1.Studies on the optimum fermentation conditions of marine actinomycete ACMA006 with antitumor activity

Objective To increase the yield of antitumor substances from marine actinomycete ACMA006 strain.Methods Fermentation conditions were studied,including combination of medium,the age of strain,ventilatory capacity and cultivation time.Results The results showed that the best medium was as following: soya powder 10g,tryptone 5g,yeast extract paste 10g,soluble starch 15g,glycerol 15g,KH2PO4 0.5g,sea water 1000mL,pH7.2.The best culture conditions were temperature 28℃,the age of the strain 6d,the liquid volume of culture medium 50mL in 150mL Triangular flask,and cultivation time 8d.Conclusion At last 78.1g?L-1 biomass was obtained and the inhibition rate on human liver cancer cells HepG2 was 93.7% when the broth was diluted 4000 times.
2.The role of D-dimer in detection and diagnosis of mycoplasma pneumonia

Objective To investigate the role of D-dimer in the diagnosis and evaluation of mycoplasma pneumonia.Methods The positive rate and concentration of D-dimer in mycoplasma pneumonia,bacterial pneumonia,viral pneumonia and healthy controls were detected and compared.The positive rate and concentration of D-dimer in patients with systemic inflammatory response syndrome(SIRS)and without SIRS were observed.Changes of D-dimer concentration in patients with mycoplasma pneumonia before and 2 weeks after treatment were observed.Results Compared with bacterial pneumonia group(16.7%),the viral pneumonia group(20.0%)and healthy control group(0.0%),the positive rate of D-dimer in mycoplasma pneumonia group(41.6%)was significantly higher(x2=5.625,4.158,17.308,all P<0.05).The positive rate of mycoplasma pneumonia group with SIRS(64.0%)was significantly higher than that in other groups(x2=17.308,P<0.05).The D-dimer levels of the mycoplasma pneumonia group [(324.4±125.3)μg/L],bacterial pneumonia group[(282.3±95.4)μg/L] and viral pneumonia group[(293.1±92.3)μg/L]were significantly higher than that in the healthy controls[(72.9±22.4)μg/L](t=10.878,11.704,12.698,all P<0.05).The concentration of D-dimer[(381.4±129.4)μg/L] in the mycoplasma pneumonia group with SIRS was significantly higher than that in the other groups(t=2.668,P<0.05).Compared with before treatment,the concentration of D-dimer after treatment in the two groups was significantly lower[(129.3±65.3)μg/L,(89.7±28.6)μg/L](t=2.582,P<0.05).Conclusion The level of D-dimer in children with mycoplasma pneumonia was significantly increased,and the severity of the disease could be reflected.

OBJECTIVE:To control scientifically the limits of stock drugs in Hospital.METHODS:The method of setting the upper and lower limit of stored drugs through Hospital Information System(HIS)and the influencing factors were discussed by taking the outpatient pharmacy of our hospital as an example.RESULTS&CONCLUSION:Working out scientific and reasonable upper and lower limit for stock drugs is helpful for the improvement of the management quality of hospital pharmacy.
4.Quantization and Enforcement of Medicine Provision Management Aim in Our Hospital

OBJECTIVE:To improve hospital's drug control level.METHODS:Quantized medicine provision management aims like stock limit,conformity rate of object-account of single category,medicine loss,mistake rate of recipe preparation,etc.were made and enforced.RESULTS:Through the above aims,medicine stock has been reduced effectively;the conformity rate of object-account has been increased;the medicine loss and incidences of mistakes in recipe preparation have been decreased.CONCLUSION:Working out feasible medicine provision management aims is an effective way to strengthen drug control.
5.The factors influencing the transfer of porcine endogenous retroviruses across the membrane in bioartificial livers

ObjectiveTo identify the factors influencing the transfer of porcine endogenous retroviruses across the membrane of a new bioartificial liver (BAL),which is a necessary caution to consider for BALs carrying porcine hepatocytes.MethodsA novel porcine BAL composed of two circuits was constructed using a plasma filter with membrane pore size of 500 nm and a plasma component separator with membrane pore sizes 10 nm,20 nm,30 nm,and 35 nm.Co-cultured cells of porcine hepatocytes and mesenchymal stem cells or single porcine hepatocytes were incubated in the bioreactors.The BAL was continuously worked for 72 hours during which the supernatant was collected from the internal and external circuits every 12 hours.PERV RNA,reverse transcriptase (RT) activity,and in vitro infectivity from the supernatant were detected.ResultsIn the plasma filter group,the PERV RNAlevels were the same in both circuits,suggesting little to no effect of the plasma filter on the PERV RNA's crossing.With plasma component separators,PERV RNA was found in the external circuits at different times without positive RT activity and HEK293 cell infection,but its level was reduced significantly.The larger the membrane pore size was,the earlier and the more RNA was detected.ConclusionsThe membrane pore size,the treatment time and the viral level in the internal circuit are contributing factors influencing the transfer of PERV RNA across the membrane in a BAL.
6.The relationship between serum copper/zinc ratio and classification as well as prognosis of Hodgkin's disease

AIM:To clarify the relationship between serum copper/zinc ratio(CZR) and histopathological classification as well as prognosis of Hodgkin's disease (HD). METHODS:The serum copper level(SCL),serum zinc level(SZL)and CZR were determined by atomic absorption spectrophotometry(AAS)in 135 patients with HD and 100 healthy individuals.RESULTS:The results showed that SCL and CZR were higher and SZL was lower in patients with HD than in healthy individuals (P<0.01). SCL and CZR were significantly higher in the patients with lymphocyte depleted type(LD)than mixed cellulal type(MC), SCL and CZR were higher in MC type than nodular sclerosis type(NS)and lymphocyte predominance type (LP)(P<0.01). It was also found that with the development of the disease, the SCL and CZR elevated but SZL dropped. And with the alleviation of the disease, the SCL and CZR rose while SZL dropped. While the illness is relieved and the SCL and CZR markedly dropped and SCZ rose. SCL and CZR were strongly correlated with histopathologic type and clinical stage of tumor.CONCLUSION:SCL, SZL and CZR are parameters useful for clarifying histopathological classification, clinical stage and prognosis of HD.
7.Effect of preoperative respiratory exercise on pulmonary function and complications in elderly patients after abdominal operation

Objective To compare and analyze the effects of preoperative respiratory function exercise on postoperative pulmonary function and complications in elderly patients who undergoing abdominal surgery . Methods A total of 92 elderly patients who underwent abdominal surgery were recruited .They were randomly divided into observation group(46 cases) and control group(46 cases) according to the random number table method . The control group was treated with routine preoperative care methods , the observation group was given preoperative respiratory exercise on the basis of the control group .The pulmonary function tests were performed pre and post surgery.Then,the postoperative pulmonary complications ,VC,FEV1,FVC,MVV and the average length of hospital stay of the two groups were recorded and compared .Results After operation,the levels of VC,FEV1,FVC and MVV in the control group were (2.48 ±0.71)L,(51.40 ±10.10)L,(1.79 ±0.17)L,(62.40 ±13.00)L/min,respectively, which in the observation group were (2.89 ±0.72) L,(63.20 ±9.60) L,(2.21 ±0.16) L and(73.30 ± 12.70)L/min,respectively,the differences between the two groups were significant (t =2.750,5.743,12.202, 4.068,all P<0.05).The average hospitalization time in the observation group was (11.62 ±1.41) d,which was significantly shorter than (15.56 ±6.23) d in the control group (t =4.163,P <0.001).The incidence rate of pulmonary infection in the control group was 26.09%,which was significantly higher than 8.69% in the observation group(χ2 =4.842,P<0.05).Conclusion The respiratory function training can effectively reduce the incidence of postoperative pulmonary complications in elderly patients with abdominal surgery ,improve the patients ' postoperative pulmonary function, and shorten the patients ' hospital recovery time .Respiratory function exercise is a simple method,and is worthy of promotion in related fields .
8.Therapeutic effect and mechanism of mesenchymal stem cells on acute liver failure

Objective To explore the therapeutic effects of soluble cytokines secreted by mesenchymal stem cells (MSCs) on acute liver failure (ALF).Methods MSCs isolated from Sprague-Dawley rats were determined by FACS analysis.Conditioned medium derived from MSCs (MSCs-CM) was collected and analyzed by a cytokine microarray.SD rats were divided into 3 groups:(1) ALF + dulbecco's modified eagle medium (DMEM) group:1 ml DMEM was injected into SD rats after D-Gal administration;(2) ALF + MSCs group:1 ml MSCs (1 × 106) was injected into SD rats after D-Gal administration;(3) ALF + MSCs-CM group:1 ml MSCs-CM was injected into SD rats after D-Gal administration.Biochemical indicators,survival rate,histology and inflammatory factors were studied.Exogenous recombinant rat IL-10,antirat IL-10 antibody and AG490 (STAT3 signaling pathway inhibitor) were administrated to explore the therapeutic mechanism of MSCs-CM.Results The respective serum biochemical indexes of ALF + DMEM group,ALF + MSCs group,and ALF + MSCs-CM group were:ALT (1 709.8 ± 372.1,865.5 ± 52.8,964.7 ± 414.6 U/L),AST (4234.0 ± 807.3,2440.8 ± 511.9,2739.8 ± 587.3 U/L),andTBil (79.3 ± 10.9,43.8 ± 7.0,61.2 ± 6.7 μg/L).The survival rates of the three groups were 10.0%,80.0%,and 70.0%,respectively.The levels of inflammatory factors in each group were IFN-γ (69.8 ± 4.7,46.4 ± 4.3,54.6 ± 2.4pg/ml),IL-1β (58.5 ± 7.6,40.5 ± 6.9,44.1 ± 6.0pg/ml),IL-6 (71.9 ± 16.1,38.4 ± 7.7,45.3 ± 9.0),and IL-10 (38.3 ± 6.0,75.4 ± 11.1,59.6 ± 11.9 pg/ml).Protein microarray results suggested that MSCs-CM expresses a variety of inflammatory-related cytokines,with IL-10 levels being most pronounced.IL-10 (ALT 1 126.9 ± 419.3 U/L,AST2370.8 ± 561.2 U/L) alone significantly reduced transaminase levels compared with ALF group (ALT 1 709.8 ± 372.1 U/L,AST 4234.0 ± 807.3 U/L),while anti-IL-10 antibody (ALT 1 568.5 ± 325.4 U/L,AST4043.7 ± 819.0 U/L) neutralized the therapeutic effect of MSCs-CM (ALT 964.7 ± 414.6 U/L,AST 2 739.8 ± 587.3 U/L).IL-10 could significantly increase the level of pSTAT3 in ALF rats (0.93 ± 0.03 vs 0.68 ± 0.01),while STAT3 inhibitor AG490 (0.84 ± 0.04) could decrease the expression of pSTAT3 and reverse the therapeutic effect of IL-10.Conclusion The factors released by MSCs,especially IL-10,have the potential therapeutic effect on ALF,and STAT3 signaling pathway may mediate the anti-inflammation effects of IL-10.
