1.Determination of Tiopronin in Tiopronin/Glucose Injection by HPLC

OBJECTIVE:To establish an HPLC method for the determination of tiopronin in Tiopronin/glucose injection.METHODS:The separation was performed on Diamonsil-C18 column with flow rate of 1 mL?min-1.The mobile phase composed of 0.07 mol?L-1 NaH2PO4.The detection wavelength was 210 nm.RESULTS:Tiopronin showed good linearity within range of 24.17~56.39 ?g?mL-1(r=0.999 6).The average recovery was 99.06% (RSD=0.7%).CONCLUSION:The method is simple,rapid,accurate and specific for the determination of tiopronin in Tiopronin/glucose injection.

3.The short-and log-term effects and side effect comparisons of concurrent chemoradiotherapy in the treatment for kazak and han nationality patients with advanced esophageal carcinoma in Xinjiang provenience

Objective To study the short-term curative effects and side effects of the three-dimensional conformal radiotherapy combined with chemotherapy in advanced esophageal cancer in Xinjiang provenience.Methods One hundred and twelve cases were divided into Kazak and Han nationality group according to the different nations.Chemotherapy was DF regime:5-Fu 1000 ms/m2+DDP 20 mg/m2,d1-4,21 days as a cycle,total 4-6 cycles.For both groups,3DCRT was carried out by conventional fractionation with total dose of 60-66 Gy/30-33frin 6-7w.Results The following-up rate was 100%.The number of pailents followed up at two years wero 23 and 27 in Kazak and Han nationality group,respectively.The immediate effective rates(CR(complete remission)+PR(partial remission))were 77%(34/44)and 85%(58/68)in ban and kazak group,respectively(X2=4.89,P=0.180).The 1-and 2-year survival rate were 75%,59%and 52%,40%(X2=1.71,P=0.191)in Kazak and Han group,respectively.The 3-4grade radioesophagitis were 5%(2/44)and 7%(5/68)in kazak group and han group,respectively(X2=1.66,P=0.435).The 3-4 grade radiopneumonia were 2%(1/44)and 1%(1/68)in kazak group and ban group,respectively(X2=0.99,P=0.608).The 3-4 grade haematolosical toxicity were9%(4/44) and 29%(20/68 in kazak and in han group,respectively(X2=6.57,P=0.037).Conclusions There was no significant difference in short-term curative effects between patients from han and kazak nationality with advanced carcinoma of esophagus who received the three-dimensional conformal radiotherapy combined with chemotherapy.but Kazak people showed better tolerance.
4.Construction and Identification of Recombinant Plasmid of p3XFLAG-CMV7-NICD1

Objective To construct the eukaryotic expression vector of Notch1intracellular domain,p3XFLAG?CMV7?NICD1,so as to prepare for the further research and exploration of effect of Notch1 on promoting epithelial?mesenchymal transition of human lens epithelial cells. Methods The cDNA fragment was reversely transcribed by RT?PCR from total RNA extracted from the SRA01/04 cells and was encoded with the specific am?plification?targeted NICD1was obtained from the SRA01/04 cells,then the cDNA fragment was inserted into p3XFLAG?CMV7 to transcribe Esche?richia coli DH5α. And the recombinant plasmid was extracted after bacterial screening by LB plating medium and confirmed by the restriction endo?nuclease digestion and DNA sequencing. Results The target gene obtained had the same molecular size as predicted. It was indicated that recom?bined p3XFLAG?CMV7 plasmid contained correct recombinant human Notch1 sequences and p3XFLAG?CMV7?NICD1 was constructed success?fully. The western blotting showed protein NICD1 expressed in SRA01/04 cells transfected with p3XFLAG?CMV7?NICD1. Conclusion The suc?cessful construction of p3XFLAG?CMV7?NICD1 will provide a foundation for a further study studies in on the effect relationship of Notch signaling pathway and in posterior capsular opacification(PCO)after cataract extraction.
5.The effects of TCF21 genes on radiation and chemotherapy sensitivity of A549 lung adenocarcinoma cells

Objective To investigate the transcription factor 21 (transcription factor 21 ,TCF21) gene on human lung cancer A549 cells sensitivity to chemotherapy .Methods Using lentivirus technology in A549 lung cancer cells highly expressed genes TCF21 ,fluorescence quantitative PCR ,Western Blot analysis were used to analyse the expression of the target gene ,MTT assay was used to detect the effect of TCF21 lung adenocarcinoma A549 cells on cisplatin (cis‐Dichlorodiamineplatinum ,DDP) chemosen‐sitivity ,and colony assay was used to detect the effect of overexpression of TCF21 lung adenocarcinoma A549 cells on radiosensitiv‐ity .Results After 72 h ,with the increasing concentration of DDP (0 ,0 .625 ,1 .250 ,2 .500 ,5 .000 ,10 .000 mg/L) ,corresponding in‐hibition rates in each group increased ,and the inhibition rate of the high expression group was significantly higher in empty vector group and untransfected group (P<0 .05) ,no significant difference between the two then;overexpression TCF21 group with drug concentration and time and increase the rate of high expression inhibition corresponding increase (P<0 .05) ;after receiving X radi‐ation ,non‐transfected group ,untransfected plus radiotherapy group ,vector group ,vector plus radiotherapy group ,high expression and high expression + radiotherapy colony formation rates were:95 .17% ± 2 .85% ,88 .20% ± 2 .03% ,93 .80% ± 4 .17% , 85 .60% ± 2 .42% ,71 .67% ± 3 .21% ,56 .00% ± 2 .65% .Conclusion TCF21 gene expression can significantly enhance the sensitiv‐ity to radiotherapy and chemotherapy DDP A549 lung cancer cells .

The endocrine system with the normal aging is characterized by a progressive loss of reserve capacity,resulting in a decreased ability to adapt to environmental demands.This loss of homeostatic regulation reflects important alterations in hormonal synthesis,metabolism,and action.However,these changes may have no significant clinical manifestations,especially under non-stress conditions.This review discusses the endocrine system in the elderly in general.
7.Effect of TCF21 on A549 human lung cancer cells in nude mice

Objective To investigate the effect of TCF21 on human non-small cell lung cancer A549 cells , and to reveal the role of TCF21 in the development of NSCLC. Methods Overexpression of TCF21 in NSCLC A549 cells was mediated by lentivirus vector. TCF21 protein expression was identified by Western blotting assay. The experimental group, negative control group and the blank control group were set up. A549 cells were subcuta-neously seeded in BALB/c nude mice on the left armpit. Results TCF21 was successful overexpressed in the ex-perimental group. Compared with the negative control group and the blank control group , the tumor growth was slow, and the final tumor volume was significantly reduced in the experimental group. Conclusion Overexpression of TCF21 can inhibit the tumor growth of NSCLC in nude mice , indicating that TCF21 may play an important role in NSCLC development.
8.A Correlation Study on Suicide Ideation,Depression and Levels of Mental Health among Middle and Primary School Teachers in Rural Areas

Objective: To investigate the relationship between suicide ideation, depression and levels of mental health,and provide basis for enhancing levels of mental health of middle and primary school teachers in rural area. Methods: Stratified sampling method was used to draw 423 middle and primary school teachers in Jiangxi province. They were investigated with self-administered questionnaire, Beck Depression Inventary(BDI) and SCL-90. Results: The rate of expressing suicide ideation was 32.2%. Multivariance logistic regression analysis revealed that the risk factors for suicide ideation were depression, anxiety, photic anxiety hostility and psychoticism. Conclusion: Suicide ideation is more common among middle and primary school teachers in rural areas. Emphasis should be placed on mental health to prevent suicide.
10.Glucagon-Like PeptideⅠand Targeted Differentiation of Stem Cells

Diabetes mellitus has become one of the diseases which threaten the heath of human being in the 21st century.A goal of research in diabetes is to find a way to increase the number of functional insulin-producing cells. Islet transplantation has been considered to be the most effective approach to cure type Ⅰ and part of type Ⅱ diabetes mellitus.This approach, however, is severely limited by an inadequate supply of donor islets available for transplantation.Moreover, recent progress of stem cells research has shown that stem cells may act as a new source of islet transplantation in diabetes mellitus treatment. Recent evidence indicates that Glucagon-Like PeptideⅠ(GLP-1) plays a very important role in targeted differentiation of stem cells into Insulin-Producing Cells and pancreatic development. GLP-1 is an intestine-derived insulinotropic hormone that stimulates glucose dependent insulin production and secretion. GLP-1 can induce differentiation of stem cells into insulin-producing cells, which is achieved by up regulation of PDX-1 expression.PDX-1 is a transcription factor critical for pancreatic development and endocrine cell neogenesis and a marker for pancreatic stem cells. These new findings suggest an approach to create Insulin-Producing cells in vitro by expanding stem/progenitor cells and then to convert them into Insulin-Producing cells by treatment with GLP-1. Thus GLP-1 may be a means by which to create Insulin-Producing cells ex vivo for transplantation into patients with insulinopenic type Ⅰ diabetes and severe forms of type Ⅱ diabetes. This article reviews recent progress about GLP-1 and targeted differentiation of stem cells induced by GLP-1.
