Objective To study the effect of alcohol on apoptosis and apoptosis-associated genes-Bcl-2 and Bax of cardiac muscle cell.Methods We established an animal experimental model by giving different concentration of alcohol to the healthy mice.Myocardial apoptosis and the expression of apoptosis-associated gene Bcl-2 and Bax were quantitatively analysed by the nuclear fast red-crystal violet staining and immunohistochemical method.Results Some nuclears of the experimental groups were stained purple and agglutinated.Counting under the high power microscope,it was found that the amount of abnormal nuclears significantly increased in alcohol-treated groups compared with the control group (P

Objective To investigate the apoptosis in neural epithelium induced by passive smoking. Methods By using the nuclear fast red-crystal violet staining method and situ TUNEL technology, the quantitative changes of apoptosis in neural epithelium in gold hamster were analyzed. Results Excessive apoptosis and less cell division were observed in neural epithelium compared with the control group. 3.Effects of Alcohol Exposure on the Expression of Bcl-2 and Bax in Embryonic Neuroepithelial Cells in Golden Hamster

Objective To explore the expression of Bcl-2 and Bax in embryonic neuroepithelial cells in Golden hamster exposured to the teratogenic dose of alcohol. Methods The experimental pregnant golden hamsters were given ethanol through intraperitoneally injected, 5.8 g/kg body weight. The Bcl-2 and Bax expressions in neuroepithelial cells of experimental and control animals were detected with immunohistochemical and image analysis at 4,8,16,24,48 and 72 h time points after treatment. Results Bcl-2 and Bax proteins distributed in nuclear membranes and cytoplasm of the neuroepithelial cells as well as mesenchymal cells. In control groups, the expressions of these two kind proteins increased as the time passed at beginning, 16 hours after, came to the summit and then descended gradually. Compared with the control, the expressions of Bcl-2 in experimental groups were down-regulated, expressions of Bax were up-regulated except at 8 h time point. Conclusion Alcohol can induce down-regulation of the expression of Bcl-2 protein and up-regulation of the expression of bax protein, which may play a an important role in regulation of apoptosis in alcohol-induced neural tube malformation.

5.The Role of Fibronectin and Lectins Binding Receptors in Development of Neural Tube Defects Induced by Passive Smoking

Objective To investigate the possible mechanisms of teratogenesis caused by passive smoking, temporospatial changes in surface carbohydrates of the embryonic neural epithelium and the mesenchyme around it were explored. Methods The pregnant golden hamsters were randomly divided into the treated group and the control groups. In the chamber made by ourself for passive smoking experiment, the animals of the treated group were exposed to smoke from embryonic day 4 to 7, 3 times per day for 4 consecutive days. At twelve o'clock and eighteen o'clock of embryonic day 8, eight o'clock of embryonic day 9 and eight o'clock of embryonic day 10 respectively, the pregnant golden hamsters of both treated group and control group were sacrificed, the uteri were removed and embedded routinely with paraffin, sectioned transversely in series. The sections of each group were stained with immunohistochemical ABC method and concannavalin agglutinin (ConA) and ulex europaeus agglutinin (UEA) were used as molecule probes to conduct the qualitative and quantitative analysis of the fibronectin and the related surface carbohydrates of the embryonic neural epithelium and the mesenchyme around it. Objective: The regeneration and repair of peripheral nerves are focuses of neurological studies.The(bFGF) may play an important role in the development of the neural system.Dexamethasone is usually employed in the management of neural injuries.The aim of this study is to establish a rat model,and investigate the effect of Dexamethasone on the expression of bFGF in the neurons of the spinal cord following sciatic nerve injury.Methods: Fifty-six male Wistar rats were randomly divided into 4 groups: a nerve injury group,with sciatic nerve injury induced by clamping the nerve in the right;a dexamethasone group,immediately injected with dexamethasone [0.5 mg/(kg/d)] in the injury side;a saline group,given saline only;and a control group,rats without sciatic nerve injury.Changes of bFGF in the motor neurons of the L4-L6 spinal cords were observed by immunohistochemistry.Results: Dexamethasone obviously upregulated the expression of bFGF in the motor neurons of the spinal cord.Conclusion: Dexamethasone can improve the expression of bFGF in the motor neurons of the spinal cord after sciatic nerve injury,and contribute to the recovery of the function of the injured neurons.
7.Study on effect of astragalus polysaccharide on peripheral circulation MDSC in lung cancer and its clinical effect

Objective To investigate the effect of Astragalus Polysaccharide on peripheral circulation myeloid derived suppressor cells (MDSC) in lung cancer and its clinical effect. Methods 100 patients with advanced non small cell lung cancer were selected and divided into 2 groups, 50 cases in the control group treated with chemotherapy aalone, 50 cases in the experiment group received injection of Astragalus Polysaccharide on the basis of the control group, peripheral blood MDSC levels, peripheral blood T lymphocyte subsets levels, serum ARG I activity, TNF-α, IL-6 levels, the clinical effect and incidence of adverse reactions were compared after the treatment. Results Compared with the control group, peripheral blood levels of MDSC (Lin-HLA DR-\CD33+\CD11b+) was lower after treatment in the experiment group , peripheral blood levels of CD3+, CD4+T lymphocytes and CD4+/CD8+ were higher, and CD8+T lymphocytes level was lower after treatment, serum levels of ARG I activity, TNF-α, IL-6 levels were lower after treatment, (P<0.05) , the total effective rate in the control group(42.0%)was lower than the experiment group(64.0%), (P<0.05), the incidence of gastrointestinal reaction(40/24), 3 ~ 4 degree of leukocyte reduction(26/12), liver function damage(19/9), renal damage(17/7) in the control group were higher than the experiment group (P<0.05). Conclusion Astragalus Polysaccharide can significantly reduce the peripheral circulation MDSC (Lin-HLA DR-\CD33+\CD11b+) level in patients with lung cancer, improve T lymphocyte immune function, inhibit the activity of serum ARG I, reduce the level of TNF-αand IL-6, effectively improve the clinical efficacy, and reduce the toxic and side effects of chemotherapy.
8.Effects of Xiaoyingqiangji Decoction on Mitochondria in Skeletal Muscle of Hyperthyroid Myopathy

Objective To observe the effect of Xiaoyingqiangji decoction on mitochondria in skeletal muscle of hyperthyroid myopathy. Methods Healthy Wistar rats were randomly divided into 4 groups: the group of hyperthyroid myopathy were given L-thyroxine intraperitoneal injection for 42 d; Chinese medicine treated group were treated with Xiaoyingqiangji decoction for 30 d after L-thyroxine injection; the saline control group were given saline solution for 30 d after L-thyroxine injection; normal control group were given saline solution. Then the skeletal muscles were obtained and the succinate dehydrogenase (SDH) were measured. The ultrastructural changes were observed with TEM. Results The activity and content of SDH in the group of hyperthyroid myopathy decreased, and the cristae of mitochondria ruptured and even disappeared. After given Chinese medicine, the activity and content of SDH increased, while the mitochondria showed normal ultrastructural appearance. Conclusion Xiaoyingqiangji decoction can improve the function of skeletal muscles in hyperthyroid myopathy by recovering enzyme activity and mitochondria structure.
9.Effect of Xiaoyingqiangjitang on the Motor End-plate of Skeletal Muscle in Hyperthyroid Myopathy

Objective To observe the effect of Xiaoyingqiangjitang on the ultrastructure of motor end-plate of skeletal muscle in rats with hyperthyroid myopathy, and changes of acetylcholin esterase (AchE).Methods The 36 healthy female Wistar rats were randomly divided into the normal control group (n=6), and model group, Chinese medicine treated group and saline control group (all n=10). All animals in the model group, Chinese medicine treated group and saline control group were treated with group given L-thyroxine intraperitoneal injection for 42 days to establish the animal model of hyperthyroid myopathy. The animals of the Chinese medicine treated group were treated with Xiaoyngqiangjitang for 30 days after L-thyroxine intraperitoneal injection. The enzyme activity of AchE and ultrastructure of motor end-plate were observed by light and electron microscopes.Results The enzyme activity and content in the group of hyperthyroid myopathy was decreased, the reaction products of AchE located in synapse became weaker and distributed heterogeneously, the junction infolding of motor end-plate became smaller and shorter, the mitochondria in axon terminal became vacuolated. After given Chinese medicine, the enzyme activity and content increased, the junction folding, the mitochondria showed normal ultrastructural appearance.Conclusion Xiaoyingqiangjitang can improve the function of skeletal muscle in hyperthyroid myopathy by recovering the enzyme activity of AchE and ultrastructures of motor end-plate.
10.Clinical Application of Whole Aorta and Coronary CT Angiography Combined with Low Radiation Dose, Low Contrast Medium Injection Rate and Dose Protocol

[Objective] To investigate the clinical value of using prospective ECG-gating Helical protocol in whole aorta and coronary arteries CT angiography combined with low tube voltage,low contrast medium injection rate and dose technology.[Methods]A total of 60 patients (heart rate≤ 75 bpm and normal heart rhythm) with suspected aortic disease or post-operation were randomly divided into 2 groups:G1 (n =30;tube voltage:80 kV;contrast medium injection rate:3 mL/s;contrast medium dose:0.65 mL/kg)and G2 (n =30;tube voltage:120 kV;contrast medium injection rate:4 mL/s;contrast medium dose:1.2 mL/kg).The two groups underwent aorta angiography with prospective ECG-gating Helical protocol by using 320-detector CT.The mean CT values and standard deviation of aorta,coronaries and adjacent adipose tissue were measured,signal to noise ratio (signal-to-noise ratio,SNR)and contrast to noise ratio (contrast-to-noise ratio,CNR) were calculated.The subjective quality scoring of 3-d post-processing images were evaluated independently by 2 senior radiologists.The mean CT values and standard deviation of aorta,coronaries and adipose tissue,SNR,CNR,subjective quality scoring,contrast medium dose and radiation dose were compared and analyzed.[Results] The abnormal and normal rate of aorta in two groups were 83.33% (25/30),16.67% (5/30) and 86.67% (26/30),13.33%(4/30);while the abnormal and normal rate of coronary arteries in two groups were 43.33% (13/30),56.67% (17/30) and 53.33%(16/30),46.67% (14/30).SNR values of ascending aorta,descending aorta,abdominal aorta,superior mesenteric artery,bilateral renal artery,left main coronary artery,left anterior descending branch,left circumflex branch,right coronary artery and CNR values of abdominal aorta,superior mesenteric artery,bilateral renal artery,left anterior descending branch,left circumflex branch had no statistical significance,while the other indicators were significant differences between the two groups.The radiation dose and the contrast medium dose of G1 was significantly lower than G2 (P < 0.05).There was no difference in the subjective image quality of 3D post-processing images between the two groups (P > 0.05).[Conclusions] Using prospective ECG-gating Helical protocol in whole aorta and coronary arteries CT angiography combined with low tube voltage,low contrast medium injection rate and low contrast medium dose technology can help to reduce radiation and contrast medium dose without impacting on the imaging quality satisfactorily.
