1.Construction of subtracted cDNA library by suppression subtracted hybridization for differentially expressed genes in Mycobacterium tuberculosis

Objective: To isolate Mycobacterium tuberculosis H37Rv and H37Ra genes especially,and to construct two cDNA libraries by using suppression subtractive hybridization (SSH).Methods: We used suppression subtractive hybridization (SSH) to clone the differential genomic genes between Mycobacterium tuberculosis virulence strain H37Rv and attenuated strain H37Ra.After two times of subtractive hybridization and two times of PCR,the products of last PCR amplification were inserted into pGEM-T Easy vector and be transformed into the E.coli DH5α and screened of blue and white clones of the transformants.The subtracted cDNA library of differentially expressed genes were identified by RT-PCR.Results:High or especially expressed genes as tester had been obtained by SSH in correctitude reaction (H37Rv as tester) and reverse reaction (H37Ra as tester),the cDNA libraries of A and B of Mycobacterium tuberculosis H37Rv and H37Ra were successfully constructed.90% of the colonies were white clones,the single band of the colonies was 75% and 80%.Conchision:The cDNA libraries of Mycobacterium tuberculosis H37Rv and H37Ra were successfully constructed using SSH technique,which lay a solid foundation for screening and cloning new specific differentially expressed genes in them.
2.Diagnosis and Management of Mondor Disease

Objective To review the epidemiology, etiology,pathology,clinical presentation,diagnosis and treatment,recurrance and prognosis of Mondor disease.Methods Literatures on Mondor disease were reviewed,and our study of Mondor disease was also included.Results Mondor disease is difficult to diagnose,but is with good prognosis in most patients.It's unclear whether Mondor disease is associated with the etiology of breast cancer.Conclusion The diagnosis and therapy of Mondor disease are important,especially when it is accompanied with breast cancer.
3.Correlation study on the ADAMTS-4 in early brain injury after experimental subarachnoid hemorrhage and blood-brain barrier permeability in rats

Objective To establish the rat model of experimental subarachnoid hemorrhage (SAH) and to detect the expression of ADAMTS-4 in early brain injury(EBI) after SAH in rats .Methods Totally 72 adult male SD rats were randomly divided into 2 groups ,the control group and SAH group .The expression of ADAMTS-4 and the content of brain Evans Blue(EB) were detected . The ADAMTS-4 inhibitor α2-macroglobulin was used to study the relationship between the expression of ADAMTS-4 and blood-brain barrier permeability .Results The expression of the mRNA and protein of ADAMTS-4 increased dramatically at 24 h and the contents of EB increased progressively at 12 h after SAH .The protein expression of ADAMTS-4 and the content of brain EB was positive correlated(r=0 .917 ,P<0 .05) .The expression of ADAMTS-4 was inhibited by α2-macroglobulin(P<0 .05) and the con-tents of EB decreased dramatically (P<0 .05) .Conclusion ADAMTS-4 may be involved in the pathological progression of early brain injury after SAH .

5.Effect of selective head cooling therapy on cerebral blood flow and oxygenation in neonates with hypoxic-ischemic brain damage

Objective To study the changes of cerebral blood flow and cerebral oxygenation during selective head cooling (SHC) in neonates with hypoxic-ischemic brain damage (HIBD). Methods Thirty-eight neonates with moderate and severe HIBD were enrolled in two groups: SHC group (n=18) and normal temperature group (n=20). The gestational age of all subjects were more than 38 weeks and Apgar score in 5 minutes after birth was less than 5 scores. Also, nerve system symptoms and electroencephalogram abnormal was found in 6 hours after birth. Neonates in SHC group had nasopharyngeal temperature maintained 34℃ and in normal trmerature guoup had rectum temperature maintained about 36℃. Seven healthy neonates were as control group. Three groups were observed 84 h. Cerebral blood flow velocity (CBFV) of middle cerebral artery (MCA) was measured with transcranial Doppler (TCD) at 6, 12, 24, 48, 72 and 84 hours after birth. Cerebral cytochrome aa3 (Cytaa3 ) was monitored continuously throughout the study using near infrared spectroscopy (NIRS). Objective:To study the mechanism of protein kinase C regulating CD44 gene expression in vascular endothelial cells.Methods:Human umbilical vein endothelial cells (HUVECs)were taken as study model.The extract of Raf-1 kinase by immunoprecipitation was used for the Western blot analysis,and its activity was determined by enhanced chemiluminescene assay.CD44 gene expression was detected with RT-PCR,and phosphorylation was measurated by autoradiography.Results:CD44 phosphorylation in HUVECs was enhanced by 10ng/ml PMA treatment as compared with untreated cells, which reached the highest level at 30 minutes. Raf-1 kinase activity increased significantly after exposure to 10ng /ml PMA, and 0.05 ?mol/L Calphostinc could inhibit the role of Protein kinase C(PKC). 7.Analysis of the Arrangement of the Military Medical Curriculum for Cadets

Objective:By analyzing the arrangement of the military medical curriculum for cadets,some advantages and disadvantages about the current course arrangements are discussed.Methods: To ensure the regain rate,188 cadets,who are undergraduates in our university,answered questionnaires in class.Then the results were analyzed by statistics.Later cadet representatives had an informal discussion with investigators.Results:According to the results,the arrangements of the military medical curriculum for cadets in the military medical universities are qualified on the whole,can meet cadets' cultivation needs.But to accommodate the requirement of military medical talented men in the 21-century,some suggestions about courses information,such as curriculum category,curriculum sequence,curriculum contents,curriculum duration and etc,are given to the related teaching departments.Meanwhile,the results of this study can provide some information for the cultivation of national defense students in the military medical university.
8.Effects of four extracts of traditional Chinese medicinal materials on the proliferation of human lung cancer A549 cell line

AIM To study the influence of four extracts of traditional Chinese medicinal materials(sodium cantharidinate, matrine, cinobufotain and sodium ferlate) on the proliferation of human lung cancer A549 cell line and breast cancer MCF7 cell line;the possible mechanism of sodium ferlate on the inhibition of A549 cells. METHODS The inhibition of cell proliferation was determined by MTT assay and cell cycle was determined by flow cytometry. RESULTS All four extracts of traditional Chinese medicinal materials showed growth inhibition to breast cancer MCF7 cells while only sodium ferlate showed inhibition to lung cancer A549 cells. Synergistic inhibition was found when sodium ferlate was combined with each of the three commonly-used chemotheraputic drugs. Sodium ferlate could induce A549 cell apoptosis. CONCULSION The inhibition of cell proliferation induced is by sodium cantharidinate, matrine, and cinobufotain is quite different between different cancer cell lines. Sodium ferlate can inhibit cancer cell proliferation and show synergistic action while combined with chemotheraputic drugs. The mechanism of sodium ferlate on A549 cell proliferation seem to be related to the cell apoptosis.
9.A Survey and Analysis of the Current Situation of Military Medical Officers' Continuing Medical Education

10.Effects of general and epidural anesthesia on the formation of deep vein thrombosis after total knee arthroplasty:a meta-analysis

BACKGROUND:Although the therapeutic effect of total knee arthroplasty for severe gonarthritis is reliable, deep vein thrombosis also occurred in many patients. Therefore, deep vein thrombosis has been a major reason for unexpected death after total knee arthroplasty. OBJECTIVE:To study the incidence of deep vein thrombosis after total knee arthroplasty with general and epidural anesthesia. METHODS:We searched“PubMed”,“Medline”,“Elsevier”,“Embase”, Cochrane library,“Wanfang”,“China National Knowledge Infrastructure”for papers published from January 2000 to March 2015. The mesh words were“total knee arthroplasty”,“TKA”,“thrombosis”,“DVT”,“randomized control ed trial”,“RCT”for studies concerning deep vein thrombosis after total knee arthroplasty with different anesthetic methods. The double blind method was used for data extraction and assessment of literature quality. Revman5.3 software was utilized to analyze the extracted data using meta-analysis. RESULTS AND CONCLUSION:There were 8 studies including 885 patients. The heterogeneity of the included studies was very low (I2=0, P=0.72). The combined OR was 0.25 (95%confidence interval:0.18, 1.35), indicating that continuous epidural anesthesia in total knee arthroplasty could apparently reduce the possible formation of postoperative deep vein thrombosis. Because of lack of the changes in coagulation factors under general anesthesia and epidural anesthesia, the mechanism underlying epidural anesthesia to reduce the incidence of deep vein thrombosis deserves further investigations. Among patients with deep vein thrombosis, continuous epidural anesthesia could diminish the possible formation of deep vein thrombosis. Thus, epidural anesthesia should be used firstly for knee replacement in patients with high risk of deep vein thrombosis.
