1.Therapeutic Effect of Modified Simiao Powder for Drug-induced Hyperuricemia

[Objective] To observe the therapeutic effect of heat-clearing and diuresis-promoting herbal medicine for hyperuricemia induced by pyrazinamide. [Methods] Eighty-six patients with pyrazinamide-induced hyperuricemia were randomized into 2 groups: group A ( n = 43) received antiphthisic drugs (including isoniazid, rifampin, pyrazinamide and ethambutol) , allopurinol tablets and Modified Simiao Powder (one dose per day); group B ( n = 43) was treated with antiphthisic drugs and allopurinol tablets. Fourteen days constituted one treatment course and the two groups were treated for 2 courses. After treatment, the therapeutic effect was assessed and the changes of blood uric acid level were observed. [Results] In group A, 28 (65.12%) were cured, 12 (27.91%) effective, 3 (6.97%) ineffective and the total effective rate was 93.02%; in group B, 16 (37.21%) were cured, 17 (39.53%) effective, 10 (23.26%) ineffective and the total effective rate was 76.74% . [摘 要] 目的:探讨程序性死亡受体-1(PD-1)单抗联合顺铂或吉西他滨在KRAS基因突变非小细胞肺癌(NSCLC)A549细胞移植瘤小鼠模型治疗中的作用。方法:构建免疫系统-肿瘤双人源化A549细胞小鼠移植瘤模型,将60只小鼠按随机数字表法分成6组(10只/组),分别为对照组(200 μL/kg PBS)、PD-1单抗组(20 mg/kg PD-1单抗)、顺铂组(3 mg/kg顺铂)、PD-1单抗+顺铂组(20 mg/kg PD-1单抗+3 mg/kg顺铂)、吉西他滨组(30 mg/kg吉西他滨)和PD-1单抗+吉西他滨组(20 mg/kg PD-1单抗+30 mg/kg吉西他滨)。TUNEL和DAPI双染色法检测移植瘤组织中细胞凋亡水平,测量移植瘤体积和质量并计算肿瘤生长抑制率,免疫组化法检测移植瘤微血管密度(MVD)。结果:成功构建免疫系统-肿瘤双人源化NSCLC A549细胞小鼠移植瘤模型,PD-1单抗+顺铂组移植瘤的细胞凋亡率、肿瘤生长抑制率均最高,移植瘤体积、质量和MVD均最小,与其他5组小鼠比较差异均有统计学意义(均P<0.05)。结论:顺铂与PD-1单抗具有协同活性,而吉西他滨拮抗PD-1单抗的治疗作用。提示PD-1单抗联合顺铂对KRAS突变NSCLC A549细胞移植瘤小鼠的疗效更好。
3.Mechanism of puerarin targeting the mitochondria to inhibit proliferation and invasion of breast cancer cells

Objective :
To investigate the effect and mechanism of puerarin on the proliferation , invasion and apop- tosis of breast cancer cell HCC1806 via network pharmacology , molecular docking and in vitro experiments .
Methods:
The data of breast cancer and mitochondrial diseases were collected from GEO database , and the differentially expressed genes were analyzed by GEO2R . Overlapping genes between breast cancer and mitochondrial database were analyzed by Venn diagram . GO and KEGG enrichment analyzed the overlapping genes . STRING and Cyto- scape analyzed overlapping gene interaction networks and screen core genes . Interaction between puerarin and core genes was docked with autodock . Cell proliferation , apoptosis and invasion capacity were measured by CCK-8 , EdU , TUNEL and Transwell experiments , mitochondrial membrane potential was measured by Mito-Tracker experi- ments and protein expression levels were measured by Western blot. Anti-tumor efficacy of puerarin was analyzed by subcutaneous xenograft of breast cancer and immunohistochemical assay in vivo .
Results :
Among 132 overlap- ping genes in breast cancer and mitochondrial disease , 10 core differentially expressed genes were selected . Among these core genes , dual serine/threonine and tyrosine protein kinase (DST) was low expressed in breast cancer tis- sues . Puerarin bound to DST , up-regulated its expression , inhibited the proliferation and invasion of HCC1806 breast cancer cells , promoted breast cell apoptosis , increased the expression levels of apoptosis-related proteins Cleaved-Caspase 3 and Bax , down-regulated the expression of anti-apoptosis protein Bcl-2 , and decreased mito- chondrial membrane potential . In vivo , puerarin inhibited the growth of breast cancer and up-regulated the expres- sion of DST in tumor tissues .
Conclusion
Puerarin inhibits the proliferation and invasion of breast cancer cells , promotes the apoptosis of cancer cells and inhibits breast cancer growth .
4.The correlation study between the serum cortisol and C type behaviors of perioperative patients with lung cancer

Objective To discuss the correlation between the serum cortisol level and C type behaviors of perioperative patients with lung cancer.Methods A total of 100 patients from the thoracic department of Anhui Provincial Hospital were enrolled as the study object.The general information,the serum cortisol level,the C type behavior of perioperative patients with lung cancer were investigated by using the General Information Questionnaire,Serum cortisol testing and C Type Behavior Scales.Results The serum cortisol level of 100 patients was (447.85±87.65) μg/L.The C Type Behaviors Scale showed that the scores of anxiety were significantly higher in these patients than in the norm,P<0.05,depression scores were significantly higher in these patients than in the norm,P<0.01,while the extroversion anger,reason,optimism and social support were significantly lower,P<0.01.The serum cortisol of patients was positively correlated with anxiety,depression and introversion anger of C type behaviors,r=0.215,0.241,0.225,P< 0.05.The serum cortisol of patients was negatively correlated with optimism and social support of C type behaviors,r=-0.223,-0.276,P<0.05.Conclusions The serum cortisol level of perioperative patients was higher than the norm,and correlated with the C type behaviors.
5.Effect on content of serum inflammatory cytokines of patients with ankylosing spondylitis in early stage treated by long snake moxibustion at Governor Vessel and functional exercise.

OBJECTIVETo explore the clinical effect and mechanism of long snake moxibustion combined with functional exercise for treatment of ankylosing spondylitis in early stage.
METHODSSeventy-eight cases of ankylosing spondylitis in early stage were randomly divided into an observation group and a control group, 39 cases in each group. The observation group was treated with long snake moxibustion from Dazhui (GV 14) to Yaoshu (GV 2) combined with functional exercise; the control group was treated with oral administration of Sulfasalazine. The effects of the two groups were observed, and contents of erythrocyte sedimentation rate (ESR), tumor necrosis factor alpha (TNF-alpha), intercellular adhesion molecule-1 (ICAM-1) were tested before and after treatment.
RESULTSThe total effective rate of 89.7% (35/39) in the observation group, was superior to that of 76.9% (30/39) in the control group (P<0.05). The contents of ESR, TNF-alpha, and ICAM-1 showed downtrend after treatment in the both groups (P<0.05, P<0.01), and the observation group decreased more obviously (all P<0.01).
CONCLUSIONLong snake moxibustion at Governor Vessel combined with functional exercise for treatment of ankylosing spondylitis in early stage, can significantly improve clinical symptoms, decrease the level of serum inflammatory cytokines, and the effect is superior to that of Sulfasalazine oral administration.

6.Study on the prevalence rate and risk factors of chronic obstructive pulmonary disease in rural community population in Hubei province.

OBJECTIVETo explore the prevalence of chronic obstructive pulmonary disease (COPD) and its risk factors in rural area in the southern part of Hubei province and to provide evidence for prevention and treatment.
METHODSUsing uniform scheme, procedures and questionnaire, a multistage survey for 1883 people in rural area in southern part of Hubei province was performed, and physical examinations and lung function tests were conducted for every participant.
RESULTSIn this survey, 186 COPD cases were identified with a overall prevalence of 9.88%. The prevalence for male was 13.68% while in female it was 6.46%. The prevalence was higher in males than in females (chi2 = 27.48, P< 0.001) and higher with the increase of age (chi2 = 79.22, P<0.001). Factors associated with COPD were identified in one-way variance model as follows: sex, age, smoking, time for cooking, using biomass fuels for in-door cooking, family history, frequent cough before age 14, low body index and having house remodelling in the last five years. Results from multivariate logistic regression analysis showed that sex (OR= 1.3010), age (OR = 1.8506), smoking (OR= 3.0118), cooking time (OR= 1.7651), family history (OR = 1.5278), frequent cough before age 14 (OR = 2.8965) were risk factors.
CONCLUSIONThe prevalence of COPD in the southern part of Hubei province was high, suggesting that comprehensive intervention measures should be taken.

7.Lipid peroxidation changes induced by dibutyl phthalate in allergic asthma mice

Background Dibutyl phthalate (DBP) is a common plasticizer in daily life and has been proved to be related to the exacerbation of allergic asthma. Domestic and foreign studies have shown that lipid peroxidation is closely related to the severity of asthma, which can be used as a basis for the diagnosis and treatment of asthma. Whether DBP can induce lipid peroxidation in allergic asthma remains to be further studied. Objective To investigate whether DBP aggravates allergic asthma by inducing lipid peroxidation in allergic asthma mice. Methods Eighty male BALB/c mice were randomly divided into 4 groups, namely control group, DBP group (40 mg·kg−1), 50 μg ovalbumin (OVA) group (allergic asthma model group), and DBP+OVA group. The DBP group and the DBP+OVA group were given DBP by gavage from Day 1 to 28, and the OVA group and the DBP+OVA group were sensitized by intraperitoneal injection of OVA, once every 3 d, a total of 5 injections, from Day 9 to 21. From Day 29 to 35, the OVA group and the DBP+OVA group were challenged by OVA atomization. After the exposure, samples of blood and lung were collected. The airway hyperresponsiveness of mice was observed by lung function analysis. The serum contents of immunoglobulin E (IgE), OVA-specific immunoglobulin E (OVA-IgE), and lung homogenate levels of interleukin 4 (IL-4) were detected by enzyme-linked immunosorbent assay (ELISA) to evaluate airway allergic inflammation. The pathological changes of lung tissues were observed after hematoxylin-eosin (HE) staining and collagen fiber (Masson) staining. The contents of reactive oxygen species (ROS), lipid ROS, glutathione peroxidase 4 (GPX4), reduced glutathione (GSH), malondialdehyde (MDA), and 4-hydroxynonenal (4-HNE) in lung homogenates were detected by ELISA to evaluate lipid peroxidation. Results The results of lung function analysis showed that compared with the control group, the inspiratory resistance (Ri) and expiratory resistance (Re) of the OVA group and the DBP+OVA group were increased, and the lung compliance (Cldyn) was decreased. The DBP + OVA group was more severe, and the difference between the OVA group and the DBP + OVA group was statistically significant (P<0.05 or P<0.01). Compared with the control group, the contents of IgE, OVA-IgE, and IL-4 in the OVA group and the DBP+OVA group were increased (P<0.05 or P<0.01), which indicated more severe allergic airway inflammation. The HE sections of the OVA group and the DBP+OVA group showed inflammatory cell infiltration around the airway, airway wall hyperplasia and thickening, and severe airway deformation, and the presentation of the DBP+OVA group was the most serious. After Masson staining, the OVA group and the DBP+OVA group showed depositions of a large number of collagen fibers, and the blue collagen fibrosis in the DBP+OVA group was even more serious. ROS, lipid ROS, MDA, and 4-HNE levels increased and GSH and GPX4 levels decreased in the OVA and DBP+OVA groups (P<0.05 or P<0.01), with the most severe effect in the DBP+OVA group. Conclusion DBP may induce lipid peroxidation in mice allergic asthma by producing excessive ROS which may aggravate the allergic asthma in mice.
8.ANA- 12 inhibits spinal inflammation and alleviates acute and chronic pain in rats by targeted blocking of BDNF/TrkB signaling.

OBJECTIVE:
To investigate the inhibitory effect of ANA-12 that blocks brain-derived neurotrophic factor (BDNF)/ tropomyosin receptor kinase B (TrkB) signaling on inflammatory pain in rats and explore the underlying mechanism.
METHODS:
Forty-two adult SD rats were randomized into BDNF-induced acute pain group (n=24) and CFA-induced chronic pain group. The former group were randomly divided into 4 subgroups, including a control group, ANA-12 treatment group, BDNF treatment group, and BDNF+ANA-12 treatment group; the latter group were subgrouped into control group, CFA treatment group (CFA) and CFA + ANA-12 treatment group. The effects of ANA-12 treatment on pain behaviors of the rats with BDNF-induced acute pain and CFA-induced chronic inflammatory pain were observed. Western blotting was used to examine TrkB signaling and expressions of microglia marker protein Iba1 and TNF-α in the spinal cord of the rats.
RESULTS:
BDNF injection into the subarachnoid space significantly increased the number of spontaneous paw withdrawal of the rats (P < 0.05), which was obviously reduced by ANA-12 treatment (P < 0.05). The rats with intraplantar injection of CFA, showed significantly increased ipsilateral mechanical stimulation sensitivity (P < 0.05), and ANA-12 treatment obviously increased the ipsilateral foot withdrawal threshold (P < 0.05). Treatment with either BDNF or CFA significantly increased the phosphorylation level of TrkB (Y705) in the spinal cord of the rats (P < 0.05), which was significantly lowered by ANA-12 treatment (P < 0.05). Treatment with BDNF and CFA both significantly up-regulated the expressions of Iba1 and TNF-α in the spinal cord (P < 0.05), but ANA-12 significantly reduced their expression levels (P < 0.05).
CONCLUSION
ANA-12 can reduce spinal cord inflammation and relieve acute and chronic pain in rats by targeted blocking of BDNF/TrkB signaling.

9.Effect of tetrandrine on bax, bcl-2 and TGF-β2 mRNA expressions in cultured human Tenon's capsule fibroblasts.

OBJECTIVETo investigate the effect of tetrandrine (Tet) on the expression of bax, bcl-2, and transforming growth factor-β2 (TGF-β2) mRNA in cultured human fibroblasts of Tenon's capsules (TCFS) and explore its possible mechanism.
METHODSThe third passage of TCFS cultured in vitro were exposed to 1×10(-5) mol/L Tet for 24 h, and real-time fluorescence quantitative PCR was used to detect the changes in the expressions of bax, bcl-2, and TGF-β2 mRNA.
RESULTSThe expression level of bax mRNA was obviously higher, while bcl-2 and TGF-β2 mRNA levels were significantly lower in Tet-treated TCFS than those in the control cells (P<0.05).
CONCLUSIONTet can inhibit the proliferation of TCFS possibly by reducing the expressions of bcl-2 and TGF-β2 mRNA, enhancing the expression of bax mRNA and inducing cell apoptosis, suggesting its potential in preventing fibrous scar formation after glaucoma filtration surgery.

10.EFFECT OF VITAMIN E,C AND ?-CAROTENE SUPPLEMENT ON CELLULAR OXIDATIVE DAMAGE IN AGED PEOPLE

Objective:To study the effect of natural antioxidant ?-carotene(?-C) combined with vitamin E and C supplement on cellular oxidative damage in aged people. Method:Three hundred subjects aged from 60 to 75y,128 males and 172 females were randomized into 5 groups:group 1(VE 200mg/d+VC 300 mg/d+?-C 16.7 mg/d) ,group 2(VE 200 mg/d+VC 300 mg/d+?-C 8.4 mg/d) ,group 3(VE 200 mg/d+VC 300 mg/d+?-C 5.6 mg/d) ,group 4(VE 200mg/d+VC 300mg/d) ,group 5(VE 5 mg/d) . The trial lasted 16 weeks. All subjects were asked to record the nutrients intake daily. The investigators followed-up and supervised the subjects irregularly. The samples of blood and urine were collected at the beginning and end of the trial. The proliferation of lymphocytes was detected by MTT and H2O2-induced erythrocyte hemolytic degrees were measured before and after the trial. Thefluidity of erythrocyte membrane was evaluated by fluorescence polarization(?) and microviscosity(?) . O6-MeG was analyzed by high performance capillary zone electrophoresis. Results:The proliferation of lymphocytes was increased and H2O2-induced erythrocyte hemolytic degrees were decreased from group 1 to group 4 at the end of trial with significant difference compared with control group 5. The ? and ? values were lower in 1 to 4 groups at the end of trial. O6-MeG was significantly lower in group 1 to 4 group at the end of trial,and group 1 lower than group 4. Conclusion:Different doses of natural antioxidant ?-carotene supplement combined with vitamin E and C could improve the function of erythrocyte and the proliferation of lymphocytes,and decrease DNA alkylating damage.
