1.Embryonic stem cell transplantation for treatment of cerebrovascular diseases

BACKGROUND:Whether embryonic stem cel transplantation can be an effective method for treatment of cerebrovascular disease has become a hot spot. OBJECTIVE:To investigate the effect and feasibility of embryonic stem cel-differentiated neural precursor cel transplantation for the treatment of cerebrovascular disease. METHODS:The rat models of Parkinson’s disease, ischemic brain injury and vascular dementia were established and the embryonic stem cells were in vitro cultured and induced to differentiate into the neural precursor cells. The embryonic stem cel-differentiated neural precursor cells were transplanted into the rat brains with corresponding cerebrovascular disease, and then the rotational behavior changes, pathological changes in the brain tissue as wel as changes in the hippocampal structure and the number of nerve cells in the rats with cerebrovascular disease were observed. RESUTLS AND CONCLUSION:After the embryonic stem cel-differentiated neural precursor cells transplanted into the rat brains with Parkinson’s disease, the frequency of apomorphine-induced rotation was decreased and in the downward trend, while the striatal dopamine levels were significantly increased. After transplantation of embryonic stem cel-differentiated neural precursor cells into the rat brains with ischemic brain injury, the cells could survive for a long time, and then migrated and distributed in the injured hippocampus to form the hippocampal structure;the cells could differentiate into neurons, and the number of nerve cells in the injured hippocampus was significantly increased. The results indicate that the transplanted glial cel line-derived neurotrophic factor gene-modified embryonic stem cells can improve the learning and memory function of vascular dementia rats, enhance neural plasticity, and induce self-directed migration and differentiation into mature neurons.
2.Clinical study on the effect of oral and acupoint application of traditional chinese medicine combined with azithromycin for the children with mycoplasma pneumonia

Objective To investigate the clinical curative effect of oral and acupoint application of traditional chinese medicine combined with Azithromycin for the children with Mycoplasma Pneumonia. Methods A total of 160 children with mycoplasma pneumonia were divided into two groups according to digital random table method, with 80 cases in each group. The patients of the two groups were given conventional treatment to relieve cough and reduce phlegm; On the basic treatment of conventional treatment, the control group were treated with azithromycin, wile the treatment group were treated with oral traditional chinese medicine Tinghuang runfei decotion and acupointion application of Tinglizi, Baijiezi, Shengnanxing, Dahuang, and Bingpian. The treatment of both groups last 3 weeks. The time of defervescence, the disappearance time of cough and pulmonaryrales of the two groups were compared, and the serum levels of inter leukin 8 (IL-8), tumor necrosis factor-a (TNF-a), thrombin regulatory proteins (TM), D-dimer were compared before and after treatment. The total effect rate and the incidence of adverse events were compared.Results The defervesce time (3.79 ± 1.68 dvs. 4.88 ± 1.61 d,t=3.846), disappearance time of cough (8.21 ± 2.42 dvs. 10.35 ± 2.60 d,t=5.389) and disappearance time of pulmonary rales (6.21 ± 1.89 dvs. 7.78 ± 2.08 d,t=4.997) in the treatment group were significantly less than those in the control group (P<0.01). The levels of IL-8 (9.98 ± 4.26 ng/Lvs.14.18 ± 4.82 ng/L, t=5.840), TNF-a (20.78 ± 5.93ng/Lvs. 26.07 ± 6.42 ng/L,t=5.414), TM (9.63 ± 2.88μg/Lvs. 13.08 ± 3.37μg/L,t=6.961), D-dimer levels (0.09 ± 0.04 ng/Lvs. 0.15 ± 0.06 ng/L,t=7.442) the treatment group were significantly better than those in the control group (P<0.01). The total effect rate in the treatment gruop was significantly higher than that of the control group (97.5%vs. 88.8%;χ2=4.783,P=0.029), but there was no significant difference in the incidence of adverse events between the two groups (16.3%vs. 10.0%;χ2=1.370,P=0.272).Conclusions Oral and acupointion of traditional chinese medicine Tinghuang runfei decotion combined with azithromycin could relieve the symptoms, improve the clincal effect and show its safety for the children with mycoplasma pneumonia.
3.Analysis of IL-17 and IL-10 expressions of patients with hepatic cystic echinococcosis in cyst stages

Objective To explore the significance of IL-17 and IL-10 in patients with different types of hepatic cystic echinococcosis ( CE ) in cyst stages , to investigate the values of evaluation of hydatid cyst activity . Methods 33 patients with hepatic hydatid disease and 21 healthy people (control group) were enrolled, divided into 4 groups, according to WHO ultrasonic classification for CE:Control group (n=21), CEⅠ,Ⅱgroup (active, n=14), CEⅢgroup (transitional, n=10), and CEⅣ,Ⅴgroup (inactive, n=9). The serum concentrations of IL-17 and IL-10 were measured by enzyme-linked immunosorbent assay. Results Serum levels of IL-17 in CEⅢgroup were significantly higher than other groups (P<0.05). There were no significant difference in serum IL-17 levels between CEⅠ,Ⅱgroup [(38.57 ± 3.83) pg/mL] and CEⅣ,Ⅴgroup [(40.22 ± 5.15) pg/mL]. Serum levels of IL-10 were significantly higher in CEⅢgroup [(14.65 ± 2.15) pg/mL] than CEⅠ,Ⅱgroup [(11.38 ± 0.71) pg/mL] (P<0.05), while CEⅠ,Ⅱgroup were significantly higher than CEⅣ,Ⅴgroup [(7.60 ± 0.46) pg/mL] (P<0.05). There were no significant difference of IL-17/IL-10 ratio between CEⅠ, Ⅱgroup (3.44 ± 1.04) and CEⅢgroup (4.60 ± 2.40), and there were no significant difference of IL-17/IL-10 ratio between CEⅢgroup and CEⅣ,Ⅴgroup (5.39 ± 1.95). IL-17/IL-10 ratio in CEⅠ, Ⅱgroup was significantly lower than CEⅣ, Ⅴgroup (P<0.05). The serum concentrations of IL-17 has no correlation with IL-10 in different cyst stages. Conclusions The synergy effect of IL-17 and IL-10 may play the certain role to promote the parasite progress. Neither IL-17 nor IL-10 level could be as an independent marker of echinococcal cysts activity, while IL-17/IL-10 ratio reflect the hydatid cyst activity in certain degree, but as a echinococcal cysts activity marker, further evidence should be needed.
4.Clinical application of primary tumor contralateral facial artery musculocutaneous flap to reconstruct oral and maxillofacial defects.

OBJECTIVEThis study aims to explore the method that uses primary tumor contralateral facial artery musculocutaneous (FAMM) flap to reconstruct defects of the tongue and floor of mouth.
METHODSSix cases were selected for the use of primary tumor contralateral FAMM flap to reconstruct tongue and floor of mouth defects after tumor resection.
RESULTSThe FAMM flap of the six cases had a long pedicle that could reach the contralateral tongue and floor of mouth. All flaps were intact until post-operation. All patients experienced post-operation complications, such as temporary facial tension and limited mouth opening, which improved after 3 months. Half a year later, the flaps still did not show signs of shrinking.
CONCLUSIONFeatures of the primary tumor contralateral FAMM flap include the tissue-like material provided for reconstructing tongue or floor of mouth defects, easy acquisition, and high survival rate with minimal donor site morbidity. As such, it is an ideal material for repairing tongue and floor of mouth defects.

5.Expression and role of β-catenin in suppression of liver regeneration in small-for-size liver graft after transplantation in rats

Objective To investigate the expression and role of β-catenin in small-for-size liver grafts during early stage of liver regeneration after liver transplantation in rats.Method The livers of male Sprague-Dawley rats were reduced to 30% or 50% of their original sizes and transplanted.The experiment was divided into 3 groups:small-for-size graft group (SSG),half-size graft group (HSG) and sham-operated group.Liver samples were harvested at various time points after transplantation (n =6 per time point) for Western blotting and immunohistochemistry.Six rats in each group were sacrificed at 3rd day after liver transplantation for estimating liver regeneration rate.Result Liver regeneration rate in SSG group was lower than that in HSG group.The expression of β-catenin was down-regulated in liver graft of both groups after being stored in cold Ringer solution for 2 h.The expression of β-catenin was significant up-regulated in HSG group from 5 min to 12 h after operation,while the down-regulated expression of β-catenin was persisted in SSG group at 5 min after operation,and mildly increased expression of β-catenin occurred at 2 h and 6 h,which was significantly lower than that in HSG group at the corresponding time points.The expression of active-β-catenin was low in each group before transplantation.Significant expression of active-β-catenin was found at 5 min in HSG group and persisted until 12 h after operation,mildly increased expression of active-β-catenin in SSG group was only found at 2 h,which was lower than that in HSG group at the same time points.Immunohistochemical staining revealed that β-catenin was mainly expressed on the hepatocyte membrane and in cytoplasm in the sham-operative group,many hepatocytes exhibited nuclear localization of β-catenin in HSG group from 5 min to 24 h,while only some hepatocytes exhibited nuclear localization of β-catenin in SSG group.The expression of Cyclin D1 in SSG group was significantly lower than that in HSG group,which was similar to the expression of C-Myc.Conclusion Attenuated activation of Wnt/β-catenin signaling and down-regulated expression of target genes during early regeneration of small-for-size liver grafts may be involved in the inhibition of liver regeneration of small for size liver grafts.
6.Application of 99TCm tracer technique in rabbit cerebral thromboembolic stroke

Objective To study the application of 99Tcm in rabbit cerebral thromboembolic stroke and thrombolysis effect of recombinant tissue plasminogen activator (rt-PA).Methods The 0.5 mL radioactive pertechnetate sodium (specification:5 mCi/2mL and radiation intensity 92.5 MBq/mL) was combined with 30 μL stannous chloride (5 mg/mL),and the 20 μL mixture was joined to whole blood,red blood cells,and plasma for labelling.Then 50 μL CaCl2 (0.5 mol/L) and bovine thrombin (50 IU/mL) were doped in mixture,and rapidly sucked into a polyethylene plastic pipe (PE80).Thrombus was formed for 2 h at 37 ℃ and cut into small pieces of 10 mm.Autologous blood clots combined with 99Tcm from external carotid artery were injected to internal carotid artery of rabbit,the radioactivity (counts per minute,CPM) was measured by gamma counting instrument,and the improvement of rt-PA 4.5 mg/kg (clinical equivalent dose) on this model was observed.Results After thromboembolism,CPM increased approximately by (5.1 ± 1.3) times,which suggested that the model was reliable.The rt-PA 4.5 mg/kg had significant progressive thrombolysis effect.Conclusion 99Tcm tracer technology could be applied to rabbit cerebral stroke model,which is stable and reliable

Objective To improve overall value of healthcare industry through setting up critical inpatient medical services strategic plan.Methods Identify major objectives which the local government expects to achieve through strategic map; Standardize inpatient output and assign weight to each group through diagonosis related groups; Translate the objectives of strategic map and result of diagnosis related group to Balanced Score-card; Finally build up strategic map and according action plans.ResultsPreliminarily established 16 objectives、23 measures and 13 tasks in four perspectives including customer,internal work flow,learning and growing and finance.ConclusionThe strategy map and the balanced score-card can help implement full strategic plan of regional inpatient medical services; DRGs is a core management tool of patient-centred service output management; Balanced Score-card is able to realize continuous improvement of Beijing inpatient medical services from macro to micro persoetive.
8.ZJU index and the risk of nonalcoholic fatty liver disease in the Uygur population in the rural area of Xinjiang Uygur Autonomous Region: A cohort study

ObjectiveTo investigate the association between ZJU index and the onset of nonalcoholic fatty liver disease (NAFLD) in the Uygur population and the value of ZJU index in predicting the risk of NAFLD. MethodsThe Uighur community of The 51st Regiment of The Third Division of Xinjiang Kashgar Corps was selected as the investigation site, and the Uygur residents who lived in this area and had an age of >18 years were selected as subjects. Follow-up studies were conducted in 2019, 2020, and 2021, and the investigation of outcomes was completed in June to August of 2021. Finally 10 597 subjects were enrolled for analysis. The Kruskal-Wallis H test was used for comparison of continuous variables between groups, and the chi-square test was used for comparison of categorical variables between groups. The subjects were divided into Q1-Q4 groups according to the level of ZJU index. The Kaplan-Meier curve was used to predict the incidence rate of NAFLD, and the Cox regression model was used to analyze the association between ZJU index and the risk of NAFLD; the area under the ROC curve (AUC) was used to evaluate the value of ZJU index in predicting the risk of NAFLD. ResultsDuring the median follow-up time of 4.92 years, the incidence rate of NAFLD was 9.4% (992/10 597) among the study population. After adjustment for multiple factors, there was a significant increase in the risk of NAFLD with the increase in ZJU index, with a hazard ratio of 2.55 (95% confidence interval [CI]: 1.60 — 4.06), 7.32 (95%CI: 4.78 — 11.20), and 21.74 (95%CI: 14.32 — 33.00), respectively (all Ptrend<0.001). The ROC curve showed that ZJU index had a higher value in predicting NAFLD (AUC=0.816), and the male subgroup had a significantly higher predictive accuracy of ZJU index than the female subgroup (AUC: 0.829 vs 0.809). ConclusionZJU index is a predictive factor for the onset of NAFLD in the Uygur population in rural areas of Xinjiang and has a good value in predicting the risk of NAFLD.
