1.Optimization of the Content Determination Conditions of Total Alkaloids from Zhuang Medicine Munronia delavayi and Comparison of the Contents in M. delavayi from Different Producing Areas

OBJECTIVE:To optimize the content determination conditions of total alkaloids from Zhuang medicine Munronia delavayi,and to determine the content of total alkaloids in M. delavayi from different producing areas. METHODS:With the con-tent of total alkaloids as index,using solvent amount,ultrasonic time,ultrasonic extraction times and pH value of buffer as fac-tors,the content determination conditions of total alkaloids from Zhuang medicine M. delavayi were optimized by orthogonal test. Optimized content determination conditions were adopted to determine the content of total alkaloids in M. delavayi from 18 produc-ing areas in different harvest time. RESULTS:The optimum content determination conditions were as follows as the amount of sol-vent(CHCl3)20 ml,ultrasonic processing for 3 times,lasting for 15 min each time,pH value of buffer 4.5. The contents of total alkaloids in M. delavayi from 18 producing areas were between 0.6-11.98 mg/g,showing great difference. M. delavayi from Long-lin county and Tianlin county harvested in Oct. had the lowest content of total alkaloids. CONCLUSIONS:Optimized content deter-mination condition can be used for the content determination of total alkaloids in Zhuang medicine M. delavayi and quality control of it. The content determination of total alkaloids in M. delavayi is related to producing area and harvest time.
2.Study on Quality Standard of Zhuang Medicine Homalocladium platycladum

OBJECTIVE:To establish quality standard for Zhuang medicine Homalocladium platycladum. METHODS:TLC method was used for qualitative identification. HPLC method was used to determine the content of quercetrin in samples:Agilent Zorbax SB-C18 column,mobile phase consisted of acetonitrile-0.1% acetic acid(20:80,V/V),flow rate of 1.0 mL/min,detection wavelength of 360 nm,column temperature of 30 ℃,injection volume of 10 μL. The contents of moisture,total ash and extract in samples were determined. referring to Chinese Pharmacopiea(2015 edition). RESULTS:TLC spots of H. platycladum were clear and well-separated without interference from negative control. The linear range of quercetin were 0.023-0.46 mg/mL(r=0.9991). RSDs of precision,stability and repeatability tests were all lower than 2.0%. The average recoveries were 98.01%-103.05%(RSD=1.71%,n=6). The contents of moisture,total ash,extract and quercetrin in samples were 6.39%-9.78%,2.82%-8.07%, 0.02%-0.27%,14.14%-28.45%,0.09%-0.50% respectively. CONCLUSIONS:Established trial can be used for quality control of Zhuang medicine H. platycladum.
3.Establishment of HPLC Fingerprint of Homalocladium platycladum and Cluster Analysis

OBJECTIVE:To establish HPLC fingerprint of Homalocladium platycladum,and to carry on the cluster analysis. METHODS:HPLC method was adopted. The determination was performed on Ultimate C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid(gradient elution)at the flow rate of 0.5 mL/min. The detection wavelength was set at 360 nm, and column temperature was 28 ℃. The sample size was 10 μL. Using quercetin as reference substance,HPLC chromatograms of 13 batches of samples were determined. The similarity evaluation was carried out by using Similarity Evaluation System for TCM Chromatographic Fingerprint(2012 edition)to determine common peak. Cluster analysis were also conducted. RESULTS:A total of 12 common peaks were established in HPLC fingerprints of 13 batches of samples. The similarity was higher than 0.90. After validation,HPLC chromatograms of 13 batches of sample were in good agreement with control fingerprint. 13 batches of medicinal materials were clustered into 2 categories;S4,S6 and S10 were clustered into one category,and the rest were a category. CONCLUSIONS:Established fingerprint and cluster analysis results can provide reference for quality evaluation of H. platycladum and its exploitation and utilization.
4.Effects of mirabilite stuck on umbilical region combined with early enteral nutrition on gastrointestinal function and serum inflammatory mediators in patients with mechanical ventilation

Objective To study the effects of mirabilite stuck on umbilical region combined with early enteral nutrition (EEN) on gastrointestinal function and serum inflammatory mediators in patients with mechanical ventilation. Methods Sixty-four patients with mechanical ventilation admitted to the Department of Intensive Care Unit of the Second Affiliated Hospital of Zhejiang Traditional Chinese Medical University from January to December 2016 were enrolled, and they were divided into an observation group and a control group by random number table, 32 cases in each group. The control group was treated with routine western medicine+ EEN, and the observation group was treated additionally with mirabilite stuck onto umbilical region on the basis of the treatment of the control group. The clinical effect was observed after 7 days of treatment. The differences in gastrointestinal function scores and the levels of serum inflammatory mediators were compared between the two groups before and after treatment.Results After treatment, gastrointestinal function scores of the diarrhea, bowel sound, abdominal distension and the levels of white blood cell count (WBC), serum procalcitonin (PCT), serum C-reactive protein (CRP) were decreased significantly in both groups compared with those before treatment, and the degrees of decreasing in above index levels after treatment in the observation group were more obvious than those in the control group [diarrhea: 0.72±0.03 vs. 1.59±0.21, bowel sounds: 0.87±0.05 vs. 1.54±0.18, abdominal distension: 0.77±0.04 vs. 1.63±0.09, WBC (×109/L): 9.87±1.25 vs. 12.46±2.04, PCT (μg/L): 5.43±1.02 vs. 9.65±1.63, CRP (mg/L): 56.87±4.52 vs. 89.43±8.24, allP < 0.05].Conclusion Applying mirabilite stuck on umbilical region combined with EEN can effectively promote the recovery of gastrointestinal function and significantly decrease the levels of serum inflammatory mediators in patients with mechanical ventilation.
5.IgE and Non-IgE mediate the activation of human mast cells

Mast cells are crucial effector cells in allergic reactions,where IgE is the most notably mechanism to trigger their degranulation and release large amounts of allergic mediators.However,IgE is not the only way to stimulate these cells to degranulate.There is a lot of stimuli,such as complement,neuropeptides,cytokines and other inflammatory products,which can directly trigger mast cell degranulation,and cause selective release of mediators.These mediators are closely related to many allergic and inflammatory diseases.The paper will focus on the activation of non-IgE stimulated human mast cells,and describe the allergic and inflammatory diseases briefly,and provide the theoretical basis for clinical detection,diagnosis and treatment.
