1.Monitoring on main hygiene indicators before and after clean operating rooms are used

Objective To compare the main hygiene indicators before and after clean operating rooms are used,evaluate the influencing factors,and find out the improvement measures.Methods In 2015,some cleaning operating rooms in Chengdu were detected,according to different service years and maintenance status,operating rooms were divided into newly-built group,replacement group,and non-replacement group,change in qualified rate of three groups of clean operating room indicators were analyzed.Results A total of 111 cleaning operating rooms were detected,including 56 newly-built operating rooms,and 55 operating rooms (24 in replacement group,31 in non-replacement group) which have been used for more than 1 years.The qualified rate of air cleanliness in newlybuilt group,replacement group,and non-replacement group were 98.21%,100.00%,and 74.19% respectively,difference among three groups was significantly(P<0.001),the qualified rate of air cleanliness in newly-built group and replacement group were both higher than non-replacement group,while newly-built group and replacement group was not significantly different (P =1.000);difference in bacterial concentration,static pressure difference,and ventilation frequency of air in operating rooms of three groups were all not statistically significant(all P>0.05).Conclusion After clean operating room have been used for one year,air cleanliness declined,there was no significant change in static pressure difference and air exchange frequency,which indicates that when concentration of airborne bacteria is qualified,risk of infection due to unqualified air cleanliness still needs to be paid attention,the replacement of high efficiency particulate air filter in clean operating rooms can significantly improve the cleanliness of operating rooms.
2.Effects of combination of ketamine and N-acetylcysteine on brain damage following cerebral ischemia /reperfusion in mice

AIM: To evaluate the effects of the combination of ketamine (KT) and N-acetylcysteine (NAC) on damage following cerebral ischemia/reperfusion in ICR mice. METHODS: Male ICR mice were randomly divided into seven groups: Sham group, NS (saline 0.1 ml?kg -1 ) group, KT (15 mg?kg -1 ) group, NAC (75 mg?kg -1 ) group, NAC+KT (75+15 mg?kg -1 ) group. (1) ICR mice underwent two hours cerebral ischemia by transient right middle cerebral artery occlusion (tMCAO) and followed 6 h and 24 h reperfusion. Then brains were prepared for the determination of the infarction volume. Before the death, neurological deficits were scored. (2) ICR mice subjected to five minutes ischemia by two common carotid arteries occlusion (2-VO) and followed 0.5 , 2 and 6 h reperfusion. Brains were prepared for the determination of the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the content of MDA. RESULTS: (1) tMCAO produced severe neurological deficits, decreased the average score and brought about large infarction volume. KT, NAC showed the improvement of the average score and reduced infarction volume to some extent, and KT+NAC improved significantly. (2) The content of the MDA, the activities of GSH-Px and SOD in 2-VO mice deteriorated sharply, KT, NAC reduced the content of the MDA, enhanced the activities of GSH-Px and SOD, NAC+KT significantly ameliorated the levels of MDA, increased the activity of SOD and GSH-Px. CONCLUSION: The damage of cerebral ischemia/reperfusion leads to the decrease of neurological score, the increase of infarction volume, the reduction of activities of SOD and GSH-Px and the elevation of MDA. KT and NAC partly relieve the damage, and NAC and KT in combination attenuates the damage more effectively.
3.Peripheral Mohs micrographic surgery for the treatment of extramammary Paget′s disease Wan Miaojian, Ma

Objective To evaluate effectiveness of peripheral Mohs micrographic surgery for the treatment of extramammary Paget′s disease (EMPD). Methods A total of 28 patients with EMPD were treated with peripheral Mohs micrographic surgery. The depth and extent of tumor infiltration were evaluated before the surgery. One day before the surgery, 20% aminolevulinic acid hydrochloride was topically applied to determine and label surgical margins under a Wood′s lamp. After fluorescence-based localization, peritumoral skin tissues were resected and underwent frozen-section examination according to the protocol for Mohs micrographic surgery. Meanwhile, the tumor was resected. After surgery, patients were followed up every 3 - 6 months to detect local recurrence and metastasis. Results Of the 28 patients, 25 were male and 3 were female. Six patients each underwent 3 sessions of frozen-section examination, and 12 patients each received 2 sessions, with an average of 1.86 sessions for each patient. During the follow-up for 5 - 72 months, local recurrence occurred in 3 cases, and 1 patient died of tumor metastasis and uremia after 2 years of follow-up. Conclusion Peripheral Mohs micrographic surgery is a time-saving and effective treatment for EMPD.
4.Effect of H2O2 stress on glutathione production by Candida utilis.

Glutathione (GSH) plays an important role in the responses of microorganisms to the environmental stimulation and stress. The effect of H2O2 stress under different fermentation time and H2O2 concentration as well as continuous stress on GSH fermentation of Candida utilis were investigated in this paper. It was found that low concentration of H202 accelerated GSH production. When treated by low concentration of H2O2 (36 mmol/L), the final concentration of GSH reached 922 mg/L and the intracellular GSH content reached 1.64%, which increased by 7% and 35% than the controls, respectively.

5.In vitro effect of diammonium glycyrrhizinate on the growth of human hair follicles and Wnt/β-catenin signaling pathway

Objective To evaluate the effect of diammonium glycyrrhizinate extracted from the Chinese traditional medicine licorice root on the growth of human hair follicles cultured in vitro,and to detect the expression of wnt/β-catenin signaling pathway-related molecules.Methods Isolated hair follicles were cultured with diammonium glycyrrhizinate at different concentrations of 0.1,0.01,0.001 and 0.000 1 μmol/L for 10 days,and the hair follicles cultured in Williams' E medium without diammonium glycyrrhizinate served as a control group.The length of hair follicles was measured under a microscope every day,the morphologic changes of hair follicles were observed,and photos were taken.Immunofluorescence assay was performed to assess the proliferation of hair matrix cells,as well as to determine the expression of β-catenin,glycogen synthase kinase 3β (GSK3β),phosphorylated GSK3β (p-GSK3β) and lymphoid enhancer factor-1 (Lef1) in the Wnt/β-catenin signaling pathway.Statistical analysis was carried out by repeated-measures analysis of variance and one-way analysis of variance.Results As repeated-measures analysis of variance showed,only 0.01 μmol/L diammonium glycyrrhetate showed significantly promotive effect on the growth of hair follicles compared with the medium alone (P < 0.05),and there were no significant differences in the length of hair follicles between the other concentration groups and the control group.Compared with the control group,the transition to the catagen phase of human hair cycle was delayed in the 0.01-μmol/L diammonium glycyrrhetate group,while it did not change in the other diammonium glycyrrhetate groups and control group.Immunofluorescence assay showed that the number of ki67-positive hair matrix cells was obviously increased in the 0.1-,0.01-,0.001-μmol/L diammonium glycyrrhizinate groups compared with the control group,while there was no difference between the 0.000 1-μmol/L diammonium glycyrrhizinate group and the control group.One-way analysis of variance revealed that the expression of β-catenin,p-GSK3β and Lef1 significantly differed among all the groups (F =12.604,16.65,15.266 respectively,P < 0.05),while no significant difference in the expression of GSK3β was found among these groups (F =1.472,P > 0.05).Least significant difference (LSD)-t test revealed that the expre-ssion of β-catenin,p-GSK3β and Lef1 in the hair matrix cells was significantly higher in the 0.1-,0.01-,0.001-μmol/L diammonium glycyrrhizinate groups than in the control group (all P < 0.05),but there was no significant difference between the 0.000 1-μmol/L diammonium glycyrrhizinate group and the control group (P > 0.05).Conclusion Diammonium glycyrrhetate at the concentration of 0.01 μmol/L shows markedly promotive effect on the in vitro growth of hair follicles,and can increase the proliferative activity of hair matrix cells and delay the transition to the catagen phase,which may be associated with the activation of Wnt/β-catenin signaling pathway.
6.Application of mechanically reinforced 45S5 Bioglass®-derived bioactive glass-ceramic porous scaffolds for bone defect repairing in rabbits.

Objective: To evaluate the application of mechanically reinforced 45S5 Bioglass®-derived glass ceramic porous scaffolds for repair of bone defect in rabbits. Methods: The BG-ZnB powders were added into the 45S5 Bioglass® powder/paraffin microsphere mixtures and were sintered at 900℃ to obtain porous scaffolds with highly bioactive BG-ZnB of 0%, 2% or 4% of mass fraction (denoted as 45S5/ZnB0, 45S5/ZnB2, 45S5/ZnB4). Phase composition, porosity and compression properties of three kinds of as-sintered scaffolds were characterized by X-ray analysis, mercury porosimetry, and mechanical test. Thirty-six male New Zealand rabbits with critical-sized femoral bone defects were randomly divided into three groups (45S5/ZnB0 group, 45S5/ZnB2 group and 45S5/ZnB4 group, 12 for each), and were implanted with three kinds of porous scaffolds respectively. X-ray, micro-CT three-dimensional reconstruction and tissue slice staining were used to detected the efficiency of bone regeneration at 6 and 16 weeks after operation. The growth of newly formed bone was observed using HE, Masson staining and EnVision method. Results: Phase compositions of 45S5/ZnB2 and 45S5/ZnB4 were the same with 45S5/ZnB0, but the average pore size and porosity of the scaffolds were decreased with the increase of BG-ZnB content. 45S5/ZnB2 and 45S5/ZnB4 scaffolds exhibited higher compressive strength, osteogenesis and trabecular density than those of the 45S5/ZnB0 scaffold (all P<0.05). With the mechanical reinforcement of BG-ZnB increased, the content of new bone, collagen type I and osteocalcin increased. Conclusion: Low-melt BG-ZnB-assisted sintering is a promising approach to improve the mechanical strength of 45S5 Bioglass®.

7.Effects of Faecalibacterium prausnitzii supernatant on Th17 cell and IL-17A in dextran sulfate sodium-induced ulcerative colitis mice.

OBJECTIVE:
To explore the protective and therapeutic effects of Faecalibacterium prausnitzii (Fp) supernatant on ulcerative colitis (UC) in mice induced by dextran sulfate sodium (DSS) and the underlying mechanisms.
METHODS:
Forty male mice were randomly divided into a control group, a model group, a treatment group and a prevention group (n=10 in each group). The colorectal histopathologic damage score (HDS) were calculated; the proportion of helper T cell (Th17) in mononuclear cells (MNC) in spleen, the levels of IL-17A and IL-6 in plasma were detected; the mRNA levels of transcription factor retinoic acid-related orphan receptor-γt (ROR-γt), interleukin (IL)-17A and IL-6 in colon mucosa tissues were also determined.
RESULTS:
Compared with the model group, the colon HDS in the treatment group and the prevention group were significantly decreased (both P<0.05), but there was no significant difference in the treatment group and the prevention group (P>0.05). The proportion of Th17 cells in spleen in the treatment group and the prevention group was also remarkably lower than that in the model group (both P<0.01). The levels of IL-17A and IL-6 in plasma in the treatment group were significantly lower than those in the model group (both P<0.05). The mRNA expression of ROR-γt, IL-17A and IL-6 in the colon mucosa tissues in the treatment group were remarkably lower than those in the model group (all P<0.05). But there was no statistic difference in the level of IL-6 in the plasma and the colon mucosa tissues between the prevention group and the model group (P>0.05).
CONCLUSION
Fp supernatant has protective and therapeutic effects on ulcerative colitis in mice induced by DSS, which might be mediated by decrease of Th17 and IL-17A levels in the plasma and the colon mucosa tissues. Fp supernatant also can decrease mice colitis by reducing IL-6 levels.

8.Association between gaseous pollutants and emergency ambulance dispatches for asthma in Chengdu, China: a time-stratified case-crossover study.

OBJECTIVES:
The association between concentrations of sulfur dioxide (SO), nitrogen dioxide (NO), carbon monoxide (CO), ozone (O), and emergency ambulance dispatches (EADs) for asthma was explored in the central Sichuan Basin of southwestern China for the first time.
METHODS:
EADs for asthma were collected from the Chengdu First-Aid Command Center. Pollutant concentrations were collected from 24 municipal environmental monitoring centers and including SO, NO, CO, daily 8-h mean concentrations of O (O-8 h), and particulate matter less than 2.5 μm in aerodynamic diameter (PM). The climatic data were collected from the Chengdu Municipal Meteorological Bureau. All data were collected from years spanning 2013-2017. A time-stratified case-crossover design was used to analyze the data.
RESULTS:
After controlling for temperature, relative humidity, and atmospheric pressure, IQR increases in SO (13 μg/m), NO (17 μg/m), and CO (498 μg/m) were associated with 18.8%, 11.5%, and 3.1% increases in EADs for asthma, respectively. The associations were strongest for EADs and SO, NO, and CO levels with 3-, 5-, and 1-day lags, respectively.
CONCLUSIONS
This study provides additional data to the limited body of literature for potential health risks arising from ambient gaseous pollutants. The results of the study suggest that increased concentrations of SO, NO, and CO were positively associated with emergency ambulance dispatches for asthma in Chengdu, China. Further studies are needed to investigate the effects of individual air pollutants on asthma.

【Objective】 To explore the efficacy and safety of "ditching and ridge removal" with 450 nm semiconductor blue laser in the treatment of large volume benign prostatic hyperplasia (BPH), in order to promote the clinical application of this method. 【Methods】 A retrospective study was conducted on 30 patients with large volume BPH treated with "ditching and ridge removal" with 450 nm semiconductor blue laser in Yingsheng Branch of Tai’an Central Hospital during Sep.and Dec.2023.The laser operation time, level of hemoglobin before and after operation, bladder irrigation time after operation, urinary catheter indwelling time, postoperative hospital stay, and intraoperative and postoperative complications were recorded.The changes of international prostate symptom score (IPSS), quality of life scale (QoL) score, maximum urinary flow rate (Qmax) and post-void residual volume (PVR) were compared before and 1 month after operation. 【Results】 The volume of prostate was (104.5±14.52) mL, the laser operation time was (20.13±2.98) min, and the bladder irrigation time was (20.27±2.56) h.The catheter was removed in all patients 2 days after operation, and all patients were discharged 3 days after operation.One month after operation, the IPSS, QoL, Qmax and PVR were significantly improved as compared with those before operation (P<0.05).No complications occurred during the follow-up. 【Conclusion】 "Ditching and ridge removal" with 450 nm semiconductor blue laser is a new, safe and effective method in the treatment of large volume BPH.
10.Comparison of clinical efficacy of transurethral blue laser vaporization of prostate and transurethral resection of prostate in the treatment of benign prostatic hyperplasia

【Objective】 To compare the clinical efficacy and safety of transurethral blue laser vaporization of prostate and transurethral resection of prostate (TURP) in the treatment of benign prostatic hyperplasia (BPH). 【Methods】 The clinical data of 142 BPH patients undergoing surgery in our hospital during Jan.and Aug.2023 were retrospectively analyzed.The patients were divided into blue laser group (n=72) and TURP group (n=70).The operation time, hemoglobin (Hb) decrease, bladder irrigation time, catheter indwelling time and postoperative hospital stay were compared between the two groups.The changes of international prostate symptom score (IPSS), quality of life score (QoL), maximum urinary flow rate (Qmax), postvoid residual volume (PVR), international index of erectile function-5 (IIEF-5) score and complications before and 6 months after operation were also compared. 【Results】 Six months after operation, the IPSS, QoL, Qmax and PVR were significantly improved in both groups (P<0.05), but there were no significant differences between the two groups (P>0.05). Compared with the TURP group, the blue laser group had shorter operation time \[(47.04±4.03) min vs.(25.46±5.84) min\], shorter postoperative bladder irrigation time \[ (54.17±4.99) h vs.(21.18±4.11) h\], shorter catheter indwelling time \[5(5, 6) d vs.2(2, 2) d\], shorter postoperative hospital stay \[7(7, 8) d vs. 4(3, 4) d\], less Hb decrease \[(9.61±2.16) g/L vs.(4.31±2.34) g/L\], and lower incidence of complications (51.4% vs.4.2%), with significant differences (P<0.05).Six months after surgery, the blue laser group had higher IIEF-5 score than that the TURP group \[(20.06±2.13) vs. (18.93±1.85)\], with significant difference (P<0.05). 【Conclusion】 Both transurethral blue laser vaporization of prostate and TURP can effectively treat BPH, but the former is more effective, for it has advantages of short recovery time, few complications and small impact on the sexual function of patients.
