1.The clinical observation of advanced non small cell lung cancer treated with Shenqifuzheng injections combined with PTX and DDP

Objective To observe the clinical effects of Shenqifuzheng injections combined with PTX and DDP on advanced non small cell lung cancers. Methods Fifty cases of advanced non small cell lung cancers were randomly divided into pure chemotherapy and chemotherapy combined with traditional Chinese medicine groups. The former was treated with 30 mg/m2 DDP from day 1 to day 3, 135 mg/m2 PTX only at the first day. The latter was treated with 250 ml Shenqifuzheng injections every day until the twentieth-first one, three days before the beginning of chemotherapy. After two periods of treatments, the therapeutic effects were evaluated, respectively. Results The recent effective ratios of chemotherapy combined with traditional Chinese medicine and pure chemotherapy groups were 48 % and 40 %, respectively. The former could ameliorate the physical status, the numbers of peripheral leucocytes, the decreases of blood platelet, the gastrointestinal reactions of subjects. Conclusion The Shenqifuzheng injections could increase the recent treatment effect of PTX combined with DDP on advanced non small cell lung cancers, effectively meliorate the clinical symptoms and alleviate the side effects of chemotherapy leading to improve the life qualities of subjects.

3.Study on relationship between serum total IgE and β《,2》-adrenoreceptor polymorphism in asthmatic patients

Objective To study the association between serum total IgE level and β2-adrenoreceptor polymorphism (β2-AR) in asthmatic patients. Methods ELISA was used to determine serum total IgE and AS-PCR was used to determine β2-adrenoreceptor polymor- phism in 44 asthmatic patients. Results In β2-AR 16 locus genotype, the distribution frequencies of Arg/Arg, Gly/Gly showed increasing tendency, whereas Arg/Gly showed decreasing tendency, in normal serum total IgE group and increased serum total IgE group. But there was no significant difference between this two groups. In β2- AR 27 locus genotype, the distribution frequency of Gln/Gln genotype accounts for 76.2% and Gln/Glu genotype for 19.0% in increased serum total IgE group, while Gln/Gln genotype accounts for 9.0% and Gln/Glu geno- type for 73.9% in normal serum total IgE group. There was significant difference between two groups ( P<0.01 ). Conclusions Serum total IgE was correlated with β2-AR 27 locus genetic polymorphism, which may contribute to understand the mechanism of asthma in the peo- ple of the Han nationality in GuiZhou.
4.ZFP580, a novel transcription factor, is involved in cardioprotection of hypoxic preconditioning against hypoxia-reoxygenation injury in myocar-dial cells

AIM:To elucidate whether ZFP580 is involved in the cardioprotective effects of hypoxic precondi-tioning (HPC) against hypoxia-reoxygenation (H/R) injury in H9c2 myocardial cells.METHODS: Rat heart-derived H9c2 cells were cultured in DMEM .H/R was induced by incubation under ischemic hypoxia for 3 h and reoxygenation for 2 h.HPC was induced by exposing the H 9c2 cells to 10 min of hypoxia and 20 min of reoxygenation for 3 cycles before H/R treatment.MTT staining and LDH leakage detection were used to evaluate the effects of HPC .Western blotting was used to detect the protein levels of ZFP580, phosphorylated ERK1/2 and cleaved caspased-3.The effects of ZFP580 overexpre-ssion or knockdown on H/R induced apoptosis were determined .RESULTS:The results of MTT staining and LDH leakage detection showed evidence of HPC cytoprotection against H /R-induced cell death in H9c2 cells.ZFP580 protein level and ERK1/2 phosphorylation were significantly increased in the HPC group compared with control group and H /R group. PD98059, an inhibitor of ERK1/2 phosphorylation , significantly suppressed the HPC-induced up-regulation of ZFP580 pro-tein expression.ZFP580 overexpression significantly inhibited apoptosis and caspase-3 activation in H9c2 cells.CON-CLUSION:HPC exhibits cytoprotection against H/R and leads to high level of ZFP 580 protein in H9c2 cells.ZFP580 is regulated by ERK1/2 activation and mediates the anti-apoptotic effect of the ERK1/2 signaling pathway in HPC cytoprotec-tion.
5.Molecular docking of anthocyanins constituents and HER-2 kinase domain.

Anthocyanins are a ubiquitous group of water-soluble plant pigments of the flavonoid family, with anticancer property through HER-2 signaling pathway. Nowadays, molecular docking plays an important role in exposing the active sites and obtaining the bioactive conformation involving protein-ligand interactions. According to the crystal structure of HER-2 kinase domain and 12 main antitumor compounds of anthocyanins as well as ATP, a molecular docking study was performed by MVD program. All 12 compounds could bind to the same cavity of HER-2 kinase domain by high affinity (MolDock Score < -105 kJ/mol for anthocyanidins, < -130 kJ/mol for anthocyanidins-glc), where hydrophobic force and hydrogen bond played key roles. Additionally, this cavity overlapped with ATP binding (MolDock Score = -161 kJ/mol) domain; the binding of anthocyanins presumably interfered the H bond formation between ATP and HER-2. These results indicate that anthocyanins may competitively bind to ATP binding site in HER-2 kinase domain by suppressing HER-2 activation and downstream signaling cascade. This may provide useful theoretical instruction for the molecular mechanism of HER-2 kinase activity inhibition by anthocyanins in cancer prevention and treatment.

7.Protective effects of melatonin on learning and memory in rats by noise stress

Objective To explore the effect of melatonin(MT) on the behavior of rats treated with noise stress and the related bio-mechanism. Methods Fifty rats were randomly divided into a blank group,two experimental groups and two control groups. The blank group was untreated. The experimental and control groups were exposed to 120dB noise stress for 1 day or 3 days, 8 hours per day, and treated with 15 mg/kg melatonin by intraperitoneal injection,or the same volume of physiological saline 30 minutes before noise stress. After noise stress,the rats' behavior was measured by open field test, learning and memory ability of rats was investigated with the method of Morris water maze and then nitric oxide (NO), superoxide dismutase (SOD) and malondialdehyde (MDA) contents in cerebral cortex and hippocampus of the rats were measured by TBA and Griess method respectively. Results No matter noise stress time was 1 day or 3 days, the excitability and explorative behavior of the 2 experimental groups(total movement distance (TMD) (1322.50 ± 504.32) cm, (1819.55 ± 458.37) cm, faster movement time (FMT) (68.49 ± 23.90) s, (87.34 ± 16.01) s, distance to center (DTC) (63.56 ± 2. 75) cm, (60. 13 ±1.87)cm, inner toriod time(ITT) (7.87 ±2.06)s,(9.60 ±2.89)s) in the open field test decreased significantly compared with those of the control group (TMD (2042.03 ± 449. 19) cm, (2325.73 ± 384.90) cm,FMT (109.32 ±21.84)s,(124.65 ± 16.74)s, DTC (58.00± 1.53)cm,(55.05 ±5.13)cm, ITT (12.84 ±3.62) s, (14.92 ± 2.75) s, P < 0. 05, P < 0.01);the escape latency of the experimental groups (( 10. 69 ±3.37) s, (18.87 ± 4.74) s) in Morris water maze was significantly shorter than that of the control group (( 23.86± 7.66)s, (33.55 ± 7.20)s, P< 0.05, P<0.01). The contents of NO or MDA in cerebral cortex and hippocampus of the experimental groups (NO in cerebral cortex (3.35 ± 0.40) μmol/gprot, (4.50 ± 0.41) μmol/gprot, NO in hippocampus (2.24 ±0.18) μmol/gprot,(3.15 ±0.21) μmol/gprot, MDA in cerebral cortex(1.34 ±0.44)nmol/mgprot, (2.39 ± 0. 18) nmol/mgprot, MDA in hippocampus (0. 13 ± 0. 07) nmol/mgprot, (0.53 ± 0. 10)nmol/mgprot) were lower than those of the control group (NO in cerebral cortex (3.35 ± 0. 40) μmol/mgprot,(5.03 ± 0.44)μmol/mgprot, NO in hippocampus (2.93 ± 0. 31) μmol/gprot, (3.38 ± 0.24) μmol/gprot, MDA in cerebral cortex (2.24 ± 0.26) nmol/mgprot, (4.21 ± 0.21) nmol/mgprot, MDA in hippocampus (0.47 ± 0.29)nmol/mgprot, (1.33 ± 0. 187) nmol/mgprot, P < 0.05, P < 0. 01) respectively and the contents of SOD in cerebral cortex and hippocampus of the experimental groups (in cerebral cortex (763.95 ± 214.36) U/mgprot, (491.33 ±35.85) U/mgprot, in hippocampus (817.02 ± 232.39) U/mgprot, (644.85 ± 28.02) U/mgprot) were higher than those of the control group(in cerebral cortex (556.50 ± 101.51) U/mgprot, (327.35 ± 30.54) U/mgprot, in hippocampus (279.74 ± 117.02) U/mgprot, (108.75 ± 15.52) U/mgprot, P < 0.05, P< 0.01) respectively. Conclusion Melatonin is effective in improving the ability of learning and memory in the rats of noise stress,possibly by inhibiting the increase of NO and MDA and increasing the SOD activity in cerebral cortex and hippocampus of the rats.
8.Comparison of therapeutic efficacy between Ivor-Lewis esophagectomy and 2-incision esophagectomy via left thoracic-cervical pathway in the treatment of middle thoracic esophageal carcinoma

Objective:To retrospectively evaluate the therapeutic efficacy of two different surgical approaches, Ivor-Lewis esophagectomy and 2-incision esophagectomy via left thoracic-cervical pathway, in the treatment of middle thoracic esophageal squamous cell carcinoma. Methods:One hundred and two patients from 167 patients with middle thoracic esophageal squamous cell carcinoma received Ivor-Lewis esophagectomy and another 65 patients received 2-incision esophagectomy via left thoracic-cervical pathway. The local recurrence rate of tumor and survival rate were calculated by using Kaplan-Meier method. The difference in the survival rate between the two surgical methods was analyzed by using log-rank test. The prognostic risk factors were assessed by COX regression analysis. Results:Peri-operative complications occurred in 35 patients (21.0%), in which the incidences of recurrent laryngeal nerve (RLN) injury and anastomotic leakage were higher in the left thoracic-cervical group (P<0.05), and the incidence of intrathoracic gastric retention was higher in the Ivor-Lewis group, but the difference was not significant. The 3-year local recurrence rate was 37.3% in the Ivor-Lewis group, and 40.0% in the left thoracic-cervical group. The difference was not significant (P>0.05). The overall 5-year survival rate of the 167 patients was 34.6%. It was 36.0% in the Ivor-Lewis group and 32.3% in the left thoracic-cervical group, respectively (P>0.05). COX regression analysis revealed that pTNM staging was the independent prognostic factor [P=0.000, HR(hazard ratio)=2.69]. Conclusion:Both Ivor-Lewis esophagectomy and 2-incision esophagectomy via left thoracic-cervical pathway are feasible alternatives in the treatment of middle thoracic esophageal squmamous cell carcinoma. We should choose the rational operative method based on the patients'individual condition.
9.Theanine affects sedative effect of pentobarbital sodium in ICR mice.

OBJECTIVETo evaluate the effects of theanine on sedative effects induced by pentobarbital sodium.
METHODThe locomotor activities of ICR mice induced by theanine (0.25, 0.5, 1.0, 2.0, 4.0 g x kg(-1)), pentobarbital sodium (5, 10 mg x kg(-1)) or the combination of both were determined with video-tracking system, and a novel index: Peripheral active time Peripheral time (PATP) was established. Hypnosis effect of combination of both was tested with right-reflex disappearance.
RESULTCompared with normal saline (74.52 +/- 20.4)%, theanine alone decreased this PATP in dose-dependent manner from (62.03 +/- 21.11)%, (56.44 +/- 21.69)%, (31.13 +/- 17.2)%, (25.06 +/- 10.03)% to (17.21 +/- 7.43)% (P>0.05, P<0.05, P<0.01, P<0.01 and P<0.01, respectively). Compared with pentobarbital sodium (5 mg x kg(-1)), between 0.25 g x kg(-1) and 1.0 g x kg(-1) theanine combined with that decreased peripheral PATP from (28.30 +/- 17.57)%, (30.64 +/- 17.21)% to (24.28 +/- 9.59)% (all P<0.01), and increased by 2.0 g x kg(-1) reversely (61.95 +/- 19.39)%. Compared with normal saline, pentobarbital sodium (10 mg x kg(-1)) and the combination with theanine decreased significantly PATP (all P<0.01). Compared with pentobarbital sodium (10 mg x kg(-1)), 0.25 g x kg(-1) theanine combined with that increased PATP [(25.37 +/- 13.68)% vs (10.08 +/- 7.98)%, P<0.01)] and 0.5 g x kg(-1), 1.0 g x kg(-1) theanine could depresse that increase [(14.56 +/-10.10)%, (8.24 +/- 4.08)% vs (10.08 +/- 7.98)%]. Total distance and peripheral active time showed the same or similar tendency in theanine alone or combination with pentobarbital sodium . Theanine enchanced hypnosis effect of pentobarbital sodium in dose-dependent manner.
CONCLUSIONTheanine can affect the sedative effect of low dose pentobarbital sodium in bidirectional action style but not change the hypnosis effect.

10.The progress and prospect of assessment measures of uterine cavity function for infertility patients

Objective:To explore the progress and prospect of evaluation methods for infertility patients.Methods:From September 2017 to December 2020,199 cases of infertility patients who have accepted mini-hysteroscopy examination in the Department of Gynecological Endocrinology,Beijing Obstetrics and Gynecology Hospital,Capital Medical University.The patients who had primary ovary insufficiency,tubal infertility were excluded;patients with hysteroscopy contradiction,such as acute or sub-acute vaginal tract inflammation,en-dometrium lesion or endometrium carcinoma,severe cardiovascular,liver or kidney insufficiency were excluded.40 patients were excluded and 15 patients lost follow up,144 patients were eventually included in the analysis.The patient's age,body mass index,infertility etiology,preoperative and postoperative stimulate ovulation cycle,pregnancy state and pregnancy time were recorded.The effect of mini-hysteroscopy on ovulation induction cycle was analyzed in pregnant patients.Results:The average age of 144 patients was 32.10±4.67 years old.96 (66.67%) patients were found to have intrauterine abnormalities,while 48 (33.33%) patients were found to have normal intrauterine cavity.48 cases of pregnancy,accounting for 33.33%;the median and interquartile ranges[M (Q1,Q3)]of ovulation induction cycles before and after surgery were respectively[3 (0,5)],[2 (1,3)],the difference was statistically significant(P < 0.05).Among the 48 pregnant patients,32 of them had no structural lesions in hysteroscopy,the ovula-tion induction cycle of these patients before and after hysteroscopy were respectively[5 (0,9)],[1 (0,3)],the difference was statistically significant (P < 0.05).Conclusion:Uterine cavity evaluation is important for the assessments of fertility.As an advanced diagnosis instrument,mini-hysteroscopy can not only timely detect the intrauterine abnormalities of patients,but also with great application value in shorten the ovulation cycle in infertility patients and increase the pregnancy rate.
