1.Determination of Peoniflorin in Bazhen(Eight Ingredients) Teabag by HPLC

OBJECTIVE: To develop an HPLC method for determination of peoniflorin in Bazhen(eight ingredients) teabag.METHODS:The samples were separated on Diamonsil C18 column(250 mm?4.6 mm,5 ?m) with acetonitrile-water(17∶83) as mobile phase under a detection wavelength of 230 nm.RESULTS: The linear range for peoniflorin was 0.105 3~ 2.106 0 ?g(r=0.999 9) and its average recovery rate was 98.58%(RSD=1.17%,n=9).CONCLUSION: The method is simple,rapid,accurate and applicable for the quality control of Bazhen(eight ingredients) teabag.
2.ESTROGEN AND PROGESTERONE RECEPTOR IN NONEPITHELIOGENIC OVARIAN TUMORS

Tissue samples from 42 patients with nonepitheliogenie ovarian tumors were assayed for estrogen receptor (ER) and progesterone receptor (PR) by Lce's fluorescent histochemical method. 2 of 4 theca cell tumor, 6 of 8 granulosa cell tumor had ER and PR. Among the 6 cases of granulosa cell tumor, 4 cases exhibited a high level of receptors (ER、PR positsve cell: 40%~50% of total tumor cells). As to teratomas, the epithelial cells of sweat glands, and cylindrical epithelia, as well as chondrocytes were ER PR positive. No ER-and PR-positive cells were found in endodermal sinus tumors, drsgerminomas and fibromas. Of 4 mataststic ovarian tumors, which were from gastrointestinal adenocarcinoma, tbe ER-and PR-positive cells were found in 1 specimen. The findings suggested that hormone-related therapy might be worthy of trying on thc patients with receptor positive granulosa cell tumor.
3.The impact of self- efficacy on negative automatic thoughts and quality of life in cancer patients

Objective To explore the impact of self-efficacy on negative automatic thoughts and quality of life in cancer patients who received chemotherapy. Methods 106 patients who diagnosed cancer and received chemotherapy were assessed by General Self-Efficacy Scale ( GSES), WHO quality of life questionnaire-brief (WHOQOL-BREF) and Automatic Thoughts Questionnaire(ATQ) . According to the score of GSES, the highest 27% were assigned to higher scores group, and the lowest 27% were assigned to lower scores group, then compared two groups' WHOQOL-BREF and ATQ with t -test and Pearson correlation analysis. Results The mean score of GSES of 106 patients was ( 22.32 ± 8.61 ) points, and the low self-confidence were 55.66%, the mean score of GSES of higher scores group was( 28.64 ± 7.64)points, and the lower scores group was (21.37 ± 5.78 )points, and the two groups have significant difference ( t = 4.18, P < 0. 01 ). The score of ATQ of higher scores group(63.71 ±7.94) was significantly lower than that of lower scores group(86.57 ±9.87), the total and all dimensions score of WHOQOL-BREF of higher scores group was higher than that of lower scores group, respectively.The score of GSES had positive correlation with the score of WHOQOL-BREF , and had negative correlation with the score of ATQ. Conclusion Cancer patients have lower level of self-efficacy, and it is closed correlated with quality of life and negative automatic thoughts.
4.The effect of family care index on incidence of post-traumatic stress disorder after the first chemotherapy of cancer patients

Objective To investigate the effect of family care on incidence of post-traumatic stress disorder after the first chemotherapy of cancer patients.Methods According to the assessment score of Family APGAR Index,67 cases of diagnosed cancer patients with chemotherapy for the first time were divided into bad family function group and good family function group.Then all the patients were assessed by Post-Traumatic Stress Disorder questionnaire(PTSD-SS) and the results were statistically analyzed.Results Among the 67 patients,the APGAR score between 0 to 6 points were 28 cases,account for 41.79% ; 7 to 10 points were 39 cases,account for 58.21%.The score of good family function group was (4.52 ± 1.27) points,the score of bad family function group was (3.86 ± 1.23 ) paints,the difference was significant ( t =2.13,P < 0.05 ).As to the PTSD-SS total score and various factors' scores,the good family function group were higher than that of bad family function group,the difference was significant (P< 0.01 or < 0.05).Cancer patients' APGAR score appeared to be negatively correlated with PTSD-SS score(P< 0.01 or < 0.05).Conclusion The family care index of cancer patients is closely related to their psychological stress,good family function is better psychological stress level is lower.
5.Clinical and Pathologic Features of Primary Hepatic Carcinoma Associated with Hepatitis B Virus

Objective To investigate the clinical and pathologic features of primary hepatic carcinoma(PHC) associated with hepatitis B virus,HBV). Methods 200 cases of the patients with PHC were diagnosed by histopathologic examination and surgery. The history of HBV infection, its infective patterns and abnomal liver function were obtained from the medical history, and the tests of liver function and HBV five markers. The pathologic features of PHC associated with HBV were determined by analysis of histopathologic changes and views of surgery. Results The markers of HBV were positive in 181 out of 200 cases with PHC, and the positive rate of HBV infection was 90.5%. The postitive rate of HBsAg, anti-HBe and anti-HBc was the highest among the five markers of HBV (50.83%). The positive rate of HBsAg and HBeAg and anti-HBc was only 4.97%. The PHC patients without history of HBV infection and asymptomatic carriers of HBV accounted for 61.33% and 6.63%, respectively. The cases with cured hepatitis and no relapse accounted for 8.29%. The cases with chronic hepatitis B and with liver cirrhosis accounted for 22.10% and 1.66%, respectively. Liver functions were slightly abnomal in approximately one-half patients with PHC. Histopathologic features were as following: HCC accounted for 91.95%, and nodular type, macro-type and small hepatic carcinomas accounted for 55.25%, 30.94% and 13.81%,respectively. The distributions of PHC were mainly in right lobe of liver. The incidence of venous cancer embolism was 12.71%. The incidence of liver cirrhosis was 83.32%. Conclusions ⑴There was relationship between HBV and PHC. The main infective pattern of HBV was the positive HBsAg, anti-HBe and anti-HBc;⑵Pathologic lesion of liver was hidden after infection of HBV;⑶The incidence of active cirrhosis was high; and ⑷There were many pathologic features in patients with PHC associated with HBV.
6.The application of magnetic resonance imaging progressive muscular dystrophy

Objective To investigate the MRI features of progressiv e mu scular dystrophy (PMD), and evaluate the diagnostic value of MRI for PMD. Methods Thirty-three biopsy-proved PMD patients underwent MRI of face, scapular, thigh, and leg, including 16 cases of Duchenne muscular dystrophy (DMD), 2 cases of Beck er′s muscular dystrophy, 5 cases of limb-girdle muscular dystrophy, and 10 c ases o f facioscapulohumeral muscular dystrophy. Spin echo sequence, fast spin echo seq uence, and STIR sequence were utilized. Results The signal inte nsity of disease d muscle with DMD, limb-girdle muscular dystrophy, and Becker′s muscular dystr op hy was hyperintense on both T_1-weighted images(T_1WI) and T_2-weighted ima ges (T_2WI). Gracilis muscle of 21 cases, sartorius muscle of 19 cases, semiten dinosus mus cle of 19 cases, and tibialis posterior muscle of 20 cases were relatively spare d. Ten cases with facioscapulohumeral muscular dystrophy displayed two kinds of a bnormal signals: hyperintense on T_2WI and hypointense on T_1WI in all 10 ca ses;hyperintense on both T_2WI and T_1WI in 7 cases. Conclusion The MRI findings in PMD show certain characteri stics: (1)the involved muscles are mainly replaced by fat. (2)On the lower extre mi ty, Gracilis muscle, sartorius muscle, semitendinosus muscle, and tibialis poste rior muscle were relatively spared in DMD, limb-girdle muscular dystrophy, and B ecker′ muscular dystrophy.(3) Facioscapulohumeral muscular dystrophy displays t wo kinds of abnormal signals which represent fat replacement and inflammatory edem a lesion. MRI can provide objective data for clinical diagnosis, therapeutic eva luation, and follow-up. It can also help to decide the accurate localizations f or biopsies.

8.Expression and significance of C/EBPα and CK10 in nasal inverted papilloma.

OBJECTIVE:
The expression of C/EBPα, CK10 in nasal inverted papilloma (NIP) were detected in the study. Further discussed their significance in genesia, development and recurrence of NIP.
METHOD:
Three groups including nasal cavity mucosae (NM 10 cases), nasal polyp (NP 20 cases) and NIP (30 cases) were selected in the study. Expretion of C/EBPα, CK10 were detected by immunohistochemisty PV-6000 method.
RESULT:
(1) The different expression of C/EBPα and CK10 in the group of NM, NP and NIP was statistically significant (P < 0.05). (2) The different expression of C/EBPα, CK10 in the group of benign NIP and NIP with atypical hyperplasia was statistically significant (P < 0.05). (3) The different expression of C/EBPα and CK10 in the group of NIP with recurrence and NIP with no recurrence was statistically significant, P < 0.05, respectively. (4) Our result indicate that the relationship of C/EBPα and CK10 (r = 0.578, P < 0.01) was direct correlation. The difference was statistically significant.
CONCLUSION
In conclusion, the present results describe C/EBPα, CK10 expression in NIP and their possible implication in the regulation of tumor growth and differentiation. C/EBPα and CK10 production may prove useful in terms of a prognostic marker for the recurrence in nasal inverted papilloma.

9.Significances of COX-2, p21, Ki-67 expression and HPV infection in nasal inverted papilloma.

OBJECTIVE:
To investigate the significance of expression of COX-2, p21, Ki67 and HPV in nasal inverted papilloma.
METHOD:
Detecting COX-2, p21, Ki-67 in 30 cases of nasal inverted papilloma (NIP), 20 cases of nasal polyps (NP) and 10 cases of normal nasal mucosa (NM) by two step immunohistochemical method, and HPV virus by flow-through hybridization method.
RESULT:
The positive expression rate of COX-2 and Ki-67 in NIP, NP and NM group was decreased in turn, COX-2 had significant difference in the groups(χ2 = 30.00, P< 0. 05); the positive expression rate of Ki-67 had significant differences between NIP and NM group (χ2 = 8. 533, P<0. 05). The expression of COX-2 in NIP tissues was positively correlate with that of Ki-67 by using Spearman rank correlation analysis (r=0.78, P<0.05). Expression of p21 were not observed in NIP group. The positive rate of HPV was 26. 67% in 30 cases of NIP, all of HPV16 type.
CONCLUSION
COX-2, Ki-67 and HPV infection have certain correlation with the occurrence of NIP. The occurrence of NIP has relationship with inflammatory reaction mediated by COX-2. Ki-67 can well reflect the proliferation activity of tumor cells, and can be used to measure the proliferation rate of nasal inverted papilloma. The COX-2 and Ki-67 have a synergistic role in the pathogenesis of NIP. p21 has no significant relationship with the incidence of NIP. HPV infection is related to the pathogenesis of NIP, but not as a;major factor in the pathogenesis of NIP.

10.Effect of siRNA targeting PML-RARa fusion gene on activity of the acute promyelocytic leukemia cell line NB4.

This study aims to investigate the effects of small interference RNA (siRNA) targeting PML-RARa mRNA on the activity of the acute promyelocytic leukemia cell line NB4. The proliferation inhibition was evaluated by MTT assay and colony-formation inhibiting test. Apoptosis was determined by flow cytometry after siRNA treatment. The results showed that the cell growth of siRNA treated group was inhibited, and the apoptosis of NB4 could be induced. The siRNA targeting PML-RARα mRNA might be a valid therapy of acute promyelocytic leukemia.
