress and biocompatibility of cerebral intravascular stent

OBJECTIVE: To investigate therapeutic progress of cerebral intravascular stent, and to evaluate biocompatlbility with host.METHODS: Articles were collected from CNKI and Medline database with the keywords of "cerebrovascular disease, stent, and therapy" in both Chinese and English from 1989 to 2009. Among 53 articles, 22 were included according to inclusion and exclusion criteria; while the included articles were summarized in the fields of therapeutic progress of cerebral intravascular stent,complication following cerebral intravascular stent implantation, and biocompatlbility of cerebral intravascular stent in order to investigate the biocompatibility of various stents.RESULTS: Cerebral intravascular stent was mainly used to treat cerebral artery stenosis, cerebral aneurysm, venous sinus stenosis, and thrombus. Complications following cerebral intravascular stent implantation included carotid sinus syndrome,hypertransfusion syndrome, cerebral angiospasm, thrombosis, and restenosis. Pre-enlargement prior to implantation in the stenotic region played an important role in avoiding deformation and displacement of stent. Restenosis correlated to stent types following cerebral intravascular stent implantation. For example, metal stent could promote thrombosis; however, polymer which had an excellent biocompatibility to vessel wall was superior to metal stent, thus it could prevent endomembrane proliferation following implantation. Metal-coated stent could inhibit aggregation of platelet; additionally, drug stent could effectively prevent restenosis via high-concentration drug release for a long term.CONCLUSION: Cerebral intravascular stent is considered as an ideal tool to treat cerebrovascular disease. Metal stent has a poor compatibility, but polymer stent, coating stent, and drug stent have a good compatibility.
7.Intermingled transplantation of allochthonous and autogenous bone in the treatment of tumor-like legion

AIM: To approach the best therapeutic measures after allochthonous intermingled with autogenous bone transplantation in children with tumor-like legion. METHODS: Forty children received bone transplantation between from March 2003 to March 2006 were selected from First Hospital Affiliated to Zhengzhou University. The guardians of Children all knew and agreed with the items. Subjects were divided into two groups according to the bone grafts with 20 children in each group: Compound transplantation group and autoplastic transplantation group. ① Subjects in compound transplantation group were transplanted with compound bone of allochthonous bone, autogenous bone and marrow with 160 thousands of gentamicin, 400 mg of vacocin and 5 mg of dexamethasone added. Allochthonous bone was obtained from dried radiosteriliaed bone, which was made by Shanxi Aorui Biomaterial Co., Ltd. Autogenous bone was gotten from the iliac bone of children. ② Subjects in autoplastic transplantation group were transplanted with simple illac bone got from subjects. Meanwhile, 16 thousands of gentamycin was adopted in the transplantation without using dexamethasone. ③ 5 mg of dexamethasone was added in to the allochthonous bone used for the surgery of subjects in compound transplantation group, and intervenous drop infusion of hormone was given to subjects after the surgery at proper dose, while children in autoplastic transplantation group received no hormone during and after the surgery. ④ Post-operation draining was performed, and the draining flow within 24 hours less than 20 mL was taken as the index of extubate. ⑤ The allochthonous bone and autoplastic bone were immersed into 160 thousands of gentamicin and 400 mg of norvancomycin used for patients in the compound transplantation group. Patients in both groups were administrated with antibiotic drugs of blue mold for fear of infection. ⑥ The duration of recovery to normal body temperature, leukocyte count and erythrocyte sedimentation rate (ESR) as well as the time of reservation of drainage tube of patients in both groups were observed after the surgery after operation, and follow-up was conducted regularly. RESULTS: Totally 40 patients were involved in the analysis of results, and no one withdrew from the study. ① The duration of recovery to normal body temperature, leukocyte count and ESR as well as the time of reservation of drainage tube of patients in compound transplantation group were longer than those in autoplastic group [(10.95?2.63,13.85?2.68,25.90?2.53,13.15?3.01) days;(6.95?1.57,8.80?1.96,15.30?2.27,6.40?1.98) days,P
8.Development of Hyperpurificatory Biosafety Cabinet

Objective To create a sterilized condition in field operation and the daily grind avoiding the intercross contamination between the experimentation and outside. Methods The air in the experimentation was filtrated by primary and advanced filter. High intense ultraviolet was used to sterilize the air, and negative compression was kept in the experiment condition. Results Hyperpurificatory biosafety cabinet guaranteed the sterilization condition in field operation and the daily grind, avoiding the intercross contamination. Conclusion Hyperpurificatory biosafety cabinet can be used in medical treatment, pharmacy, food industry, electrical industry, instrumentation, bioengineering, scientific tests, especially biology species identification in biological warfare.

10.Reversal of drug resistance of trastuzumab-resistant human breast cancer cells by recombinant adenovirus carrying PTEN gene

Objective:To study the role of adenoviral-mediated PTEN gene transfection in reversing drug resistance of trastuzumab-resistant human breast cancer.Methods:Recombinant adenoviruses carrying PTEN gene(AdPTEN) were constructed and transfected into trastuzumab-resistant human breast cancer cell line BT474.Proliferation and apoptosis of BT474 cells treated with AdPTEN and trastuzumab were measured by MTT assay and FCM.Time course of BT474 cells apoptosis induced by AdPTEN was analyzed by detection of DNA fragmentation.Western blotting was employed to measure phosphorylated-Akt expression levels in AdPTEN treated BT474 cells.The nude mice model of BT474 cells was employed to observe the effects of AdPTEN and trastuzumab on trastuzumab-resistant human breast cancer in vivo.The expression of PTEN gene,cancer cells apoptosis and ultrastructural changes were evaluated after the mice were sacrificed.Results:We successfully constructed recombinant AdPTEN;and the titer of the recombinant adenovirus was about 4.2?1011TCID50/ml.PTEN gene expression was identified by PCR,RT-PCR and Western blotting assay.Combinatorial AdPTEN and trastuzumab significantly suppressed the proliferation of BT474 cells and induced the apoptosis.The percentage of apopotosis cells treated with AdPTEN was(20.7?5.82)%,companied by cells cycle arrest in G1 phrase(P
