1.The value of detection of copeptin and procalcitonin in early diagnosis and prognosis of cerebral hemorrhage-associated pulmonary infection

Objective To investigate that the relationship of serum concentration of copeptin ,procalcitonin(PCT )and early diagnosis ,prognosis in patients with cerebral hemorrhage‐associated pulmonary infection .Methods One hundred and twenty pa‐tients with acute cerebral hemorrhage ,acute cerebral hemorrhage‐associated pulmonary infection and 60 healthy individuals (the control group) admitted in our hospital from January 2012 to December 2013 were selected and detected for serum copeptin and procalcitonin .The differences of serum copeptin ,procalcitonin levels were compared in controls ,in patients with cerebral hemor‐rhage and cerebral hemorrhage‐associated pulmonary infection and their correlation was analyzed .Results The levels of serum copeptin in the cerebral hemorrhage‐associated pulmonary infection were significantly higher than that in control group and the cer‐ebral hemorrhage (P<0 .05) .The levels of serum procalcitonin in control group and the cerebral hemorrhage were significantly lower than that in the cerebral hemorrhage‐associated pulmonary infection ,the levels of serum C‐reactive protein ,copeptin ,procalci‐tonin and the APACHEⅡ scores of the patients with survival group were significantly lower than those with non‐survival group (P<0 .05) .Conclusion The levels of serum copeptin and procalcitonin are correlated intimately with cerebral hemorrhage‐associat‐ed pulmonary infection .They are important clinical parameters to reflect the early diagnosis and prognosis of cerebral hemorrhage‐associated pulmonary infection .
2.Clinical significance and the expression of NT-proBNP and copeptin in patients with acute cerebral hemorrhage

Objective To investigate the relationship of plasma concentration of NT-proBNP,copeptin and glasgow coma scale(GCS)scores,hematoma volumes in patients with acute cerebral hemorrhage.Methods 109 patients with acute cerebral hem-orrhage(the cerebral hemorrhage group)and 32 healthy individuals (the control group)admitted in our hospital from December 2011 to June 2013 were selected and detected for plasma NT-proBNP and copeptin.The levels of NT-proBNP,copeptin,glasgow co-ma scale(GCS)scores and hematoma volumes were compared between the two groups.Results The levels of plasma NT-proBNP and copeptin in the cerebral hemorrhage group were significantly higher than that in control group(P <0.05).The levels of plasma NT-proBNP and copeptin were significantly increased with the severity and the hematoma volume of the acute cerebral hemorrhage. The levels of NT-proBNP and copeptin are positively correlated with hematoma volumes(r=0.63,r=0.58,P <0.01)and negative-ly correlated with Glasgow Coma Scale(GCS)scores(r=-0.52,r=-0.46,P <0.01).Conclusion The levels of NT-proBNP and copeptin are positively correlated with hematoma volumes and negatively correlated with glasgow coma scale(GCS)scores.They are important clinical parameters to reflect the severity and hematoma volumes of the acute cerebral hemorrhage.
3. Platelet-rich plasma combined with microfracture versus microfracture in the treatment of knee cartilage lesions: A meta-analysis

OBJECTIVE: A large number of recent studies reported that microfracture combined with platelet-rich plasma has a good effect on osteochondral injury. This study systematically evaluated the clinical effect of arthroscopic platelet-rich plasma combined with microfracture versus microfracture in the treatment of knee cartilage lesions. METHODS: A randomized controlled trial of arthroscopic platelet-rich plasma combined with microfracture in the treatment of knee cartilage lesions was searched in CNKI, Wanfang Database, VIP Database, Chinese Biomedical Literature Database, PubMed, Embase and Cochrane Library. The date was from January 2001 to March 2020. The data were extracted and the quality of the included literature was evaluated, and the Rev-Man5.3 software was used for meta-analysis. RESULTS: (1) A total of seven randomized controlled trials were included, and the modified Jadad scale results showed that the scores of four studies were ≥ 4, which were high-quality literatures, and the scores of three studies were < 4, which were low-quality literatures. (2) Meta-analysis showed that the visual analogue scale score at 6 and 12 months, IKDC score at 12 and 24 months, Lysholm score at 12 and 24 months and WOMAC score at 12 months in the platelet-rich plasma combined with microfracture group were better than those in the microfracture group [MD=-0.40, 95%CI(-0.75,-0.05), Z=2.26, P=0.02; MD=-0.59, 95%CI(-0.86,-0.32), Z=4.30, P < 0.000 1; MD=7.70, 95%CI(4.22, 11.18), Z=4.34, P < 0.000 1; MD=10.99, 95%CI(6.79, 15.19), Z=5.13, P < 0.000 01; MD=4.98, 95%CI(2.31, 7.64), Z=3.66, P=0.000 3; MD=5.35, 95%CI(1.94, 8.77), Z=3.07, P=0.002; MD=-9.30, 95%CI(-16.50, 2.11), Z=2.53, P=0.01]. There was no significant difference in visual analogue scale score 24 months after operation and IKDC score 6 months after operation between the two groups (P > 0.05). CONCLUSION: Platelet-rich plasma combined with microfracture for knee cartilage lesions can reduce the pain in the short and medium terms, enhance the joint function in the medium and long terms, and improve the postoperative experience. However, due to the limited number of included articles, more large-sample and high-quality randomized controlled trials are needed to provide further effective data.
4.The development and application of a quantitative ELISA detection method for enterovirus 71 antigen

ObjectiveTo develop an ELISA method for quantitative determination of enterovirus 71 (EV71) antigen.The method can be applied to detect EV71 antigen contents and analyze the correlation between immunogenicity and immunoprotection.It also can be used for tissue culture infective dose( TCID50 ) assay.MethodsA double antibody sandwich ELISA method was developed for quantitative determination of EV71 antigen on the basis of the high-affinity neutralizing monoclonal antibodies ( K8G2 and Y8H2-HRP).This method was compared with microscopic observation for the detection of EV71 TCID50.The correlation was analyzed between the specific activity of EV71 antigen and the EV71 neutralizing antibody titer in immune serum.ResultsThe linear range of this method was 0.125-4.0 U/ml and the R2 value was 0.9911.The reagent did not react with other antigens except EV71 antigen.The recovery ratio of this method was 0.89-1.16.The coefficient of variation was less than 15%.The heat recovery rate was above 85% when the reagent was in 37℃ for 9 days.There was a good correlation in TCID50 of EV71 between this method and microscopic observation,r=0.990.The specific activity of EV71 antigen had positive correlation with the neutralization titer of immune serum in 21 EV71 strains,r=0.930.ConclusionThe quantitative ELISA method for EV71 antigen was developed,which could be used to detect EV71 antigen contents and analyze TCID50.The specific activity of EV71 antigen detected by the method could be used to evaluate the immunoprotection of the vaccine potency test.
5.Effect of serum from acute myocardial infarction rat on inducing rat bone marrow mesenchymal stem cells into cardiomyocytes

AIM: To induce rat bone marrow mesenchymal stem cells (BMSC) into cardiomyocytes and investigate the influence of serum coming from acute myocardial infarction (AMI) rat on the procedure. METHODS: The passage 3 BMSC were divided into six groups: groupⅠwas control group; groupⅡwas induced with 5-azacytidine; group Ⅲ was induced with 5-azacytidine and serum from AMI rat; group Ⅳ was induced with 5-azacytidine and serum from normal rat; group V and group Ⅵ were induced with serum from AMI rat or normal rat. The cardiac troponin T, GATA-4 and desmin were detected 30 days after induction. RESULTS: After inducing by 5-azacytidine, 5-azacytidine and two kinds of serum, some cells in the three groups differentiated into cardiac like cells. The expressions of cardiac troponin T, GATA-4 and desmin were positive in cells differentiated from BMSC. The troponin T expression in control group and group inducing by AMI serum alone were negative but GATA-4 and desmin expressed weakly. Some cells induced with 5-azacytidine and serum were slowly beating 2 weeks after induction, but the cells induced with 5-azacytidine alone was not beating.CONCLUSION: Serum from AMI can not induce BMSC to differentiate into cardiomyocytes, but it promotes BMSC differentiate into cardiomyocytes induced by 5-azacytidine and facilitate the differentiated cells to mature.
