1.Sub-hypothermia Treatment on Patients With Diffusing Axonal Injury

Objective To study the therapeutic effect of sub-hypothermia on 80 patients with diffusing axonal injury (DAI) and its prognosis.Methods All 80 patients with DAI were randomly divided into the sub-hypothermia group(40 cases)and the control group(40 cases). All patients'vital sign,intracranial pressure (ICP) and blood sugar were measured. 40 cases of the patients with DAI were treated by sub-hypothermia, the rectal temperate (RT) reached 32 0℃~35 0℃,sub-hypothermia lasted for 1~7 days; the control group was treated routine. According to Glasgow Outcome Scale (GOS), the prognosis of the patients was evaluated 3 months later.Results In comparison with the control group, increased ICP and raised blood sugar in sub-hypothermia group were significantly decreased (P
2.Clinical analysis of 18 patients with chronic expanding intracerebral hematoma

Objective To summarize the diagnostic and therapeutic experiences of 18 patients with chronic expanding intracerebral hematoma.Methods Clinical data of 18 patients with chronic expanding intracerebral hematoma was analyzed retrospectively.Results Most of the lesions were misdiagnosed preoperatively as cerebral neoplasms.Among 18 cases, 15 cases were cured, 2 cases had severe disability, and one case died after operation.Conclusions Chronic expanding intracerebral hematoma must be considered when encapsulated mass lesion is present in the brain. The ultimate diagnosis is confirmed at surgery and by histological analysis.
3.Neuroendoscopic neurosurgy for primary intraventricular hemorrhage

Objective To investigate the new therapy for primary intraventricular hemorrhage (PIH).Methods 31 cases of PIH were treated by rigid neuroendoscopy.The cases were composed of 16 cases of single intraventricular hemorrhage,14 cases of double intraventricular hemorrhage and 1 case the third-fourth intraventricular hemorrhage.24 cases were accompanied with acute obstructive hydrocephalus.Results Duration of operation lasted from 30 minutes to 60 minutes, with an average time of 43.4 minutes.More than 90 percent of hemorrhage in 24 cases with single or double intraventricular hemorrhage were evacuated. 50 percent to 90 percent of hemorrhage were evacuated in other 7 cases. Neurological status was obviously improved and improved in 25 cases, no change in 4 cases and dead in 2 cases.Only one patient developed hydrocephalus.Conclusion Neuroendoscopic neurosurgy for PIH was characterized by visualized manipulation, shorten operative time,minimal invasion, effective hemorrhage evacuation and excellent post-operative outcomes.
4.Endoscopic neurosurgery for the treatment of septum pellucidum cysts

Objective To discuss the techniques and significance of neuroendoscopic treatment for symptomatic septum pellucidum cysts. Methods A total of 12 patients with symptomatic septum pellucidum cysts were given a cyst-lateral ventricle ventriculostomy via a transcallosal and transventricular approach by using a neuroendoscope 4.0 mm in external diameter(Storz,Germany).Headache,epilepsy,and mental symptoms were prominent clinical manifestations.Two patients were complicated with hydrocephalus. Results Improvement of symptoms was achieved postoperatively in all the 12 patients.Out of 6 patients with a major sign of intracranial hypertension,symptoms completely disappeared in 4 patients and improved in 2 patients.Four patients with epilepsy attacks had no recurrence.A follow-up was made in 12 patients for 6~24 months(mean,18 months).Re-examinations of CT or MRI showed the cysts had decreased in size by 90% in 2 patients.The lesions completely disappeared in 2 patients with hydrocephalus. Conclusions Neuroendoscopic cyst-lateral ventricle ventriculostomy for septum pellucidum cysts is a safe,effective,and minimally invasive operation.
5.Effects of Aurora A silence by RNA interference on the apoptosis and proliferation of glioma cells

Objective To investigate the inhibitory effect of RNA interference on the expression of Aurora A in U251 cells, and the influence on proliferation and apoptosis of U251 cells. Methods The siRNA specific for Aurora A was synthesized and transfected into U251 cells in vitro. Aurora A mRNA expression and protein content were detected by RT-PCR and Western blotting respectively. The cell proliferation and apoptosis were observed by methyl thiazolyl tetrazolium(MTT) and flow cytometry(FCM). Transmission electron microscope was used to observe the ultrastructural changes of U251 cells. Results After transfection, the expression level of Aurora A mRNA was significantly decreased(P<0.01), and the protein content of Aurora A was also obviously reduced. The inhibitory rate of cell proliferation reached up to 67.57% 72 hours after transfection, which was significantly higer than that of normal control group(P<0.01). The apoptosis rate of U251 cells was significantly increased from (3.69±0.87)% to (15.34±2.16)% (P<0.01). Under the transmission electron microscope, it was observed that the U251 cells showed typical morphologic changes of apoptosis after transfection, such as karyopyknosis, chromatin condensation and margination, intracytoplasmic vacuoles formed, and apoptotic bodies formed. Conclusion The expression of Aurora A gene can be inhibited by siRNA successfully, and it results in the suppression of cell growth and induce apoptosis of human glioma cells in vitro. Aurora A may become a new target for the gene therapy of gliomas.
6.Curcumin induces U251 cell apoptosis via alteration of expression of FAK and activation of caspase

Aim To study the mechanism of curcumin induced U251 cell apoptosis. Methods U251 cells were treated with 20~100?mol?L~ -1 curcumin for 24 h and the growth inhibition rates of U251 cells were measured with MTT method. MTT method was used to measure the caspase inhibitors effect on curcumin too. Cell apoptosis was inspected with flow cytometry (FCM) and observed using electron microscope. The protein levels of FAK in U251 cells were observed by SP immunocytochemistry. The activity of caspase-3 was measured by spectrofluorometry. Results Curcumin inhibited the proliferation of U251 cells,induced apoptosis of U251 cells. At the same time the protein levels of FAK in U251 cells decreased and the activity of caspase-3 increased significantly. The apoptosis of U251 cells was partially reversed by caspase inhibitors. Conclusion Curcumin induced apoptosis via inhibition of expression of FAK and activation of caspase-3.
7.The influence of CM pathway AEA in human glioma U251 cell proliferation and apoptosis

Objective To investigate the role of ceramide pathway in cell proliferation and early apoptosis induction in U251 glioma cell after cannabinoid receptora agent anandmide(AEA)treatment.Methods U251 gliom cells were treated with AEA(1-10 μmol/L),Ceramide(5-20 μmol/L) and fumonisin B1 (FB1) (10 μmol/L) pretreatment.The growth inhibition rate of U251 was investigated by MTT assay.The early events of the apoptosis were measured by flow cytometry using annexin-V/propium iodide(PI) double staining method.Results Different concentrations of AEA inhibited the proliferation of human glioma U251 cells,and had synergistic effect with CM by FB1(10 μmol/L)pretreatment for 24 h.After exposure to AEA(10 μmol/L)for 24 h,U251 gliomacells could undergo the early cell apoptosis which was affected by FB1(10 μmol/L).Conclusion AEA through the CM de novo synthesis pathway,and CM concentration was lazy in collaboration,thus inhibiting human glioma U251 cell proliferation and induce early apoptosis.

Objective To study the effects of localized thermochemotherapy on cellular proliferation and the expression of PCNA and IGF-I protein of C6 gliomas in rat, and explore the possibility of developing new therapeutic method of gliomas. Methods C6 glioma cells were injectcd subcutaneously to rats. The rats were randomly assigned to 4 groups,and treatment were initiated on day 22 after tumor inoculation. Prior to treatment, the subcutaneous gliomas were examined to verify tumor size, which were ranged 1.5 to 2.0 cm. Then localized thermochemotherapy, hyperthermia and chemotherapy were given, respectively. The size and weight of subcutaneous tumors were measured. PCNA and IGF-I protein expression were detected by the technique of S-P immunohistochemistry. Glioma cells proliferation was detected by HE staining method and electron-microscopic observation. 9.Drug-resistance of Pseudomonas aeruginosa:An Analysis of 286 Strains

OBJECTIVE To analyze the resistance of Pseudomonas aeruginosa (PAE) isolated from clinical specimen and provide the guidance for the clinical treatment. METHODS The P. aeruginosas infection status from Jun 2005 to Dec 2007 was reviewed retrospectively,and the results of susceptibility test in 286 strains of PAE were analyzed. RESULTS The drug-resistance rates to gentamicin,cefotaxime,and ceftriaxone in PAE were all above 60.0%,and that to cefoperazone/sulbactam,piperacillin/tazobactam sodium,amikacin and levofloxacin showing all higher sensitivity. The resistance rates to meropenem and imipenem were 17.1% and 18.5%,respectively. CONCLUSIONS P. aeruginosa is one of the main pathogenic bacteria in nosocomial infection. It's very important to strengthen the monitoring of drug-resistance of PAE and rationally antibiotics usage.
10.Drug-resistance and Gene erm Existing in Meticillin-resistant Staphylococcus aureus

OBJECTIVE To investigate the resistance of meticillin-resistant Staphylococcus aureus(MRSA) and the occurrence of gene erm.METHODS ATB Staph and microdilute tests were performed to detect the susceptibility to 15 kinds of antibiotics in 50 strains of the S.aureus(SAU).Gene erm of these strains was detected by polymerase chain reaction(PCR).RESULTS There were no strains resistant to vancomycin,teicoplanin,fusidic acid and quinupristin-dalfopristin in 42 strains of MRSA detected.There were no strains sensitive to penicillin,oxacillin,gentamicin,tetracycline,ciprofloxacin and levofloxacin.Thirty-five strains habored gene erm in 42 strains of MRSA.The positive rate of gene erm was 83.3%.CONCLUSIONS The multiple-resistance of the MRSA is a serious issue.The resistance to erythromycin in MRSA is mediated by gene erm which encodes the methylase and changes the target site of drug action.
